    Ocurrencia de anchoa adulta en Cataluña (Mediterráneo noroccidental) en relación a las condiciones superficiales del mar

    Generalized additive and generalized additive threshold models were used to study the relationship between landings per unit effort (LPUE) of anchovy, Engraulis encrasicolus, during the spawning season (May-October) from 2000-2010, and environmental variables, using sea surface data derived from satellite imagery (temperature, salinity, chlorophyll a, and meridional and zonal velocity current) in three fishing zones defined along the Catalan coast. The configuration of the environment where spawning aggregations occur affects early life stages and therefore the future demographic structure of the population. It is therefore fundamental to define the environmental conditions and their variations during the spawning season. Our results show that the low salinity in the Northern and Central sector and the velocity of the zonal and meridional currents in the Central and Southern sector, respectively, implicated in retention processes, increase LPUE during the spawning period. Temperature was related to LPUE in the Southern and in the Northern sectors, in both of which a non-linear positive effect with a local maximum peak at lower temperature values was present. However, in the Northern sector, this relationship held only for the period before 2007. After 2007 the decrease in preferred temperature suggests a reduction of the thermal window in which adult spawner aggregations occur. In agreement with previous studies on this species, the relationships were non-linear, stressing the importance of the match in timing and location between favourable conditions and spawning period as a crucial event for understanding the dynamics of small pelagics populations.Con el objeto de estudiar la relación entre descargas por unidad de esfuerzo (LPUE) de la anchoa, Engraulis encrasicolus, durante la época de puesta (Mayo-Octubre) y variables ambientales marinas obtenidas por observación de satélite (temperatura, salinidad, clorofila-a, velocidad de corriente zonal y meridional) se analizaron series de datos para el periodo 2000-2010 mediante modelos aditivos generalizados, con y sin umbral, en tres zonas de pesca de la costa catalana. La configuración del ambiente en que ocurren las agregaciones de puesta afecta a los estadios tempranos y, por consiguiente, a la estructura demográfica futura de las poblaciones. Por lo tanto es de gran importancia definir las condiciones ambientales y sus variaciones durante la época de puesta. Los resultados del análisis muestran que bajas salinidades en los sectores norte y central, y las velocidades zonal y meridional de la corriente, en los sectores central y sur respectivamente, implicadas en los procesos de retención hacen aumentar la LPUE durante el periodo de puesta. La temperatura se relacionó con la LPUE en los sectores sur y norte. En ambas zonas se observó un efecto positivo y no lineal con un pico máximo a valores de temperatura bajos. Sin embargo, en el sector norte esta relación se mantuvo solamente hasta 2007. Después de 2007, la disminución en la temperatura preferencial sugiere una reducción en la ventana térmica en la que ocurren las agregaciones de puesta. Las relaciones obtenidas son no lineales, coincidiendo con estudios previos en esta especie, haciendo resaltar la importancia del ajuste espacio-temporal entre condiciones favorables y periodo de puesta como evento crucial para entender la dinámica poblacional de los pequeños pelágicos

    Estudio de la acción de una construcción peptídica dendrimérica como vacuna contra la fiebre aftosa en bovinos

