Development of substrates for reconstructed human dermis based on polyhydroxyalkanoates (PHAs) and FucoPol
Skin tissue engineering represents an important tool in various fields such as cosmetic testing
and regenerative medicine. The possibility of reproducing in vitro the fundamental properties of in
vivo skin introduces new opportunities for creating knowledge. One of the techniques employed is
the production of scaffolds to harbor cells, allowing for their proliferation and differentiation into
functional tissues. These scaffolds must have certain characteristics, such as biocompatibility,
biodegradability, appropriate physico-chemical and mechanical properties, ease of production and
cost-effectiveness, all dependent on the material chosen. Thus, polyhydroxyalkanoates (PHAs)
represent a potential scaffold material due to all of their relevant characteristics for biomedical
applications. Also, the exopolysaccharide FucoPol can be incorporated with PHA as a blend or
coating, possibly introducing bioactivity to the scaffolds. Therefore, the aim of this work was the
production of scaffolds for reconstruction of human dermis, through the emulsion templating method,
resulting in porous constructs that were tested with human dermal fibroblasts (HDFn).
The scaffolds obtained in this work were based on P(HBHVHHx), a PHA composed of 51 wt%
3-hydroxybutyrate, 18 wt% 3-hydroxyvalerate and 31 wt% 3-hydroxyhexanoate, and had a porous
structure with some interconnected pores. The scaffolds produced through emulsion with water had
mechanical, thermal and physical properties comparable with the scaffolds produced through
emulsion with FucoPol solution, both of them suitable for skin tissue engineering.
These scaffolds were tested with HDFn to assess their bioactivity. In both the cells were able to
adhere, proliferate and differentiate, showing possible extracellular matrix (ECM) deposition. The
eight-day assay revealed increased cellular growth and organized ECM deposition in the scaffolds
with FucoPol compared to the ones without. The fourteen-day assay confirmed pore geometry as the
limiting factor for the full invasion of the scaffolds. This work demonstrated the potential of naturally
derived P(HBHVHHx)/FucoPol scaffolds for the reconstruction of human dermis.A engenharia de tecidos da pele representa uma ferramenta importante em várias áreas, como
a cosmética e a medicina regenerativa. A possibilidade de reproduzir in vitro as propriedades
fundamentais da pele in vivo, introduz oportunidades para criar conhecimento. Uma das técnicas
aplicada é o desenvolvimento de scaffolds para cultura celular, permitindo a sua proliferação e
diferenciação em tecidos funcionais. Estes scaffolds devem possuir certas características, tais como
biocompatibilidade, biodegradibilidade, propriedades físico-químicas e mecânicas apropriadas,
facilidade de produção e custo-eficácia, todas dependentes do material escolhido. Assim, os
polihidroxialcanoatos (PHAs) representam um potencial material para este efeito, devido a todas as
suas características relevantes para aplicações biomédicas. Adicionalmente, o exopolissacárido
FucoPol pode ser incorporado como uma mistura ou revestimento do scaffold de PHA, possivelmente
introduzindo bioatividade. O objetivo deste trabalho foi produzir scaffolds para reconstrução da
derme humana, através da técnica de emulsão, resultando em estruturas porosas que foram
testadas in vitro com fibroblastos humanos (HDFn).
Os scaffolds de PHA produzidos, com composição monomérica 51 wt% 3-hidroxibutirato, 18
wt% 3-hidroxivalerato e 31 wt% 3-hidroxihexanoato, resultaram numa estrutura porosa com alguns
poros interconectados. A emulsão com água resultou em scaffolds com propriedades mecânicas,
térmicas e físicas comparáveis aos obtidos através de emulsão com solução aquosa de FucoPol,
sendo ambos adequados para a aplicação pretendida.
Estes scaffolds foram testados com HDFn para avaliar a bioactividade. Em ambos as células
conseguiram aderir, proliferar e diferenciar, com possível deposição de matriz extracelular (ECM). O
ensaio de oito dias revelou um aumento no crescimento celular e deposição organizada de ECM nos
scaffolds com FucoPol em detrimento dos scaffolds sem FucoPol. O ensaio de catorze dias
confirmou a geometria dos poros como sendo o fator limitante para a invasão completa dos scaffolds.
Este trabalho demonstrou o potencial de scaffolds P(HBHVHHx)/FucoPol para a reconstrução de
derme humana
Antecedentes e determinantes da construção social da reputação do negociador
A reputação como factor determinante para um negociador e para a negociação está
muitíssimo pouco estudado.Neste sentido é relevante perceber um pouco mais acerca do
mesmo. Este projecto de investigação pretende, por essa razão, aprofundar o estudo da
importância da reputação e do seu conhecimento prévio, em processos de negociação.
