    Encapsulación de la piperine presente en la especie Piper tuberculatum utilizando vesículas multilamelares y determinación de su poder antioxidante

    The piperine is an alkaloid present in the vegetal species pipilongo (Piper tuberculatum). Antioxidant properties are associated to this plant. It was encapsulated in multilamellar vesicles (MLV). For that, the validation of the extraction methodology through calibration curves of standard piperine by high resolution liquid chromatography (HPLC) was performed, using a column of C18, mobile phase MetOH: H2O (70:30) and detected by UV to 340nm with a fluency rate of 0.7 mL/ min. The lineal and non-lineal ranges were found between 1 and 300 ppm, with R2 of 0.995. During the extraction of the pipilongo fruit, the effect of two factors, solvents and ultrasound power, was studied, for the optimization of conditions and higher extraction of piperine in pipilongo. The maximum concentration of piperine (246.50 ppm) was found, using ethanol as extraction solvent. The piperine was isolated from its extract, using a solution of KOH with 10 % ethanol. It was purified and characterized by melting point, IR, GC-MS y RMN 1H. Once the extract was obtained, it was proceeded to the formation of liposomes. The phosphatidylcholine and cholesterol vesicles were prepared with extract and with piperine at different concentrations and hydrated with buffer pH 7.40. The average size of the particles formed, measured by dynamic light scattering (DLS), was in a range of 299.7–531.9 nm. For the study of antioxidant activity, a solution of DPPH 0.2 mM was prepared and a reaction was carried out with the extract and piperine liposomes and their corresponding free forms. The IC50 calculated for the extract and piperine were 2.8±0.2 and 21333±1499 ppm, respectively. These values of IC50 were compared against a standard of reference Quercetin, prepared under the same conditions. The test with DPPH showed that the released form possesses greater antioxidant activity while in encapsulation form, it attenuates and stabilizes this capacity.La piperina, alcaloide presente en la especie vegetal pipilongo (Piper tuberculatum) a la cual se le asocian propiedades antioxidantes, fue encapsulada en vesículas multilamelares (MLV). Para ello se realizó la validación de la metodología de extracción a través de curvas de calibración de piperina estándar por cromatografía líquida de alta resolución (HPLC) utilizando una columna C18, fase móvil MetOH:H2O (70:30) y detectada por UV a 340 nm con una tasa de flujo de 0.7 mL/min. El rango lineal y linealidad fueron halladas entre 1 a 300 ppm, con R2 de 0.995. En la extracción del fruto de pipilongo se estudió el efecto de dos factores, disolventes y potencia de ultrasonido, para la optimización de condiciones y mayor extracción de piperina en pipilongo. La máxima concentración de piperina (246.50 ppm) fue encontrada en las condiciones de ultrasonido (US 20 KHz a 175W, 40 min) utilizando etanol como disolvente de extracción. Del extracto, se aisló la piperina utilizando una solución de KOH 10% etanólico. Fue purificada y caracterizada mediante punto de fusión, IR, GC-MS y RMN 1H. Una vez obtenido el extracto, se procedió a la formación de liposomas. Las vesículas de fosfatidilcolina y colesterol fueron preparadas con extracto y con piperina a diferentes concentraciones e hidratadas con buffer PBS a pH 7.40. El tamaño promedio de las partículas formadas, medido por dispersión de luz dinámica (DLS), estuvo en un rango de 299.7–531.9 nm. Para el estudio de la actividad antioxidante, se preparó una solución de DPPH 0.2 mM y se llevó a reacción con los liposomas de extracto y de piperina y sus correspondientes formas libres durante 60 min, medidos a 520 nm. El IC50 calculado para el extracto y la piperina fueron 2.8±0.2 y 21333±1499 ppm, respectivamente. Estos valores de IC50 fueron comparados frente a un estándar de referencia preparado bajo las mismas condiciones, quercetina (IC50 2.5±0.1 ppm). El ensayo con DPPH mostró que la forma liberada posee mayor actividad antioxidante mientras que en la encapsulación atenúa y estabiliza esta capacidad

    Renal volume of five-year-old preterm children are not different than full-term controls

    Objective: In previous studies, smaller renal volumes were reported in prematurely born infants, however, these renal volumes were not corrected for body surface area, the main determinant of renal size. Given the rapid growth of the renal cortex after premature birth, the authors hypothesized that corrected volumes would not differ from healthy controls. Methods: Ambispective cohort study with prospective follow-up of prematurely born babies in a large specialized center and retrospectively recruited healthy control group. Children were assessed for renal length and renal volumes at age 5 by three independent ultrasonographers. Detailed anthropometry, blood pressure and renal function were also obtained. Age independent z-scores were calculated for all parameters and compared using descriptive statistics. Results: Eighty-nine premature study participants (median 32 weeks gestational age) and 33 healthy controls (median 38 weeks gestational age) were studied. Study participants did not differ in age, sex, Afro-Colombian descent, height, blood pressure, serum creatinine, or new Schwartz eGFR. Premature study participants had a significantly lower weight (17.65 ± 2.93 kg) than controls (19.05 ± 2.81 kg, p = 0.0072) and lower body surface area. The right renal volumes were significantly smaller (39.4 vs 43.4 mL), but after correction for body surface area, the renal volume and renal length z-scores were identical for both kidneys (mean right kidney -0.707 vs -0.507; mean left kidney -0.498 vs -0.524, respectively). Conclusion: Renal volumes need to be corrected to body surface area. After correction for body surface area, 5-year-old healthy and prematurely born children have comparable renal volumes

