    Victorian index of estuary condition : recommended themes and measures

    Deakin University was engaged by the Department of Sustainability and Environment to develop an Index of Estuary Condition (IEC) for evaluating the environmental condition of Victorian estuaries. The Index will ultimately complement the existing Index of Stream Condition (ISC) by providing a consistent statewide assessment of the environmental condition of estuaries. This will better enable:&bull; Estuarine condition to be reported at regional, state and national levels.&bull; Prioritisation of resource allocation.&bull; Strategic evaluation of management interventions in estuaries.Workshops involving participants with expertise in a variety of disciplines were convened to integrate learnings from assessment programs currently being developed interstate and overseas. This report synthesises and builds on the output from those workshops which: &bull; Identified key components (themes) of estuaries that contribute to estuarine condition. &bull; Contributed to development of a broad conceptual model for Victorian estuaries &bull; Identified possible measures of each themeIn keeping with the sub-indices of the ISC, six themes were identified for use in the IEC: Physical form, Hydrology, Water quality, Sediment, Flora and Fauna. Several measures within each theme are recommended to assess estuary condition.Implementation of particular measures in the IEC partly depends on the investment required to both collect and interpret the required data. With regard to data collection, each measure was assessed according to whether there is an established sampling procedure, how frequently data need to be collected and the level of expertise required for collection and processing. For interpreting the data, measures were scored on whether baseline condition is established and whether descriptions and scores are developed which reflect the extent of deviation from that condition. These scores were used to indicate which measures are feasible to implement immediately and which require further investigation. A trial of the recommended IEC measures in a selection of estuaries is recommended as it would provide an opportunity to test the measures and their suitability for application statewide. It is suggested that the trial is conducted in estuaries subject to various levels of threats within each of the four estuary classes described by Barton et al. (2008)<br /

    Estuary environmental flows assessment methodology for Victoria

    This report sets out a method to determine the environmental water requirements of estuaries in Victoria. The estuary environmental flows assessment method (EEFAM) is a standard methodology which can be applied consistently across Victorian estuaries.The primary objective of EEFAM is to define a flow regime to maintain or enhance the ecological health of an estuary. The method is used to inform Victorian water resource planning processes.The output of EEFAM is a recommended flow regime for estuaries. This recommendation is developed from the known dependence of the estuary&rsquo;s flora, fauna, biogeochemical and geomorphological features on the flow regime. EEFAM is an evidence-based methodology. This bottom-up or &lsquo;building block&rsquo; approach conforms to the asset-based approach of the Victorian River Health Strategy and regional river health strategies.EEFAM is based on and expands on FLOWS, the Victorian method for determining environmental water requirements in rivers. The list of tasks has been modified and re-ordered in EEFAM to reflect environmental and management issues specific to estuaries. EEFAM and FLOWS can be appliedsimultaneously to a river and its estuary as part of a whole-of-system approach to environmental flow requirements. Like the FLOWS method, EEFAM is modular, and additional components can be readily incorporated

    Enhanced Cellular Immunity in Shrimp (Litopenaeus vannamei) after ‘Vaccination’

