    Production and characterization of Orpinomyces mutant xylanases with improved temperature and pH stabilities

    The error-prone PCR technique has been widely used in order to obtain thermostable enzymes more suitable for industrial conditions. The Orpinomyces xynA mutant library allowed the selection of four thermostable mutants (M1-M4). Molecular dynamics (MD) predicted an N-terminal tail as being a destabilizing structural region and allowed further enhancing of the mutant xylanases thermostability. Thus, removal of the 27 amino acid residues enabled an increase in the enzyme half-life values (t1/2). However, besides the improved thermostability, the large enzyme production and high catalytic performance are also relevant for the biotechnological application of enzymes. During the mutant enzymes production in E. coli, the IPTG induction protocol allowed high expression levels of soluble and active xylanases. The mutant xylanases without the 27 amino acid residues showed improved thermostability and the shorter versions of M2 and M4 (named as SM2 and SM4) also presented a good performance in more extreme pH conditions

    A WORLDWIDE 3D GCP DATABASE INHERITED FROM 20 YEARS OF MASSIVE MULTI-SATELLITE OBSERVATIONS

    Abstract. High location accuracy is a major requirement for satellite image users. Target performance is usually achieved thanks to either specific on-board satellite equipment or an auxiliary registration reference dataset. Both methods may be expensive and with certain limitations in terms of performance. The Institut national de l'information géographique et forestière (IGN) and Airbus Defence and Space (ADS) have worked together for almost 20 years, to build reference data for improving image location using multi-satellite observations. The first geometric foundation created has mainly used SPOT 5 High Resolution Stereoscopic (HRS) imagery, ancillary Ground Control Points (GCP) and Very High Resolution (VHR) imagery, providing a homogenous location accuracy of 10m CE90 almost all over the world in 2010.Space Reference Points (SRP) is a new worldwide 3D GCP database, built from a plethoric SPOT 6/7 multi-view archive, largely automatically processed, with cloud-based technologies. SRP aims at providing a systematic and reliable solution for image location (Unmanned Aerial Vehicle, VHR satellite imagery, High Altitudes Pseudo-Satellite…) and similar topics thanks to a high-density point distribution with a 3m CE90 accuracy.This paper describes the principle of SRP generation and presents the first validation results. A SPOT 6/7 smart image selection is performed to keep only relevant images for SRP purpose. The location of these SPOT 6/7 images is refined thanks to a spatiotriangulation on the worldwide geometric foundation, itself improved where needed. Points making up the future SRP database are afterward extracted thanks to classical feature detection algorithms and with respect to the expected density. Different filtering methods are applied to keep the best candidates. The last step of the processing chain is the formatting of the data to the delivery format, including metadata. An example of validation of SRP concept and specification on two tests sites (Spain and China) is then given. As a conclusion, the on-going production is shortly presented

    The use of tire rubber in the production of high-performance concrete

    The advances in concrete technology lead to the search for alternative materials that provide improvements in concrete properties while at the same time collaborating with sustainable practices in construction. In this study, the influence of the incorporation of waste tire rubber on the mechanical properties of high-performance concrete was discussed. The waste rubber from the tire retreading process was used in partial substitution of the fine aggregate (sand) in the percentages of 7.5%, 15% and 30% with respect to the mass of the sand. For the characterization of the concrete, the following tests were carried out: water absorption, void index, specific density, compressive strength, flexural tensile strength, modulus of elasticity and microscopy analysis.     Cooperation between NMDA-Type Glutamate and P2 Receptors for Neuroprotection during Stroke: Combining Astrocyte and Neuronal Protection