    The major leucyl aminopeptidase of Trypanosoma cruzi (LAPTc) assembles into a homohexamer and belongs to the M17 family of metallopeptidases

    <p>Abstract</p> <p>Background</p> <p>Pathogens depend on peptidase activities to accomplish many physiological processes, including interaction with their hosts, highlighting parasitic peptidases as potential drug targets. In this study, a major leucyl aminopeptidolytic activity was identified in <it>Trypanosoma cruzi</it>, the aetiological agent of Chagas disease.</p> <p>Results</p> <p>The enzyme was isolated from epimastigote forms of the parasite by a two-step chromatographic procedure and associated with a single 330-kDa homohexameric protein as determined by sedimentation velocity and light scattering experiments. Peptide mass fingerprinting identified the enzyme as the predicted <it>T. cruzi </it>aminopeptidase EAN97960. Molecular and enzymatic analysis indicated that this leucyl aminopeptidase of <it>T. cruzi </it>(LAPTc) belongs to the peptidase family M17 or leucyl aminopeptidase family. LAPTc has a strong dependence on neutral pH, is mesophilic and retains its oligomeric form up to 80°C. Conversely, its recombinant form is thermophilic and requires alkaline pH.</p> <p>Conclusions</p> <p>LAPTc is a 330-kDa homohexameric metalloaminopeptidase expressed by all <it>T. cruzi </it>forms and mediates the major parasite leucyl aminopeptidolytic activity. Since biosynthetic pathways for essential amino acids, including leucine, are lacking in <it>T. cruzi</it>, LAPTc could have a function in nutritional supply.</p

    Covid-19 in Children and the Influence on the Employment Activity of Their Female Caregivers: A Cross Sectional Gender Perspective Study

    INTRODUCTION: During the COVID-19 pandemic, women disproportionately assume more unpaid activities, affecting their employment. OBJECTIVE: Describe the influence of COVID-19 on the employment of caregivers of children and adolescents from a gender perspective. METHODS: Cross-sectional study in three high-complexity hospitals in Bogotá, Colombia from April 2020 to June 2021. A subsample of the FARA cohort was taken, including those patients with a positive test for SARS-COV2. We took as our analysis category children older than 8 years and younger than 18 years who had a positive SARS-COV2 test, as well as, caregivers of all children with a positive SARS-COV2 test. This subsample was drawn from the FARA cohort. A survey was applied to them. We carried out a descriptive and stratified analysis by age group, educational, and socioeconomic level. RESULTS: We included 60 surveys of caregivers and 10 surveys of children. The main caregiver in 94.8% of the cases was a female. At the beginning of the pandemic, 63.3% of the caregivers were employed, and 78.9% of those lost their employment. The vast majority of these caregiver were women (96.6%, CONCLUSION: Caregivers of children with COVID-19 with low educational levels and lower socioeconomic conditions, as well as those with children under 5 years showed greater likelihood of employment loss between the interviewed subsample

