    The relationship between insulin binding, insulin activation of insulin-receptor tyrosine kinase, and insulin stimulation of glucose uptake in isolated rat adipocytes

    We have studied the relationship between insulin activation of insulin-receptor kinase and insulin stimulation of glucose uptake in isolated rat adipocytes. Glucose uptake was half-maximally or maximally stimulated, respectively, when only 4% or 14% of the maximal kinase activity had been reached. To investigate this relationship also under conditions where the insulin effect on activation of receptor kinase was decreased, the adipocytes were exposed to 10 microM-isoprenaline alone or with 5 micrograms of adenosine deaminase/ml. An approx. 30% (isoprenaline) or approx. 50% (isoprenaline + adenosine deaminase) decrease in the insulin effect on receptor kinase activity was found at insulin concentrations between 0.4 and 20 ng/ml, and this could not be explained by decreased insulin binding. The decreased insulin-effect on kinase activity was closely correlated with a loss of insulin-sensitivity of glucose uptake. Moreover, our data indicate that the relation between receptor kinase activity and glucose uptake (expressed as percentage of maximal uptake) remained unchanged. The following conclusions were drawn. (1) If activation of receptor kinase stimulates glucose uptake, only 14% of the maximal kinase activity is sufficient for maximal stimulation. (2) Isoprenaline decreases the coupling efficiency between insulin binding and receptor-kinase activation, this being accompanied by a corresponding decrease in sensitivity of glucose uptake. (3) Our data indicate that the signalling for glucose uptake is closely related to receptor-kinase activity, even when the coupling efficiency between insulin binding and kinase activation is altered. They thus support the hypothesis that receptor-kinase activity reflects the signal which originates from the receptor and which is transduced to the glucose-transport system

    The ubiquitous proteases ÎĽ- and m-calpain are Ca2+-dependent cysteine endopeptidases. Besides involvement in a variety of physio(patho)logical processes, recent studies suggest a pivotal role of calpains in differentiation of hematopoietic cells and tumor cell invasion. However, the precise actions of calpains and their endogenous inhibitor, calpastatin, in these processes are only partially understood. Here we have studied the role of the calpain/calpastatin system in the invasion of leukemic cells under basal and differentiationstimulating conditions. To further differentiate the human leukaemic cell line THP-1 (monocytic), the cells were treated for 24 hours with the differentiationstimulating reagents phorbol 12-myristate 13-acetate (PMA) and dimethyl sulfoxide (DMSO). Macrophage and granulocytelike differentiation was confirmed by induction of vimentin expression as well as by microscopic and fluorescence assisted cytometric analysis. Extracellular matrix (ECM) invasion of both the basal and differentiation stimulated cells in a Matrigel assay was inhibited by preincubation of the cells with the specific calpain inhibitor CP1B for 24 hours. Inhibition of invasiveness correlated with decreased mRNA expression and secretion of the matrix metalloproteinases MMP-2 and MMP-9. In contrast, addition of CP1B only during the invasion process did neither influence transmigration nor MMP release. This is the first report showing that the calpain/calpastatin system mediates MMPmRNA expression of the leukemic THP-1 cells and as a consequence their invasiveness

    We present a theoretical and experimental study of the ultra-narrow bandwidth Faraday anomalous dispersion optical filter (FADOF) operating at the rubidium D1 line (795 nm). This atomic line gives better performance than other lines for the main FADOF figures of merit, e.g. simultaneously 71% transmission, 445 MHz bandwidth and 1.2 GHz equivalent noise bandwidth.Comment: 3 pages, 2 figures.     Elevated UV-B radiation reduces genome stability in plants

    Long-term depletion of the stratospheric ozone layer contributes to an increase in terrestrial solar ultraviolet-B radiation1,2,3^{1,2,3}. This has deleterious effects on living organisms, such as DNA damage4,5^{4,5}. When exposed to elevated ultraviolet-B radiation (UV-B; 280–315 nm), plants display a wide variety of physiological and morphological responses characterized as acclimation and adaptation6^{6}. Here we show, using special sun simulators, that elevated solar UV-B doses increase the frequency of somatic homologous DNA rearrangements in Arabidopsis and tobacco plants. Increases in recombination are accompanied by a strong induction of photolyase and Rad51 gene expression. These genes are putatively involved in major DNA repair pathways, photoreactivation and recombination repair7,8^{7,8}. In mutant Arabidopsis plants that are deficient in photoreactivating ultraviolet-induced cyclobutane pyrimidine dimers, recombination under elevated UV-B regimes greatly exceeds wild-type levels. Our results show that homologous recombination repair pathways might be involved in eliminating UV-B-induced DNA lesions in plants. Thus, increases in terrestrial solar UV-B radiation as forecasted for the early 21st century may affect genome stability in plants

    Tracking system options for future altimeter satellite missions

    Follow-on missions to provide continuity in the observation of the sea surface topography once the successful TOPEX/POSEIDON (T/P) oceanographic satellite mission has ended are discussed. Candidates include orbits which follow the ground tracks of T/P GEOSAT or ERS-1. The T/P precision ephemerides, estimated to be near 3 cm root-mean-square, demonstrate the radial orbit accuracy that can be achieved at 1300 km altitude. However, the radial orbit accuracy which can be achieved for a mission at the 800 km altitudes of GEOSAT and ERS-1 has not been established, and achieving an accuracy commensurate with T/P will pose a great challenge. This investigation focuses on the radial orbit accuracy that can be achieved for a mission in the GEOSAT orbit. Emphasis is given to characterizing the effects of force model errors on the estimated radial orbit accuracy, particularly those due to gravity and drag. The importance of global, continuous tracking of the satellite for reduction in these sources of orbit error is demonstrated with simulated GPS tracking data. A gravity tuning experiment is carried out to show how the effects of gravity error may be reduced. Assuming a GPS flight receiver with a full-sky tracking capability, the simulation results indicate that a 5 cm radial orbit accuracy for an altimeter satellite in GEOSAT orbit should be achievable during low-drag atmospheric conditions and after an acceptable tuning of the gravity model
