Low Ten-eleven-translocation 2 (tet2) Transcript Level Is Independent Of Tet2 Mutation In Patients With Myeloid Neoplasms

We have recently identified reduced TET2 mRNA expression in myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML), which is associated with a poor overall survival in MDS. We herein aimed to investigate TET2 mutations and their impact on TET2 expression in a cohort of patients with myeloid neoplasms, including MDS and AML patients. Findings: TET2 mutations were observed in 8 out of 19 patients (42 %) with myeloid neoplasms. The TET2 expression profile was similar between in wild type and in TET2 mutated patients. Gap modification of atomically thin boron nitride by phonon mediated interactions
A theory is presented for the modification of bandgaps in atomically thin
boron nitride (BN) by attractive interactions mediated through phonons in a
polarizable substrate, or in the BN plane. Gap equations are solved, and gap
enhancements are found to range up to 70% for dimensionless electron-phonon
coupling \lambda=1, indicating that a proportion of the measured BN bandgap may
have a phonon origin

Allelic frequencies and statistical data obtained from 12 codis STR loci in an admixed population of the Brazilian Amazon
The allelic frequencies of 12 short tandem repeat loci were obtained from a sample of 307 unrelated individuals living in Macapá, a city in the northern Amazon region, Brazil. These loci are the most commonly used in forensics and paternity testing. Based on the allele frequency obtained for the population of Macapá, we estimated an interethnic admixture for the three parental groups (European, Native American and African) of, respectively, 46%, 35% and 19%. Comparing these allele frequencies with those of other Brazilian populations and of the Iberian Peninsula population, no significant distances were observed. The interpopulation genetic distances (FST coefficients) to the present database ranged from FST = 0.0016 between Macapá and Belém to FST = 0.0036 between Macapá and the Iberian Peninsula
Predictors of response to TNF blockers in patients with polyarticular psoriatic arthritis
Psoriatic arthritis (PsA) is a chronic inflammatory
rheumatic disease with a broad clinical spectrum. PsA
can affect the axialskeleton, peripheral joints, entheses,
synovial sheaths of tendons, skin, nails and extra-articular
organs. Tumour necrosis factor alpha blockers
(TNF blockers) were a breakthrough development in
the treatment of PsA. Identifying predictors of response
to biological therapiesin patients with PsA is of utmost
importance, especially in view of the costs and potential
side effects of these agents. The aims of the present
study were to determine baseline predictive factors of
response to biological therapies, at 3 and 6 months, in
PsA patients with polyarticular involvement (with or
without axial involvement). Data were collected from
the RheumaticDiseases Portuguese Register(Reuma.pt).
Eligible patients had to be anti-TNF-naive at baseline
and to have at least 3 months of follow-up after the beginning
of TNF blocker therapy. Only patients with information
on at least one of the response measures (at
3 or 6 months of follow-up) were included in the analysis.
Univariable logistic regression analysis of potential
baseline predictors of European League Against Rheu-matism (EULAR) good clinical response, EULAR good/
/moderate response, 28-joint Disease Activity Score
with three variables including the erythrocyte sedimentation
rate (DAS28-3V-ESR) remission and Health
Assessment Questionnaire (HAQ) response were performed.
Multivariable logistic regression using a forward
selection procedure was used until the best-fit
model was obtained, taking confounding effects into
account. A total of 180 patients were eligible for the
study (mean age 52 years, 54% women). In multivariable
analysis at 3 months, females were less likely to attain
a good EULAR response [OR=0.082 (95%
CI=0.024, 0.278)], a DAS28-3V-ESR remission
[OR=0.083 (95% CI=0.017, 0.416)], a moderate or
good EULAR response [OR=0.091 (95% CI=0.011,
0.091)] and a HAQ response [OR=0.074 (95%
CI=0.009, 0.608)]. At 6 months, female gender was
also less likely to achieve a good EULAR response
[OR=0.060 (95% CI=0.011, 0.325)], DAS28-3V-ESR
remission [OR=0.060 (95% CI=0.012, 0.297)], and a
HAQ response [OR=0.138 (95% CI= 0.029, 0.654)]. In
this study we found that gender was the most consistent
predictor of response to TNF blocker therapy in
patients with polyarticular PsA, with females having a
lower probability ofresponse compared to males. These
findings suggest that gender-related biochemical, hormonal
and psychological factors could play an importantrole
in the response to TNF blockertherapy in PsA
Atypical audiovisual speech integration in infants at risk for autism
