    ANN Based Virtual Classification Model for Discriminating Active and Inactive Withanolide E Analogs against Human Breast Cancer Cell Line MCF-7

    Withanolides are a group of natural C-28 steroids built on an ergostane skeleton and classified into two major groups according to their structural skeleton: (a) compounds with a beta-oriented side chain and (b) compounds with an alpha-oriented side chain. Withanolide E represents one of the members of the latter group. Classification of active compounds on the basis of pharmacophore against specific cancer cell line poses a serious concern at the primary stage of virtual screening. To overcome this problem we have developed an artificial neural network based virtual screening model for discriminating active and non-active Withanolide-E-like derivatives or analogs against human breast cancer cell line MCF-7. In the present work, a 2D chemical descriptors ensemble pharmacophore has been modelled on the basis of withanolide E structural featured molecules. The ANN structure activity based classification model could be useful for identification of active withanolide analogs as anticancer leads against MCF-7. This model can be used for predicting possible growth inhibitory concentration (logGI50) against breast cancer cell line MCF-7.     Soil quality of Bhilwara District (Rajasthan) in relation to pisciculture

    The present paper deals with the physico-chemical data of 13 rainfed and drainable dry bundhs of Bhilwara District (India). All the dry bundhs studied showed a slightly alkaline pH ranging from 7.5 to 8.5. Electrical conductivity ranged from 2 to 8 millimhos/cm. Organic carbon in sediment fluctuated from 0.30 to 0.75%. Nitrogen, phosphate and potassium levels were fairly good, at 30 to 50 and 24 to 36 mg/100 g of soil respectively. Based on these data it was inferred that these dry bundhs were highly productive and suitable for freshwater fish culture

    Management of sheath blight in rice through application of Validamycin, Trichoderma harzianum and Pseudomonas fluorescence

    Eleven different combinations of Trichoderma harzianum, Psedumonas fluorescens and Validamycin 3L, were applied at different crop stages viz. seed, seedling, tillering and symptoms initiation along with one application of Carbendazim as standard control to test their efficacy against sheath blight of rice caused by Rhizoctonia solani under field condition. One application of Carbendazim @ 0.1% at the time of symptoms initiation (with 81.36 percent reduction of disease incidence, 83.17 percent reduction in disease severity and 62.92 percent inhancement in yield) and seed treatment with Trichoderma harzianum and seedling root dipping with Pseudomonas fluorescens and two spraying of Validamycin i.e. at tillering and symptoms initiation stages (with 69.10 percent reduction of disease incidence,83.17 percent reduction in disease severity and 62.49 percent inhancement in yield) were found to be equally and highly effective in reducing the sheath blight severity and in increasing rice grain yield also. One spraying of Validamycin at symptoms initiation stage was comparatively less effective (29.54 per cent reduction in disease incidence,53.46 per cent reduction in disease severity and 47.76 per cent enhancement in yield ) than one spraying of Carbendazim at symptoms initiation stage

    Effects of Deep Cryogenic Treatment on Wear Resistance & Life of Nickel Coated Flywheel

    The influence of deep cryogenic treatment on wear resistance and life of nickel coated flywheel was studied. Nickel was deposited on to the flywheel workpiece by plating method. The nickel coated pieces were tested at various loads and speeds on a pin-on-disc apparatus, different loading conditions were used in order to compare the wear rate. The pin-on-disc gave ample results on the wear rate of the coating. Then the coated pieces were cryogenically treated at -196? at various time interval of 8,16 & 24 hours with constant supply of liquid nitrogen. Later the cryogenic treated specimen was tested for wear rates to compare the wear rate results to ultimately find the wear before and after cryogenic treatment. The microstructures of cryogenic and conventionally treated samples were analyzed.     Chiral restoration effects on the shear viscosity of a pion gas

