Luscofusco is a children’s daycare that offers an alternative to the traditional school for children 2 to 6 years old. We began our adventure in Oza, a small village in the province of A Coruña, restoring an old rural school which had been closed years ago. Inspired by philosophies as Montessori, Pestalozzi, Malaguzzi our goal is to recover the essence of the old rural schools, and at the same time energize the area in which we are located. We are a new concept of preschool in which our role is to guide children as they develope cognitive, motor, emotional, affective, inter and intrapersonal skills through play, coexistence and experience. All this, in a carefully prepared family atmosphere situated in a natural environment. We base our work on respect for kids, their interests and their motivation to turn them into the protagonists of their learning. We assume a role of mediators in this process, use a non-directive approach and offer individual attention.peerReviewe

RNA-binding protein immunoprecipitation as a tool to investigate plant miRNA processing interference by regulatory proteins of diverse origin

Background: Due to the nature of viral RNA genomes, RNA viruses depend on many RNA-binding proteins (RBP) of viral and host origin for replication, dissemination and evasion of host RNA degradation pathways. Some viruses interfere with the microRNA (miRNA) pathway to generate better fitness. The development of an adjusted, reliable and sensitive ribonucleoprotein immunoprecipitation (RIP) assay is needed to study the interaction between RBP of different origin (including viral origin) and miRNA precursors. The method could be further applied to transiently expressed heterologous proteins in different plant species. Results: Here we describe a modified RIP assay applied to nuclear epitope-tagged proteins of heterologous origin and transiently expressed in Nicotiana benthamiana. The assay includes a combination of optimized steps as well as the careful selection of control samples and rigorous data analysis. It has proven efficient to detect and quantify miRNA processing intermediates associated with regulatory proteins. Conclusions: The RIP method described here provides a reliable tool to study the interaction of RBPs, such as transiently expressed regulatory proteins with lowly represented host RNA, as is the case of miRNA precursors. This modified method was efficiently adjusted to recover nuclear proteins and reduce unspecific background. The purification scheme optimized here for GFP-tagged proteins can be applied to a wide array of RBPs. The subsequent application of next-generation sequencing technologies will permit to sequence and characterize all RNA species bound in vivo by a given RBP.Fil: Marmisollé, Facundo Ernesto.

This paper shows the results of research conducted during the first half of 2016, related to the dynamics of underground economies drop by drop in the trade sector San Francisco neighborhood of the town of Ciudad Bolivar in Bogota, with the aim to analyze and contextualize how this problem and related crimes (the use of threat and aggression as a means to demand payment of money) which affect the coexistence and security. This, considering that the informal credit and illegal lenders are not only an economic problem, by restricting the normal development of productive, social activities, to influence the determinants of welfare and security and peaceful coexistence, since in most cases illegal lenders use their status to launder money from illicit drug trade in large quantities and also use violence and intimidation to demand payment of the lease payments of credits granted. On the other hand, the dynamics of the economies submerged dropwise has gradually linked to problems such as prostitution and drug trafficking in small quantities and also has shown that collectors are linked to several cases of murder, extortion, robbery, personal injury, threats and intimidation of its customers, mostly traders

This research project aims to create awareness, among landscape architects, on the importance of ecological design. Bioclimatic knowledge contributes to enhance the coexistence between natural and built environments, resulting in healthy places with high quality of life. The study examines ways of graphically representing climate information, to facilitate the understanding and application of bioclimatic design strategies for human comfort. Cache Valley, Utah is used as an example to demonstrate how extensive data can be represented graphically in a more readable and useful manner for practicing professionals. Temperature, humidity, wind and solar information were analyzed and converted into graphics through methods of bioclimatic analysis. The outcome is a recollection of graphic design guidelines to specific climate conditions, in this case Cache Valley, Utah. The resulting design guidelines will aid professionals to design sensitive spaces that minimize energy consumption while maximizing human comfort

Developing a strain improvement system for the entomopathogenic fungus Beauveria basssiana: a way to get better biocontrol agents?

Biocontrol agents (BCAs) based on entomopathogenic fungi (EPF) are playing an increasing role in Integrated Pest Management programmes. At present, the commercially available EPF consist of wild type strains isolated from nature, however, there is potential to breed more effective strains by recombining wild types with complementary characteristics. EPF Beauveria bassiana represents one of the most important organisms used to kill arthropod pests, as they occurred naturally in the environment, do not leave residual activity, are safer for human manipulation and they usually show a high virulence. The aim of this research was to develop a system to improve strains of the entomopathogenic fungus B. bassiana through genetic recombination. A group of 50 Beauveria strains were genotyped using multi-locus sequencing and mating gene analysis, and then phenotyped with respect to their virulence against Diamondback moth (DBM), thermal biology, tolerance of UV light, and conidial production. A phylogenetic analysis identified two different Beauveria species within the fungal collection: B. bassiana (78%) and Beauveria pseudobassiana (16%). Seven strains from different places of origin were selected with phenotypes such as tolerance to UV-B radiance, thermotolerance, virulence and potentially compatible mating types for parasexual recombination studies. Spontaneously generated nitrate non-utilizing (nit) mutants were produced from these strains using a potassium chlorate-amended selective medium and 35 vegetative compatibility groups were determined within the 50 isolates of Beauveria spp. Recombination by hyphal fusion and protoplast fusion proved to be feasible and was observed in two out of three crosses. Only one cross (X2) showed higher radial growth than the parental strains between 20 and 30 °C. Nine fungal strains were selected to investigate the potential for inducing sexual recombination by pairing complementary mating types on three different media that have probed work well in other fungal species (oatmeal agar, malt extract agar, and Czapek Dox agar +/- biotin). After six months, both in vitro and in vivo assays led to the generation of structures resembling synnemata, however no fruiting bodies nor other clear sexual structures were observed. No relationship was found between the geographical origin of the strains and their tolerance to temperature or UV-B light, suggesting that micro-environmental conditions can play a more important role in the development of determined traits of organisms sourced from specific ecosystems than the latitude or altitude of sampling locations. This study provides a significant amount of information describing several methodologies for parasexual and sexual recombination of this fungus, expanding the current knowledge of this valuable EPF