    Environmental health at La Chaparrala subbasin, Colombia 2015

    RESUMEN: Objetivo Evaluar condiciones de salud ambiental subcuenca la Chaparrala, Andes-Colombia 2015. Método Estudio descriptivo de corte transversal, se indagó sobre condiciones sanitarias y ambientales de las viviendas, entorno inmediato y prejuicios acerca de la salud ambiental, en encuesta a 117 familias asentadas sobre eje principal de la quebrada y se analizó calidad fisicoquímica del agua. Resultados Predominó el hombre como jefe de hogar, cuatro personas en promedio por vivienda, 69 urbanas y 48 rurales, con permanencia mayor a 20 años en las primeras y menor a cinco años en las segundas. La mayoría, propias con escritura, con conexión 75 % al acueducto y 73 % al alcantarillado. En zona urbana predominó techo en losa y piso en baldosa, en la rural, teja de barro y piso en cemento y paredes en adobe en ambas zonas. El agua de la quebrada cumplió parámetros fisicoquímicos establecidos en el reglamento técnico del sector de agua potable y saneamiento básico, y resolución No. 2115 de 2007, excepto Turbiedad y Nitritos y se hallaron ocurrencias y concurrencias sobre salud ambiental. Discusión Las características sociodemográficas y condiciones ambientales y sanitarias de las viviendas y sus alrededores, son similares a las reportadas en encuesta de demografía y salud 2010, y en Encuesta Nacional de Salud (ENDS) 2007. La quebrada puede seguir siendo fuente de abastecimiento para consumo humano con tratamiento convencional y como concurrencia, la salud ambiental con enfoque hacia el cuidado y protección del ambiente que difiere del concepto de Organización Mundial de la Salud (OMS).ABSTRACT: Objective To assess environmental health conditions at La Chaparrala subbasin, in the Andes Mountains of Colombia, during 2015. Method Descriptive cross-sectional study on sanitary and environmental conditions of the dwellings, as well as on immediate environment, and environmental health prejudices. A survey was applied to 117 families settled in the main axis of the creek. The physicochemical quality of the water was analyzed. Results Men were predominant as head of household, with an average of four people per house. 69 of the houses were in the urban area, while 48 were rural; the permanence was greater than 20 years for the first, and less than five years for the second. Most of the houses are owned with deeds, and 75 % of them had a connection to the aqueduct and 73 % to the sewer. The houses in the urban area were predominantly made of tile roof and tile floor, while rural houses were made of mud tile and concrete floor. Both types of construction had adobe walls. The water from the creek complied with the physicochemical parameters established in the technical regulation of the sector for drinking water and basic sanitation, and with resolution No. 2115 of 2007. Non-compliance was observed in turbidity and nitrite levels, and occurrences and concurrences on environmental health were found. Discussion The socio-demographic characteristics and environmental and health conditions of the dwellings and their surroundings are similar to those reported in Encuesta de Demografía y Salud 2010 (Demographic and Health Survey) and Encuesta Nacional de Salud 2007 (2007 National Health Survey). The creek may continue to be a source for human consumption with conventional treatment and environmental health directed to care and protect the environment, differing from the concept of World Health Organization (WHO)

    Urinary KIM-1 is not correlated with gestational age among 5-year-old children born prematurely

    BackgroundPreterm birth is associated with decreased nephron endowment. Currently, there is no reliable non-invasive biomarker to identify or monitor decreased nephron number in at-risk patients. Urinary Kidney Injury Molecule-1 (KIM-1) is a biomarker of acute and chronic renal injury. We measured urinary KIM-1 among a wide array of other potential biomarkers.MethodsWe conducted an ambispective cohort study of 5-years-old children born prematurely and healthy controls identified from city schools. Detailed anthropometrics, renal ultrasound dimensions, and biochemical parameters were measured. Urinary KIM-1 was measured using Luminex® technology. Age independent z-scores were calculated and compared. Spearman correlations were used for estimating the association between measures and KIM-1.ResultsWe enrolled 129 children, 97 (75.2%) born pre-term and 32 (24.8%) healthy controls born at full-term. Pre-term patients had significantly lower weight and body surface area than controls. Pre-term patients and controls did not differ in current age, sex, race, height, blood pressure, urinary sodium, fractional sodium excretion, serum creatinine and estimated GFR. All spearman correlation between KIM-1 and gestational age, renal and serum measurements were weak without statistical significanceConclusionIn 5-year-old children born prematurely, KIM-1 was not correlated with gestational age. Further prospective studies need to confirm this finding

    Factores de riesgo para trastornos de la alimentación en los alumnos de la Universidad de Manizales

    ResumenObjetivo: Determinar la frecuencia de los factores de riesgo para trastornos de la alimentaci&oacute;n en estudiantes de la Universidad de Manizales. Materiales y m&eacute;todos: De 3,610 estudiantes regulares de la Universidad&nbsp; de Manizales se tom&oacute; una muestra de 165 estudiantes, quienes respondieron un cuestionario integrado compuesto por variables demogr&aacute;ficas, las escalas de Zung para ansiedad o depresi&oacute;n,el Eating Disorderrs Inventory (EDI2) y el Apgar familiar; adem&aacute;s se tomaron las medidas de peso, talla, &iacute;ndice de masa corporal y pliegue en el antebrazo. Resultado: Un12.7% de la poblaci&oacute;n present&oacute; factor de riesgo positivo para trastornos de la conducta alimentar&iacute;a, 17.3% de la poblaci&oacute;n femenina y el 3.8% de la poblaci&oacute;n masculina; las personas con mayor factor de riesgo fueron las mujeres en la facultad de Comunicaci&oacute;n Social y Periodismo (24.1%). Se determin&oacute; una&nbsp; relaci&oacute;n significativa entre la variable impulso por la delgadez con la ansiedad o depresi&oacute;n. El mayor &iacute;ndice de masa corporal y de porcentaje de grasa est&aacute; asociado con un aumentoen la variable impulso por la delgadez. Conclusiones:Se encontraron factores de riesgo asociados con trastornos de la alimentaci&oacute;n.
