Transcriptomic fingerprinting of Pseudomonas putida under alternative physiological regimes
Pseudomonas putida KT2440 is a metabolically versatile soil bacterium useful both as a model biodegradative organism and as a host of catalytic activities of biotechnological interest. In this report, we present the high-resolution transcriptome of P. putida cultured on different carbon sources as revealed by deep sequencing of the corresponding RNA pools. Examination of the data from growth on substrates that are processed through distinct pathways (glucose, fructose, succinate and glycerol) revealed that > 20% of the P. putida genome is differentially expressed depending on the ensuing physiological regime. Changes affected not only metabolic genes but also a suite of global regulators e.g. the rpoS sigma subunit of RNAP, various cold-shock proteins and the three HU histone-like proteins. Specifically, the genes encoding HU subunit variants hupA, hupB and hupN drastically altered their expression levels (and thus their ability to form heterodimeric combinations) under the diverse growth conditions. Furthermore, we found that two small RNAs, crcZ and crcY, known to inhibit the Crc protein that mediates catabolite repression in P. putida, were both down-regulated by glucose. Desenvolvimento, produtividade e atributos qualitativos de cultivares de cana-de-açúcar fertirrigadas por gotejamento subsuperficial
The present study aimed to evaluate the development, yield and quality of four sugarcane cultivars fertigated by subsurface drip system. The experiment was carried out in Campinas-SP, Brazil, from January 2012 to November 2013, with the cultivars SP79-1011, IACSP94-2101, IACSP94-2094 and IACSP95-5000 subjected to daily irrigations. The irrigations depths were applied to bring soil moisture to field capacity. Soil moisture was monitored using soil moisture probes. Samples were collected along the crop cycle in order to evaluate crop development and yield, at the end of the first and second ratoons. Stalk height showed good correlation for the estimation of crop yield, with R2 equal to or higher than 0.96. The cultivar IACSP95-5000 showed the highest yield in the first ratoon. In the second ratoon the highest yield was observed in IACSP94-2101, followed by IACSP95-5000 and SP79-1011. Considering the yield results associated with the technological analysis, such as soluble solids content and apparent sucrose, the cultivar IACSP95-5000 excelled the others in the cultivation under subsurface drip irrigation.Objetivou-se, no presente estudo, avaliar o desenvolvimento, a produtividade e a qualidade de quatro cultivares de canas-de-açúcar fertirrigadas por gotejamento subsuperficial. O experimento foi realizado em Campinas, SP, Brasil, no período de janeiro de 2012 a novembro de 2013, com as cultivares SP79-1011, IACSP94-2101, IACSP94-2094 e IACSP95-5000 com irrigações diárias, sendo a lâmina aplicada para repor a umidade do solo à capacidade de campo. Para tanto foram utilizadas sondas visando à medição da umidade do solo enquanto ao longo dos ciclos foram realizadas biometrias e estimativas da produtividade nas socas. A produtividade final foi estimada no fim de cada soca. A altura dos colmos apresentou boa correlação para estimativa da produtividade da cultura com R2 igual ou maior que 0,96. A maior produtividade na primeira soca foi observada na cultivar IACSP95-5000 e na segunda soca a IACSP94-2101 foi a cultivar mais produtiva seguida das IACSP95-5000 e SP79-1011. Considerando os resultados de produtividade associados à análise tecnológica, como os teores de sólidos solúveis e sacarose aparente, a IACSP95-5000 se sobressaiu no cultivo com irrigação por gotejamento subsuperficial.20652553

Adequação do mosto de uvas Niagara Rosada para vinificação através de desidratação parcial dos cachos
This study aimed to verify the influence of partial dehydration of Niagara Rosada grape clusters in physicochemical quality of the pre- fermentation must. In Brazil, during the winemaking process it is common to need to adjust the grape must when the physicochemical characteristics of the raw material are insufficient to produce wines in accordance with the Brazilian legislation for classification of beverages, which establishes the minimum alcohol content of 8.6 % for the beverage to be considered wine. Therefore, given that the reduction in the water content of grape berries allows the concentration of chemical compounds present in its composition, especially the concentration of total soluble solids, we proceeded with the treatments that