Baseando-nos na literatura percebemos que o sexo do negociador e a partilha de informação
são importantes na construção de uma impressão. Esta investigação pretendeu testar o efeito
do sexo do negociador e da partilha de informação na percepção da reputação do negociador.
Por outro lado pretendemos investigar também se a motivação social de quem observa é um
factor importante na construção da reputação de um negociador. Utilizámos simulações de
negociação gravadas em vídeo e um questionário para aferir o efeito das variáveis em 120
participantes. Concluímos que existe um efeito da partilha de informação e da motivação
social do observador na construção da reputação do negociador. Porém, não se encontrou
efeitos significativamente relevantes do efeito do sexo do negociador na percepção da
reputação.Reputation as a determining factor to a negotiator and to negotiation processes is well
understudied. This makes it relevant to investigate more on this subject. This investigation
project intends to dig a little deeper on the importance of reputation and its prior knowledge
on negotiation processes.
Basing on literature, we understood that bargainer´s gender and information sharing
are two important pieces when forming an impression of someone. This project explored the
effect of gender and information sharing as antecedents of reputation. Furthermore, we intend
to investigate if the observer´s social motives are an underlying factor in constructing one’s
reputation.
We used video simulated negotiations and a questionnaire to test the variables on 120
participants. The results suggest that there is an effect of information sharing and of
observer’s social motives on negotiator’s reputation. However, there was no statisticaly
significant influence of gender on reputation
Inflammatory biomarkers and perinatal depression: a systematic review
This article is a preprint and has not been peer-reviewed [what does this mean?]. It reports new medical research that has yet to be evaluated and so should not be used to guide clinical practice.Background: Approximately 10 to 20% of pregnant women worldwide experience perinatal depression (PND), a depressive episode with onset during pregnancy or after childbirth. We performed a systematic review to identify, summarize and discuss
studies on inflammatory biomarkers described in relation to PND.
Methods: Inclusion criteria defined the selection of observational studies written in
English, French, Spanish or Portuguese, that evaluate analytical levels of inflammatory
molecules (protein levels) in biological fluids in women, with a diagnosis of depression
using ICD/DSM diagnostic criteria or depressive symptoms assessed by standardized
psychometric instruments, during pregnancy and/or postpartum. Case reports,
experimental studies, reviews, qualitative analysis, meta-analysis, gray literature or
replicated data were excluded. Three electronic databases were used for search
(Pubmed, Web of Science and PsychInfo) and quality assessment of selected studies
were performed using the Newcastle-Ottawa Scale. Data extraction included study
design; number of subjects; obstetric information; tools and timepoints of depression
and inflammatory markers assessment.
Results: 56 studies where the major aim was to analyze the association between
depression and inflammatory biomarkers during pregnancy and postpartum period were
included in this systematic review. Overall, the findings of our systematic review lend
support to the hypothesis that several inflammatory markers may be associated with
peripartum depressive symptoms. The associations were somewhat different looking at
pregnancy compared to the delivery time-point and postpartum, and mainly referred to
increased levels of IL-6, IL-8, CRP and TNF-α among depressed.
Discussion: Our results revealed high heterogeneity in relation to the timing of
biological sampling for markers, as well as timing and instruments used for depression
assessment within the perinatal period for the different studies. Studies differed also in relation to use of biomarkers or depression as exposure and outcome respectively, and
whether these were addressed at the same timepoint or separate ones.
Given the high burden of PND on women, children and families, it is crucial to try to
harmonize methods used in related studies, in order to be able to pool results that could
give us insights into the pathophysiological mechanisms behind how the immune
system and PND are connected; this could have great impact on early detection,

© 2023 by the authors.This work focused on the development of porous scaffolds based on biocomposites comprising two biodegradable and biocompatible biopolymers: a terpolyester, poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-3-hydroxyhexanoate) (PHBHVHHx), and the bacterial polysaccharide FucoPol. The PHBHVHHx terpolymer was composed of 3-hydroxybutyrate (55 wt%), 3-hydroxyvalerate (21 wt%), and 3-hydroxyhexanoate (24 wt%). This hydrophobic polyester has low crystallinity and can form elastic and flexible films. Fucopol is a fucose-containing water-soluble polysaccharide that forms viscous solutions with shear thinning behavior and has demonstrated emulsion-forming and stabilizing capacity and wound healing ability. Emulsion-templating was used to fabricate PHA-based porous structures in which FucoPol acted as a bioemulsifier. Compared with the scaffolds obtained from emulsions with only water, the use of FucoPol aqueous solutions resulted in structures with improved mechanical properties, namely higher tensile strength (4.4 MPa) and a higher Young’s Modulus (85 MPa), together with an elongation at break of 52%. These features, together with the scaffolds’ high porosity and pore interconnectivity, suggest their potential to sustain cell adhesion and proliferation, which is further supported by FucoPol’s demonstrated wound healing ability. Therefore, the developed PHBHVHHx:FucoPol scaffolds arise as innovative porous bioactive structures with great potential for use in tissue engineering applications.publishersversionpublishe
