H19 long noncoding RNA controls the mRNA decay promoting function of KSRP
Long noncoding RNAs (lncRNAs) interact with protein factors to regulate different layers of gene expression transcriptionally or posttranscriptionally. Here we report on the functional consequences of the unanticipated interaction of the RNA binding protein K homology-type splicing regulatory protein (KSRP) with the H19 lncRNA (H19). KSRP directly binds to H19 in the cytoplasm of undifferentiated multipotent mesenchymal C2C12 cells, and this interaction favors KSRP-mediated destabilization of labile transcripts such as myogenin. AKT activation induces KSRP dismissal from H19 and, as a consequence, myogenin mRNA is stabilized while KSRP is repurposed to promote maturation of myogenic microRNAs, thus favoring myogenic differentiation. Our data indicate that H19 operates as a molecular scaffold that facilitates effective association of KSRP with myogenin and other labile transcripts, and we propose that H19 works with KSRP to optimize an AKT-regulated posttranscriptional switch that controls myogenic differentiation
Molecular epidemiology and case-control approaches for management of an outbreak of hepatitis A in Liguria, Italy
Introduction and methods. Hepatitis A remains an important
public health problem in low endemicity areas, because of the
social and economic high burden of cyclical outbreaks. In this
study we described an outbreak of HAV infection occurred in the
city of Genoa and in its proximity and the viral circulation in the
post-epidemic period. In order to identify risk factors associated to
the illness and to determine the source of infection and the dynamics
of virus evolution, we conducted an epidemiological and molecular
investigation by a case-control study and by sequence analysis of
high variable regions of the genome.
Results. From May to October 2005, 58 HAV hepatitis cases were
notified. The case-control study showed that beach establishment
attending is strongly associated with HAV hepatitis (OR = 24.5,
p-value inf. 0.01), at multivariate analysis. The profile of epidemic
curve, the clinical onset of primary cases who occurred in few
weeks and the geographic distribution of cases clearly indicated
a common exposure to a point source: the outbreak can be probably
associated with a contaminated food product dispensed in
the affected area.
The outbreak has been mainly caused by a single variant, confirming
the common exposure to a point source; this variant previously circulated
within homosexual man (MSM) network in Northern Europe.
During the outbreak and in the following months, different variants
originating from Southeast Asia, Southern America and Northern
Africa, have co-circulated: all these cases were related to international
travel and none of these had determined secondary cases.
Discussion. The epidemiological picture of hepatitis A in Liguria is
characterized by a wide heterogeneity of circulating HAV strains.
This pattern could be associated with the increase of imported
cases and transmission within network of persons with similar risk
factors. Molecular approach coupled to descriptive and analytical
epidemiological studies appeared un-replaceable tools for management
and control of HAV outbreaks, because of their capacity
to recognize infection origin, transmission patterns and dynamics
of virus evolution
A posteriori error analysis and adaptive non-intrusive numerical schemes for systems of random conservation laws
In this article we consider one-dimensional random systems of hyperbolic
conservation laws. We first establish existence and uniqueness of random
entropy admissible solutions for initial value problems of conservation laws
which involve random initial data and random flux functions. Based on these
results we present an a posteriori error analysis for a numerical approximation
of the random entropy admissible solution. For the stochastic discretization,
we consider a non-intrusive approach, the Stochastic Collocation method. The
spatio-temporal discretization relies on the Runge--Kutta Discontinuous
Galerkin method. We derive the a posteriori estimator using continuous
reconstructions of the discrete solution. Combined with the relative entropy
stability framework this yields computable error bounds for the entire
space-stochastic discretization error. The estimator admits a splitting into a
stochastic and a deterministic (space-time) part, allowing for a novel
residual-based space-stochastic adaptive mesh refinement algorithm. We conclude
with various numerical examples investigating the scaling properties of the
residuals and illustrating the efficiency of the proposed adaptive algorithm
Displacement power spectrum measurement of a macroscopic optomechanical system at thermal equilibrium
The mirror relative motion of a suspended Fabry-Perot cavity is studied in
the frequency range 3-10 Hz. The experimental measurements presented in this
paper, have been performed at the Low Frequency Facility, a high finesse
optical cavity 1 cm long suspended to a mechanical seismic isolation system
identical to that one used in the VIRGO experiment. The measured relative
displacement power spectrum is compatible with a system at thermal equilibrium
within its environmental. In the frequency region above 3 Hz, where seismic
noise contamination is negligible, the measurement distribution is stationary
and Gaussian, as expected for a system at thermal equilibrium. Through a simple
mechanical model it is shown that: applying the fluctuation dissipation theorem
the measured power spectrum is reproduced below 90 Hz and noise induced by

Nutraceutical approach for the management of cardiovascular risk \u2013 a combination containing the probiotic Bifidobacterium longum BB536 and red yeast rice extract: results from a randomized, double-blind, placebo-controlled study
BACKGROUND:
Probiotics incorporated into dairy products have been shown to reduce total (TC) and LDL cholesterolemia (LDL-C) in subjects with moderate hypercholesterolemia. More specifically, probiotics with high biliary salt hydrolase activity, e.g. Bifidobacterium longum BB536, may decrease TC and LDL-C by lowering intestinal cholesterol reabsorption and, combined with other nutraceuticals, may be useful to manage hypercholesterolemia in subjects with low cardiovascular (CV) risk. This study was conducted to evaluate the efficacy and safety of a nutraceutical combination containing Bifidobacterium longum BB536, red yeast rice (RYR) extract (10\u2009mg/day monacolin K), niacin, coenzyme Q10 (Lactoflorene Colesterolo\uae). The end-points were changes of lipid CV risk markers (LDL-C, TC, non-HDL-cholesterol (HDL-C), triglycerides (TG), apolipoprotein B (ApoB), HDL-C, apolipoprotein AI (ApoAI), lipoprotein(a) (Lp(a), proprotein convertase subtilisin/kexin type 9 (PCSK9)), and of markers of cholesterol synthesis/absorption.
