    Local threats and potential infectious hazards to maned wolves (Chrysocyon brachyurus) in the southeastern Argentine Chaco

    A near-threatened species, the maned wolf ’ s ( Chrysocyon brachyurus ) range extends from central Brazil, northern and eastern Bolivia, and southeastern Peru to the southeast of the Gran Chaco ecoregion and adjacent scrubland in Argentina. Rural domestic dogs under ownership may pose threats to maned wolves as reservoir hosts of multiple pathogens. A serologic survey of rural domestic dogs and maned wolves was conducted to document exposure to canine pathogens in southeastern Santiago del Estero, Argentina, with a parallel questionnaire of knowledge, attitudes, and perception of local villagers in relation to maned wolves. The three maned wolves examined had evidence of exposure to canine adenovirus, canine distemper virus, canine parvovirus, Leptospira interrogans spp., and Dirofilaria immitis . Dogs had very high seroprevalence for Neospora caninum and canine coronavirus (67%), canine adenovirus (59%), D. immitis (58%), canine distemper virus (57%), Toxoplasma gondii (33%), and L. interrogans spp. (20%). Antibodies for canine parvovirus, Brucella canis and Trypanosoma cruzi were rare or absent. Multiple logistic regression analysis showed that dog seroprevalence was not significantly associated with age, sex, and function for any of the pathogens investigated. The frequency distribution of seroreactivity to specific pathogens per dog was highly aggregated. Villagers considered that illegal trade, hunting, vehicular collisions, and interactions with dogs were the most serious local threats for maned wolves. The multiple threats faced by the maned wolf populations in southeastern Santiago del Estero suggest that their populations are unlikely to remain viable unless specific protection measures are taken.Fil: Orozco, Maria Marcela.     Vesicants are potent blistering agents. The prototype vesicant is sulphur mustard gas, first used in World War I, which still has no effective antidote. We used a mustard gas surrogate 2-chloroethyl ethyl sulphide (CEES) to study the ability of resveratrol (RES) and pterostilbene (PTS), two well-established stilbene antioxidants, ebselen (EB-1), an organoselenium compound, and three EB-1 analogues (EB-2, EB-3, and EB-4) to reduce CEES toxicity in human epidermoid carcinoma cells (A-431). Following a 24-hour incubation of a toxic concentration of CEES (1000 μmol L-1), we used the MTT [3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide] test to analyse cell viability. Different concentrations of test antioxidants alone (15 μmol L-1, 30 μmol L-1 or 60 μmol L-1) did not decrease cell viability. Treatment with CEES and test antioxidants for 24 h showed that only EB-1 and its analogues EB-2, EB-3, and EB-4 but not the stilbene compounds could rescue the cells from death. EB-1 and EB-4 were the most effective at reducing CEES cytotoxicity and did so in a concentration-dependent manner, while EB-2 and EB-3 demonstrated the least protective effect. In summary, the data described herein indicate that organoselenium antioxidants, especially EB-4, may prove useful as countermeasures to blistering agents.Plikavci izazivaju izražene mjehuriće na koži. Najpoznatiji je svakako sumporni iperit, koji se prvi put uporabio u 1. svjetskom ratu i do današnjega dana nema djelotvornog protuotrova. U ispitivanju smo rabili zamjenu za iperit, 2-kloroetil etilni sulfi d (CEES) da bismo testirali sposobnost resveratrola (RES) i pterostilbena (PTS), dvaju poznatih stilbenskih antioksidansa, organoselenijeva spoja epselena (EB-1) te njegovih triju analoga (EB-2, EB-3 i EB-4) da smanji toksičnost CEES-a u humanih stanica epidermoidnog karcinoma (A-431). Vijabilnost stanica testirali smo s pomoću 3-(4,5-dimetiltiazol-2-il)-2,5-difeniltetrazol bromida (MTT) nakon 24-satne inkubacije s toksičnom koncentracijom CEES-a (1000 μmol L-1). Antioksidansi davani sami u različitim koncentracijama (15 μmol L-1, 30 μmol L-1, odnosno 60 μmol L-1) nisu smanjili vijabilnost stanica. Dvadesetčetverosatna primjena CEES-a i testiranih antioksidansa pokazala je da samo EB-1 i njegovi analozi EB-2, EB-3 i EB-4 mogu spriječiti smrt stanica, ali ne i stilbenski spojevi. EB-1 i EB-4 pokazali su se najdjelotvornijima u ublažavanju toksičnosti CEES-a u skladu s koncentracijom, dok su se EB-2 i EB-3 pokazali najmanje djelotvornima.     Features of collisionless turbulence in the intracluster medium from simulated Faraday rotation maps - II. The effects of instabilities feedback

