    Ganglioside synthesis by plasma membrane-associated sialyltransferase in macrophages

    Gangliosides are constituents of the mammalian cell membranes and participate in the inflammatory response. However, little is known about the presence and enzymatic activity of ganglioside sialyltransferases at the cell surface of macrophages, one of the most important immune cells involved in the innate inflammatory process. In the present study, using biochemical and fluorescent microscopy approaches, we found that endogenous ST8Sia-I is present at the plasma membrane (ecto-ST8Sia-I) of murine macrophage RAW264.7 cells. Moreover, ecto-ST8Sia-I can synthetize GD3 ganglioside at the cell surface in lipopolysaccharide (LPS)-stimulated macrophages even when LPS-stimulated macrophages reduced the total ST8Sia-I expression levels. Besides, cotreatment of LPS with an inhibitor of nitric oxide (NO) synthase recovered the ecto-ST8Sia-I expression, suggesting that NO production is involved in the reduction of ST8Sia-I expression. The diminution of ST8Sia-I expression in LPS-stimulated macrophages correlated with a reduction of GD3 and GM1 gangliosides and with an increment of GD1a. Taken together, the data supports the presence and activity of sialyltransferases at the plasma membrane of RAW264.7 cells. The variations of ecto-ST8Sia-I and ganglioside levels in stimulated macrophages constitutes a promissory pathway to further explore the physiological role of this and others ganglioside metabolism-related enzymes at the cell surface during the immune response.Fil: Vilcaes, Aldo Alejandro.     Project overview and update on WEAVE: the next generation wide-field spectroscopy facility for the William Herschel Telescope

    We present an overview of and status report on the WEAVE next-generation spectroscopy facility for the William Herschel Telescope (WHT). WEAVE principally targets optical ground-based follow up of upcoming ground-based (LOFAR) and space-based (Gaia) surveys. WEAVE is a multi-object and multi-IFU facility utilizing a new 2-degree prime focus field of view at the WHT, with a buffered pick-and-place positioner system hosting 1000 multi-object (MOS) fibres, 20 integral field units, or a single large IFU for each observation. The fibres are fed to a single spectrograph, with a pair of 8k(spectral) x 6k (spatial) pixel cameras, located within the WHT GHRIL enclosure on the telescope Nasmyth platform, supporting observations at R~5000 over the full 370-1000nm wavelength range in a single exposure, or a high resolution mode with limited coverage in each arm at R~20000.     Objective: To present the presentation of primary genetic neuropathies. Case presentation: We present the case of a young adult patient from a rural area with onset in adolescence of difficulty running and multiple falls associated with decreased strength, on physical examination hand with contractures, muscle atrophy and decreased reflexes, electromyography is sent with generalized peripheral polyneuropathy and mixed symmetrical motor and sensory conduction block in all nerves explored, genetic study is sent with a diagnostic report of Charcot Marie Tooth disease. Conclusions: It is of great importance for the primary care physician the knowledge of the most frequent primary genetic neuropathy for its adequate diagnostic approach through the use of physical examination and genetic support.Objetivo:  Dar a conocer la presentación de las neuropatías genéticas primarias Presentación del caso: Se presenta el caso de un paciente adulto joven de zona rural con cuadro de inicio en la adolescencia de dificultad para correr y múltiples caídas asociado a disminución de fuerza ,al examen físico mano con contracturas, atrofia muscular y disminución de reflejos ,se envía electromiografía con polineuropatía periférica generalizada y simétrica mixta con bloqueo de conducción motora y sensitiva en todos los nervios explorados, se envía estudio genético con reporte diagnóstico de Enfermedad de Charcot Marie Tooth . Conclusiones: Es de gran importancia para el médico de atención primaria el conocimiento de la neuropatía genética primaria más frecuente para su adecuado enfoque diagnostico mediante el uso de el examen físico y el apoyo genético