    Excitotoxicity is the principle mechanism of acute injury during stroke. It is defined as the unregulated accumulation of excitatory neurotransmitters such as glutamate within the extracellular space, leading to over-activation of receptors, ionic disruption, cell swelling, cytotoxic Ca2+ elevation and a feed-forward loop where membrane depolarisation evokes further neurotransmitter release. Glutamate-mediated excitotoxicity is well documented in neurons and oligodendrocytes but drugs targeting glutamate excitotoxicity have failed clinically which may be due to their inability to protect astrocytes. Astrocytes make up ~50% of the brain volume and express high levels of P2 adenosine triphosphate (ATP)-receptors which have excitotoxic potential, suggesting that glutamate and ATP may mediate parallel excitotoxic cascades in neurons and astrocytes, respectively. Mono-cultures of astrocytes expressed an array of P2X and P2Y receptors can produce large rises in [Ca2+]i; mono-cultured neurons showed lower levels of functional P2 receptors. Using high-density 1:1 neuron:astrocyte co-cultures, ischemia (modelled as oxygen-glucose deprivation: OGD) evoked a rise in extracellular ATP, while P2 blockers were highly protective of both cell types. GluR blockers were only protective of neurons. Neither astrocyte nor neuronal mono-cultures showed significant ATP release during OGD, showing that cell type interactions are required for ischemic release. P2 blockers were also protective in normal-density co-cultures, while low doses of combined P2/GluR blockers where highly protective. These results highlight the potential of combined P2/GluR block for protection of neurons and glia.</jats:p

    Aortic thrombus in a patient with myeloproliferative thrombocytosis, successfully treated by pharmaceutical therapy: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Thrombosis in myeloproliferative thrombocytosis occurs usually in the microvessels and medium-sized arteries and veins and only rarely in the aorta. Aortic thrombosis is usually treated with thrombectomy. Reported here is a rare case that was treated pharmacologically.</p> <p>Case presentation</p> <p>A 60-year-old Japanese woman presented with numbness of both lower extremities. Her platelet count was 1787 × 10<sup>3</sup>/μl. Through bone marrow examination, we diagnosed her condition as myelodysplastic and/or myeloproliferative disorder-unclassifiable. Abdominal ultrasonography and computed tomographic scan revealed aortic thrombosis. Her platelet count was controlled with hydroxyurea and ranimustine. Aspirin and ticlopidine improved the numbness in both lower limbs on the second day. Aortic thrombosis was not observed in a computed tomographic scan on the seventh day.</p> <p>Conclusion</p> <p>For aortic thrombosis, surgical management is usually adopted, but pharmacological management is also an option because of its immediate curative effects.</p

    The P2Y1 receptor is involved in the maintenance of glucose homeostasis and in insulin secretion in mice

    Pancreatic β cells express several P2 receptors including P2Y1 and the modulation of insulin secretion by extracellular nucleotides has suggested that these receptors may contribute to the regulation of glucose homeostasis. To determine whether the P2Y1 receptor is involved in this process, we performed studies in P2Y1 mice. In baseline conditions, P2Y1-mice exhibited a 15% increase in glycemia and a 40% increase in insulinemia, associated with a 10% increase in body weight, pointing to a role of the P2Y1 receptor in the control of glucose metabolism. Dynamic experiments further showed that P2Y1-mice exhibited a tendency to glucose intolerance. These features were associated with a decrease in the plasma levels of free fatty acid and triglycerides. When fed a lipids and sucrose enriched diet for 15 weeks, the two genotypes no longer displayed any significant differences. To determine whether the P2Y1 receptor was directly involved in the control of insulin secretion, experiments were carried out in isolated Langerhans islets. In the presence of high concentrations of glucose, insulin secretion was significantly greater in islets from P2Y1-mice. Altogether, these results show that the P2Y1 receptor plays a physiological role in the maintenance of glucose homeostasis at least in part by regulating insulin secretion

    TRY plant trait database - enhanced coverage and open access

    Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives

    Mitotic Spindle Orients Perpendicular to the Forces Imposed by Dynamic Shear

    Orientation of the division axis can determine cell fate in the presence of morphogenetic gradients. Understanding how mitotic cells integrate directional cues is therefore an important question in embryogenesis. Here, we investigate the effect of dynamic shear forces on confined mitotic cells. We found that human epithelial cells (hTERT-RPE1) as well as MC3T3 osteoblasts align their mitotic spindle perpendicular to the external force. Spindle orientation appears to be a consequence of cell elongation along the zero-force direction in response to the dynamic shear. This process is a nonlinear response to the strain amplitude, requires actomyosin activity and correlates with redistribution of myosin II. Mechanosteered cells divide normally, suggesting that this mechanism is compatible with biological functions