Antioxidant and antimycotic activities of two native lavandula species from portugal
The antioxidant and antimycotic activities of the essential oils and extracts of two native Portuguese Lavandula species, L. stoechas subsp. luisieri and L. pedunculata, were evaluated by in vitro assays. The total phenolics and flavonoids content were also determined. The antioxidant potential was assessed through DPPH radical scavenging, inhibition of lipid peroxidation (ILP), and DNA protection assays. All samples displayed a high DPPH scavenging activity, some of them showing concentration dependence. The majority of the samples were also able to inhibit lipid peroxidation. A strong correlation was observed between the results of DPPH and ILP assays and the flavonoids content of the samples. In the DNA protection assay, all the extracts were able to preserve DNA integrity. The antimycotic activity was performed against twelve fungi belonging to Basidiomycota and Ascomycota Divisions. L. stoechas subsp. luisieri exhibited the broadest activity spectra. L. pedunculata extracts were active against five fungi. Cryptococcus neoformans was the most sensitive, being inhibited by all the extracts. Our results led to the conclusion that L. stoechas subsp. luisieri and L. pedunculata can be useful as new sources of natural antioxidants and antimycotic agents, providing a possible valorization of the existing biodiversity and resources of Portuguese flora
Depressão e ideação suicida na adolescência: implementação e avaliação de um programa de intervenção
Objective: To develop, implement and evaluate a suicide prevention program aimed at adolescents.Material and Methods: A quasi-experimental study, before and after, with adolescents from an educational institution in São Luís, Maranhão, Brazil. The study followed the stages of development, implementation and evaluation of a suicide prevention intervention program. Three assessment instruments were administered before and after the intervention: Beck's Suicidal Ideation Scale; Beck's Depression Inventory and Beck's Hopelessness Scale. Results: Before the intervention, 102 adolescents participated, 30 (29.4%) had suicidal ideation and depressive symptoms. After the intervention the instruments were applied to the 30 selected adolescents, 12 (40.0%) continued with ideation. Prior to intervention, the average Depression Inventory was 23.83 and then 7.17 (p <0.0001). As for hopelessness, the average obtained before was 7.23 and then 2.17 (p <0.0001); Regarding suicidal ideation, the average obtained before was 10.50 and then 2.57 (p <0.0001). Conclusion: There was a decrease in depressive symptoms, hopelessness and suicidal ideation after the implementation of the elaborated intervention program.Objetivo: Desarrollar, implementar y evaluar un programa de prevención del suicidio dirigido a adolescentes.Materiales y Métodos: Estudio cuasi experimental, del tipo antes y después, con adolescentes de una institución educativa en São Luís, Maranhão, Brasil. El estudio siguió las etapas de desarrollo, implementación y evaluación de un programa de intervención para la prevención del suicidio. Se administraron tres instrumentos de evaluación antes y después de la intervención: la escala de ideación suicida de Beck; el Inventario de Depresión de Beck y la Escala de Desesperanza de Beck.Resultados: Antes de la intervención, participaron 102 adolescentes, 30 (29.4%) tenían ideación suicida y síntomas depresivos. Después de la intervención, los instrumentos se aplicaron a los 30 adolescentes seleccionados, 12 (40.0%) continuaron con la ideación. Antes de la intervención, el Inventario de depresión promedio fue de 23.83 y luego de 7.17 (p <0.0001). En cuanto a la desesperanza, el promedio obtenido antes fue 7.23 y luego 2.17 (p <0.0001); Con respecto a la ideación suicida, el promedio obtenido antes fue de 10.50 y luego de 2.57 (p <0.0001).Conclusión: Hubo una disminución en los síntomas depresivos, la desesperanza y la ideación suicida después de la implementación del elaborado programa de intervención.Objetivo: Analisar os resultados do desenvolvimento e implementação de um programa de prevenção do suicidio dirigido a adolescentes.Materiais e Métodos: Estudo quase experiemntal, do tipo antes e depois, com adolescentes de uma Instituição de Ensino em São Luís, Maranhão, Brasil. O estudo seguiu as etpas de desenvolvimento, implementação e avaliação de um programa de intervenção direcionado a prevenção do suicídio. Health vulnerability in border region and Social Skills in adolescents
Introdução: Estudo buscou analisar a associação da vulnerabilidade em saúde na região de fronteira e as habilidades sociais de adolescentes.
Métodos: Estudo de coorte transversal realizado com 722 adolescentes. A relação da vulnerabilidade com as Habilidades Sociais foi analisada no grupo geral e estratificada por gênero. O Teste Chi-quadrado e a Regressão Logística Binomial foram usados.