    We investigate the shear viscosity of a pion gas in relativistic kinetic theory, using the Nambu-Jona-Lasinio model to construct the pion mass and the pi-pi interaction at finite temperature. Whereas at low temperatures the scattering properties and, hence, the viscosity are in agreement with lowest-order chiral perturbation theory, we find strong medium modifications in the crossover region. Here the system is strongly coupled and the scattering lengths diverge, similarly as for ultra-cold Fermi gases at a Feshbach resonance. As a consequence, the ratio eta/s is found to be strongly reduced as compared to calculations without medium-modified masses and scattering amplitudes.     Salt reduction is important for reducing hypertension and the risk of cardiovascular events, nevertheless worldwide salt intakes are above recommendations. Consequently strategies to reduce intake are required, however these require an understanding of salt intake behaviours to be effective. As limited information is available on this, an international study was conducted to derive knowledge on salt intake and associated behaviours in the general population. An online cohort was recruited consisting of a representative sample from Germany, Austria, United States of America, Hungary, India, China, South Africa, and Brazil (n=6987; aged 18-65. years; age and gender stratified). Participants completed a comprehensive web-based questionnaire on salt intake and associated behaviours. While salt reduction was seen to be healthy and important, over one third of participants were not interested in salt reduction and the majority were unaware of recommendations. Salt intake was largely underestimated and people were unaware of the main dietary sources of salt. Participants saw themselves as mainly responsible for their salt intake, but also acknowledged the roles of others. Additionally, they wanted to learn more about why salt was bad for health and what the main sources in the diet were. As such, strategies to reduce salt intake must raise interest in engaging in salt reduction through improving understanding of intake levels and dietary sources of salt. Moreover, while some aspects of salt reduction can be globally implemented, local tailoring is required to match level of interest in salt reduction. These findings provide unique insights into issues surrounding salt reduction and should be used to develop effective salt reduction strategies and/or policies. © 2013 The Authors

    Human and murine fibroblast single cell transcriptomics reveals fibroblast clusters are differentially affected by ageing, and serum cholesterol

    Aims Specific fibroblast markers and in-depth heterogeneity analysis are currently lacking, hindering functional studies in cardiovascular diseases (CVDs). Here, we established cell-type markers and heterogeneity in murine and human arteries and studied the adventitial fibroblast response to CVD and its risk factors hypercholesterolaemia and ageing. Methods and results Murine aorta single-cell RNA-sequencing analysis of adventitial mesenchymal cells identified fibroblast-specific markers. Immunohistochemistry and flow cytometry validated platelet-derived growth factor receptor alpha (PDGFRA) and dipeptidase 1 (DPEP1) across human and murine aorta, carotid, and femoral arteries, whereas traditional markers such as the cluster of differentiation (CD)90 and vimentin also marked transgelin+ vascular smooth muscle cells. Next, pseudotime analysis showed multiple fibroblast clusters differentiating along trajectories. Three trajectories, marked by CD55 (Cd55+), Cxcl chemokine 14 (Cxcl14+), and lysyl oxidase (Lox+), were reproduced in an independent RNA-seq dataset. Gene ontology (GO) analysis showed divergent functional profiles of the three trajectories, related to vascular development, antigen presentation, and/or collagen fibril organization, respectively. Trajectory-specific genes included significantly more genes with known genome-wide associations (GWAS) to CVD than expected by chance, implying a role in CVD. Indeed, differential regulation of fibroblast clusters by CVD risk factors was shown in the adventitia of aged C57BL/6J mice, and mildly hypercholesterolaemic LDLR KO mice on chow by flow cytometry. The expansion of collagen-related CXCL14+ and LOX+ fibroblasts in aged and hypercholesterolaemic aortic adventitia, respectively, coincided with increased adventitial collagen. Immunohistochemistry, bulk, and single-cell transcriptomics of human carotid and aorta specimens emphasized translational value as CD55+, CXCL14+ and LOX+ fibroblasts were observed in healthy and atherosclerotic specimens. Also, trajectory-specific gene sets are differentially correlated with human atherosclerotic plaque traits. Conclusion We provide two adventitial fibroblast-specific markers, PDGFRA and DPEP1, and demonstrate fibroblast heterogeneity in health and CVD in humans and mice. Biological relevance is evident from the regulation of fibroblast clusters by age and hypercholesterolaemia in vivo, associations with human atherosclerotic plaque traits, and enrichment of genes with a GWAS for CVD
