ATG5 is essential for ATG8-dependent autophagy and mitochondrial homeostasis in Leishmania major
Macroautophagy has been shown to be important for the cellular remodelling required for Leishmania differentiation. We now demonstrate that L. major contains a functional ATG12-ATG5 conjugation system, which is required for ATG8-dependent autophagosome formation. Nascent autophagosomes were found commonly associated with the mitochondrion. L. major mutants lacking ATG5 (Δatg5) were viable as promastigotes but were unable to form autophagosomes, had morphological abnormalities including a much reduced flagellum, were less able to differentiate and had greatly reduced virulence to macrophages and mice. Analyses of the lipid metabolome of Δatg5 revealed marked elevation of phosphatidylethanolamines (PE) in comparison to wild type parasites. The Δatg5 mutants also had increased mitochondrial mass but reduced mitochondrial membrane potential and higher levels of reactive oxygen species. These findings indicate that the lack of ATG5 and autophagy leads to perturbation of the phospholipid balance in the mitochondrion, possibly through ablation of membrane use and conjugation of mitochondrial PE to ATG8 for autophagosome biogenesis, resulting in a dysfunctional mitochondrion with impaired oxidative ability and energy generation. The German Young Olympic Athletes' Lifestyle and Health Management Study (GOAL Study): design of a mixed-method study
<p>Abstract</p> <p>Background</p> <p>In order to perform at top levels, elite athletes have to both protect and risk their health at the same time. Adolescent elite athletes have the additional challenge of coping with substantial physical, psychological and social transformations. The contradictory phenomenon of protecting and risking the adolescent athletes' health in sports challenges the development of health promotion and protection strategies. The GOAL Study (German Young Olympic Athletes' Lifestyle and Health Management Study) analyzes the individual and organizational management of health in adolescent elite sports.</p> <p>Methods/design</p> <p>We combine quantitative and qualitative approaches in a mixed-method study. This allows us to gather a broad range of representative information on squad athletes from all Olympic disciplines as well as in-depth information on four selected Olympic disciplines (artistic gymnastics, biathlon, handball and wrestling). Within the quantitative section we attempt to identify the young athletes' health and nutrition behavior, their subjective health state and their lay health representations, health-related social networks, and structures of medical attendance. 1138 national team level athletes born between 1992 and 1995 from 51 Olympic disciplines responded to the questionnaire (response rate: 61,75%). The qualitative section investigates the meaning and relevance of health and nutrition within the athletes' sports specific surroundings, the impact of biographic backgrounds on individual health behavior, and sports specific cultures of health, nutrition and risk. We interviewed 24 athletes and 28 coaching and medical experts, and carried out 14 multi-day participant observations at training sessions and competitions.</p> <p>Conclusions</p> <p>The studies' results will serve as the basis for developing tailored health promotion strategies to be in cooperation with German elite sports associations.</p
Should the Arteriovenous Fistula Be Created before Starting Dialysis?: A Decision Analytic Approach
Background: An arteriovenous fistula (AVF) is considered the vascular access of choice, but uncertainty exists about the\ud
optimal time for its creation in pre-dialysis patients. The aim of this study was to determine the optimal vascular access\ud
referral strategy for stage 4 (glomerular filtration rate ,30 ml/min/1.73 m2) chronic kidney disease patients using a decision\ud
analytic framework.\ud
Methods: A Markov model was created to compare two strategies: refer all stage 4 chronic kidney disease patients for an\ud
AVF versus wait until the patient starts dialysis. Data from published observational studies were used to estimate the\ud
probabilities used in the model. A Markov cohort analysis was used to determine the optimal strategy with life expectancy\ud
and quality adjusted life expectancy as the outcomes. Sensitivity analyses, including a probabilistic sensitivity analysis, were\ud
performed using Monte Carlo simulation.\ud
Results: The wait strategy results in a higher life expectancy (66.6 versus 65.9 months) and quality adjusted life expectancy\ud
(38.9 versus 38.5 quality adjusted life months) than immediate AVF creation. It was robust across all the parameters except\ud
at higher rates of progression and lower rates of ischemic steal syndrome.\ud
Conclusions: Early creation of an AVF, as recommended by most guidelines, may not be the preferred strategy in all predialysis\ud
patients. Further research on cost implications and patient preferences for treatment options needs to be done\ud
before recommending early AVF creation
