    Expanding the Chemical Space of Withaferin A by Incorporating Silicon to Improve its Clinical Potential on Human Ovarian Carcinoma Cells

    Ovarian cancer represents the seventh most commonly diagnosed cancer worldwide. Herein, we report on the development of a withaferin A (WA)-silyl ether library with 30 analogues reported for the first time. Cytotoxicity assays on human epithelial ovarian carcinoma cisplatin-sensitive and -resistant cell lines identified eight analogues displaying nanomolar potency (IC50 ranging from 1 to 32 nM), higher than that of the lead compound and reference drug. This cytotoxic potency is also coupled with a good selectivity index on a nontumoral cell line. Cell cycle analysis of two potent analogues revealed cell death by apoptosis without indication of cell cycle arrest in G0/G1 phase. The structure–activity relationship and in silico absorption, distribution, metabolism, and excretion studies demonstrated that the incorporation of silicon and a carbonyl group at C-4 in the WA framework enhances potency, selectivity, and drug likeness. These findings reveal analogues 22, 23, and 25 as potential candidates for clinical translation in patients with relapsed ovarian cancer

    Evaluation of Convergent, Discriminant, and Criterion Validity of the Cuestionario Burnout Granada-University Students

    Burnout is a health problem that affects professionals and students or professionals in training, especially those in health areas. For this reason, it is necessary that it is properly identified to prevent the impact it can have on the work and personal areas of the people who suffer from it. The aim of this work is to study the convergent, discriminant, and criterion validity of the Cuestionario Burnout Granada-University Students. The sample consisted of 463 undergraduate nursing students, selected by non-probabilistic convenience sampling, who participated voluntarily and anonymously in the study. The mean age of the participants was 21.9 (5.12) years, mostly female (74.1%), single (95.8%), and childless (95.6%). Information was collected face-to-face, and the instruments were completed on paper. Comparisons were made in the three dimensions of burnout of the CBG-USS between students with and without burnout, finding statistically significant differences in all three dimensions: Emotional Exhaustion (p < 0.001, d = 0.674), Cynicism (p < 0.001, d = 0.479), and Academic Efficacy (p < 0.001, d = −0.607). The Cuestionario Burnout Granada-University Students presents adequate reliability and validity indices, which demonstrates its usefulness in the identification of burnout. This syndrome has traditionally been measured in professionals, but students also present burnout, so it is necessary to have specific burnout instruments for students, since the pre-work situation and stressors of students are different from those of workers. In order to work on the prevention of university burnout, it is essential to have specific instruments for professionals in training that help in the detection of students with burnout

    We describe three new species of terrestrial frogs Pristimantis from the evergreen montane forests surrounding the Tungurahua Volcano in the Upper Pastaza River Basin, eastern foothills of Ecuador.The first species is distinguished by the distinctive shape of its digits (elongated, narrow and only slightly wider than finger discs); the second presents cranial crest and body size smaller than 31 mm; and the third is spiny and exhibits a distinctive pattern with red-wine finely delineated by cream.Describimos tres nuevas especies de ranas terrestres Pristimantis de los bosques siempreverde montanos circundantes al Volcán Tungurahua en la cuenca alta del Rio Pastaza, estribaciones orientales de Ecuador.     A pilot study applying the plant Anchored Hybrid Enrichment method to New World sages (Salvia subgenus Calosphace; Lamiaceae)