METHODS:
A 12-week randomized, parallel, double-blind, placebo-controlled study. Thirty-three subjects (18-70\u2009years) in primary CV prevention and low CV risk (SCORE: 0-1% in 24 and 2-4% in 9 subjects; LDL-C: 130-200\u2009mg/dL) were randomly allocated to either nutraceutical (N\u2009=\u200916) or placebo (N\u2009=\u200917).
RESULTS:
Twelve-week treatment with the nutraceutical combination, compared to placebo, significantly reduced TC (-\u200916.7%), LDL-C (-\u200925.7%), non-HDL-C (-\u200924%) (all p\u2009<\u20090.0001), apoB (-\u200917%, p\u2009=\u20090.003). TG, HDL-C, apoAI, Lp(a), PCSK9 were unchanged. Lathosterol:TC ratio was significantly reduced by the nutraceutical combination, while campesterol:TC ratio and sitosterol:TC ratio did not change, suggesting reduction of synthesis without increased absorption of cholesterol. No adverse effects and a 97% compliance were observed.
CONCLUSIONS:
A 12-week treatment with a nutraceutical combination containing the probiotic Bifidobacterium longum BB536 and RYR extract significantly improved the atherogenic lipid profile and was well tolerated by low CV risk subjects.

Skeleton based cage generation guided by harmonic fields
International audienceWe propose a novel user-assisted cage generation tool. We start from a digital character and its skeleton, and create a coarse control cage for its animation. Our method requires minimal interaction to select bending points on the skeleton, and computes the corresponding cage automatically. The key contribution is a volumetric field defined in the interior of the character and embedding the skeleton. The integral lines of such field are used to propagate cutting surfaces from the interior of the character to its skin, and allow us to robustly trace non-planar cross sections that adapt to the local shape of the character. Our method overcomes previous approaches that rely on the popular (but tedious and limiting) cutting planes. We validated our software on a variety of digital characters. Our final cages are coarse yet entirely compliant with the structure induced by the underlying skeleton, enriched with the semantics provided by the bending points selected by the user. Automatic placement of bending nodes for a fully automatic caging pipeline is also supported
Sensitivity Studies for Third-Generation Gravitational Wave Observatories
Advanced gravitational wave detectors, currently under construction, are
expected to directly observe gravitational wave signals of astrophysical
origin. The Einstein Telescope, a third-generation gravitational wave detector,
has been proposed in order to fully open up the emerging field of gravitational
wave astronomy. In this article we describe sensitivity models for the Einstein
Telescope and investigate potential limits imposed by fundamental noise
sources. A special focus is set on evaluating the frequency band below 10Hz
where a complex mixture of seismic, gravity gradient, suspension thermal and
radiation pressure noise dominates. We develop the most accurate sensitivity
model, referred to as ET-D, for a third-generation detector so far, including
the most relevant fundamental noise contributions.Comment: 13 pages, 7 picture
Virgo calibration and reconstruction of the gravitational wave strain during VSR1
Virgo is a kilometer-length interferometer for gravitational waves detection
located near Pisa. Its first science run, VSR1, occured from May to October
2007. The aims of the calibration are to measure the detector sensitivity and
to reconstruct the time series of the gravitational wave strain h(t). The
absolute length calibration is based on an original non-linear reconstruction
of the differential arm length variations in free swinging Michelson
configurations. It uses the laser wavelength as length standard. This method is
used to calibrate the frequency dependent response of the Virgo mirror
actuators and derive the detector in-loop response and sensitivity within ~5%.
The principle of the strain reconstruction is highlighted and the h(t)
systematic errors are estimated. A photon calibrator is used to check the sign
of h(t). The reconstructed h(t) during VSR1 is valid from 10 Hz up to 10 kHz
with systematic errors estimated to 6% in amplitude. The phase error is
estimated to be 70 mrad below 1.9 kHz and 6 micro-seconds above.Comment: 8 pages, 8 figures, proceedings of Amaldi 8 conference, to be