were formed by the combination of two temperatures (T1-37.1ºC and T2-22.9 ºC) two air speeds (S1: 1.79 m s-1 and S2: 3.21 m s-1) and a control (T0) that has not gone through the dehydration treatment. Analysis of pH, Total Titratable Acidity (TTA) were performed in mEq L-1, Total Soluble Solids (TSS) in ºBrix, water content on a dry basis and Concentration of Phenolic Compounds (CPC) in mg of gallic acid per 100g of must. The average comparison test identified statistically significant modifications for the adaptation of must for winemaking purposes, having the treatment with 22.9 ºC and air speed of 1.79 m s-1 shown the largest increase in the concentration of total soluble solids, followed by the second best result for concentration of phenolic compounds.3418692Este estudo teve como objetivo verificar a influência da desidratação parcial de cachos de uva Niagara Rosada na qualidade físico-química do mosto pré-fermentativo. No Brasil, durante o processo de vinificação, é comum a necessidade de adequação do mosto de uvas quando as características físico-químicas da matéria-prima são insuficientes para a obtenção de vinhos em conformidade com a legislação brasileira para classificação de bebidas, a qual estabelece o teor alcoólico mínimo de 8,6% para que a bebida seja considerada vinho. Portanto, tendo em vista que a redução do teor de água das bagas da uva possibilita a concentração de compostos químicos presentes em sua composição, principalmente a concentração de sólidos solúveis totais, procedeu-se aos tratamentos que foram constituídos pela combinação de duas temperaturas (T1- 37,1 ºC e T2 - 22,9 ºC), duas velocidades de ar (V1- 1,79 m s-1 e V2-3,21 m s-1) e uma testemunha (T0), que não passou pelo tratamento de desidratação. Foram realizadas análises de pH, Acidez Total Titulável (ATT) em mEq L-1, Sólidos Solúveis Totais (SST) em ºBrix, teor de água em base seca e Concentração de Compostos Fenólicos (CCF) em mg de ácido gálico por 100 g de mosto. O teste de comparação de médias identificou modificações estatisticamente significativas para a adequação do mosto no intuito de vinificação, tendo o tratamento com 22,9 ºC e velocidade de ar de 1,79 m s-1 apresentado o maior incremento na concentração de sólidos solúveis totais, seguido do segundo melhor resultado para a concentração de compostos fenólicos.||This study aimed to verify the influence of partial dehydration of Niagara Rosada grape clusters in physicochemical quality of the pre- fermentation must. DESiRED -- Dynamic, Enhanced, and Smart iRED: A P4-AQM with Deep Reinforcement Learning and In-band Network Telemetry
Active Queue Management (AQM) is a mechanism employed to alleviate transient
congestion in network device buffers, such as routers and switches. Traditional
AQM algorithms use fixed thresholds, like target delay or queue occupancy, to
compute random packet drop probabilities. A very small target delay can
increase packet losses and reduce link utilization, while a large target delay
may increase queueing delays while lowering drop probability. Due to dynamic
network traffic characteristics, where traffic fluctuations can lead to
significant queue variations, maintaining a fixed threshold AQM may not suit
all applications. Consequently, we explore the question: \textit{What is the
ideal threshold (target delay) for AQMs?} In this work, we introduce DESiRED
(Dynamic, Enhanced, and Smart iRED), a P4-based AQM that leverages precise
network feedback from In-band Network Telemetry (INT) to feed a Deep
Reinforcement Learning (DRL) model. This model dynamically adjusts the target
delay based on rewards that maximize application Quality of Service (QoS). We
evaluate DESiRED in a realistic P4-based test environment running an MPEG-DASH
service. Our findings demonstrate up to a 90x reduction in video stall and a
42x increase in high-resolution video playback quality when the target delay is

Subglottic stenosis as a clinical manifestation of Wegener's granulomatosis in adolescents: report of a case and review of literature
