    Stability of nuclear and mitochondrial reference genes in selected tissues of the Ambrosia beetle Xylosandrus germanus

    SIMPLE SUMMARY: The ambrosia beetle Xylosandrus germanus (Blandford) is a destructive wood-boring insect of horticultural tree crops. A fungal mutualist is cultivated within host trees that provides the sole source of nutrition for the larvae and adults of this beetle. Female X. germanus adults use a pouch-like structure (i.e., mycangium) to maintain and transport spores of their fungal mutualist. To facilitate future studies examining gene expression of X. germanus’ mycangium, the identification of stable genes unaffected by experimental treatments is needed to provide a standard reference during gene expression studies. Selected tissue types were dissected from laboratory-reared and field-collected specimens of the ambrosia beetle X. germanus to evaluate the stability of five reference genes, namely, 28S ribosomal RNA (28S rRNA), arginine kinase (AK), carbamoyl-phosphate synthetase 2-aspartate transcarbamylase-dihydroorotase (CAD), mitochondrial cytochrome oxidase 1 (CO1), and elongation factor-1α (EF1α). The reference genes CO1 and AK were identified as primary and secondary reference genes. By contrast, EF1α was considered unsuitable for use as a reference gene during gene expression studies with X. germanus. These results will aid in normalizing the expression of target genes during studies with X. germanus. ABSTRACT: The fungus-farming ambrosia beetle Xylosandrus germanus (Blandford) uses a pouch-like structure (i.e., mycangium) to transport spores of its nutritional fungal mutualist. Our current study sought to identify reference genes necessary for future transcriptome analyses aimed at characterizing gene expression within the mycangium. Complementary DNA was synthesized using selected tissue types from laboratory-reared and field-collected X. germanus consisting of the whole body, head + thorax, deflated or inflated mycangium + scutellum, inflated mycangium, and thorax + abdomen. Quantitative reverse-transcription PCR reactions were performed using primers for 28S ribosomal RNA (28S rRNA), arginine kinase (AK), carbamoyl-phosphate synthetase 2-aspartate transcarbamylase-dihydroorotase (CAD), mitochondrial cytochrome oxidase 1 (CO1), and elongation factor-1α (EF1α). Reference gene stability was analyzed using GeNorm, NormFinder, BestKeeper, ΔCt, and a comprehensive final ranking by RefFinder. The gene CO1 was identified as the primary reference gene since it was generally ranked in first or second position among the tissue types containing the mycangium. Reference gene AK was identified as a secondary reference gene. In contrast, EF1α was generally ranked in the last or penultimate place. Identification of two stable reference genes will aid in normalizing the expression of target genes for subsequent gene expression studies of X. germanus’ mycangium

    The COVID-19 pandemic has had major impacts on PK-16 education in the United States. In- person teaching and learning opportunities have been disrupted and as a result, schools and institutions of higher education (IHE) have resorted to creative solutions to adjust and adapt to remote learning and social distancing. In particular, teacher preparation programs have wrestled with the limitations put in place by IHE as well as PK-12 school districts while trying to maintain high quality field experiences for pre-service teachers. In this action research report, we examine the implementation of a hybrid field experience model in a teacher preparation program at a four year university, and the perspectives of pre-service teacher participants and faculty supervisors engaged in this model

    Pan-European grading scales: lessons from national systems and the ECTS

    This article assesses the impact of the Bologna Process on the grading schemes of EU member countries. In light of some problems regarding the implementation of the European Credit Transfer system (ECTS), the author proposes further reforms and offers some elements of a unified grading system for European higher education. The author explores the variation among Europe’s grading systems and the resulting lessons learned are shared here. Lastly, this article also argues that principles of justice and fairness, deemed central to academic freedom, are best upheld by the use of a unified grading system at national and European levels

    Can demographic histories explain long-term isolation and recent pulses of asymmetric gene flow between highly divergent grey fox lineages?

