    Nanoliter-scale, regenerable ion sensor: Sensing with surface functionalized microstructured optical fiber

    Femtosecond laser written Bragg gratings have been written in exposed-core microstructured optical fibers with core diameters ranging from 2.7 ÎŒm to 12.5 ÎŒm and can be spliced to conventional single mode fiber. Writing a Bragg grating on an open core fiber allows for real-time refractive index based sensing, with a view to multiplexed biosensing. Smaller core fibers are shown both experimentally and theoretically to provide a higher sensitivity. A 7.5 ÎŒm core diameter fiber is shown to provide a good compromise between sensitivity and practicality and was used for monitoring the deposition of polyelectrolyte layers, an important first step in developing a biosensor.Sabrina Heng, Mai-Chi Nguyen, Roman Kostecki, Tanya M.     Enzyme detection by surface plasmon resonance using specially engineered spacers and plasmonic labelling

    Surface Plasmon Resonance (SPR) is a powerful label free optical biosensing technology that relies on the measurement of the refractive index or change of mass in close vicinity of the sensor surface. Therefore, there is an experimental limitation in the molecular weight of the molecule that can be detected and consequently small molecules are intrinsically more difficult to detect using SPR. One approach to overcoming this limitation is to first adsorb smaller molecules onto the sensor surface, and to follow this by using their higher molecular weight antibodies counterparts which ensure the specificity (and are easier to detect via SPR due to their higher weight). Although this has been demonstrated with some success, it is not applicable in every case and some biomolecules such as enzyme are still difficult to detect due to their specific reactivity (enzymatic reaction). In this paper, we present a powerful new method that utilises specifically engineered spacers attached on one end to the sensor surface and on the other end to a nanoparticle that behaves as a plasmonic label. These spacers are design to specifically react with the biomolecule to be detected and release the (relatively large) plasmonic label, which in turn results in a measurable SPR shift (which is much larger than the shift that would have been associated with the binding of the relatively small biomolecule). As a proof of concept, this approach was used within a recently developed new form of SPR optical fibre sensor which relies on the measurement of the re-emitted light by surface scattering of the plasmonic wave rather than transmission through the fibre was used to detect an enzyme.     Microstructured optical fiber-based biosensors: reversible and nanoliter-scale measurement of zinc ions

    Published: May 6, 2016Sensing platforms that allow rapid and efficient detection of metal ions would have applications in disease diagnosis and study, as well as environmental sensing. Here, we report the first microstructured optical fiber-based biosensor for the reversible and nanoliter-scale measurement of metal ions. Specifically, a photoswitchable spiropyran Zn(2+) sensor is incorporated within the microenvironment of a liposome attached to microstructured optical fibers (exposed-core and suspended-core microstructured optical fibers). Both fiber-based platforms retains high selectivity of ion binding associated with a small molecule sensor, while also allowing nanoliter volume sampling and on/off switching. We have demonstrated that multiple measurements can be made on a single sample without the need to change the sensor. The ability of the new sensing platform to sense Zn(2+) in pleural lavage and nasopharynx of mice was compared to that of established ion sensing methodologies such as inductively coupled plasma mass spectrometry (ICP-MS) and a commercially available fluorophore (Fluozin-3), where the optical-fiber-based sensor provides a significant advantage in that it allows the use of nanoliter (nL) sampling when compared to ICP-MS (mL) and FluoZin-3 (ÎŒL). This work paves the way to a generic approach for developing surface-based ion sensors using a range of sensor molecules, which can be attached to a surface without the need for its chemical modification and presents an opportunity for the development of new and highly specific ion sensors for real time sensing applications.Sabrina Heng, Christopher A.     Exposed core microstructured optical fiber surface plasmon resonance biosensor

    Surface Plasmon Resonance (SPR) scattering offers significant advantages compared to traditional reflectivity measurements, essentially turning a non-radiative process into a radiative one. Recently, we have shown that SPR scattering can be used in an optical fiber, enabling higher signal to noise ratio, reduced dependence on the metallic thickness as well as the unique capability of multiplexed detection with a single fiber. Here we report a novel SPR scattering based sensor fabricated based on an exposed-core silica Microstructured Optical Fiber (MOF). This MOF presents a structure with a relatively small core (Ø = 10ÎŒm), exposed along the whole fiber length. This exposed core MOF allows for fabrication of SPR supporting metallic thin films directly onto the fiber core offering the new prospect of exploiting SPR in a waveguide structure that supports only a relatively small number of guided optical modes, with a structure that offers ease of fabrication and handling. A thin silver film of 50 nm thickness was deposited onto the fiber core by thermal evaporation. The significant surface roughness of the prepared metallic coatings facilitates strong scattering of the light wave coupled into the surface plasmon. Performance characteristics of the new exposed core fiber sensor were compared to those of a large bare core silica fiber (Ø = 140ÎŒm). Although sensitivity of both sensors was comparable (around 2500nm/RIU), full width at half maximum (FWHM) of the SPR peaks for the new exposed core fiber sensor decreased by a factor of 3 offering an significant enhancement in the detection limit of the new sensing platform in addition to the prospect of a sensor with a lower detection volume.Elizaveta Klantsataya, Alexandre François, Agnieszka Zuber, Valeria Torok, Roman Kostecki and Tanya M.     ‘Do i care?’ young adults' recalled experiences of early adolescent overweight and obesity: a qualitative study