Wegener's Granulomatosis is a well-characterized systemic vasculitis and necrotising granulomatous inflammation of the upper, lower respiratory tracts and the kidneys. The initial manifestations of the disease usually occur in patients over than twenty years old. The otolaryngologic symptoms like rhinorrhea, recurrent sinusitis and epistaxis are commonly present in early course of the disease. It seems that subglottic stenosis is correlated to Wegener's granumatosis in adolescents. We describe a case of a patient that developed subglottic stenosis associated with Wegener's granulomatosis in childhood during the course of the disease.A Granulomatose de Wegener é uma patologia caracterizada por vasculite sistêmica e inflamação granulomatosa necrotizante que compromete o trato respiratório alto, pulmões e rins, cujas manifestações iniciais, na maioria das vezes, ocorre em maiores de 20 anos de idade. As queixas clínicas otorrinolaringológicas estão, com freqüência, presentes nas fases iniciais da doença, sendo rinite, sinusopatia de repetição e epistaxe as mais comuns. A estenose subglógica parece estar correlacionada com a Granulomatose de Wegener em adolescentes. Principales promotores utilizados en la transformación genética de plantas
El conocimiento pleno de los promotores determina el éxito en la obtención de nuevos cultivares de plantas a través de técnicas biotecnológicas, ya que dicha secuencia del ADN regula la transcripción de otras regiones adyacentes o cercanas, encontrándose los siguientes promotores: constitutivos, tejido-específicos o estadio-específicos, inducibles y sintéticos. En esta revisión se resume de manera precisa los conceptos, ventajas y limitaciones de los distintos tipos de promotores, con ejemplos claros de ello.Palabras clave: promotor, biotecnología vegetal, transcripción genética.Full knowledge of promoters determines success in obtaining new plant cultivars through biotechnology techniques. This DNA sequence regulates the transcription of adjacent or nearby regions, which are mainly constitutive, tissue-specific or stage-specific, inducible and synthetic. Genomic analysis of four strains of Corynebacterium pseudotuberculosis bv. Equi isolated from horses showing distinct signs of infection.
The genomes of four strains (MB11, MB14, MB30, and MB66) of the species Corynebacterium pseudotuberculosis biovar equi were sequenced on the Ion Torrent PGM platform, completely assembled, and their gene content and structure were analyzed. The strains were isolated from horses with distinct signs of infection, including ulcerative lymphangitis, external abscesses on the chest, or internal abscesses on the liver, kidneys, and lungs. The average size of the genomes was 2.3 Mbp, with 2169 (Strain MB11) to 2235 (Strain MB14) predicted coding sequences (CDSs). An optical map of the MB11 strain generated using the KpnI restriction enzyme showed that the approach used to assemble the genome was satisfactory, producing good alignment between the sequence observed in vitro and that obtained in silico. In the resulting Neighbor-Joining dendrogram, the C. pseudotuberculosis strains sequenced in this study were clustered into a single clade supported by a high bootstrap value. The structural analysis showed that the genomes of the MB11 and MB14 strains were very similar, while the MB30 and MB66 strains had several inverted regions. The observed genomic characteristics were similar to those described for other strains of the same species, despite the number of inversions found. These genomes will serve as a basis for determining the relationship between the genotype of the pathogen and the type of infection that it causes
The Advertisement Calls and Distribution of Two Sympatric Species of Chiasmocleis (Méhely 1904) (Anura, Microhylidae, Gastrophryninae) from the Atlantic Forest
The advertisement calls of Chiasmocleis cordeiroi and C. crucis are described for populations from the municipalities of Igrapiúna and Camacan, respectively, state of Bahia, Brazil. Both calls consist of multipulsed notes produced in series. Differences between the two calls are: dominant frequency, higher in C. cordeiroi (range 4500–4898 Hz; C. crucis range 4069–4435 Hz); note rate, higher in C. cordeiroi (range 6.20–7.46 s/note; C. crucis range 5.17–5.59 s/note); pulse rate, higher in C. cordeiroi (151.82–194.83 s/note; C. crucis range 125.30– 142.12 s/note); and the structure of the modulation patterns of the notes. Moreover, the advertisement calls of C. crucis and C. cordeiroi are more similar than the calls of all syntopic congeners. Furthermore, the current distribution of both species was extended
Genome Sequence of Corynebacterium pseudotuberculosis MB20 bv. equi Isolated from a Pectoral Abscess of an Oldenburg Horse in California.
The genome of Corynebacterium pseudotuberculosis MB20 bv. equi was sequenced using the Ion Personal Genome Machine (PGM) platform, and showed a size of 2,363,089 bp, with 2,365 coding sequences and a GC content of 52.1%. These results will serve as a basis for further studies on the pathogenicity of C. pseudotuberculosis bv. equi
