    Infrared chemical imaging through nondegenerate two-photon absorption in silicon-based cameras

    Chemical imaging based on mid-infrared (MIR) spectroscopic contrast is an important technique with a myriad of applications, including biomedical imaging and environmental monitoring. Current MIR cameras, however, lack in performance and are much less affordable compared to mature Si-based devices, which operate in the visible and near-infrared. Here we demonstrate fast MIR chemical imaging through non-degenerate two-photon absorption (NTA) in a standard Si-based charge-coupled device (CCD). We show that wide-field MIR images can be obtained at 100 ms exposure times using picosecond pulse energies of only a few fJ per pixel through NTA directly on the CCD chip. Because this on-chip approach does not rely on phase-matching, it is alignment-free and does not necessitate complex post-processing of the images. We emphasize the utility of this technique through chemically selective MIR imaging of polymers and biological samples, including MIR videos of moving targets, physical processes and live nematodes

    Amphidromy Links a Newly Documented Fish Community of Continental Australian Streams, to Oceanic Islands of the West Pacific

    BACKGROUND: Indo-Pacific high island streams experience extreme hydrological variation, and are characterised by freshwater fish species with an amphidromous life history. Amphidromy is a likely adaptation for colonisation of island streams following stochastic events that lead to local extirpation. In the Wet Tropics of north-eastern Australia, steep coastal mountain streams share similar physical characteristics to island systems. These streams are poorly surveyed, but may provide suitable habitat for amphidromous species. However, due to their ephemeral nature, common non-diadromous freshwater species of continental Australia are unlikely to persist. Consequently, we hypothesise that coastal Wet Tropics streams are faunally more similar, to distant Pacific island communities, than to nearby faunas of large continental rivers. METHODS/PRINCIPAL FINDINGS: Surveys of coastal Wet Tropics streams recorded 26 species, 10 of which are first records for Australia, with three species undescribed. This fish community is unique in an Australian context in that it contains mostly amphidromous species, including sicydiine gobies of the genera Sicyopterus, Sicyopus, Smilosicyopus and Stiphodon. Species presence/absence data of coastal Wet Tropics streams were compared to both Wet Tropics river networks and Pacific island faunas. ANOSIM indicated the fish fauna of north-eastern Australian coastal streams were more similar to distant Pacific islands (R = 0.76), than to nearby continental rivers (R = 0.98). MAIN CONCLUSIONS/SIGNIFICANCE: Coastal Wet Tropics streams are faunally more similar to distant Pacific islands (79% of species shared), than to nearby continental fauna due to two factors. First, coastal Wet Tropics streams lack many non-diadromous freshwater fish which are common in nearby large rivers. Second, many amphidromous species found in coastal Wet Tropics streams and Indo-Pacific islands remain absent from large rivers of the Wet Tropics. The evolutionary and conservation significance of this newly discovered Australian fauna requires clarification in the context of the wider amphidromous fish community of the Pacific

    Amino Acid Repeats Cause Extraordinary Coding Sequence Variation in the Social Amoeba Dictyostelium discoideum

    Protein sequences are normally the most conserved elements of genomes owing to purifying selection to maintain their functions. We document an extraordinary amount of within-species protein sequence variation in the model eukaryote Dictyostelium discoideum stemming from triplet DNA repeats coding for long strings of single amino acids. D. discoideum has a very large number of such strings, many of which are polyglutamine repeats, the same sequence that causes various human neurological disorders in humans, like Huntington's disease. We show here that D. discoideum coding repeat loci are highly variable among individuals, making D. discoideum a candidate for the most variable proteome. The coding repeat loci are not significantly less variable than similar non-coding triplet repeats. This pattern is consistent with these amino-acid repeats being largely non-functional sequences evolving primarily by mutation and drift

    Costs of formal and informal care at home for people with dementia: ‘expert panel’ opinions from staff and informal carers

    Effective home support in dementia is key in delaying nursing home admission. However, home support is frequently not tailored to the individual needs of people with dementia. Staff allocating home support services may not identify important care needs, which only be recognised by informal carers. The purpose of this study was to explore the balance of informal and formal home support and their associated costs from the perspectives of both informal carers and paid staff. Five case vignettes of people with dementia were designed based on an existing English data set from a European study into transition into long-term care (the RightTimePlaceCare programme), representing 42 per cent of the English sample. In total, 14 informal carers and 14 paid staff were consulted in separate groups, as expert panels, regarding their recommendations for home care services for each vignette. Care recommendations of carers and staff were costed based on nationally available unit costs and compared. Informal carers allocated fewer hours of care than staff. Personal and domestic home care and day care centres were the most frequently recommended formal services by both groups, and some vignettes of people with dementia were recommended for care home admission. The ratio of costs of informal versus formal support was relatively equal for paid staff, yet unbalanced from the perspectives of informal carers with a greater proportion of formal care costs. Recommendations from this study can help shape dementia care to be more tailored to the individual needs of people with dementia and their carers

    Zwicky Transient Facility constraints on the optical emission from the nearby repeating FRB 180916.J0158+65

