    An extracellular transglutaminase is required for apple pollen tube growth

    An extracellular form of the calcium-dependent protein-crosslinking enzyme TGase (transglutaminase) was demonstrated to be involved in the apical growth of Malus domestica pollen tube. Apple pollen TGase and its substrates were co-localized within aggregates on the pollen tube surface, as determined by indirect immunofluorescence staining and the in situ cross-linking of fluorescently labelled substrates. TGase-specific inhibitors and an anti-TGase monoclonal antibody blocked pollen tube growth, whereas incorporation of a recombinant fluorescent mammalian TGase substrate (histidine-tagged green fluorescent protein:His6– Xpr–GFP) into the growing tube wall enhanced tube length and germination, consistent with a role of TGase as a modulator of cell wall building and strengthening. The secreted pollen TGase catalysed the cross-linking of both PAs (polyamines) into proteins (released by the pollen tube) and His6-Xpr-GFP into endogenous or exogenously added substrates. A similar distribution of TGase activitywas observed in planta on pollen tubes germinating inside the style, consistent with a possible additional role for TGase in the interaction between the pollen tube and the style during fertilization

    Genetic and morphometric variation in Schwarziana quadripunctata and Schwarziana mourei (Hymenoptera: Apidae: Meliponini)

    Schwarziana bees are a ground-nesting stingless bee distributed in the Neotropical region. Schwarziana quadripunctata was the first described and the most studied species of this genus. Now, there are four valid species of Schwarziana bees, but it has been suggested that the diversity of this taxon may be higher, due to undescribed cryptic species. In this study, we investigated the populational diversity of S. quadripunctata using workers collected at 11 localities in Brazil (from the Northeast to South region). We also included one population of S. mourei (collected in São Paulo state, 2 nests). We analysed the bees using geometric morphometrics and molecular analyses amplifying mtDNA cytochrome oxidase I (COI) and 16S to access the diversity among the populations. From the results of geometric morphometrics, the Mahalanobis distances between S. mourei and S. quadripunctata are greater than those distances among S. quadripunctata populations. A similar scenario can also be observed looking to the phylogenetic tree generated by the molecular markers. Morphometry and molecular markers data showed significant association with geographic distance, indicating the existence of intrapopulation variation in S. quadripunctata. Our hypothesis was supported, that the populations of S. quadripunctata showed differences in haplotypic diversity. Overall, these analyses revealed a moderate level of intraspecific variation among S. quadripunctata populations and discriminated well the species S. quadripunctata from S. mourei

    Citrus allergy from pollen to clinical symptoms

    Allergy to citrus fruits is often associated with pollinosis and sensitization to other plants due to a phenomenon of cross-reactivity. The aims of the present study were to highlight the cross-reactivity among citrus and the major allergenic pollens/fruits, throughout clinical and molecular investigations, and to evaluate the sensitization frequency to citrus fruits in a population of children and adults with pollinosis. We found a relevant percentage of sensitisation (39%) to citrus fruits in the patients recruited and in all of them the IgE-mediated mechanism has been confirmed by the positive response to the prick-to-prick test. RT-PCR experiments showed the expression of Cit s 1, Cit s 3 and a profilin isoform, already described in apple, also in Citrus clementine pollen. Data of multiple sequence alignments demonstrated that Citrus allergens shared high percentage identity values with other clinically relevant species (i.e. Triticum aestivum, Malus domestica), confirming the possible cross-allergenicity citrus/grasses and citrus/apple. Finally, a novelty of the present work has been the expression of two phospholipaseA2 isoforms (PLA2 \u3b1 and \u3b2) in Citrus as well as in Triticum pollens; being PLA2 able to generate pro-inflammatory factors, this enzyme could participate in the activation of the allergenic inflammatory cascade

    Some adults of Pseudimares aphrodite H. Asp?ck et U. Asp?ck, 2009 were observed and photographed while attracted by light in Southern Morocco, in August 2009 and 2011. Only the typus of this species, a male, was known previously from South Morocco too. Moreover the genus Pseudimares Kimmins, 1933 is perhaps the most enigmatic taxon among Neuroptera Myrmeleontidae.     Variation in plasma oxidative status and testosterone level in relation to egg-eviction effort and age of brood-parasitic common cuckoo nestlings

