    Tujuan dari penelitian ini adalah untuk mengetahui pengaruh motivasi kerja dan disiplin kerja pada karyawan Perusahaan Fitness terhadap kualitas layanan yang diberi­kan. Jenis penelitian ini adalah penelitian kuantitatif kau­sal. Penelitian ini memakai teknik analisis PLS. Populasi dari pe­ne­li­­tian ini adalah jumlah member di Perusahaan Fitness dan jumlah sampel berjumlah 99. Ha­­sil menunjukkan bahwa motivasi kerja dan disiplin kerja memiliki pe­nga­­ruh terhadap kualitas layanan

    “Bicara Karya” as a news feature program created as a new show that can be a source of information and education about the creative industry in the city of Semarang. The program elevates creative industry activities from making, success story, to barriers that occur in a creative industry. Packed with a personal explanation of someone who has expertise in their fields, discussed with a light language but not leaving the essence. In the “Bicara Karya”, The program directors are technically responsible for programming programs both visualizing the manuscript and communicating what they see to the entire production crew (camera person, scriptwriter, voice over, editor, etc.) to realize quality and in accordance with the event's concept and producer's direction. . After going through the pre-production stage, the production process, post production, the work is shown on Cakra Semarang TV every week, starting from 25 April 2017 until 18 July 2017 every Tuesday at 18.00 WIB. Through this work, it is hoped that the public will get educational program to educate and increase the audience information about the creative industry in Semarang and add insight to the viewers. Through the television program "Biacara Karya" which aired on Cakra Semarang TV is a show that raised about the industry keratif, the program director has completed its duties to run for 13 episodes without re-run and certainly not leaving the essence to be conveyed by packing it into an educative and motivating program but not being patronizing

    Semakin pesatnya pertumbuhan suatu kota pada umumnya diiringi dengan kebijakan pengembangan wilayah dari pemerintahan kota tersebut salah satunya adalah pembangunan perumahan bagi penduduk perkotaan. Sebagai suatu hunian perumahan memiliki hubungan keterkaitan dengan lingkungan sekitarnya.Perumahan merupakan suatu wilayah hunian yang dapat berkembang seiring dengan dinamika para penghuninya.Pengembangan perumahan dapat mempengaruhi perkembangan wilayah sekitarnya melalui berbagai aktifitas pembangunan / pengembangan kawasan itu sendiri ataupun melalui aktifitas warganya.Yang menjadi fokus dari penelitian ini adalah mengetahui bagaimana pengembangan perumahan terencana serta menganalisis pengaruh yang ditimbulkannya terhadap perkembangan wilayah di Kecamatan Mapanget. Lingkup dari penelitian ini adalah Kecamatan Mapanget Kota Manado sedangkan yang menjadi metode dalam penelitian ini adalah deskriptif kuantitatif dengan menggunakan analisis time series, skala guttman, nearest neighbor dan regresi linier. Berdasarkan hasil penelitian dapat disimpulkan bahwa bahwa perumahan memberikan pengaruh yang sangat besar bagi perkembangan di Kecamatan Mapanget, hal ini dapat dilihat dari analisis skalogram yang menunjukkan pusat pelayanan di di Kecamatan Mapanget selalu berubah – ubah serta pola permukiman di Kecamatan Mapanget yang telah mengalami perubahan dari pola berkelompok menjadi acak dalam kurun waktu 6 tahun sedangkan untuk analisis regresi menunjukkan jumlah perumahan memberi pengaruh yang KUAT bagi fasilitas umum dan fasilitas sosial namun terdapat beberapa variabel yang secara statistik  nilai  probabilitasnya > 0,05 maka pengaruh yang diberikan dianggap tidak signifikan.   Kata Kunci : Perumahan, Kecamatan Mapange

    Sempadan dan pertikaian pemilikan tanah adat di Sarawak

    Pertikaian pemilikan tanah adat secara jelasnya berpunca daripada tiga faktor utama iaitu status, kesahihan pemilikan dan keluasan sebenar tanah yang dimiliki. Konflik masyarakat berhubung dengan perkara tersebut boleh dikaitkan dengan cara informasi tanah diperoleh. Kebanyakan pemilik tanah adat mendapatkan informasi tanah milik mereka secara lisan iaitu melalui informasi mulut ke mulut. Hal ini akhirnya menjadi satu cabaran yang besar kepada masyarakat apabila timbul pelbagai anggapan dan intepretasi yang berbeza berkait dengan status dan kesahihan tanah adat yang dimiliki. Justeru kajian ini menelit i maklumbalas pemilik tanah adat berhubung dengan cara penentuan sempadan tanah dalam kalangan masyarakat Bidayuh di daerah Serian Sarawak. Selain itu kajian juga memberikan perhatian terhadap pertikaian persempadanan tanah adat serta impilkasinya dalam kalangan pemilik tanah adat. Maklumat yang berkait dengan tujuan kajian telah diperoleh melalui FGD serta temuselidik mendalam dengan pemilik tanah adat di kawasan kajian. Hasil kajian mendapati wujud beberapa cara penentuan sempadan dalam kalangan pemilik tanah. Sehubungan itu, dapat dirumuskan bahawa pertikaian tanah di kawasan kajian banyak didorong oleh isu status dan kesahihan pemilikan tanah yang dimiliki oleh pemilik tanah adat. Bagi mengurangkan konflik maka dicadangkan agar tanah-tanah yang ada dibuat sukatan oleh juruukur bertauliah. Apabila tanah telah disukat, pemilik tanah akan memperoleh surat hakmilik tanah yang lebih jelas dari segi status dan kesasihan pemilikan