    Foot-and-mouth disease (FMD) is a highly contagious disease of cloven-hoofed animals, and probably the most important cattle disease in terms of economic impact. Control of FMD is achieved by using vaccines containing the chemically inactivated virus; this requires the production of large volumes of the virus, the inactivation of the virus, antigen concentration and purification with the risk of virus release during vaccine production. Other vaccine problems include: (i) lack of long - term protection, (ii) thermal instability of the vaccine, etc. The use of synthetic peptides offers advantages over conventional vaccines. Dendrimeric (PD) peptides are chemically synthesized molecules with a well-defined structure, and at the same time very versatile. They consist of the union of branched structures and the presence of multiple immunogenic sites. The aim of the study was to evaluate the immunogenicity and protection induced by FMD virusderived dendrimeric peptides: B2T and B4T containing a T [3A (aa 21-35)] epitope] and two or four copies of a B epitope [site A VP1 (aa 136 to 154)] in cattle. Bovines were vaccinated with 2 mg of B4T (n = 4) or B2T (n = 4) (days 0 and 20) intramuscularly and 2 animals were used as negative control. At 38 days post-vaccination (dpv), antibody titers against FMD virus (ELISA) in B4T and B2T groups were: 3.5 ± 1.0 and 4.1 ± 1.0 respectively (the predominant isotype was IgG1). The α-peptide antibodies were also increased in both groups. The neutralization indexes were ≥ 1.8. The proliferative lymphocyte response was positive (IE ≥ 3) in animals vaccinated with B2T and B4T when the mononuclear cells were “in vitro” re-stimulated with peptides; an increase of IFNγ was also detected in the supernatant of these cells when stimulated with inactivated FMD virus. At 42 dpv the bovines were challenged with active FMDV. Two animals in the B4T group and 2 in the B2T group delayed the onset of aphthous symptoms, while only one animal in each group was fully protected. In conclusion, the protection induced by B4T and B2T was low but the highest level of protection was observed in animals that presented higher cellular and humoral responses against FMD virus.La Fiebre aftosa (FA) es una enfermedad muy contagiosa de los animales de pezuña hendida y es probablemente la enfermedad más importante en términos de impacto económico. El control se realiza mediante el uso de vacunas que contienen el virus inactivado químicamente. Esto requiere de la producción de grandes volúmenes de virus, su inactivación y posterior concentración, y de la purificación del antígeno. Por esta causa, ha aumentado la preocupación por cuestiones vinculadas a la bioseguridad, como el riesgo de que se produzca “escape” del virus durante la producción. Otros problemas de las vacunas incluyen: i) la falta de protección a largo plazo, ii) inestabilidad térmica de la vacuna, etc. El uso de péptidos sintéticos ofrece ventajas sobre las vacunas convencionales. Los péptidos dendrímeros (PD) son moléculas políméricas de síntesis química con estructuras bien definidas y muy versátiles. Consisten en la unión de estructuras ramificadas y la presencia de múltiples sitios inmunogénicos. El objetivo del estudio fue evaluar la inmunogenicidad y la protección inducida por los péptidos dendrímeros derivado del virus de la FA: B2T y B4T que contienen un epítope T [3A (aa 21-35)] y dos o cuatro copias de un epítope B [sitio A de VP1 (aa 136 a 154)] en bovinos. Se vacunaron bovinos con 2 mg de B4T (n = 4) o B2T (n = 4) (días 0 y 20) por vía intramuscular y 2 animales se usaron como controles negativos. A los 38 días postvacunación (dpv), los títulos de anticuerpos contra el virus de la FA (ELISA) en el grupo de B4T y B2T fueron de 3,5 ± 1,0 y 4,1 ± 1,0 respectivamente (el isotipo predominante fue IgG1). Los anticuerpos α–péptido también se vieron incrementados en ambos grupos. Los índices de neutralización fueron ≥ 1,8. La respuesta linfo proliferativa fue positiva (IE= o > a 3) en los animales vacunados con B2T y B4T cuando las células mononucleares fueron reestimuladas in vitro con los péptidos. También se detectó incremento de IFNγ en el sobrenadante de estas células cuando fueron estimuladas con virus de la FA inactivado. A los 42 dpv los bovinos fueron desafiados con virus de la FA activo. Dos animales en el grupo B4T y 2 en el grupo B2T retrasaron la aparición de los signos clínicos de aftosa, mientras que solo un animal de cada grupo estuvo completamente protegido. A partir de estos resultados se ha concluido que la protección inducida por B4T y B2T fue baja, pero se observó el mayor nivel de protección en los animales que presentaron mayores respuestas específicas celular y humoral frente a VFA

    Patterns of use, effectiveness and safety of gadolinium contrast agents: a European prospective cross-sectional multicentre observational study

    Background: The EU gadolinium-based contrast agents (GBCA) market has changed in recent years due to the European Medicines Agency decision to suspend the marketing authorisation of linear GBCA and the marketing authorisation of new generic macrocyclic GBCA. The study aims to understand the patterns of (GBCA) use, and to study the effectiveness and safety of GBCA in routine practice across Europe. Methods: Prospective, cross-sectional, multicentre, observational study in patients undergoing contrast-enhanced magnetic resonance. Reported usage patterns included indication, referral and examination details. Assessment of effectiveness included changes in radiological diagnosis, diagnostic confidence and image quality. Safety data were collected by spontaneous patient adverse event (AE) reporting. Results: 2118 patients were included from 8 centres across 5 European countries between December 2018 and November 2019. Clariscan, Dotarem (gadoteric acid), Gadovist (gadobutrol) and ProHance (gadoteridol) were utilised in 1513 (71.4%), 356 (16.8%), 237 (11.2%) and 12 (0.6%) patients, respectively. Most were performed in CNS-related indications (46.2%). Mean GBCA doses were 0.10 mmol/kg body weight, except for Gadovist (mean 0.12 mmol/kg). GBCA use increased confidence in diagnosis in 96.2% of examinations and resulted in a change in radiological diagnosis in 73.9% of patients. Image quality was considered excellent or good in 96.1% of patients and across all GBCA. Four patients reported AEs (0.19%), with only 1 (0.05%) considered serious. Conclusions: This European study confirmed that GBCAs are used appropriately in Europe for a wide range of indications. The study demonstrated a significant increase in diagnostic confidence after GBCA use and confirmed the good safety profile of GBCAs, with comparable results for all agents used

