    Effects of the use of raw or cooked chickpeas and the sausage cooking time on the quality of a lamb-meat, olive-oil emulsion-type sausage

    Producción CientíficaReformulation of cooked sausages using high-protein plant-based food such as chickpea as meat extenders and vegetable oils to replace animal fat can be a suitable approach to promote the consumption of smaller portions of meat. The pre-processing of chickpea and the sausage cooking intensity can potentially affect the quality of reformulated sausages. In this study, an emulsion-type sausage made with lamb meat, chickpea and olive oil was prepared in triplicate following three different formulations containing the same targeted levels of protein (8.9%), lipids (21.5%), and starch (2.9%): control sausage (CON; control, without chickpea), and raw (RCP) and cooked chickpea (CCP) sausages (both with 7% chickpea). Sausages were cooked at 85 °C for two heating times (40 min or 80 min) and were analysed for weight loss, emulsion stability, colour, texture, lipid oxidation and volatile composition. Compared to CON sausages, the use of raw chickpea reduced the elasticity and significantly increased lipid oxidation during the sausage-making process resulting in changes in the volatile composition. The use of previously cooked chickpea, however, resulted in the sausages having greater cooking loss, hardness and chewiness than CON sausages, while there was no difference in lipid oxidation, and differences in volatile compounds were scarce. The reformulation with cooked chickpea could provide a sausage with more similarity to the CON sausage. The extended heating time of 80 min at 85 °C did not significantly affect the quality traits in either CON or reformulated sausages except for a higher cooking loss.Universidad Nacional Mayor de San Marcos, Project (A2008001m

    In a greenhouse experiment Brachiaria brizantha Stapf, cv. Marandu was grown in a nutrient solution. The treatments were a complete nutrient solution, nutrient solutions with the omissions of each macronutrient (N, P, K, Ca, Mg and S) and a control (with the omission of all macronutrients). Plants were harvested 50 days after transplanting. Dry matter yield, tiller number and plant height were recorded, and nutrient concentrations in shoot and root were determined. Omissions of N and P were the most limiting for plant growth, whereas K and Ca did not affect grass growth and tillering. Mg and S omissions resulted in significant decreases in the plant parameters. Great decreases in plant nutrient concentrations were verified in the treatments with nutrient omissions in solution. Visual symptoms of deficiencies were observed for each nutrient omission treatment, with those of N and P being the first and that of Ca being the latest shown by the plants. Studies on forms and sources of nutrients for nutrient solution to be used with this grass are suggested.Em experimento de casa de vegetação, cultivou-se Brachiaria brizantha Stapf. cv. Marandu em solução nutritiva, tendo como tratamentos a solução completa, as omissões individuais de N, P, K, Ca, Mg, S e a testemunha (com omissão dos macronutrientes). Fez-se o acompanhamento da manifestação dos sintomas visuais de deficiência e, 50 dias após o transplante, avaliou-se a produção de matéria seca (MS), número de perfilhos (NP), a altura das plantas (H), bem como a composição mineral da parte aérea e das raízes. As omissões de N e P limitaram o crescimento, ao passo que a omissão de K não afetou a produção de MS e NP. A omissão de Ca não trouxe, para o período considerado, restrições na produção de MS, NP e H. Nas omissões de Mg e S as plantas tiveram fraco desempenho, superando apenas os tratamentos onde foram omitidos o N e o P. A composição mineral da parte aérea só apresentou diferenças acentuadas nos teores para os nutrientes omitidos. Sintomas visuais de deficiência foram observados para todas as omissões, sendo os primeiros manifestados através da omissão do N e do P e os últimos, através da omissão do Ca. São sugeridos estudos para melhor definição do balanço entre formas e fontes de nutrientes para a solução nutritiva quando utilizada no cultivo de plantas de Brachiaria brizantha cv. Marandu

    New locus underlying auriculocondylar syndrome (ARCND): 430 kb duplication involving TWIST1 regulatory elements

    Background Auriculocondylar syndrome (ARCND) is a rare genetic disease that affects structures derived from the first and second pharyngeal arches, mainly resulting in micrognathia and auricular malformations. To date, pathogenic variants have been identified in three genes involved in the EDN1-DLX5/6 pathway (PLCB4, GNAI3 and EDN1) and some cases remain unsolved. Here we studied a large unsolved four-generation family. Methods We performed linkage analysis, resequencing and Capture-C to investigate the causative variant of this family. To test the pathogenicity of the CNV found, we modelled the disease in patient craniofacial progenitor cells, including induced pluripotent cell (iPSC)-derived neural crest and mesenchymal cells. Results This study highlights a fourth locus causative of ARCND, represented by a tandem duplication of 430 kb in a candidate region on chromosome 7 defined by linkage analysis. This duplication segregates with the disease in the family (LOD score=2.88) and includes HDAC9, which is located over 200 kb telomeric to the top candidate gene TWIST1. Notably, Capture-C analysis revealed multiple cis interactions between the TWIST1 promoter and possible regulatory elements within the duplicated region. Modelling of the disease revealed an increased expression of HDAC9 and its neighbouring gene, TWIST1, in neural crest cells. We also identified decreased migration of iPSC-derived neural crest cells together with dysregulation of osteogenic differentiation in iPSC-affected mesenchymal stem cells. Conclusion Our findings support the hypothesis that the 430 kb duplication is causative of the ARCND phenotype in this family and that deregulation of TWIST1 expression during craniofacial development can contribute to the phenotype.Molecular Technology and Informatics for Personalised Medicine and Healt

    On the reliability of a simple method for scoring phenotypes to estimate heritability: A case study with pupal color in Heliconius erato phyllis, Fabricius 1775 (Lepidoptera, Nymphalidae)

    In this paper, two methods for assessing the degree of melanization of pupal exuviae from the butterfly Heliconius erato phyllis, Fabricius 1775 (Lepidoptera, Nymphalidae, Heliconiini) are compared. In the first method, which was qualitative, the exuviae were classified by scoring the degree of melanization, whereas in the second method, which was quantitative, the exuviae were classified by optical density followed by analysis with appropriate software. The heritability (h2) of the degree of melanization was estimated by regression and analysis of variance. The estimates of h 2 were similar with both methods, indicating that the qualitative method could be particularly suitable for field work. The low estimates obtained for heritability may have resulted from the small sample size (n = 7-18 broods, including the parents) or from the allocation-priority hypothesis in which pupal color would be a lower priority trait compared to morphological traits and adequate larval development

    Non-existence and uniqueness results for supercritical semilinear elliptic equations

    Non-existence and uniqueness results are proved for several local and non-local supercritical bifurcation problems involving a semilinear elliptic equation depending on a parameter. The domain is star-shaped but no other symmetry assumption is required. Uniqueness holds when the bifurcation parameter is in a certain range. Our approach can be seen, in some cases, as an extension of non-existence results for non-trivial solutions. It is based on Rellich-Pohozaev type estimates. Semilinear elliptic equations naturally arise in many applications, for instance in astrophysics, hydrodynamics or thermodynamics. We simplify the proof of earlier results by K. Schmitt and R. Schaaf in the so-called local multiplicative case, extend them to the case of a non-local dependence on the bifurcation parameter and to the additive case, both in local and non-local settings.Comment: Annales Henri Poincar\'e (2009) to appea