    It has long been viewed that invertebrates rely exclusively upon a wide variety of innate mechanisms for protection from disease and parasite invasion and lack any specific acquired immune mechanisms comparable to those of vertebrates. Recent findings, however, suggest certain invertebrates may be able to mount some form of specific immunity, termed ‘specific immune priming’, although the mechanism of this is not fully understood (see Textbox S1). In our initial experiments, either formalin-inactivated Vibrio harveyi or sterile saline were injected into the main body cavity (haemocoel) of juvenile shrimp (Litopenaeus vannamei). Haemocytes (blood cells) from V. harveyi-injected shrimp were collected 7 days later and incubated with a 1∶1 mix of V. harveyi and an unrelated Gram positive bacterium, Bacillus subtilis. Haemocytes from ‘vaccinated’ shrimp showed elevated levels of phagocytosis of V. harveyi, but not B. subtilis, compared with those from saline-injected (non-immunised) animals. The increased phagocytic activity was characterised by a significant increase in the percentage of phagocytic cells. When shrimp were injected with B. subtilis rather than vibrio, there was no significant increase in the phagocytic activity of haemocytes from these animals in comparison to the non-immunised (saline injected) controls. Whole haemolymph (blood) from either ‘immunised’ or non-immunised’ shrimp was shown to display innate humoral antibacterial activity against V. harveyi that was absent against B. subtilis. However, there was no difference in the potency of antibacterial activity between V. harveyi-injected shrimp and control (saline injected) animals showing that ‘vaccination’ has no effect on this component of the shrimp's immune system. These results imply that the cellular immune system of shrimp, particularly phagocytosis, is capable of a degree of specificity and shows the phenomenon of ‘immune priming’ reported by other workers. However, in agreement with other studies, this phenomenon is not universal to all potential pathogens

    Albedo and Reflection Spectra of Extrasolar Giant Planets

    We generate theoretical albedo and reflection spectra for a full range of extrasolar giant planet (EGP) models, from Jovian to 51-Pegasi class objects. Our albedo modeling utilizes the latest atomic and molecular cross sections, a Mie theory treatment of extinction by condensates, a variety of particle size distributions, and an extension of the Feautrier radiative transfer method which allows for a general treatment of the scattering phase function. We find that due to qualitative similarities in the compositions and spectra of objects within each of five broad effective temperature ranges, it is natural to establish five representative EGP albedo classes: a ``Jovian'' class (Teff≲150_{\rm eff} \lesssim 150 K; Class I) with tropospheric ammonia clouds, a ``water cloud'' class (Teff∼250_{\rm eff} \sim 250 K; Class II) primarily affected by condensed H2_2O, a ``clear'' class (Teff≳350_{\rm eff} \gtrsim 350 K; Class III) which lacks clouds, and two high-temperature classes: Class IV (900 K ≲\lesssim Teff_{\rm{eff}} ≲\lesssim 1500 K) for which alkali metal absorption predominates, and Class V (Teff_{\rm{eff}} ≳\gtrsim 1500 K and/or low surface gravity (≲\lesssim 103^3 cm s−2^{-2})) for which a high silicate layer shields a significant fraction of the incident radiation from alkali metal and molecular absorption. The resonance lines of sodium and potassium are expected to be salient features in the reflection spectra of Class III, IV, and V objects. We derive Bond albedos and effective temperatures for the full set of known EGPs and explore the possible effects of non-equilibrium condensed products of photolysis above or within principal cloud decks.     Targeted cell imaging properties of a deep red luminescent iridium(III) complex conjugated with a c-Myc signal peptide

    A nuclear localisation sequence (NLS) peptide, PAAKRVKLD, derived from the human c-Myc regulator gene, has been functionalised with a long wavelength (λex = 550 nm; λem = 677 nm) cyclometalated organometallic iridium(III) complex to give the conjugate Ir-CMYC. Confocal fluorescence microscopy studies on human fibroblast cells imaged after 18–24 h incubation show that Ir-CMYC concentrations of 80–100 μM promote good cell uptake and nuclear localisation, which was confirmed though co-localisation studies using Hoechst 33342. In comparison, a structurally related, photophysically analogous iridium(III) complex lacking the peptide sequence, Ir-PYR, showed very different biological behaviour, with no evidence of nuclear, lysosomal or autophagic vesicle localisation and significantly increased toxicity to the cells at concentrations >10 μM that induced mitochondrial dysfunction. Supporting UV-visible and circular dichroism spectroscopic studies show that Ir-PYR and Ir-CMYC display similarly low affinities for DNA (ca. 103 M−1), consistent with electrostatic binding. Therefore the translocation and nuclear uptake properties of Ir-CMYC are attributed to the presence of the PAAKRVKLD nuclear localisation sequence in this complex

