    Stereocilia Membrane Deformation: Implications for the Gating Spring and Mechanotransduction Channel

    AbstractIn hair cells, although mechanotransduction channels have been localized to tips of shorter stereocilia of the mechanically sensitive hair bundle, little is known about how force is transmitted to the channel. Here, we use a biophysical model of the membrane-channel complex to analyze the nature of the gating spring compliance and channel arrangement. We use a triangulated surface model and Monte Carlo simulation to compute the deformation of the membrane under the action of tip link force. We show that depending on the gating spring stiffness, the compliant component of the gating spring arises from either the membrane alone or a combination of the membrane and a tether that connects the channel to the actin cytoskeleton. If a bundle is characterized by relatively soft gating springs, such as those of the bullfrog sacculus, the need for membrane reinforcement by channel tethering then depends on membrane parameters. With stiffer gating springs, such as those from rat outer hair cells, the channel must be tethered for all biophysically realistic parameters of the membrane. We compute the membrane forces (resultants), which depend on membrane tension, bending modulus, and curvature, and show that they can determine the fate of the channel

    Confirmatory factor analysis of the Child Feeding Questionnaire among low-income African American families of preschool children

    This study examined the factor structure for three of the Child Feeding Questionnaire (CFQ) subscales, a widely used measure of parental feeding practices, among 296 low-income parents of African American preschool children. Confirmatory factor analysis showed an overall poor fit among CFQ subscales; Restriction, Pressure to Eat, and Concern about Child Weight, (χ2, (df = 87 = 300.249, CFI = 1.00, NNFI = 1.07, RMSEA = .091). Additionally, Cronbach’s Alpha coefficients for 2 of the three subscales were below acceptable recommendations (Restriction = 0.69; Pressure to Eat = 0.58). These results suggest further psychometric clarification is needed to understand commonly reported feeding practice constructs among low-income African American mothers of preschool aged children

    Estimating the Spatial Extent of Bottom-Water Hypoxia and Habitat Degradation in a Shallow Estuary

    Bottom-water hypoxia (≤ 2 mg 1-1 dissolved oxygen [DO]) greatly modifies the benthic habitat of estuaries, depending upon spatial extent, duration, and frequency. Bottom-water hypoxia often develops under conditions of density stratification, which inhibits vertical mixing, and warm temperatures, which enhance biological oxygen demand. Long-term, mid-channel data from the Neuse River Estuary in North Carolina permitted evaluation of how stratification and temperature combined to affect DO concentrations at the bottom. Salinity stratification (DS) and water temperature (T) explained respectively 30 and 23% of the variance in bottom-water DO concentrations. The amount of salinity stratification required to induce bottom-water hypoxia declined with increasing water temperature. About 80% of observed hydrographic profiles exhibited bottom hypoxia when DS exceeded 5 psu and T exceeded 20°C. Using cross-channel hydrographic surveys as verification, we derived a general set of methods to estimate the lateral extent of low-DO bottom water from midchannel hydrographic profiles. The method involves cross-estuary and along-estuary extrapolation based on assumption of a flat oxycline. Occasional violation of this assumption resulted in modest overestimation in cross-channel extent of low DO. Application of this method produced estimates ranging from 0 to 116 km2 of bottom area (0 to 42% of the estuarine study area) exposed to hypoxia over all sample dates in summer 1997. The maximal bottom area exposed to hypoxia corresponded closely with an independent estimate of the area (100 km2) that experienced almost complete mortality of Macoma spp. clams, the key benthic resource for demersal fishes and crabs. Consequently, mid-channel hydrographic profiles taken along the mid-channel of the estuary can be employed to assess the spatial scale of bottom habitat degradation due to hypoxia

    Development of Standard Methods to Estimate Manure Production and Nutrient Characteristics From Dairy Cattle

    Total collection data from many universities were pooled for statistical analyses to evaluate existing data from dairy animals and determine if one or more modifications were needed for Standard Table D384.1. Many of these studies were carried out to evaluate nutritional characteristics associated with different diets. The data collected were sufficient to evaluate total manure, total and volatile solids, and N excretion values. Some experiments had sufficient number of samples analyzed to evaluate P, K, Ca, Mg, Na, Cl, S, and micro-elements. Statistical analyses were conducted to determine if a single column value was appropriate and define the regression equations necessary to estimate excretions if the assumptions of the column inputs were not met. The results indicate that separate classifications are needed for replacement heifers and mature animals. Final classifications for replacement heifers included: milk fed calves, weaned calves weighing less than 274 kg, heifers weighing between 273 and 613 kg, and veal calves. Additionally, classifications for lactating and dry animals are needed. Previously, the estimates for manure production and nutrient excretion were based on body weight. These findings indicate that a better predictor for lactating cattle is daily milk production instead of body weight. Milk production drives feed intake in the lactating animal. It is most appropriate that estimated manure and nutrient excretion values reflect the relationship between feed intake, milk production (nutrient utilization) and nutrient excretion

