Cloning and expression of the Bacillus amyloliquefaciens transglutaminase gene in E. coli using a bicistronic vector construction
Transglutaminases (TGases) are a class of transferases widely used in the food and biotechnology industries. In this work, we describe the production of recombinant Bacillus amyloliquefaciens TGase in Escherichia coli, obtaining the protein in its soluble and active form. In order to reduce TGase activity inside host cells and consequently its toxicity, we constructed a bicistronic plasmid containing the B. amyloliquefaciens TGase gene fused to the inhibitory Streptomyces caniferus prodomain. To make the enzyme active and avoid the need of prodomain removal in vitro, we also cloned the 3C protease gene into the same plasmid. After a fast single-step purification protocol, we obtained a partially purified recombinant TGase with 37 mU/mg protein activity, that crosslinked bovine serum albumin (BSA). This is the first report on the expression of B. amyloliquefaciens TGase in E. coli in its mature and active form
A Contribuição do Estágio no Entendimento do Papel do Professor de Química
Resumo: As tendências contemporâneas de ensino vêm sendo consideradas essenciais para a produção de aprendizagens significativas. Considerando a importância do uso dessas tendências na construção de significados, o presente trabalho envolveu a inserção de professores em formação na docência e a avaliação compartilhada da prática docente, aproximando os saberes teóricos e práticos. Percebemos que esses professores desenvolveram aulas nas quais houve grande interação com os estudantes, abrindo espaço para a participação, mas tiveram dificuldades com o estabelecimento de um discurso dialógico. Na reflexão sobre a ação, percebemos evolução das concepções sobre o papel do professor e dos estudantes na dinâmica da sala de aula
Prevalence of Lens Opacity in Interventional Cardiologists and Professional Working in the Hemodynamics in Brazil
Abstract Background: Posterior subcapsular cataract is a tissue reaction commonly found among professionals exposed to ionizing radiation. Objective: To assess the prevalence of cataract in professionals working in hemodynamics in Brazil. Methods: Professionals exposed to ionizing radiation (group 1, G1) underwent slit lamp examination with a biomicroscope for lens examination and compared with non-exposed subjects (group 2, G2). Ophthalmologic findings were described and classified by opacity degree and localization using the Lens Opacities Classification System III. Both groups answered a questionnaire on work and health conditions to investigate the presence of risk factors for cataract. The level of significance was set at 5% (p < 0.05). Results: A total of 112 volunteers of G1, mean age of 44.95 (±10.23) years, and 88 volunteers of G2, mean age of 48.07 (±12.18) years were evaluated; 75.2% of G1 and 85.2% of G2 were physicians. Statistical analysis between G1 and G2 showed a prevalence of posterior subcapsular cataract of 13% and 2% in G1 and G2, respectively (0.0081). Considering physicians only, 38% of G1 and 15% of G2 had cataract, with the prevalence of posterior subcapsular cataract of 13% and 3%, respectively (p = 0.0176). Among non-physicians, no difference was found in the prevalence of cataract (by types). Conclusions: Cataract was more prevalent in professionals exposed to ionizing radiation, with posterior subcapsular cataract the most frequent finding
Morphogenetic characteristics and management of Tanzania grass Características morfogenéticas e manejo de capim-tanzânia
The objective of this work was to evaluate the effect of grazing interval and period of evaluation over tissue turnover in Tanzania grass pastures (Panicum maximum cv. Tanzania) and to ascertain if herbage accumulation rate can be used as a criterion to establish a defoliation schedule for this grass in Southeast of Brazil. A randomized block design with a split-plot arrangement was used. The effect of three grazing intervals was evaluated within seven periods between October 1995 and September 1996. Responses monitored were leaf and stem elongation rates, leaf senescence rate, stem length, and tiller density. Net herbage accumulation rate was calculated using tissue turnover data. The grazing intervals for Tanzania grass should be around 38 days between October and April (spring and early autumn) and 28 days during the reproductive phase of the grass (April/May). Between May and September (late autumn and winter), grazing interval should be around 48 days. Herbage accumulation rate is not a good criterion to establish defoliation time for Tanzania grass. Studies on the effects of stem production in grazing efficiency, animal intake and forage quality are needed to improve Tanzania grass management.<br>O objetivo deste trabalho foi avaliar o efeito do intervalo entre pastejos e do período de avaliação sobre algumas características morfogenéticas do capim-tanzânia (Panicum maximum cv. Tanzânia) e verificar se o manejo desse capim pode ser baseado na taxa de acúmulo de matéria seca na região Sudeste do Brasil. Foi utilizado um delineamento de blocos completos ao acaso com parcelas subdivididas no tempo. Foi avaliado o efeito de três intervalos entre pastejos e de sete períodos de avaliação entre outubro de 1995 e setembro de 1996 sobre: taxa de alongamento foliar e dos colmos, taxa de senescência foliar, altura dos colmos e densidade populacional de perfilhos. Essas informações permitiram determinar a taxa líquida de acúmulo de matéria seca. O capim-tanzânia deve ser pastejado com cerca de 38 dias no período de outubro a abril, com 28 dias na fase reprodutiva (abril/maio) e com aproximadamente 48 dias entre maio e setembro. O intervalo entre pastejos do capim-tanzânia não deve ser estabelecido apenas com base na taxa de acúmulo de matéria seca. São necessários estudos sobre as interações entre produção de hastes e eficiência de pastejo, consumo e qualidade de forragem, a fim de se determinar melhor o manejo desse capim
Chronic total occlusion percutaneous coronary intervention in Latin America
Objectives To report clinical, angiographic characteristics, outcomes, and predictors of unsuccessful procedures in patients who underwent chronic total occlusion (CTO) percutaneous coronary interventions (PCI) in Latin America. Background CTO PCI has been increasingly performed worldwide, but there is a lack of information in this region. Methods An international multicenter registry was developed to collect data on CTO PCI performed in centers in Latin America. Patient, angiographic, procedural and outcome data were evaluated. Predictors of unsuccessful procedures were assessed by multivariable analysis. Results We have included data related to 1,040 CTO PCIs performed in seven countries in Latin America (Argentina, Brazil, Chile, Colombia, Ecuador, Mexico, and Puerto Rico). The mean age was 64 +/- 10 years, and CTO PCI was performed mainly for angina control (81%) or treatment of a large ischemic area (30%). Overall technical success rate was 82.5%, and it was achieved with antegrade wire escalation in 81%, antegrade dissection/re-entry in 8% and with retrograde techniques in 11% of the successful procedures. Multivariable analysis identified moderate/severe calcification, a blunt proximal cap and a previous attempt as independent predictors of unsuccessful procedures. In-hospital major adverse cardiovascular events (MACE) occurred in 3.1% of the cases, death in 1% and cardiac tamponade in 0.9% Conclusions CTO PCI in Latin America has been performed mainly for ischemia relief. Procedures were associated with a success rate above 80% and low incidence of MACE. Predictors of unsuccessful procedures were similar to those previously reported in the literature.Brazilian Society of Interventional Cardiolog