    Structural and infrared absorption properties of self-organized InGaAs GaAs quantum dots multilayers

    Self-organized InGaAs/GaAs quantum dots (QDs) stacked multilayers have been prepared by solid source molecular beam epitaxy. Cross-sectional transmission electron microscopy shows that the InGaAs QDs are nearly perfectly vertically aligned in the growth direction [100]. The filtering effect on the QDs distribution is found to be the dominant mechanism leading to vertical alignment and a highly uniform size distribution. Moreover, we observe a distinct infrared absorption from the sample in the range of 8.6-10.7 mu m. This indicates the potential of QDs multilayer structure for use as infrared photodetector

    Analysis of host responses to Mycobacterium tuberculosis antigens in a multi-site study of subjects with different TB and HIV infection states in sub-Saharan Africa.

    BACKGROUND: Tuberculosis (TB) remains a global health threat with 9 million new cases and 1.4 million deaths per year. In order to develop a protective vaccine, we need to define the antigens expressed by Mycobacterium tuberculosis (Mtb), which are relevant to protective immunity in high-endemic areas. METHODS: We analysed responses to 23 Mtb antigens in a total of 1247 subjects with different HIV and TB status across 5 geographically diverse sites in Africa (South Africa, The Gambia, Ethiopia, Malawi and Uganda). We used a 7-day whole blood assay followed by IFN-γ ELISA on the supernatants. Antigens included PPD, ESAT-6 and Ag85B (dominant antigens) together with novel resuscitation-promoting factors (rpf), reactivation proteins, latency (Mtb DosR regulon-encoded) antigens, starvation-induced antigens and secreted antigens. RESULTS: There was variation between sites in responses to the antigens, presumably due to underlying genetic and environmental differences. When results from all sites were combined, HIV- subjects with active TB showed significantly lower responses compared to both TST(-) and TST(+) contacts to latency antigens (Rv0569, Rv1733, Rv1735, Rv1737) and the rpf Rv0867; whilst responses to ESAT-6/CFP-10 fusion protein (EC), PPD, Rv2029, TB10.3, and TB10.4 were significantly higher in TST(+) contacts (LTBI) compared to TB and TST(-) contacts fewer differences were seen in subjects with HIV co-infection, with responses to the mitogen PHA significantly lower in subjects with active TB compared to those with LTBI and no difference with any antigen. CONCLUSIONS: Our multi-site study design for testing novel Mtb antigens revealed promising antigens for future vaccine development. The IFN-γ ELISA is a cheap and useful tool for screening potential antigenicity in subjects with different ethnic backgrounds and across a spectrum of TB and HIV infection states. Analysis of cytokines other than IFN-γ is currently on-going to determine correlates of protection, which may be useful for vaccine efficacy trials

We infected human monocyte derived macrophages (hMDMs) with different species of mycobacteria, in increasing order of pathogenicity, i.e. M. smegmatis, M. bovis BCG, and M. tuberculosis R179 that had been cultured in the absence of detergents. RNA was isolated post-infection and transcriptomic analysis using amplicons (Ampliseq) revealed 274 differentially expressed genes (DEGs) across three species, out of which we selected 19 DEGs for further validation. We used qRT-PCR to confirm the differential expression of 19 DEGs. We studied biological network through Ingenuity Pathway Analysis® (IPA) which revealed up-regulated pathways of the interferon and interleukin family related to the killing of M. smegmatis. Apart from interferon and interleukin family, we found one up-regulated (EIF2AK2) and two down-regulated (MT1A and TRIB3) genes as unique potential targets found by Ampliseq and qRT-PCR which may be involved in the intracellular mycobacterial killing. The roles of these genes have not previously been described in tuberculosis. Multiplex ELISA of culture supernatants showed increased host immune response toward M. smegmatis as compared to M. bovis BCG and M.tb R179. These results enhance our understanding of host immune response against M.tb infection.Publisher's versio