    Statistical analysis of Faraday rotation measure (RM) maps of the intracluster medium (ICM) of galaxy clusters provides a unique tool to evaluate some spatial features of the magnetic fields there. Its combination with numerical simulations of magnetohydrodynamic (MHD) turbulence allows the diagnosis of the ICM turbulence. Being the ICM plasma weakly collisional, the thermal velocity distribution of the particles naturally develops anisotropies as a consequence of the large-scale motions and the conservation of the magnetic moment of the charged particles. A previous study (Paper I) analysed the impact of large-scale thermal anisotropy on the statistics of RM maps synthesized from simulations of turbulence; these simulations employed a collisionless MHD model that considered a tensor pressure with uniform anisotropy. In this work, we extend that analysis to a collisionless MHD model in which the thermal anisotropy develops according to the conservation of the magnetic moment of the thermal particles. We also consider the effect of anisotropy relaxation caused by the microscale mirror and firehose instabilities. We show that if the relaxation rate is fast enough to keep the anisotropy limited by the threshold values of the instabilities, the dispersion and power spectrum of the RM maps are indistinguishable from those obtained from collisional MHD. Otherwise, there is a reduction in the dispersion and steepening of the power spectrum of the RM maps (compared to the collisional case). Considering the first scenario, the use of collisional MHD simulations for modelling the RM statistics in the ICM becomes better justified.Fil: Lima, R. Santos.     Circulating Galectin-1 and Galectin-3 in sera from patients with systemic sclerosis: associations with clinical features and treatment

    Systemic Sclerosis (SSc) is a rheumatic disease characterized by fibrosis, microvascular damage and immune dysregulation. Two major subsets, limited cutaneous systemic sclerosis (lcSSc) and diffuse cutaneous systemic sclerosis (dcSSc) can be defined, according to the extent of skin involvement. Increasing evidence indicates a role for galectins in immune and vascular programs, extracellular matrix remodeling and fibrosis, suggesting their possible involvement in SSc. Here, we determined serum levels of galectin (Gal)-1 and Gal-3 in 83 SSc patients (dcSSc n = 17; lcSSc n = 64; ssSSc n = 2), and evaluated their association with clinical manifestations of the disease. Patients with dcSSc showed lower Gal-3 levels, compared to lcSSc (p = 0.003), whereas no considerable difference in Gal-1 levels was detected between groups. Remarkably, higher concentrations of Gal-1 were associated with the presence of telangiectasias (p = 0.015), and higher concentrations Gal-3 were associated with telangiectasias (p = 0.021), diarrhea (p = 0.039) and constipation (p = 0.038). Moreover, lower Gal-3 levels were associated with the presence of tendinous retractions (p = 0.005). Patients receiving calcium blockers (p = 0.048), methotrexate (p = 0.046) or any immunosuppressive treatment (p = 0.044) presented lower concentrations of Gal-3 compared to those not receiving such treatments. The presence of telangiectasia and the type of SSc maintained their statistical association with Gal-3 (β 0.25; p = 0.022 and β 0.26; p = 0.017, respectively) in multiple linear regression models. In conclusion, serum levels of Gal-3 are associated with clinical manifestations of SSc. Among them, the presence of telangiectasias could be explained by the central role of this lectin in the vascularization programs.Fil: Sundblad, Victoria.     Merida virus, a putative novel rhabdovirus discovered in Culex and Ochlerotatus spp. mosquitoes in the Yucatan Peninsula of Mexico