    The discovery rate of fast radio bursts (FRBs) is increasing dramatically thanks to new radio facilities. Meanwhile, wide-field instruments such as the 47 deg2^2 Zwicky Transient Facility (ZTF) survey the optical sky to study transient and variable sources. We present serendipitous ZTF observations of the CHIME repeating source FRB 180916.J0158+65, that was localized to a spiral galaxy 149 Mpc away and is the first FRB suggesting periodic modulation in its activity. While 147 ZTF exposures corresponded to expected high-activity periods of this FRB, no single ZTF exposure was at the same time as a CHIME detection. No >3σ>3\sigma optical source was found at the FRB location in 683 ZTF exposures, totalling 5.69 hours of integration time. We combined ZTF upper limits and expected repetitions from FRB 180916.J0158+65 in a statistical framework using a Weibull distribution, agnostic of periodic modulation priors. The analysis yielded a constraint on the ratio between the optical and radio fluences of η≲200\eta \lesssim 200, corresponding to an optical energy Eopt≲3×1046E_{\rm opt} \lesssim 3 \times 10^{46} erg for a fiducial 10 Jy ms FRB (90% confidence). A deeper (but less statistically robust) constraint of η≲3\eta \lesssim 3 can be placed assuming a rate of r(>5r(>5 Jy ms)= hr−1^{-1} and 1.2±1.11.2\pm 1.1 FRB occurring during exposures taken in high-activity windows. The constraint can be improved with shorter per-image exposures and longer integration time, or observing FRBs at higher Galactic latitudes. This work demonstrated how current surveys can statistically constrain multi-wavelength counterparts to FRBs even without deliberately scheduled simultaneous radio observation.Comment: Accepted for publication in ApJL, 9 pages, 4 figures, 1 tabl

    Costs of reproduction are present but latent in eusocial bumblebee queens

    Background: The standard evolutionary theory of ageing proposes that ageing occurs because of a trade-off between reproduction and longevity. Eusocial insect queens exhibit positive fecundity-longevity associations and so have been suggested to be counter-examples through not expressing costs of reproduction and through remodelling conserved genetic and endocrine networks regulating ageing and reproduction. If so, eusocial evolution from solitary ancestors with negative fecundity-longevity associations must have involved a stage at which costs of reproduction were suppressed and fecundity and longevity became positively associated. Using the bumblebee (Bombus terrestris), we experimentally tested whether queens in annual eusocial insects at an intermediate level of eusocial complexity experience costs of reproduction, and, using mRNA-seq, the extent to which they exhibit a remodelling of relevant genetic and endocrine networks. Specifically, we tested whether costs of reproduction are present but latent, or whether a remodelling of relevant genetic and endocrine networks has already occurred allowing queens to reproduce without costs. Results: We experimentally increased queens’ costs of reproduction by removing their eggs, which caused queens to increase their egg-laying rate. Treatment queens had significantly reduced longevity relative to control queens whose egg-laying rate was not increased. Reduced longevity in treatment queens was not caused by increased worker-to-queen aggression or by increased overall activity in queens. In addition, treatment and control queens differed in age-related gene expression based on mRNA-seq in both their overall expression profiles and the expression of ageing-related genes. Remarkably, these differences appeared to occur principally with respect to relative age, not chronological age. Conclusions: This study represents the first simultaneously phenotypic and transcriptomic experimental test for a longevity cost of reproduction in eusocial insect queens. The results support the occurrence of costs of reproduction in annual eusocial insects of intermediate social complexity and suggest that reproductive costs are present but latent in queens of such species, i.e. that these queens exhibit condition-dependent positive fecundity-longevity associations. They also raise the possibility that a partial remodelling of genetic and endocrine networks underpinning ageing may have occurred in intermediately eusocial species such that, in unmanipulated conditions, age-related gene expression depends more on chronological than relative age

    Developmental diet alters the fecundity-longevity relationship and age-related gene expression in Drosophila melanogaster

    The standard evolutionary theory of aging predicts a negative relationship (trade-off) between fecundity and longevity. However, in principle, the fecundity-longevity relationship can become positive in populations in which individuals have unequal resources. Positive fecundity-longevity relationships also occur in queens of eusocial insects such as ants and bees. Developmental diet is likely to be central to determining trade-offs as it affects key fitness traits, but its exact role remains uncertain. For example, in Drosophila melanogaster, changes in adult diet can affect fecundity, longevity, and gene expression throughout life, but it is unknown how changes in developmental (larval) diet affect fecundity-longevity relationships and gene expression in adults. Using D. melanogaster, we tested the hypothesis that varying developmental diet alters the directionality of fecundity-longevity relationships in adults, and characterised associated gene expression changes. We reared larvae on low (20%), medium (100%), and high (120%) yeast diets, and transferred adult females to a common diet. We measured fecundity and longevity of individual adult females and profiled gene expression changes with age. Adult females raised on different larval diets exhibited fecundity-longevity relationships that varied from significantly positive to significantly negative, despite minimal differences in mean life-time fertility or longevity. Treatments also differed in age-related gene expression, including for aging-related genes. Hence the sign of fecundity-longevity relationships in adult insects can be altered and even reversed by changes in larval diet quality. By extension, larval diet differences may represent a key mechanistic factor underpinning positive fecundity-longevity relationships observed in species such as eusocial insects

    Towards Efficient Detection of Small Near-Earth Asteroids Using the Zwicky Transient Facility (ZTF)

    We describe ZStreak, a semi-real-time pipeline specialized in detecting small, fast-moving near-Earth asteroids (NEAs) that is currently operating on the data from the newly-commissioned Zwicky Transient Facility (ZTF) survey. Based on a prototype originally developed by Waszczak et al. (2017) for the Palomar Transient Factory (PTF), the predecessor of ZTF, ZStreak features an improved machine-learning model that can cope with the 10×10\times data rate increment between PTF and ZTF. Since its first discovery on 2018 February 5 (2018 CL), ZTF/ZStreak has discovered 4545 confirmed new NEAs over a total of 232 observable nights until 2018 December 31. Most of the discoveries are small NEAs, with diameters less than ∼100\sim100 m. By analyzing the discovery circumstances, we find that objects having the first to last detection time interval under 2 hr are at risk of being lost. We will further improve real-time follow-up capabilities, and work on suppressing false positives using deep learning.Comment: PASP in pres
