    A host of traveling waves in a model of three-dimensional water-wave dynamics

    We describe traveling waves in a basic model for three-dimensional water-wave dynamics in the weakly nonlinear long-wave regime. Small solutions that are periodic in the direction of translation (or orthogonal to it) form an infinite-dimensional family. We characterize these solutions through spatial dynamics, by reducing a linearly ill-posed mixed-type initial-value problem to a center manifold of infinite dimension and codimension. A unique global solution exists for arbitrary small initial data for the two-component bottom velocity, specified along a single line in the direction of translation (or orthogonal to it).     Effect of aliskiren on post-discharge outcomes among diabetic and non-diabetic patients hospitalized for heart failure: insights from the ASTRONAUT trial

    Aims The objective of the Aliskiren Trial on Acute Heart Failure Outcomes (ASTRONAUT) was to determine whether aliskiren, a direct renin inhibitor, would improve post-discharge outcomes in patients with hospitalization for heart failure (HHF) with reduced ejection fraction. Pre-specified subgroup analyses suggested potential heterogeneity in post-discharge outcomes with aliskiren in patients with and without baseline diabetes mellitus (DM). Methods and results ASTRONAUT included 953 patients without DM (aliskiren 489; placebo 464) and 662 patients with DM (aliskiren 319; placebo 343) (as reported by study investigators). Study endpoints included the first occurrence of cardiovascular death or HHF within 6 and 12 months, all-cause death within 6 and 12 months, and change from baseline in N-terminal pro-B-type natriuretic peptide (NT-proBNP) at 1, 6, and 12 months. Data regarding risk of hyperkalaemia, renal impairment, and hypotension, and changes in additional serum biomarkers were collected. The effect of aliskiren on cardiovascular death or HHF within 6 months (primary endpoint) did not significantly differ by baseline DM status (P = 0.08 for interaction), but reached statistical significance at 12 months (non-DM: HR: 0.80, 95% CI: 0.64-0.99; DM: HR: 1.16, 95% CI: 0.91-1.47; P = 0.03 for interaction). Risk of 12-month all-cause death with aliskiren significantly differed by the presence of baseline DM (non-DM: HR: 0.69, 95% CI: 0.50-0.94; DM: HR: 1.64, 95% CI: 1.15-2.33; P < 0.01 for interaction). Among non-diabetics, aliskiren significantly reduced NT-proBNP through 6 months and plasma troponin I and aldosterone through 12 months, as compared to placebo. Among diabetic patients, aliskiren reduced plasma troponin I and aldosterone relative to placebo through 1 month only. There was a trend towards differing risk of post-baseline potassium ≥6 mmol/L with aliskiren by underlying DM status (non-DM: HR: 1.17, 95% CI: 0.71-1.93; DM: HR: 2.39, 95% CI: 1.30-4.42; P = 0.07 for interaction). Conclusion This pre-specified subgroup analysis from the ASTRONAUT trial generates the hypothesis that the addition of aliskiren to standard HHF therapy in non-diabetic patients is generally well-tolerated and improves post-discharge outcomes and biomarker profiles. In contrast, diabetic patients receiving aliskiren appear to have worse post-discharge outcomes.     <p>We study asymptotic stability of solitary wave solutions in the one-dimensional Benney--Luke equation, a formally valid approximation for describing two-way water-wave propagation. For this equation, as for the full water-wave problem, the classic variational method for proving orbital stability of solitary waves fails dramatically due to the fact that the second variation of the energy-momentum functional is infinitely indefinite. We establish nonlinear stability in energy norm under the spectral stability hypothesis that the linearization admits no nonzero eigenvalues of nonnegative real part. We then verify this hypothesis for waves of small energy.</p

    Upland habitat loss as a threat to Pantanal wetlands

    The fate of Pantanal, one of the world's largest, most pristine and diverse wetlands, stands in the balance. The most recent (2014) and comprehensive land-cover change assessment of the upper Paraguay River Basin (UPRB), comprising both lowlands (Pantanal floodplain) and their surrounding upland savannas (Cerrado plateaus), shows the extent of decline of native vegetation (Fig. 1). Around 80% of the Pantanal floodplain native vegetation remains (Fig. 1), and over 60% of its Cerrado plateaus have been converted into pasture and croplands (SOS-Pantanal et al. 2015). The most worrying aspect is the fast rate of land clearing during the last 30 years (Supporting Information). In fact, the Cerrado is experiencing higher native vegetation conversion rates than Amazon and Atlantic Forest ecosystems in recent years but is still largely unprotected (Overbeck et al. 2015)

    The precise timing of flowering in adverse environments is critical for plants to secure reproductive success. We report a mechanism in Arabidopsis (Arabidopsis thaliana) controlling the time of flowering by which the S-acylation-dependent nuclear import of the protein SALT OVERLY SENSITIVE3/CALCINEURIN B-LIKE4 (SOS3/CBL4), a Ca2+-signaling intermediary in the plant response to salinity, results in the selective stabilization of the flowering time regulator GIGANTEA inside the nucleus under salt stress, while degradation of GIGANTEA in the cytosol releases the protein kinase SOS2 to achieve salt tolerance. S-acylation of SOS3 was critical for its nuclear localization and the promotion of flowering, but partly dispensable for salt tolerance. SOS3 interacted with the photoperiodic flowering components GIGANTEA and FLAVIN-BINDING, KELCH REPEAT, F-BOX1 and participated in the transcriptional complex that regulates CONSTANS to sustain the transcription of CO and FLOWERING LOCUS T under salinity. Thus, the SOS3 protein acts as a Ca2+- and S-acylation-dependent versatile regulator that fine-tunes flowering time in a saline environment through the shared spatial separation and selective stabilization of GIGANTEA, thereby connecting two signaling networks to co-regulate the stress response and the time of flowering.ISSN:1040-4651ISSN:1531-298XISSN:1532-298

    TLR7 promotes smoke-induced experimental lung damage through the activity of mast cell tryptase

    : Toll-like receptor 7 (TLR7) is known for eliciting immunity against single-stranded RNA viruses, and is increased in both human and cigarette smoke (CS)-induced, experimental chronic obstructive pulmonary disease (COPD). Here we show that the severity of CS-induced emphysema and COPD is reduced in TLR7-deficient mice, while inhalation of imiquimod, a TLR7-agonist, induces emphysema without CS exposure. This imiquimod-induced emphysema is reduced in mice deficient in mast cell protease-6, or when wild-type mice are treated with the mast cell stabilizer, cromolyn. Furthermore, therapeutic treatment with anti-TLR7 monoclonal antibody suppresses CS-induced emphysema, experimental COPD and accumulation of pulmonary mast cells in mice. Lastly, TLR7 mRNA is increased in pre-existing datasets from patients with COPD, while TLR7+ mast cells are increased in COPD lungs and associated with severity of COPD. Our results thus support roles for TLR7 in mediating emphysema and COPD through mast cell activity, and may implicate TLR7 as a potential therapeutic target
