    Tato bakalářská práce se zabývá euroskepticismem v České republice a zájmy české ekonomiky. Hlavním cílem práce je identifikovat projevy euroskepticismu v České republice a zjistit, je-li opozice politických stran založena na faktech či zda vychází pouze ze stranické ideologie a jsou-li tyto názory v zájmu české ekonomiky. Práce je rozdělena do tří kapitol. V první kapitole se práce věnuje typologiím euroskepticismu a vývoji evropské ekonomické integrace. Druhá kapitola sleduje vliv členství v EU na českou ekonomiku. Třetí kapitola se věnuje euroskepticismu v ČR a kritickému zhodnocení jeho projevů v oblasti otázky přijetí eura a imigrace.This bachelor’s thesis deals with Euroscepticism in the Czech Republic and the influence of the EU on the Czech economy. The main goal of this bachelor’s thesis is to identify manifestations of Euroscepticism in the Czech Republic and to find out if the opposition to the EU of political parties is based on facts or only on party ideology and if these views are in the best interest of the Czech economy. The thesis is divided into three chapters. The first chapter deals with the typologies of Euroscepticism and the development of European economic integration. The second chapter examines the impact of EU membership on the Czech economy. The third chapter deals with Euroscepticism in the Czech Republic and critically evaluates how Euroscepticism manifests in the light of euro adoption and immigration

    Title: Case study of physiotherapy treatment of the patient with the diagnosis of ankylosing spondylitis. Objective: The main target of this bachelor's thesis was to take up with the issue of the ankylosing spondylitis, as the rheumatic disease and to outline the possibilities of its treatment. The next target was the elaboration of the case study of the patient with this diagnose, followed by the appraisal of the therapy's effect. Methods: In the general part is concerned with the Ankylosing spondylitis issue, namely with its symptoms, manifestations, methods of examination and a subsequent treatment of this disease. The second, special section of this bachelor thesis is focused on the individual patient with ankylosing spondylitis, physiotheapy assesment, subsequent therapy and the final effect of it. Results: The results of the physiotherapeutic care of the patient were observed to be positive in objective (clinical) and subjective way. The painful situations became milder and the patient assumed a positive approach to the kinesitherapy, which she didn't have till now.     Téma mé bakalářské práce je Koordinovaný vizuální styl neziskové organizace. Vybrala jsem si neziskovou organizaci Motýl o. s. v Plzni. Mým cílem bylo vytvořit zcela novou identitu, která by byla svěží, působivá a aktuální. Začala jsem s navržením loga a základního manuálu. Poté jsem se zaměřila na tiskoviny a nakonec jsem pracovala na katalogu, letáčcích a propagačních materiálech.ObhájenoMy topic for bachelor work is Coordinated visual identity of a nonprofit organization. I choose a nonprofit organization Motýl o. s. in Pilsen. I wanted to create a completely new identity for them that would be lush, impressive and contemporary. I started with a logo and basic manual. Next I created mercantile printed matter and in the end I designed brochures, leaflets and promotional materials

    Genetic Resistance to Avian Leukosis Viruses Induced by CRISPR/Cas9 Editing of Specific Receptor Genes in Chicken Cells

    Avian leukosis viruses (ALVs), which are pathogens of concern in domestic poultry, utilize specific receptor proteins for cell entry that are both necessary and sufficient for host susceptibility to a given ALV subgroup. This unequivocal relationship offers receptors as suitable targets of selection and biotechnological manipulation with the aim of obtaining virus-resistant poultry. This approach is further supported by the existence of natural knock-outs of receptor genes that segregate in inbred lines of chickens. We used CRISPR/Cas9 genome editing tools to introduce frame-shifting indel mutations into tva, tvc, and tvj loci encoding receptors for the A, C, and J ALV subgroups, respectively. For all three loci, the homozygous frame-shifting indels generating premature stop codons induced phenotypes which were fully resistant to the virus of respective subgroup. In the tvj locus, we also obtained in-frame deletions corroborating the importance of W38 and the four amino-acids preceding it. We demonstrate that CRISPR/Cas9-mediated knock-out or the fine editing of ALV receptor genes might be the first step in the development of virus-resistant chickens

    Retroviruses and retrovirus-derived vectors integrate nonrandomly into the genomes of host cells with specific preferences for transcribed genes, gene-rich regions, and CpG islands. However, the genomic features that influence the transcriptional activities of integrated retroviruses or retroviral vectors are poorly understood. We report here the cloning and characterization of avian sarcoma virus integration sites from chicken tumors. Growing progressively, dependent on high and stable expression of the transduced v-src oncogene, these tumors represent clonal expansions of cells bearing transcriptionally active replication-defective proviruses. Therefore, integration sites in our study distinguished genomic loci favorable for the expression of integrated retroviruses and gene transfer vectors. Analysis of integration sites from avian sarcoma virus-induced tumors showed strikingly nonrandom distribution, with proviruses found prevalently within or close to transcription units, particularly in genes broadly expressed in multiple tissues but not in tissue-specifically expressed genes. We infer that proviruses integrated in these genomic areas efficiently avoid transcriptional silencing and remain active for a long time during the growth of tumors. Defining the differences between unselected retroviral integration sites and sites selected for long-terminal-repeat-driven gene expression is relevant for retrovirus-mediated gene transfer and has ramifications for gene therapy

    Genetic Diversity of NHE1, Receptor for Subgroup J Avian Leukosis Virus, in Domestic Chicken and Wild Anseriform Species

    <div><p>J subgroup avian leukosis virus (ALV-J) infects domestic chicken, jungle fowl, and turkey and enters the host cell through a receptor encoded by <i>tvj</i> locus and identified as Na+/H+ exchanger 1 (NHE1). The resistance to ALV-J in a great majority of examined galliform species was explained by deletions or substitutions of the critical tryptophan 38 in the first extracellular loop of NHE1, and genetic polymorphisms around this site predict the susceptibility or resistance of a given species or individual. In this study, we examined the NHE1 polymorphism in domestic chicken breeds and documented quantitative differences in their susceptibility to ALV-J <i>in vitro</i>. In a panel of chicken breeds assembled with the aim to cover the maximum variability encountered in domestic chickens, we found a completely uniform sequence of NHE1 extracellular loop 1 (ECL1) without any source of genetic variation for the selection of ALV-J-resistant poultry. In parallel, we studied the natural polymorphisms of NHE1 in wild ducks and geese because of recent reports on ALV-J positivity in feral Asian species. In anseriform species, we demonstrate a specific and highly conserved critical ECL1 sequence without any homologue of tryptophan 38 in accordance with the resistance of duck cells to prototype ALV-J. Last, we demonstrated that the new Asian strains of ALV-J have not evolved their envelope glycoprotein to the entry the duck cells. Our results contribute substantially to the current discussion of possible heterotransmission of ALV-J and its spill-over into the wild ducks and geese.</p></div
