    (In-)Approximability Results for Interval, Resource Restricted, and Low Rank Scheduling

    We consider variants of the restricted assignment problem where a set of jobs has to be assigned to a set of machines, for each job a size and a set of eligible machines is given, and the jobs may only be assigned to eligible machines with the goal of makespan minimization. For the variant with interval restrictions, where the machines can be arranged on a path such that each job is eligible on a subpath, we present the first better than 2-approximation and an improved inapproximability result. In particular, we give a (2-1/24)-approximation and show that no better than 9/8-approximation is possible, unless P=NP. Furthermore, we consider restricted assignment with R resource restrictions and rank D unrelated scheduling. In the former problem, a machine may process a job if it can meet its resource requirements regarding R (renewable) resources. In the latter, the size of a job is dependent on the machine it is assigned to and the corresponding processing time matrix has rank at most D. The problem with interval restrictions includes the 1 resource variant, is encompassed by the 2 resource variant, and regarding approximation the R resource variant is essentially a special case of the rank R+1 problem. We show that no better than 3/2, 8/7, and 3/2-approximation is possible (unless P=NP) for the 3 resource, 2 resource, and rank 3 variant, respectively. Both the approximation result for the interval case and the inapproximability result for the rank 3 variant are solutions to open challenges stated in previous works. Lastly, we also consider the reverse objective, that is, maximizing the minimal load any machine receives, and achieve similar results

    Scheduling with Many Shared Resources

    Consider the many shared resource scheduling problem where jobs have to be scheduled on identical parallel machines with the goal of minimizing the makespan. However, each job needs exactly one additional shared resource in order to be executed and hence prevents the execution of jobs that need the same resource while being processed. Previously a (2m/(m+1))(2m/(m+1))-approximation was the best known result for this problem. Furthermore, a 6/56/5-approximation for the case with only two machines was known as well as a PTAS for the case with a constant number of machines. We present a simple and fast 5/3-approximation and a much more involved but still reasonable 1.5-approximation. Furthermore, we provide a PTAS for the case with only a constant number of machines, which is arguably simpler and faster than the previously known one, as well as a PTAS with resource augmentation for the general case. The approximation schemes make use of the N-fold integer programming machinery, which has found more and more applications in the field of scheduling recently. It is plausible that the latter results can be improved and extended to more general cases. Lastly, we give a 5/4ε5/4 - \varepsilon inapproximability result for the natural problem extension where each job may need up to a constant number (in particular 33) of different resources

    Abnormal P300 in people with high risk of developing psychosis

    Background Individuals with an “at-risk mental state” (or “prodromal” symptoms) have a 20–40% chance of developing psychosis; however it is difficult to predict which of them will become ill on the basis of their clinical symptoms alone. We examined whether neurophysiological markers could help to identify those who are particularly vulnerable. Method 35 cases meeting PACE criteria for the at-risk mental state (ARMS) and 57 controls performed an auditory oddball task whilst their electroencephalogram was recorded. The latency and amplitude of the P300 and N100 waves were compared between groups using linear regression. Results The P300 amplitude was significantly reduced in the ARMS group [8.6 ± 6.4 microvolt] compared to controls [12.7 ± 5.8 microvolt] (p < 0.01). There were no group differences in P300 latency or in the amplitude and latency of the N100. Of the at-risk subjects that were followed up, seven (21%) developed psychosis. Conclusion Reduction in the amplitude of the P300 is associated with an increased vulnerability to psychosis. Neurophysiological and other biological markers may be of use to predict clinical outcomes in populations at high risk

    Fully-Featured Anonymous Credentials with Reputation System

    We present CLARC\mathsf{CLARC} (Cryptographic Library for Anonymous Reputation and Credentials), an anonymous credentials system (ACS) combined with an anonymous reputation system. Using CLARC\mathsf{CLARC}, users can receive attribute-based credentials from issuers. They can efficiently prove that their credentials satisfy complex (access) policies in a privacy-preserving way. This implements anonymous access control with complex policies. Furthermore, CLARC\mathsf{CLARC} is the first ACS that is combined with an anonymous reputation system where users can anonymously rate services. A user who gets access to a service via a credential, also anonymously receives a review token to rate the service. If a user creates more than a single rating, this can be detected by anyone, preventing users from spamming ratings to sway public opinion. To evaluate feasibility of our construction, we present an open-source prototype implementation

    Alternative splicing of Tcf7l2 transcripts generates protein variants with differential promoter-binding and transcriptional activation properties at Wnt/β-catenin targets

    Alternative splicing can produce multiple protein products with variable domain composition from a single gene. The mouse Tcf7l2 gene is subject to alternative splicing. It encodes TCF4, a member of the T-cell factor (TCF) family of DNA-binding proteins and a nuclear interaction partner of β-catenin which performs essential functions in Wnt growth factor signalling. Multiple TCF4 isoforms, potentially exhibiting cell-type-specific distribution and differing in gene regulatory properties, could strongly influence tissue-specific Wnt responses. Therefore, we have examined mouse Tcf7l2 splice variants in neonatal tissues, embryonic stem cells and neural progenitors. By polymerase chain reaction amplification, cloning and sequencing, we identify a large number of alternatively spliced transcripts and report a highly flexible combinatorial repertoire of alternative exons. Many, but not all of the variants exhibit a broad tissue distribution. Moreover, two functionally equivalent versions of the C-clamp, thought to represent an auxiliary DNA-binding domain, were identified. Depending upon promoter context and precise domain composition, TCF4 isoforms exhibit strikingly different transactivation potentials at natural Wnt/β-catenin target promoters. However, differences in C-clamp-mediated DNA binding can only partially explain functional differences among TCF4 variants. Still, the cell-type-specific complement of TCF4 isoforms is likely to be a major determinant for the context-dependent transcriptional output of Wnt/β-catenin signalling

    Exploring Predictors of Outcome in the Psychosis Prodrome: Implications for Early Identification and Intervention

    Functional disability is a key component of many psychiatric illnesses, particularly schizophrenia. Impairments in social and role functioning are linked to cognitive deficits, a core feature of psychosis. Retrospective analyses demonstrate that substantial functional decline precedes the onset of psychosis. Recent investigations reveal that individuals at clinical-high-risk (CHR) for psychosis show impairments in social relationships, work/school functioning and daily living skills. CHR youth also demonstrate a pattern of impairment across a range of cognitive domains, including social cognition, which is qualitatively similar to that of individuals with schizophrenia. While many studies have sought to elucidate predictors of clinical deterioration, specifically the development of schizophrenia, in such CHR samples, few have investigated factors relevant to psychosocial outcome. This review integrates recent findings regarding cognitive and social-cognitive predictors of outcome in CHR individuals, and proposes potential directions for future research that will contribute to targeted interventions and improved outcome for at-risk youth

    Trophic macrophages in development and disease

    Specialized phagocytes are found in the most primitive multicellular organisms. Their roles in homeostasis and in distinguishing self from non-self have evolved with the complexity of organisms and their immune systems. Equally important, but often overlooked, are the roles of macrophages in tissue development. As discussed in this Review, these include functions in branching morphogenesis, neuronal patterning, angiogenesis, bone morphogenesis and the generation of adipose tissue. In each case, macrophage depletion impairs the formation of the tissue and compromises its function. I argue that in several diseases, the unrestrained acquisition of these developmental macrophage functions exacerbates pathology. For example, macrophages enhance tumour progression and metastasis by affecting tumour-cell migration and invasion, as well as angiogenesis