    HTLV-1 infection in solid organ transplant donors and recipients in Spain

    HTLV-1 infection is a neglected disease, despite infecting 10-15 million people worldwide and severe illnesses develop in 10% of carriers lifelong. Acknowledging a greater risk for developing HTLV-1 associated illnesses due to immunosuppression, screening is being widely considered in the transplantation setting. Herein, we report the experience with universal HTLV testing of donors and recipients of solid organ transplants in a survey conducted in Spain. All hospitals belonging to the Spanish HTLV network were invited to participate in the study. Briefly, HTLV antibody screening was performed retrospectively in all specimens collected from solid organ donors and recipients attended since the year 2008. A total of 5751 individuals were tested for HTLV antibodies at 8 sites. Donors represented 2312 (42.2%), of whom 17 (0.3%) were living kidney donors. The remaining 3439 (59.8%) were recipients. Spaniards represented nearly 80%. Overall, 9 individuals (0.16%) were initially reactive for HTLV antibodies. Six were donors and 3 were recipients. Using confirmatory tests, HTLV-1 could be confirmed in only two donors, one Spaniard and another from Colombia. Both kidneys of the Spaniard were inadvertently transplanted. Subacute myelopathy developed within 1 year in one recipient. The second recipient seroconverted for HTLV-1 but the kidney had to be removed soon due to rejection. Immunosuppression was stopped and 3 years later the patient remains in dialysis but otherwise asymptomatic. The rate of HTLV-1 is low but not negligible in donors/recipients of solid organ transplants in Spain. Universal HTLV screening should be recommended in all donor and recipients of solid organ transplantation in Spain. Evidence is overwhelming for very high virus transmission and increased risk along with the rapid development of subacute myelopathy

    Este título, sugestivo por los impactos durante la situación de la Covid 19 en el mundo, y que en Colombia lastimosamente han sido muy críticos, permiten asumir la obligada superación de tensiones sociales, políticas, y económicas; pero sobre todo científicas y tecnológicas. Inicialmente, esto supone la existencia de una capacidad de la sociedad colombiana por recuperar su estado inicial después de que haya cesado la perturbación a la que fue sometida por la catastrófica pandemia, y superar ese anterior estado de cosas ya que se encontraban -y aún se encuentran- muchos problemas locales mal resueltos, medianamente resueltos, y muchos sin resolver: es decir, habrá que rediseñar y fortalecer una probada resiliencia social existente - producto del prolongado conflicto social colombiano superado parcialmente por un proceso de paz exitoso - desde la tecnociencia local; como lo indicaba Markus Brunnermeier - economista alemán y catedrático de economía de la Universidad de Princeton- en su libro The Resilient Society…La cuestión no es preveerlo todo sino poder reaccionar…aprender a recuperarse rápido.This title, suggestive of the impacts during the Covid 19 situation in the world, and which have unfortunately been very critical in Colombia, allows us to assume the obligatory overcoming of social, political, and economic tensions; but above all scientific and technological.     El objetivo de esta memoria es realizar un estudio de nuestra ley de matrimonio civil, con el fin de determinar e identificar sus principios informadores, los cuales serán para estos efectos, el de intervención judicial a favor del matrimonio, el derecho a contraer matrimonio, el interés superior de los hijos y del cónyuge más débil. En la investigación se expone, lo entendido por parte de nuestra doctrina por cada uno de estos principios fundamentales en sus diferentes perspectivas, enfatizando en el principio del interés superior de los hijos y del cónyuge más débil y su aplicación en diferentes cuerpos legales nacionales, convención sobre los derechos del niño y jurisprudencia. El resultado de esta investigación será establecer la interpretación y aplicación por parte de nuestros Tribunales de Familia de este principio, en diferentes materias propias de su competencia.     Role of plasma-membrane-bound sialidase NEU3 in clathrin-mediated endocytosis