    <p>Objective: Individual behaviour change to reduce obesity requires awareness of, and concern about, weight. This paper therefore describes how young adults, known to have been overweight or obese during early adolescence, recalled early adolescent weight-related awareness and concerns. Associations between recalled concerns and weight-, health- and peer-related survey responses collected during adolescence are also examined.</p> <p>Design: Qualitative semi-structured interviews with young adults; data compared with responses to self-report questionnaires obtained in adolescence.</p> <p>Participants: A total of 35 participants, purposively sub-sampled at age 24 from a longitudinal study of a school year cohort, previously surveyed at ages 11, 13 and 15. Physical measures during previous surveys allowed identification of participants with a body mass index (BMI) indicative of overweight or obesity (based on British 1990 growth reference) during early adolescence. Overall, 26 had been obese, of whom 11 had BMI99.6th centile, whereas 9 had been overweight (BMI=95th–97.9th centile).</p> <p>Measures: Qualitative interview responses describing teenage life, with prompts for school-, social- and health-related concerns. Early adolescent self-report questionnaire data on weight-worries, self-esteem, friends and victimisation (closed questions).</p> <p>Results: Most, but not all recalled having been aware of their overweight. None referred to themselves as having been obese. None recalled weight-related health worries. Recollection of early adolescent obesity varied from major concerns impacting on much of an individual's life to almost no concern, with little relation to actual severity of overweight. Recalled concerns were not clearly patterned by gender, but young adult males recalling concerns had previously reported more worries about weight, lower self-esteem, fewer friends and more victimisation in early adolescence; no such pattern was seen among females. Conclusion: The popular image of the unhappy overweight teenager was not borne out. Many obese adolescents, although well aware of their overweight recalled neither major dissatisfaction nor concern. Weight-reduction behaviours are unlikely in such circumstances.</p&gt

    Boronate probes for the detection of hydrogen peroxide release from human spermatozoa

    Human spermatozoa are compromised by production of reactive oxygen species (ROS) and detection of ROS in spermatozoa is important for the diagnosis of male infertility. Probes 2',7'-dichlorohydrofluorescein diacetate (DCFH), dihydroethidium (DHE) and MitoSOX red (MSR) are commonly used for detecting ROS by flow cytometry, however these probes lack sensitivity to hydrogen peroxide (H2O2), which is particularly damaging to mammalian sperm cells. This study reports the synthesis and use of three aryl boronate probes, peroxyfluor-1 (PF1), carboxy peroxyfluor-1 (CPF1) and a novel probe 2(2-ethoxyethoxy)ethoxy peroxyfluor-1 (EEPF1) in human spermatozoa. PF1 and EEPF1 were found to be effective in detecting H2O2 and peroxynitrite (ONOO(-)) produced by spermatozoa when stimulated with menadione or 4-hydroxynonenal. EEPF1 was more effective at detection of ROS in spermatozoa than DCFH, DHE and MSR; furthermore it distinguished poorly motile sperm as shown by greater ROS production. EEPF1 should therefore have a significant role in diagnosis of oxidative stress in male infertility, cryopreservation, age, lifestyle and exposure to environmental toxicants.Malcolm S.     The Effect of a Brief Salivary α-Amylase Exposure During Chewing on Subsequent in Vitro Starch Digestion Curve Profiles

    There is inconsistency between current in vitro digestion methods with regard to accommodation of a (salivary) α-amylase exposure during the oral phase. The effect of a salivary α-amylase pre-exposure on subsequent in vitro starch digestion curve profiles for various foods was investigated. Foods were chewed, expectorated and the boluses left to rest for 0–15 min. During pancreatic digestion, aliquots were taken and hydrolysis curves constructed for comparison against those of the same foods comminuted with a manually-operated chopper, hence spared exposure to saliva. Hydrolysate aliquots taken at T0 (time zero) of the digestion of chewed samples contained higher levels of glucose and dextrins compared with chopped samples. Pancreatin activity immediately overwhelmed differences in sugar released due to salivary amylase activity. Within 10 min no differences were detectable between hydrolysis curves for chewed and chopped foods. Salivary amylase pretreatment does not contribute to the robustness or relative accuracy of in vitro methods