The language difficulties often seen in individuals with autism might stem from an inability to integrate audiovisual information, a skill important for language development. We investigated whether 9-month-old siblings of older children with autism, who are at an increased risk of developing autism, are able to integrate audiovisual speech cues. We used an eye-tracker to record where infants looked when shown a screen displaying two faces of the same model, where one face is articulating/ba/and the other/ga/, with one face congruent with the syllable sound being presented simultaneously, the other face incongruent. This method was successful in showing that infants at low risk can integrate audiovisual speech: they looked for the same amount of time at the mouths in both the fusible visual/ga/− audio/ba/and the congruent visual/ba/− audio/ba/displays, indicating that the auditory and visual streams fuse into a McGurk-type of syllabic percept in the incongruent condition. It also showed that low-risk infants could perceive a mismatch between auditory and visual cues: they looked longer at the mouth in the mismatched, non-fusible visual/ba/− audio/ga/display compared with the congruent visual/ga/− audio/ga/display, demonstrating that they perceive an uncommon, and therefore interesting, speech-like percept when looking at the incongruent mouth (repeated ANOVA: displays x fusion/mismatch conditions interaction: F(1,16) = 17.153, p = 0.001). The looking behaviour of high-risk infants did not differ according to the type of display, suggesting difficulties in matching auditory and visual information (repeated ANOVA, displays x conditions interaction: F(1,25) = 0.09, p = 0.767), in contrast to low-risk infants (repeated ANOVA: displays x conditions x low/high-risk groups interaction: F(1,41) = 4.466, p = 0.041). In some cases this reduced ability might lead to the poor communication skills characteristic of autism
Qualidade de vida nas pessoas com esquizofrenia: a influência das características sociodemográficas e clínicas e da satisfação com o suporte social
Objective: To evaluate the relationship of sociodemographic and clinical characteristics and satisfaction with social support with the quality of life of schizophrenic patients.
Methodology: This study included a sample of 268 participants. An interview was conducted to obtain sociodemographic and clinical data, supplemented with two assessment tools used to evaluate quality of life (World Health Organization Quality of Life instrument-Abbreviated version – WHOQOL-Bref) and satisfaction with social support (Social Support Satisfaction Scale – SSSS). Descriptive and inferential analyses were performed.
Results: Most individuals were male (63.4%), with a mean age of 45.4 years, single (85.4%), living with their family (62.3%) and unemployed (90.3%). As for clinical characteristics, most had the disease for less than 20 years (50.7%), and 55.6% had at least one hospitalization within the last 5 years. Being employed and having had no hospitalization within the last 5 years were positively correlated with one or more WHOQOL-Bref domains. The results of the variables intimacy (p<0.001) and satisfaction with friends (p<0.001) were independently related to the total WHOQOL-Bref score.
Conclusion: Having a job, having had no hospitalization within the last 5 years and having greater satisfaction with social support are factors that positively influence quality of life among schizophrenics. It is therefore crucial that the psychosocial rehabilitation of patients with schizophrenia take these factors into account, increasing the support network, preventing relapses and promoting occupational activities.Objetivo: Avaliar a relação entre as características sociodemográficas e clínicas e a satisfação com o suporte social com a qualidade de vida dos doentes com esquizofrenia.
Métodos: A amostra do estudo é de 268 participantes. Foi realizada uma entrevista para obter os dados sociodemográficos e clínicos e aplicados dois questionários para avaliar a qualidade de vida (World Health Organization Quality of Life instrument-Abbreviated version – WHOQOL-Bref) e a satisfação com o suporte social (Escala de Satisfação com o Suporte Social – ESSS). Foram efetuadas análises descritivas e inferenciais.
Resultados: A maioria dos indivíduos era do gênero masculino (63,4%), com uma média de idade de 45,4 anos, solteiros (85,4%), vivendo com a família (62,3%) e desempregados (90,3%). Relativamente às características clínicas, 50,7% tinham a doença há menos de 20 anos, e 55,6% estiveram internados pelo menos uma vez nos últimos 5 anos. Os resultados demonstraram que estar empregado e não ter sido internado nos últimos 5 anos estão positivamente relacionados com um ou mais domínios da WHOQOL-Bref. A satisfação com a intimidade (p<0,001) e a satisfação com os amigos (p<0,001) foram independentemente associados ao escore total da WHOQOL-Bref.
Conclusão: Ter emprego, não ter hospitalizações nos últimos 5 anos e estar satisfeito com o suporte social são fatores que influenciam positivamente a qualidade de vida dos doentes com esquizofrenia. Por conseguinte, é crucial que esses fatores sejam levados em conta nos programas de reabilitação com o suporte social, aumentando a rede de suporte, evitando recaídas e promovendo atividades ocupacionais.publishe