    Hsp72 is targeted to the mitotic spindle by Nek6 to promote K-fiber assembly and mitotic progression

    Hsp70 proteins represent a family of chaperones that regulate cellular homeostasis and are required for cancer cell survival. However, their function and regulation in mitosis remain unknown. In this paper, we show that the major inducible cytoplasmic Hsp70 isoform, Hsp72, is required for assembly of a robust bipolar spindle capable of efficient chromosome congression. Mechanistically, Hsp72 associates with the K-fiber-stabilizing proteins, ch-TOG and TACC3, and promotes their interaction with each other and recruitment to spindle microtubules (MTs). Targeting of Hsp72 to the mitotic spindle is dependent on phosphorylation at Thr-66 within its nucleotide-binding domain by the Nek6 kinase. Phosphorylated Hsp72 concentrates on spindle poles and sites of MT-kinetochore attachment. A phosphomimetic Hsp72 mutant rescued defects in K-fiber assembly, ch-TOG/TACC3 recruitment and mitotic progression that also resulted from Nek6 depletion. We therefore propose that Nek6 facilitates association of Hsp72 with the mitotic spindle, where it promotes stable K-fiber assembly through recruitment of the ch-TOG-TACC3 complex

    Potential for Mineable Bedrock in the Marion 30 X 60 minute quadrangle

    The Ohio Department of Natural Resources (ODNR), Division of Geological Survey has completed a reconnaissance map showing areas of mineable bedrock, including shale, limestone, and dolomite, likely covered by thin surficial materials (glacial drift) in the Marion, Ohio, 30 x 50-minute (1:1,100,000-scale) quadrangle. The main purpose of this map was to create a reconnaissance-level map that shows the potential for mining carbonate and shale bedrock in this quadrangle. We sought to create this map from as many existing ODNR Division of Geological Survey maps and GIS datasets as possible. The map shows areas of surficial materials in increments of 10 ft and totaling less than 40 ft overlying Silurian- and Devonian-age dolomite and limestone, and it also shows a limited area in the easternmost portion of the quadrangle where surficial materials (totaling less than 20 ft) overlay potential Devonian-age shale resources.United States Geological Survey: National Cooperative Geologic Mapping Program, Great Lakes Geologic Mapping Coalitio

    Marine Fisheries Stock Assessment Improvement Plan: report of the National Marine Fisheries Service National Task Force for Improving Fish Stock Assessments

    This report argues for greatly increased resources in terms of data collection facilities and staff to collect, process, and analyze the data, and to communicate the results, in order for NMFS to fulfill its mandate to conserve and manage marine resources. In fact, the authors of this report had great difficulty defining the "ideal" situation to which fisheries stock assessments and management should aspire. One of the primary objectives of fisheries management is to develop sustainable harvest policies that minimize the risks of overfishing both target species and associated species. This can be achieved in a wide spectrum of ways, ranging between the following two extremes. The first is to implement only simple management measures with correspondingly simple assessment demands, which will usually mean setting fishing mortality targets at relatively low levels in order to reduce the risk of unknowingly overfishing or driving ecosystems towards undesirable system states. The second is to expand existing data collection and analysis programs to provide an adequate knowledge base that can support higher fishing mortality targets while still ensuring low risk to target and associated species and ecosystems. However, defining "adequate" is difficult, especially when scientists have not even identified all marine species, and information on catches, abundances, and life histories of many target species, and most associated species, is sparse. Increasing calls from the public, stakeholders, and the scientific community to implement ecosystem-based stock assessment and management make it even more difficult to define "adequate," especially when "ecosystem-based management" is itself not well-defined. In attempting to describe the data collection and assessment needs for the latter, the authors took a pragmatic approach, rather than trying to estimate the resources required to develop a knowledge base about the fine-scale detailed distributions, abundances, and associations of all marine species. Thus, the specified resource requirements will not meet the expectations of some stakeholders. In addition, the Stock Assessment Improvement Plan is designed to be complementary to other related plans, and therefore does not duplicate the resource requirements detailed in those plans, except as otherwise noted