    To avoid competition for parental care, brood-parasitic Common Cuckoo (Cuculus canorus) nestlings evict all of the host's eggs and nestlings within a few days after hatching. Little is known about the physiological effects of eviction behavior on the cuckoo nestling's oxidative balance or about age-related variation in plasma oxidative status and testosterone level of developing birds. We examined whether the cuckoo nestling's plasma oxidative status was related to prior effort in eviction and quantified variation in the level of reactive oxygen metabolites, of nonenzymatic antioxidant capacity, and of testosterone concentration in plasma at various phases of the cuckoo's development. Levels of both reactive oxygen metabolites and antioxidant capacity were greater in older than in younger nestlings, suggesting that younger nestlings effectively counterbalance their increased production of free radicals, whereas, near fledging, levels of reactive oxygen metabolites increase despite improved antioxidant capacity. Possibly, overall energy expenditure increases with age and elevates the production of reactive oxygen species to a rate higher than what the antioxidant system could eliminate. Plasma testosterone level was the highest at nestlings' intermediate phase of growth. High levels of testosterone may be required during the period of fastest growth, and when the growth rate levels off near fledging, testosterone levels may also decline. Cuckoo chicks that evicted more host eggs from steeper nests had higher plasma levels of reactive oxygen metabolites shortly after the eviction period, suggesting that eviction is costly in terms of an increased level of oxidative stress. Para evitar la competencia por el cuidado parental, los polluelos parásitos de nidada de Cuculus canorus desalojan todos los huevos y los polluelos del hospedador a los pocos días después de la eclosión. Se sabe poco sobre los efectos fisiológicos del comportamiento de desalojo en el balance oxidativo de los polluelos de C. canorus o sobre la variación en el estatus oxidativo del plasma y el nivel de testosterona relacionado con la edad de las aves en desarrollo. Examinamos si el estatus oxidativo del plasma de los polluelos de C. canorus se relacionaba con un esfuerzo previo de desalojo y cuantificamos la variación en el nivel de metabolitos reactivos de oxígeno, la capacidad antioxidante no enzimática y la concentración de testosterona en el plasma en varias fases del desarrollo de C. canorus. Tanto los niveles de metabolitos reactivos de oxígeno como la capacidad antioxidante fueron superiores en los polluelos de mayor edad que en los más jóvenes, lo que sugiere que los polluelos de menor edad contrarrestan eficazmente el aumento de la producción de radicales libres, mientras que, cuando se apróximan al abandono del nido, los niveles de metabolitos reactivos de oxígeno aumentan a pesar de una mejora en la capacidad antioxidante. Posiblemente, el gasto total de energía se incrementa con la edad, elevándose la producción de formas reactivas de oxígeno a una tasa mayor de la que el sistema antioxidante puede eliminar. El nivel de testosterona en el plasma fue máximo en la fase intermedia del crecimiento de los polluelos. Pueden requerirse altos niveles de testosterona durante el período de mayor crecimiento y, cuando la tasa de crecimiento se estabiliza cerca del abandono del nido, los niveles de testosterona también podrián disminuir. Los polluelos de C. canorus que desalojaron más huevos del hospedador en nidos con una estructura más empinada tuvieron niveles de plasma de metabolitos reactivos de oxígeno en plasma más altos poco después del período de desalojo, sugiriendo que el desalojo es costoso en términos de un incremento en el nivel de estrés oxidativo

    Study of mural paintings by Fulvio Pennacchi in Sao Paulo City by mineralogical techniques

    Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP)The present research deals with two mural paintings made in 1947 with the fresco technique by Fulvio Pennacchi in the Catholic Chapel of the Hospital das Clinicas (Sao Paulo City, Brazil), namely the Virgin Annunciation and the Supper at Emmaus. This study regards the materials and painting techniques used by the artist, based on historical research, on in situ observations and laboratory analytical techniques (stereomicroscopy, scanning electron microscopy with an energy dispersive spectrometer, X-ray diffractometry, electron microprobe, images obtained with UV-light), aiming to improve the methods of characterization of objects of our cultural heritage, and to enhance its preservation accordingly. The results lead to the identification of the plaster components and of distinct layers in the frescoes, besides further information on grain size, impurities and textures, composition of pigments, and features of deterioration, such as efflorescences. The degree of degradation of the murals painting was assessed by this way. Our data suggest that a single layer of plaster was used by Pennacchi, as a common mortar with fine- and medium-grained aggregates.