    Secondary contact zones between deeply divergent, yet interfertile, lineages provide windows into the speciation process. North American grey foxes (Urocyon cinereoargenteus) are divided into western and eastern lineages that diverged approximately 1 million years ago. These ancient lineages currently hybridize in a relatively narrow zone of contact in the southern Great Plains, a pattern more commonly observed in smaller-bodied taxa, which suggests relatively recent contact after a long period of allopatry. Based on local ancestry inference with whole-genome sequencing (n = 43), we identified two distinct Holocene pulses of admixture. The older pulse (500–3500 YBP) reflected unidirectional gene flow from east to west, whereas the more recent pulse (70–200 YBP) of admixture was bi-directional. Augmented with genotyping-by-sequencing data from 216 additional foxes, demographic analyses indicated that the eastern lineage declined precipitously after divergence, remaining small throughout most of the late Pleistocene, and expanding only during the Holocene. Genetic diversity in the eastern lineage was highest in the southeast and lowest near the contact zone, consistent with a westward expansion. Concordantly, distribution modelling indicated that during their isolation, the most suitable habitat occurred far east of today\u27s contact zone or west of the Great Plains. Thus, long-term isolation was likely caused by the small, distant location of the eastern refugium, with recent contact reflecting a large increase in suitable habitat and corresponding demographic expansion from the eastern refugium. Ultimately, long-term isolation in grey foxes may reflect their specialized bio-climatic niche. This system presents an opportunity for future investigation of potential pre- and post-zygotic isolating mechanisms

    Mitigation of Iron and Aluminum Powder Deflagrations via Active Explosion Suppression in a 1 m3 Sphere Vessel

    Combustible metal dust explosions continue to present a significant threat to metal handling and refining industries. Addition of noncombustible inert material to combustible dust mixtures, through either premixing or high-rate injection as the incipient flame front begins to develop, is common practice for preventative inhibition or explosion protection via active suppression, respectively. Metal dusts demonstrate an extremely reactive explosion risk due to amplified heat of combustion, burning temperature, flame speed, explosibility parameters (KSt and Pmax), and ignition sensitivity. Inhibition efficiency of suppressant agents used for active mitigation is shown to be reliant on fuel explosibility, discrete burning mechanism, and combustion temperature range and thus may be increasingly variable depending on the fuel in question. For this reason, mitigation of metal powder deflagrations at moderate total suppressed pressures (relative to the overall strength of the enclosure) and at low agent concentrations remains challenging. This paper reviews recent metal dust suppression testing in a Fike Corporation’s 1 m3 sphere combustion chamber and evaluates the efficacy of multiple suppression agents (sodium bicarbonate [SBC], sodium chloride [Met-L-X], and monoammonium phosphate [MAP]) for the mitigation of iron and aluminum powder deflagrations at suspended fuel concentrations of 2250 and 500 g/m3, respectively

The \emph{size-Ramsey number} \sr(H) of a graph HH is defined to be \sr(H)=\min\{|E(G)|\colon G\rightarrow (H)_2\}. Answering a question of Conlon, we prove that, for every fixed~kk, we have \sr(P_n^k)=O(n), where~PnkP_n^k is the kkth power of the nn-vertex path PnP_n (i.e., the graph with vertex set V(Pn)V(P_n) and all edges {u,v}\{u,v\} such that the distance between uu and vv in PnP_n is at most kk).     Identification of regenerating island-derived protein 3E in dogs

    Regenerating islet-derived protein (REG) 1A (aka pancreatic stone protein) and REG3A (aka pancreatitis-associated protein) are upregulated in humans with sepsis, pancreatitis, and gastrointestinal diseases, but little is known about this protein family in dogs. Our aim was to identify REG1 and REG3 family members in dogs. REG-family genes were computationally annotated in the canine genome and proteome, with verification of gene expression using publicly available RNA-seq data. The presence of the protein in canine pancreatic tissue and plasma was investigated with Western blot and immunohistochemistry, using anti-human REG1A and REG3A antibodies. Protein identity was confirmed with mass spectrometry. Two members of the REG3 subfamily were found in the canine genome, REG3E1 and REG3E2, both encoding for the same 176 AA protein, subsequently named REG3E. Anti-human REG3A antibodies demonstrated cross-reactivity with the canine REG3E protein in pancreas homogenates. In canine plasma, a protein band of approximately 17 kDa was apparent. Mass spectrometry confirmed this protein to be the product of the two annotated REG3E genes. Strong immunoreactivity to anti-human REG3A antibodies was found in sections of canine pancreas affected with acute pancreatitis, but it was weak in healthy pancreatic tissue. Recombinant canine REG3E protein underwent a selective trypsin digestion as described in other species. No evidence for the presence of a homolog of REG1A in dogs was found in any of the investigations. In conclusion, dogs express REG3E in the pancreas, whose role as biomarker merits further investigations. Homologs to human REG1A are not likely to exist in dogs