    Protection induced by a glycoprotein E-deleted bovine herpesvirus type 1 marker strain used either as an inactivated or live attenuated vaccine in cattle

    BACKGROUND: Bovine herpesvirus type 1 (BoHV-1) is the causative agent of respiratory and genital tract infections; causing a high economic loss in all continents. Use of marker vaccines in IBR eradication programs is widely accepted since it allows for protection of the animals against the disease while adding the possibility of differentiating vaccinated from infected animals.The aim of the present study was the development and evaluation of safety and efficacy of a glycoprotein E-deleted (gE-) BoHV-1 marker vaccine strain (BoHV-1ΔgEβgal) generated by homologous recombination, replacing the viral gE gene with the β-galactosidase (βgal) gene. RESULTS: In vitro growth kinetics of the BoHV-1ΔgEβgal virus was similar to BoHV-1 LA. The immune response triggered by the new recombinant strain in cattle was characterized both as live attenuated vaccine (LAV) and as an inactivated vaccine. BoHV-1ΔgEβgal was highly immunogenic in both formulations, inducing specific humoral and cellular immune responses. Antibody titers found in animals vaccinated with the inactivated vaccine based on BoHV-1ΔgEβgal was similar to the titers found for the control vaccine (BoHV-1 LA). In the same way, titers of inactivated vaccine groups were significantly higher than any of the LAV immunized groups, independently of the inoculation route (p < 0.001). Levels of IFN-γ were significantly higher (p < 0.001) in those animals that received the LAV compared to those that received the inactivated vaccine. BoHV-1ΔgEβgal exhibited an evident attenuation when administered as a LAV; no virus was detected in nasal secretions of vaccinated or sentinel animals during the post-vaccination period. BoHV-1ΔgEβgal, when used in either formulation, elicited an efficient immune response that protected animals against challenge with virulent wild-type BoHV-1. Also, the deletion of the gE gene served as an immunological marker to differentiate vaccinated animals from infected animals. All animals vaccinated with the BoHV-1ΔgE βgal strain were protected against disease after challenge and shed significantly less virus than control calves, regardless of the route and formulation they were inoculated. CONCLUSIONS: Based on its attenuation, immunogenicity and protective effect after challenge, BoHV-1ΔgEβgal virus is an efficient and safe vaccine candidate when used either as inactivated or as live attenuated forms.Fil: Romera, Sonia.     Semiconductor pixel detectors for digital mammography

    Abstract We present some results obtained with silicon and gallium arsenide pixel detectors to be applied in the field of digital mammography. Even though GaAs is suitable for medical imaging applications thanks to its atomic number, which allows a very good detection efficiency, it often contains an high concentrations of traps which decrease the charge collection efficiency (CCE). So we have analysed both electrical and spectroscopic performance of different SI GaAs diodes as a function of concentrations of dopants in the substrate, in order to find a material by which we can obtain a CCE allowing the detection of all the photons that interact in the detector. Nevertheless to be able to detect low contrast details, efficiency and CCE are not the only parameters to be optimized; also the stability of the detection system is fundamental. In the past we have worked with Si pixel detectors; even if its atomic number does not allow a good detection efficiency at standard thickness, it has a very high stability. So keeping in mind the need to increase the Silicon detection efficiency we performed simulations to study the behaviour of the electrical potential in order to find a geometry to avoid the risk of electrical breakdown

    A New Cage-Like Particle Adjuvant Enhances Protection of Foot-and-Mouth Disease Vaccine