    Sequences corresponding to a putative, novel rhabdovirus [designated Merida virus (MERDV)] were initially detected in a pool of Culex quinquefasciatus collected in the Yucatan Peninsula of Mexico. The entire genome was sequenced, revealing 11 798 nt and five major ORFs, which encode the nucleoprotein (N), phosphoprotein (P), matrix protein (M), glycoprotein (G) and RNA-dependent RNA polymerase (L). The deduced amino acid sequences of the N, G and L proteins have no more than 24, 38 and 43 % identity, respectively, to the corresponding sequences of all other known rhabdoviruses, whereas those of the P and M proteins have no significant identity with any sequences in GenBank and their identity is only suggested based on their genome position. Using specific reverse transcription-PCR assays established from the genome sequence, 27 571 C. quinquefasciatus which had been sorted in 728 pools were screened to assess the prevalence of MERDV in nature and 25 pools were found positive. The minimal infection rate (calculated as the number of positive mosquito pools per 1000 mosquitoes tested) was 0.9, and similar for both females and males. Screening another 140 pools of 5484 mosquitoes belonging to four other genera identified positive pools of Ochlerotatus spp. mosquitoes, indicating that the host range is not restricted to C. quinquefasciatus. Attempts to isolate MERDV in C6/36 and Vero cells were unsuccessful. In summary, we provide evidence that a previously undescribed rhabdovirus occurs in mosquitoes in Mexico.The authors thank Valeria Bussetti for expert technical assistance.     Identification and antimicrobial susceptibility of Streptomyces and other unusual Actinobacteria clinical isolates in Spain

    Two hundred and eighty-six isolates from human clinical samples were identified between 1996 and 2019 as belonging to 8 families, 19 genera and 88 species of Actinobacteria. The most identified genera were Streptomyces (182 strains from 45 species), Actinomadura (29 strains, 5 species), Nocardiopsis (21 strains, 6 species) and Dietzia (18 strains, 5 species). The rest of the identified genera (15) contained 27 species with 36 isolates. Of the species studied, only 13/88 had been documented previously as isolates from clinical samples, and in some cases, as true pathogens. In this sense, a literature review of the species found in infections or in clinical samples without clear involvement in pathology has been carried out. Finally, the susceptibility to 8 antimicrobial agents has been studied. Streptomyces showed high resistance (80.8%) against cefotaxime and cotrimoxazole (55.5%), and no isolate resistance to amikacin and linezolid have been found. Lower percentages of resistance have been found in other genera, except in Dietzia (100% against cotrimoxazole and 44.4% against erythromycin). The greatest resistance in these genera was to cotrimoxazole (29.8) and erythromycin (27,9%), and no resistance to linezolid has been found in these genera. In Microbispora, Nonomuraea and Umezawaea, no resistant isolates have been found against any antibiotic studied. Only 3/104 isolates were resistant to amikacin in Amycolatopsis, Crossiella, and Micromonosopora. One isolate of Amycolatopsis was resistant to imipenem.S

    The Gemini NICI Planet-Finding Campaign: The Frequency of Giant Planets Around Debris Disk Stars

    We have completed a high-contrast direct imaging survey for giant planets around 57 debris disk stars as part of the Gemini NICI Planet-Finding Campaign. We achieved median H-band contrasts of 12.4 mag at 0.5" and 14.1 mag at 1" separation. Follow-up observations of the 66 candidates with projected separation < 500 AU show that all of them are background objects. To establish statistical constraints on the underlying giant planet population based on our imaging data, we have developed a new Bayesian formalism that incorporates (1) non-detections, (2) single-epoch candidates, (3) astrometric and (4) photometric information, and (5) the possibility of multiple planets per star to constrain the planet population. Our formalism allows us to include in our analysis the previously known Beta Pictoris and the HR 8799 planets. Our results show at 95% confidence that 5MJup planet beyond 80 AU, and 3MJup planet outside of 40 AU, based on hot-start evolutionary models. We model the population of directly-imaged planets as d^2N/dMda ~ m^alpha a^beta, where m is planet mass and a is orbital semi-major axis (with a maximum value of amax). We find that beta 1.7. Likewise, we find that beta < -0.8 and/or amax < 200 AU. If we ignore the Beta Pic and HR 8799 planets (should they belong to a rare and distinct group), we find that 3MJup planet beyond 10 AU, and beta < -0.8 and/or alpha < -1.5. Our Bayesian constraints are not strong enough to reveal any dependence of the planet frequency on stellar host mass. Studies of transition disks have suggested that about 20% of stars are undergoing planet formation; our non-detections at large separations show that planets with orbital separation > 40 AU and planet masses > 3 MJup do not carve the central holes in these disks.Comment: Accepted to ApJ on June 24, 2013. 67 pages, 17 figures, 12 table