Multimodal Stimulation of Colorado Potato Beetle Reveals Modulation of Pheromone Response by Yellow Light
Orientation of insects to host plants and conspecifics is the result of detection and integration of chemical and physical cues present in the environment. Sensory organs have evolved to be sensitive to important signals, providing neural input for higher order multimodal processing and behavioral output. Here we report experiments to determine decisions made by Colorado potato beetle (CPB), Leptinotarsa decemlineata, in response to isolated stimuli and multimodal combinations of signals on a locomotion compensator. Our results show that in complete darkness and in the absence of other stimuli, pheromonal stimulation increases attraction behavior of CPB as measured in oriented displacement and walking speed. However, orientation to the pheromone is abolished when presented with the alternative stimulation of a low intensity yellow light in a dark environment. The ability of the pheromone to stimulate these diurnal beetles in the dark in the absence of other stimuli is an unexpected but interesting observation. The predominance of the phototactic response over that to pheromone when low intensity lights were offered as choices seems to confirm the diurnal nature of the insect. The biological significance of the response to pheromone in the dark is unclear. The phototactic response will play a key role in elucidating multimodal stimulation in the host-finding process of CPB, and perhaps other insects. Such information might be exploited in the design of applications to attract and trap CPB for survey or control purposes and other insect pests using similar orientation mechanisms

Evolution of Salmonella enterica Virulence via Point Mutations in the Fimbrial Adhesin
Whereas the majority of pathogenic Salmonella serovars are capable of infecting many different animal species, typically producing a self-limited gastroenteritis, serovars with narrow host-specificity exhibit increased virulence and their infections frequently result in fatal systemic diseases. In our study, a genetic and functional analysis of the mannose-specific type 1 fimbrial adhesin FimH from a variety of serovars of Salmonella enterica revealed that specific mutant variants of FimH are common in host-adapted (systemically invasive) serovars. We have found that while the low-binding shear-dependent phenotype of the adhesin is preserved in broad host-range (usually systemically non-invasive) Salmonella, the majority of host-adapted serovars express FimH variants with one of two alternative phenotypes: a significantly increased binding to mannose (as in S. Typhi, S. Paratyphi C, S. Dublin and some isolates of S. Choleraesuis), or complete loss of the mannose-binding activity (as in S. Paratyphi B, S. Choleraesuis and S. Gallinarum). The functional diversification of FimH in host-adapted Salmonella results from recently acquired structural mutations. Many of the mutations are of a convergent nature indicative of strong positive selection. The high-binding phenotype of FimH that leads to increased bacterial adhesiveness to and invasiveness of epithelial cells and macrophages usually precedes acquisition of the non-binding phenotype. Collectively these observations suggest that activation or inactivation of mannose-specific adhesive properties in different systemically invasive serovars of Salmonella reflects their dynamic trajectories of adaptation to a life style in specific hosts. In conclusion, our study demonstrates that point mutations are the target of positive selection and, in addition to horizontal gene transfer and genome degradation events, can contribute to the differential pathoadaptive evolution of Salmonella
Climate simulations for 1880-2003 with GISS modelE
We carry out climate simulations for 1880-2003 with GISS modelE driven by ten
measured or estimated climate forcings. An ensemble of climate model runs is
carried out for each forcing acting individually and for all forcing mechanisms
acting together. We compare side-by-side simulated climate change for each
forcing, all forcings, observations, unforced variability among model ensemble
members, and, if available, observed variability. Discrepancies between
observations and simulations with all forcings are due to model deficiencies,
inaccurate or incomplete forcings, and imperfect observations. Although there
are notable discrepancies between model and observations, the fidelity is
sufficient to encourage use of the model for simulations of future climate
change. By using a fixed well-documented model and accurately defining the
1880-2003 forcings, we aim to provide a benchmark against which the effect of
improvements in the model, climate forcings, and observations can be tested.
Principal model deficiencies include unrealistically weak tropical El Nino-like
variability and a poor distribution of sea ice, with too much sea ice in the
Northern Hemisphere and too little in the Southern Hemisphere. The greatest
uncertainties in the forcings are the temporal and spatial variations of