    Foot-and-Mouth Disease (FMD) is an acute viral disease that causes important economy losses. Vaccines with new low-cost adjuvants that stimulate protective immune responses are needed and can be assayed in a mouse model to predict their effectiveness in cattle. Immunostimulant Particle Adjuvant (ISPA), also known as cage-like particle adjuvant, consisting of lipid boxes of dipalmitoyl-phosphatidylcholine, cholesterol, sterylamine, alpha-tocopherol, and QuilA saponin, was shown to enhance protection of a recombinant vaccine against Trypanosoma cruzi in a mouse model. Thus, in the present work, we studied the effects on the magnitude and type of immunity elicited in mice and cattle in response to a vaccine based on inactivated FMD virus (iFMDV) formulated with ISPA. It was demonstrated that iFMDV–ISPA induced protection in mice against challenge and elicited a specific antibody response in sera, characterized by a balanced Th1/Th2 profile. In cattle, the antibody titers reached corresponded to an expected percentage of protection (EPP) higher than 80%. EPP calculates the probability that livestock would be protected against a 10,000 bovine infectious doses challenge after vaccination. Moreover, in comparison with the non-adjuvanted iFMDV vaccine, iFMDV–ISPA elicited an increased specific T-cell response against the virus, including higher interferon gamma (IFNγ)+/CD8+ lymphocyte production in cattle. In this work, we report for first time that an inactivated FMDV serotype A vaccine adjuvanted with ISPA is capable of inducing protection against challenge in a murine model and of improving the specific immune responses against the virus in cattle.Fil: Bidart, Juan Esteban.     Bovine Dendritic Cell Activation, T Cell Proliferation and Antibody Responses to Foot-And-Mouth Disease, Is Similar With Inactivated Virus and Virus Like Particles

    Foot-and-mouth disease (FMD) is a highly contagious disease of cloven-hoofed animals that causes severe economic losses in the livestock industry. Currently available vaccines are based on the inactivated FMD virus (FMDV). Although inactivated vaccines have been effective in controlling the disease, they have some disadvantages. Because of these disadvantages, investigations are being made to produce vaccines in low containment facilities. The use of recombinant empty capsids (also referred as Virus Like Particles, VLPs) has been reported to be a promising candidate as a subunit vaccine because it avoids the use of virus in the vaccine production and conserves the conformational epitopes of the virus. Mignaqui and collaborators have produced recombinant FMDV empty capsids from serotype A/ARG/2001 using a scalable technology in mammalian cells that elicited a protective immunity against viral challenge in a mouse model. However, further evaluation of the immune response elicited by these VLPs in cattle is required.     MANα1-2MAN decorated liposomes enhance the immunogenicity induced by a DNA vaccine against BoHV-1

    New technologies in the field of vaccinology arise as a necessity for the treatment and control of many diseases. Whole virus inactivated vaccines and modified live virus ones used against Bovine Herpesvirus-1 (BoHV-1) infection have several disadvantages. Previous works on DNA vaccines against BoHV-1 have demonstrated the capability to induce humoral and cellular immune responses. Nevertheless, ‘naked’ DNA induces low immunogenic response. Thus, loading of antigen encoding DNA sequences in liposomal formulations targeting dendritic cell receptors could be a promising strategy to better activate these antigen-presenting cells (APC). In this work, a DNA-based vaccine encoding the truncated version of BoHV-1 glycoprotein D (pCIgD) was evaluated alone and encapsulated in a liposomal formulation containing LPS and decorated with MANα1-2MAN-PEG-DOPE (pCIgD-Man-L). The vaccinations were performed in mice and bovines. The results showed that the use of pCIgD-Man-L enhanced the immune response in both animal models. For humoral immunity, significant differences were achieved when total antibody titres and isotypes were assayed in sera. Regarding cellular immunity, a significant increase in the proliferative response against BoHV-1 was detected in animals vaccinated with pCIgD-Man-L when compared to the response induced in animals vaccinated with pCIgD. In addition, upregulation of CD40 molecules on the surface of bovine dendritic cells (DCs) was observed when cells were stimulated and activated with the vaccine formulations. When viral challenge was performed, bovines vaccinated with MANα1-2MAN-PEG-DOPE elicited better protection which was evidenced by a lower viral excretion. These results demonstrate that the dendritic cell targeting using MANα1-2MAN decorated liposomes can boost the immunogenicity resulting in a long-lasting immunity. Liposomes decorated with MANα1-2MAN-PEG-DOPE were tested for the first time as a DNA vaccine nanovehicle in cattle as a preventive treatment against BoHV-1.     Study of the response of a single photon counting system in various experimental conditions for low-energy radiological applications

    In low-energy radiological applications the capability to have reproducible image uniformity and good detection efficiency are relevant issues. A single photon counting system based on a 700 μm thick silicon pixel detector, to ensure a good detection efficiency, and connected to the Medipix2 read-out chip is a good candidate to realize a low-noise detection system with a stable response. We present the experimental results obtained with this system. The performaces obtained with a variable thickness phantom are also reported