    Synthetic routes, characterization and photophysical properties of luminescent, surface functionalized nanodiamonds

    The functionalization of small diameter (ca. 50 nm) polycarboxylated nanodiamond particles using amide coupling methodologies in both water and acetonitrile solvent has been investigated. In this manner, the surfaces of nanodiamond particles were adorned with different luminescent moieties, including a green fluorescent 1,8-naphthalimide species (Nap-1), and a red emitting ruthenium(II) tris-bipyridine complex (Ru-1), as well as dual functionalization with both luminophores. Comprehensive characterization of the surface functionalized nanodiamonds has been achieved using a combination of dynamic light scattering, nanoparticle tracking analysis, transmission electron microscopy, X-ray photoelectron spectroscopy, zeta potential measurements, microwave plasma atomic emission spectroscopy and time-resolved photophysics. The tendency of the functionalized nanodiamonds to aggregate reflects the degree of surface substitution, yielding small aggregates with typical particle sizes ca. 150 nm. This is likely to be driven by the reduction of the zeta potential, concomitant with the conversion of surface charged carboxylate groups to neutral amide functions. The results show that luminescent nanodiamond materials can be synthesised with tuneable photophysical properties

    End-of-life care in UK care homes: a systematic review of the literature

    Background The number of people requiring end-of-life care provision in care homes has grown significantly. There is a need for a systematic examination of individual studies to provide more comprehensive information about contemporary care provision. Aim The aim of this study was to systematically review studies that describe end-of-life care in UK care homes. Method A systematic PRISMA review of the literature published between 2008 and April 2017 was carried out. A total of 14 studies were included in the review. Results A number of areas of concern were identified in the literature in relation to the phases of dying during end-of-life care: end-of-life pre-planning processes; understandings of end-of-life care; and interprofessional end-of-life care provision. Conclusions Given that the problems identified in the literature concerning end-of-life care of residents in care homes are similar to those encountered in other healthcare environments, there is logic in considering how generalised solutions that have been proposed could be applied to the specifics of care homes. Further research is necessary to explore how barriers to good end-of-life care can be mitigated, and facilitators strengthened

    Proline-rich tyrosine kinase 2 mediates gonadotropin-releasing hormone signaling to a specific extracellularly regulated kinase-sensitive transcriptional locus in the luteinizing hormone beta-subunit gene

    G protein-coupled receptor regulation of gene transcription primarily occurs through the phosphorylation of transcription factors by MAPKs. This requires transduction of an activating signal via scaffold proteins that can ultimately determine the outcome by binding signaling kinases and adapter proteins with effects on the target transcription factor and locus of activation. By investigating these mechanisms, we have elucidated how pituitary gonadotrope cells decode an input GnRH signal into coherent transcriptional output from the LH β-subunit gene promoter. We show that GnRH activates c-Src and multiple members of the MAPK family, c-Jun NH(2)-terminal kinase 1/2, p38MAPK, and ERK1/2. Using dominant-negative point mutations and chemical inhibitors, we identified that calcium-dependent proline-rich tyrosine kinase 2 specifically acts as a scaffold for a focal adhesion/cytoskeleton-dependent complex comprised of c-Src, Grb2, and mSos that translocates an ERK-activating signal to the nucleus. The locus of action of ERK was specifically mapped to early growth response-1 (Egr-1) DNA binding sites within the LH β-subunit gene proximal promoter, which was also activated by p38MAPK, but not c-Jun NH(2)-terminal kinase 1/2. Egr-1 was confirmed as the transcription factor target of ERK and p38MAPK by blockade of protein expression, transcriptional activity, and DNA binding. We have identified a novel GnRH-activated proline-rich tyrosine kinase 2-dependent ERK-mediated signal transduction pathway that specifically regulates Egr-1 activation of the LH β-subunit proximal gene promoter, and thus provide insight into the molecular mechanisms required for differential regulation of gonadotropin gene expression
