    The cross-sectional and prospective associations between sleep characteristics and adiposity in toddlers: Results from the GET UP! Study

    Background: The associations between sleep characteristics and adiposity in children under three years are not fully understood yet. Objective: The objective of the study is to examine the cross-sectional and prospective associations between sleep characteristics and adiposity in toddlers over a 12-month period. Methods: Participants were 202 toddlers from the GET-UP! Study. Sleep duration, sleep timing, and sleep variability were assessed using 24-hour accelerometry for seven consecutive days. Height and weight were measured, and BMI z scores were calculated. Linear mixed models were performed to examine the cross-sectional and prospective associations between sleep characteristics and adiposity, with adjustments for clustering effects and demographic factors. Results: Total sleep duration was negatively associated with higher adiposity cross-sectionally (B = −0.12; 95% CI: −0.23, −0.01;.033) but not prospectively (B = 0.01; 95% CI: −0.13, 0.10;.843). Nap duration was prospectively associated with higher levels of adiposity (B = 0.41; 95% CI: 0.14, 0.68;.003). Sleep variability and sleep timing were not associated with concurrent or subsequent adiposity. Conclusion: Although sleep duration is an important factor associated with obesity in toddlerhood, the potential effects of different types of sleep duration may vary. While longer total sleep duration may protect children from increasing adiposity, longer nap duration seems to be risk factor. As evidence in this age group is scarce, more research is needed to confirm this finding

    Variation of Burkholderia cenocepacia cell wall morphology and mechanical properties during cystic fibrosis lung infection, assessed by atomic force microscopy

    The influence that Burkholderia cenocepacia adaptive evolution during long-term infection in cystic fibrosis (CF) patients has on cell wall morphology and mechanical properties is poorly understood despite their crucial role in cell physiology, persistent infection and pathogenesis. Cell wall morphology and physical properties of three B. cenocepacia isolates collected from a CF patient over a period of 3.5 years were compared using atomic force microscopy (AFM). These serial clonal variants include the first isolate retrieved from the patient and two late isolates obtained after three years of infection and before the patient's death with cepacia syndrome. A consistent and progressive decrease of cell height and a cell shape evolution during infection, from the typical rods to morphology closer to cocci, were observed. The images of cells grown in biofilms showed an identical cell size reduction pattern. Additionally, the apparent elasticity modulus significantly decreases from the early isolate to the last clonal variant retrieved from the patient but the intermediary highly antibiotic resistant clonal isolate showed the highest elasticity values. Concerning the adhesion of bacteria surface to the AFM tip, the first isolate was found to adhere better than the late isolates whose lipopolysaccharide (LPS) structure loss the O-antigen (OAg) during CF infection. The OAg is known to influence Gram-negative bacteria adhesion and be an important factor in B. cenocepacia adaptation to chronic infection. Results reinforce the concept of the occurrence of phenotypic heterogeneity and adaptive evolution, also at the level of cell size, form, envelope topography and physical properties during long-term infection

    The N terminus of the peroxisomal cycling receptor, Pex5p, is required for redirecting the peroxisome-associated peroxin back to the cytosol

    Most newly synthesized peroxisomal matrix proteins are transported to the organelle by Pex5p, a remarkable multidomain protein involved in an intricate network of transient protein-protein interactions. Presently, our knowledge regarding the structure/function of amino acid residues 118 to the very last residue of mammalian Pex5p is quite vast. Indeed, the cargo-protein receptor domain as well as the binding sites for several peroxins have all been mapped to this region of Pex5p. In contrast, structural/functional data regarding the first 117 amino acid residues of Pex5p are still scarce. Here we show that a truncated Pex5p lacking the first 110 amino acid residues (DeltaN110-Pex5p) displays exactly the peroxisomal import properties of the full-length peroxin implying that this N-terminal domain is involved neither in cargo-protein binding nor in the docking/translocation step of the Pex5p-cargo protein complex at the peroxisomal membrane. However, the ATP-dependent export step of DeltaN110-Pex5p from the peroxisomal membrane is completely blocked, a phenomenon that was also observed for a Pex5p version lacking just the first 17 amino acid residues but not for a truncated protein comprising amino acid residues 1-324 of Pex5p. By exploring the unique properties of DeltaN110-Pex5p, the effect of temperature on the import/export kinetics of Pex5p was characterized. Our data indicate that the export step of Pex5p from the peroxisomal compartment ( in contrast with its insertion into the organelle membrane) is highly dependent on the temperature

Foi possível delinear três aspectos da questão: (a) O conceito é mencionado em cerca de 5% dos artigos analisados; (b) O uso varia da simples menção até uma discussão mais profunda; (c) Em alguns casos, mesmo quando não mencionado, é possível identificar o conceito como referência. Isso sugere que a noção de “indústria cultural” se manteve pertinente apesar das mudanças sociais e tecnológicas na própria comunicação

    The genome sequence of the highly acetic acid-tolerant zygosaccharomyces bailii-derived interspecies hybrid strain ISA1307, isolated from a sparkling wine plant

    In this work, it is described the sequencing and annotation of the genome of the yeast strain ISA1307, isolated from a sparkling wine continuous production plant. This strain, formerly considered of the Zygosaccharomyces bailii species, has been used to study Z. bailii physiology, in particular, its extreme tolerance to acetic acid stress at low pH. The analysis of the genome sequence described in this work indicates that strain ISA1307 is an interspecies hybrid between Z. bailii and a closely related species. The genome sequence of ISA1307 is distributed through 154 scaffolds and has a size of around 21.2 Mb, corresponding to 96% of the genome size estimated by flow cytometry. Annotation of ISA1307 genome includes 4385 duplicated genes (~90% of the total number of predicted genes) and 1155 predicted single-copy genes. The functional categories including a higher number of genes are 'Metabolism and generation of energy', 'Protein folding, modification and targeting' and 'Biogenesis of cellular components'. The knowledge of the genome sequence of the ISA1307 strain is expected to contribute to accelerate systems-level understanding of stress resistance mechanisms in Z. bailii and to inspire and guide novel biotechnological applications of this yeast species/strain in fermentation processes, given its high resilience to acidic stress. Belém, PA, Brasil.O presente trabalho enfatiza resultados recentes sobre eco-epidemiologia da febre amarela, obtidos em duas localidades ecologicamente muito diferentes: Barcarena (PA), situada na floresta amazônica densa, e a região de Campo Grande (MS), coberta de cerrado atravessado por florestas de galeria na região Centro-oeste. No primeiro local foi isolada uma amostra a partir de um lote de 6 Haemagogus janthinomys. Na região de Campo Grande, 2. 480 mosquitos antropófilos foram coletados, dos quais cerca de 40% eram vetores potenciais da FA. Essas espécies, classificadas por ordem decrescente de abundãncia, eram: Ae. scapularis, Sa. chloropterus, Hg. janthinomys, Hg. leucocelaenus, Hg. spegazinii e Sa. soperi. Quatro amostras de FA foram isolados a partir de Hg. janthinomys, uma de Sa. chloropterus (primeiro registro na América do Sul), e uma de Sa. soperi (primeiro registro). As taxas mínimas de infecção variaram segundo os locais coleta, mas foram sempre alta comparação com dados anteriores. A taxa média de sobrevivência diária do Hg. janthinomys é igual a 0,9635, permitindo estimar o seu valor na ocasião em que os caso humanos mais recentes foram provavelmente contaminados. deduziu-se que as epizootias foram muito intensas e mais ou menos simultâneas nos dois locais considerados. O maior problema a ser resolvido é o modo de re-introdução do vírus, ou sua sobreviv~encia, em cada região sob estudo