    Gangliosides are sialic acid-containing glycosphingolipids mainly expressed at the outer leaflet of the plasma membrane. Sialidase NEU3 is a key enzyme in the catabolism of gangliosides with its up-regulation having been observed in human cancer cells. In the case of CME (clathrin-mediated endocytosis), although this has been widely studied, the role of NEU3 and gangliosides in this cellular process has not yet been established. In the present study, we found an increased internalization of Tf (transferrin), the archetypical cargo for CME, in cells expressing complex gangliosides with high levels of sialylation. The ectopic expression of NEU3 led to a drastic decrease in Tf endocytosis, suggesting the participation of gangliosides in this process. However, the reduction in Tf endocytosis caused by NEU3 was still observed in glycosphingolipid-depleted cells, indicating that NEU3 could operate in a way that is independent of its action on gangliosides. Additionally, internalization of α2-macroglobulin and low-density lipoprotein, other typical ligands in CME, was also decreased in NEU3-expressing cells. In contrast, internalization of cholera toxin β-subunit, which is endocytosed by both clathrin-dependent and clathrin-independent mechanisms, remained unaltered. Kinetic assays revealed that NEU3 caused a reduction in the sorting of endocytosed Tf to early and recycling endosomes, with the Tf binding at the cell surface being also reduced. NEU3-expressing cells showed an altered subcellular distribution of clathrin adaptor AP-2 (adaptor protein 2), but did not reveal any changes in the membrane distribution of clathrin, PtdIns(4,5)P2 or caveolin-1.     Repopulation and culture of the pacific calico scallops in Bahia Concepcion, Baja California Sur, Mexico

    The Pacific Calico scallop, Argopecten ventricosus (Sowerby), is the most important commercial species in Bahia Concepcion. Catches declined steadily from 1991 to 1993 as a result of overexploitation, forcing the adoption of management measures. The Government has now established regulations for exploitation and scallop culture development. In 1994, Centro Interdisciplinario de Ciencias Marinas (CICIMAR) and the Fisheries Department of the Mexican Government initiated research on the feasibility of increasing scallop biomass through a reseeding programme to protect artisanal fishing. The use of onion bags to collect spat in Bahia Concepcion (1988-1994) demonstrated that settlement was cumulative from January to March. At Punta Coloradito, there were two peaks, in January and early March. Commercial collectors were deployed in February at El Indio and in April at El Remate. In June, about one million juvenile scallops were placed in an enclosure, held until mid-August and then released. Mass mortality (>80%) of the scallops was observed in September, increasing to 99% in October. The high water temperature and predators may be responsible for the mortalities

    Background and Objectives: This European Group for Blood and Marrow Transplantation (EBMT) multicentre randomized phase III study was designed to assess the safety and efficacy of CD34+ selection in newly diagnosed myeloma patients undergoing autologous transplantation. Design and Methods: One hundred and eleven patients responsive to initial chemotherapy were randomized to receive CD34+ selected (arm A) or unselected PBPC (arm B) after conditioning with high-dose melphalan and TBI. ASO-PCR was used to assess purging efficacy and reinfused tumor load. Tumor load could be assessed in 59 patients. Results: CD34+ selection gave a median tumor cell depletion of 2.2 logs (0.77-5.96). No tumor cells were detected in products infused in 17/26 (A) and 5/33 (B) patients. The five year overall survival (OS), event free survival (EFS) and relapse rate (RR) were 51%, 20% and 80% in arm A and 45%, 18% and 80% in arm B respectively with no significant difference between the two groups. Thirteen patients in arm A and 2 in arm B experienced episodes of serious early infection (p=0.02). There were 3 early transplant related deaths in A but none in B. Interpretation and Conclusions: Despite significant tumor cell reduction, CD34+ selection does not reduce RR and increases the risk of severe post-transplant infections. There was also no difference in RR between patients in either arm who received grafts with detectable tumor cells and those receiving grafts with no detectable tumor cells, suggesting that reinfused tumor cells may not be the main cause of relapse after autologous transplant in myeloma. ©2007 Ferrata Storti Foundation.SCOPUS: ar.jinfo:eu-repo/semantics/publishe