Resultados: Demonstrou-se uma associação entre fatores de risco em saúde e as habilidades sociais de adolescentes, sendo relevantes: 1) O consumo de álcool, sentimento de solidão e envolvimento em brigas com o auto-controle; 2) Indicadores mais amplos de saúde sexual com as habilidades sociais; 3) O bullying, regulação parental com a empatia, e por fim 4) A atividade
física, regulação parental com a desenvoltura social.
Conclusões: O estudo demonstra a associação entre o repertório de habilidades sociais e a vulnerabilidade em saúde de adolescentes em região de fronteira, apontando evidências de programas para futuras pesquisas.Introduction: The study sought to analyze the association between health vulnerability in the border
region and the social skills of adolescents. Methods: Cross-sectional cohort study carried out with 722 adolescents. The relationship between vulnerability and Social Skills was analyzed in the general group and stratified by gender. Chi-square Test and Binomial Logistic Regression were used. Results: An association was demonstrated between health risk factors and adolescents' social
skills, the following being relevant: 1) Alcohol consumption, feelings of loneliness and involvement in fights with self-control; 2) Broader indicators of sexual health with social skills; 3) Bullying, parental regulation with empathy, and finally 4) Physical activity, parental regulation with social resourcefulness.
Conclusions: The study demonstrates the association between the repertoire of social skills and the
health vulnerability of adolescents in the border region, pointing to evidence of programs to instrument future research
Intervenções em competências sociais com adolescentes e o desenvolvimento de atitudes positivas em situação de vulnerabilidade: uma revisão sistemática
O objectivo deste trabalho foi avaliar evidências
científicas oriundas de estudos de intervenções em
competências sociais com adolescentes para o desenvolvimento
de atitudes positivas em situação de
vulnerabilidade. Os estudos foram identificados a
partir de buscas nas bases de dados LILACS, PubMed,
PsycINFO e SCOPUS no período de 2008 até 2018. Os
critérios de selecção foram publicações em inglês,
português ou espanhol, estudos primários de intervenção
em competências sociais e participantes adolescentes.
A revisão incluiu 17 estudos relacionados
a intervenções em competências sociais utilizando
como estratégias instruções e actividades semanais,
instruções e grupos com interacção social, somente
instruções e sistema mobile coach. As intervenções
em competências sociais com adolescentes em situação
de vulnerabilidade são estratégias eficazes
para o desenvolvimento de atitudes positivas. Estes
resultados fornecem um guia baseado em evidências
para investigadores, profissionais e instituições para
a prática em prevenção e promoção a saúde de adolescentesEl objetivo de este trabajo fue evaluar la evidencia

Estos hallazgos proporcionan una guía basada en
evidencia para investigadores, profesionales e instituciones

Os guardiões da horta
“Os Guardiões da Horta” consiste num conjunto de oficinais mensais (ano letivo 22/23) desenhadas, montadas e dinamizados por STOL-Science Through Our Lives/Dep. Biologia-UMinho (STOL) e que vão ao encontro do interesse do Colégio D. Pedro V (DPV) em explorar a sua horta de um modo consistente, investigativo e com aquisição prática de conhecimentos enunciados nas aprendizagens essenciais do 1º Ciclo. Todas as oficinas tiveram como ponto de partida a recolha das conceções prévias dos alunos e versaram os seguintes temas gerais: as formigas, as sementes, o solo, plantar e semear, os polinizadores.
Cada oficina teve início com a leitura expressiva de uma história alusiva ao tema (ou visualisada em vídeo) e avançou para a sua exploração hands on e usando imagens, artefatos e modelos tridimensionais. Quando a pesquisa não retribuia uma história suficientemente adequada à temática, ela era escrita de raíz (como aconteceu com a oficina dedicada ao solo e seu pH). A título de exemplo passamos a referir a organização da primeira oficina dedicada às formigas e que se iniciou com a fábula A Cigarra e a Formiga. Aspectos explorados posteriormente passaram pela(s): morfologia das formigas, sua longevidade, relações que estabelecem com outros organismos, sua nutrição e a forma como procuram o alimento, organização e hierarquia de um formigueiro, suas funções numa horta, entre muitos outros. O entusiasmo foi tanto que o DPV chegou a adquirir um formigueiro em acrílico para observação continuada de uma colónia de formigas Messor barbarus, por parte dos alunos, prolongando a exploração da temática para além da duração da oficina.
Como conclusão e inserida no projeto de sala, ultimou-se este projeto com uma visita às Lagoas de Bertiandos para observação da fauna e flora locais, estendendo esta iniciativa de educação ambiental para além dos limites do colégio onde, em “terreno não familiar”, as crianças espontaneamente aplicavam conceitos e conhecimentos abordados nas oficinas