    Coping Responses During the COVID-19 Pandemic: A Cross-Cultural Comparison of Russia, Kyrgyzstan, and Peru

    Background. The COVID-19 pandemic has subjected people around the world to severe stress, evoking a variety of coping responses. Coping responses can be broadly classified into four strategies: 1) problem-focused coping; 2) emotion-focused coping; 3) socially supported coping; and 4) avoidance. While there is a wide variability of individual coping responses, to some extent they are also culturally specific. Objective. This study aimed to compare the differences in the prevalence and factor structure of coping responses during COVID-19 pandemic in three countries: Russia, Kyrgyzstan, and Peru. Design. The sample included 501 participants from Russia, 456 participants from Kyrgyzstan, and 354 participants from Peru. The mean age of participants was 28 years in Russia (SD = 13.5); 24 years in Kyrgyzstan (SD = 10.0); and 30 years in Peru (SD = 12.3). In Russia and Kyrgyzstan, coping strategies were assessed with an abbreviated Russian adaptation of the COPE (Coping Orientations to Problems Experienced) questionnaire. In Peru, coping responses were assessed using the Spanish version of the Brief COPE questionnaire. The average scores from fifteen COPE scales were used as the input data for linear modelling and factor analysis. Results. The coping scores varied substantially within each country. Differences between countries accounted for 17.7% of the total variability in religious coping; 15.8% in acceptance; 13.9% in mental disengagement; and less than 7% in the other coping strategies. No difference in the prevalence of coping responses was found between Russian and Kyrgyz participants after accounting for age and gender. In all three countries the coping responses were associated with the same four coping domains: problem-focused coping, socially supported coping, avoidance, and emotion-focused coping. Four factors explained up to 44% of the total variation in the COPE scores. Religious coping and mental disengagement were classified into different coping domains in the three countries. Conclusion. The results suggest that during the COVID-19 pandemic, people from different countries apply the full range of coping responses within the four universal coping strategies. Religious coping and mental disengagement differed the most across the countries, suggesting that some coping behaviors can take on different roles within the system of coping responses to stressful events. We attribute these differences to differing cultural and socioeconomic characteristics, and the different measures taken by governments in response to COVID-19

    Detection of RNA from a Novel West Nile-like Virus and High Prevalence of an Insect-specific Flavivirus in Mosquitoes in the Yucatan Peninsula of Mexico

    As part of our ongoing surveillance efforts for West Nile virus (WNV) in the Yucatan Peninsula of Mexico, 96,687 mosquitoes collected from January through December 2007 were assayed by virus isolation in mammalian cells. Three mosquito pools caused cytopathic effect. Two isolates were orthobunyaviruses (Cache Valley virus and Kairi virus) and the identity of the third infectious agent was not determined. A subset of mosquitoes was also tested by reverse transcription-polymerase chain reaction (RT-PCR) using WNV-, flavivirus-, alphavirus-, and orthobunyavirus-specific primers. A total of 7,009 Culex quinquefasciatus in 210 pools were analyzed. Flavivirus RNA was detected in 146 (70%) pools, and all PCR products were sequenced. The nucleotide sequence of one PCR product was most closely related (71-73% identity) with homologous regions of several other flaviviruses, including WNV, St. Louis encephalitis virus, and Ilheus virus. These data suggest that a novel flavivirus (tentatively named T\u27Ho virus) is present in Mexico. The other 145 PCR products correspond to Culex flavivirus, an insect-specific flavivirus first isolated in Japan in 2003. Culex flavivirus was isolated in mosquito cells from approximately one in four homogenates tested. The genomic sequence of one isolate was determined.
