Decreased activity of inducible nitric oxide synthase type 2 and modulation of the expression of glutathione S-transferase alpha, bcl-2, and metallothioneins during the differentiation of CaCo-2 cells.

Reactive oxygen species modulate the cell growth of a wide variety of mammalian cells. To determine whether oxidative metabolism is altered during the differentiation process, we studied the expression of pro- and antioxidant proteins in proliferating and differentiated CaCo-2 cells, a human colon adenocarcinoma cell line. Nitric oxide synthase type 2 (iNOS) produces nitric oxide (NO). Depending on its rate of synthesis, NO may either promote cellular and DNA damage or reduce the ability of other free radicals to induce cell injury. Using Western and Northern blot analysis and arginine conversion assay, we demonstrate that the expression of iNOS decreases when cells undergo differentiation. This biological event entails a diminished production of NO metabolites and correlates with the loss of activation of soluble guanylate cyclase activity. In differentiated cells, a 2-fold down-regulation of the nuclear factor kappa B activity was observed, suggesting that nuclear factor kappa B could be one of the iNOS gene regulatory factors in the CaCo-2 model. In parallel, we studied the expression of other antioxidant proteins including glutathione S-transferase alpha (GST alpha), bcl-2, and the metallothioneins (MTs). We show that the protein levels of GST alpha and MT increase during the differentiation of CaCo-2 cells, whereas bcl-2 levels decrease. Our investigation indicates that the expression of iNOS, GST alpha, bcl-2, and MT is associated with the enterocytic differentiation. The shift in the expression of specific antioxidant genes during CaCo-2 cell differentiation may occur to avoid alterations in the cell redox potential

Production of individual marine organic aggregates using paramagnetic microspheres : a new tool for examining microbial associations with aggregates

We describe a new method to produce marine aggregates from natural organic material based on the sticking properties of transparent exopolymeric particles. Seawater samples were prescreened and ultrafiltered to concentrate the 30 kDa to 10 mu m size fraction. First, we produced small magnetizable aggregates by combining glass microfibers and paramagnetic 1-mu m beads with the organic matter present in the concentrated solution. The second step involved clustering the small aggregates into a single macro-aggregate, using a small ring-shaped magnet as an aggregation nucleus. Viral and bacterial densities, determined after dissolution of the newly formed aggregates with methanol, averaged 13.8 x 10(6) +/- 3.6 x 10(6) vir. agg.(-1) and 4.1 x 10(6) +/- 1.1 x10(6) bact. agg.(-1). Bacterial respiration and production measurements of single aggregates averaged 8.47 +/- 1.72 nmol O-2 agg.(-1) h(-1) and 1.54 +/- 0.45 ng C agg.(-1) h(-1), respectively. Particulate organic carbon and nitrogen content of the newly formed macro-aggregates averaged 31.92 +/- 2.67 mu g C agg.(-1) and 3.44 +/- 0.43 mu g N agg.(-1), respectively. This approach allows the concentration and isolation of the organic matter precursors that compose natural aggregates and provides a simple protocol for recombining those precursors into single newly formed macro-aggregates, which can then be easily manipulated for further investigation. This method is a new tool for investigations into the interactions between microorganisms and marine aggregates and their implications at the ecosystem level, but also into the interactions between aggregates and dissolved organic or inorganic substances

The article is aimed to propose a simulating model of the creation of the innovational regional center to examine the advantages and innovational possibilities in the regional economy. The popular software “I think” which has become widely used in modeling different similar economic processes, is used for the development of the simulation model. This gives the possibility to take into consideration various factors of influence on these processes and calculate their consequences with the option of their gradual correction and working out the models in the real practice

Do the levels of industrial pollutants influence the distribution and abundance of dinoflagellate cysts in the recently-deposited sediment of a Mediterranean coastal ecosystem?

We studied the relationships between sediment industrial pollutants concentrations, sediment characteristics and the dinoflagellate cyst abundance within a coastal lagoon by investigating a total of 55 sampling stations within the Bizerte lagoon, a highly anthropized Mediterranean ecosystem. The sediment of Bizerte lagoon is char- acterized by a high dinocyst abundance, reaching a maximum value of 2742 cysts·g−1 of dry sediment. The investigated cyst diversity was characterized by the presence of 22 dominant dinocyst morphotypes belonging to 11 genera. Two dinoflagellate species dominated the assemblage: Alexandrium pseudogonyaulax and Protoperidinium claudicans, representing 29 to 89% and 5 to 38% of the total cyst abundance, respectively, depending on the station. Seven morphotypes belonging to potentially toxic species were detected, including Alexandrium minutum, A. pseudogonyaulax, Alexandrium catenella/tamarense species complex, Lingulodinium polyedrum, Gonyaulax cf. spinifera complex, Prorocentrum micans and Protoceratium reticulatum. Pearson correlation values showed a positive correlation (α = 0.05) between cyst abundance and both water content and fine silt sediment content. Clustering revealed that the highest abundance of cysts corresponds to stations presenting the higher amounts of heavy metals. The simultaneous autoregressive model (SAM) highlighted a significant correlation (α = 0.05) between cyst accumulation and two main factors: sediment water content and sediment content for several heavy metals, including Hg, Cd, Cu, Ni and Cr. These results suggest that the degree of heavy metal pollution could influence cyst accumulation patterns.peer-reviewe