    We conducted a pilot study using Anchored Hybrid Enrichment to resolve relationships among a mostly Neotropical sage lineage that may have undergone a recent evolutionary radiation. Conventional markers (ITS, trnL-trnF and trnH-psbA) have not been able to resolve the relationships among species nor within portions of the backbone of the lineage. We sampled 12 representative species of subgenus Calosphace and included one species of Salvia´s s.l. closest relative, Lepechinia, as outgroup. Hybrid enrichment and sequencing were successful, yielding 448 alignments of individual loci with an average length of 704. bp. The performance of the phylogenomic data in phylogenetic reconstruction was superior to that of conventional markers, increasing both support and resolution. Because the captured loci vary in the amount of net phylogenetic informativeness at different phylogenetic depths, these data are promising in phylogenetic reconstruction of this group and likely other lineages within Lamiales. However, special attention should be placed on the amount of phylogenetic noise that the data could potentially contain. A prior exploration step using phylogenetic informativeness profiles to detect loci with sites with disproportionately high substitution rates (showing "phantom" spikes) and, if required, the ensuing filtering of the problematic data is recommended. In our dataset, filtering resulted in increased support and resolution for the shallow nodes in maximum likelihood phylogenetic trees resulting from concatenated analyses of all the loci. Additionally, it is expected that an increase in sampling (loci and taxa) will aid in resolving weakly supported, short deep internal branches.Fil: Fragoso Martínez, Itzi.     We describe a new species of terrestrial frog Pristimantis from the western slopes of the Ecuadorian Andes, in the province of Pichincha, Ecuador. The species is a member of the P. devillei species-group and is characterized by having cranial crests with high frontoparietals, concealed tympanum, glandular dorsolateral folds and flanks, and subconical tubercles on the upper eyelid, ulnar region and heel.Describimos una nueva especie de rana terrestre Pristimantis de las estribaciones occidentales de los Andes de Ecuador, en la provincia de Pichincha, Ecuador.     Personality Factors as Predictors in Burnout Level Changes for Surgical Area Nurses

    Surgical area nurses provide comprehensive care to patients throughout the surgical process. Increases in life expectancy lead to the appearance and development of diseases, translating into an increase in the number of necessary interventions. Increases in the workload can be another risk factor for the development of burnout in professionals in this area. Knowledge of psychological and personality-related variables provides relevant information of level changes in the dimensions of burnout syndrome. Three logistic regression models, based on a cross-sectional study with 214 nurses working in the surgical area in the Andalusian Health Service, Spain, were built for each dimension. These models included different variables related to depression and personality, with some being significant at the population level and consequently true risk or protection factors. Friendliness, responsibility and extraversion are protection factors for the personal accomplishment dimension, whilst neuroticism is a risk factor for this dimension. Friendliness is also a protection factor for depersonalization, whilst depression is a risk factor. Finally, neuroticism, responsibility and depression are risk factors for the emotional exhaustion dimension of burnout. These findings provide relevant information that makes anticipation of this syndrome in this group easier

    DDR1 and Its Ligand, Collagen IV, Are Involved in In Vitro Oligodendrocyte Maturation

    Discoidin domain receptor 1 (DDR1) is a tyrosine kinase receptor expressed in epithelial cells from different tissues in which collagen binding activates pleiotropic functions. In the brain, DDR1 is mainly expressed in oligodendrocytes (OLs), the function of which is unclear. Whether collagen can activate DDR1 in OLs has not been studied. Here, we assessed the expression of DDR1 during in vitro OL differentiation, including collagen IV incubation, and the capability of collagen IV to induce DDR1 phosphorylation. Experiments were performed using two in vitro models of OL differentiation: OLs derived from adult rat neural stem cells (NSCs) and the HOG16 human oligodendroglial cell line. Immunocytofluorescence, western blotting, and ELISA were performed to analyze these questions. The differentiation of OLs from NSCs was addressed using oligodendrocyte transcription factor 2 (Olig2) and myelin basic protein (MBP). In HOG16 OLs, collagen IV induced DDR1 phosphorylation through slow and sustained kinetics. In NSC-derived OLs, DDR1 was found in a high proportion of differentiating cells (MBP+/Olig2+), but its protein expression was decreased in later stages. The addition of collagen IV did not change the number of DDR1+/MBP+ cells but did accelerate OL branching. Here, we provide the first demonstration that collagen IV mediates the phosphorylation of DDR1 in HOG16 cells and that the in vitro co-expression of DDR1 and MBP is associated with accelerated branching during the differentiation of primary OLs

    Rediscovery of the Endangered Carchi Andean Toad, \u3cem\u3eRhaebo colomai\u3c/em\u3e (Hoogmoed, 1985), in Ecuador, with Comments on Its Conservation Status and Extinction Risk

    Since 1984 there have been no records of Rhaebo colomai (Hoogmoed, 1985) within the territory of Ecuador. This species was known from 2 localities in the province of Carchi, northwestern Ecuador, and the department of Nariño, southwestern Colombia, which were reported in 1979 and 2015, respectively. We report the recent sightings of R. colomai at 3 new localities in Ecuador and discuss and evaluate this species’ extinction risk and conservation status
