    Hydration status and fluid balance of elite European youth soccer players during consecutive training sessions

    The objective of the study was to investigate the hydration status and fluid balance of elite European youth soccer players during three consecutive training sessions. Fourteen males (age 16.9 ± 0.8 years, height 1.79 ± 0.06 m, body mass (BM) 70.6 ± 5.0 kg) had their hydration status assessed from first morning urine samples (baseline) and pre- and post-training using urine specif-ic gravity (USG) measures, and their fluid balance calculated from pre- to post-training BM change, corrected for fluid intake and urine output. Most participants were hypohydrated upon waking (USG >1.020; 77% on days 1 and 3, and 62% on day 2). There was no significant difference between first morning and pre-training USG (p = 0.11) and no influence of training session (p = 0.34) or time (pre- vs. post-training; p = 0.16) on USG. Significant BM loss occurred in sessions 1-3 (0.69 ± 0.22, 0.42 ± 0.25, and 0.38 ± 0.30 kg respectively, p < 0.05). Mean fluid intake in sessions 1-3 was 425 ± 185, 355 ± 161, and 247 ± 157 ml, respectively (p < 0.05). Participants replaced on average 71.3 ± 64.1% (range 0-363.6%) of fluid losses across the three sessions. Body mass loss, fluid intake, and USG measures showed large inter-individual variation. Elite young European soccer players likely wake and present for training hypohydrat-ed, when a USG threshold of 1.020 is applied. When training in a cool environment with ad libitum access to fluid, replacing ~71% of sweat losses results in minimal hypohydration (<1% BM). Consumption of fluid ad libitum throughout training ap-pears to prevent excessive (≥2% BM) dehydration, as advised by current fluid intake guidelines. Current fluid intake guide-lines appear applicable for elite European youth soccer players training in a cool environment

    The influence of serial carbohydrate mouth rinsing on power output during a cycle sprint

    The objective of the study was to investigate the influence of serial administration of a carbohydrate (CHO) mouth rinse on performance, metabolic and perceptual responses during a cycle sprint. Twelve physically active males (mean (± SD) age: 23.1 (3.0) years, height: 1.83 (0.07) m, body mass (BM): 86.3 (13.5) kg) completed the following mouth rinse trials in a randomized, counterbalanced, double-blind fashion; 1. 8 x 5 second rinses with a 25 ml CHO (6% w/v maltodextrin) solution, 2. 8 x 5 second rinses with a 25 ml placebo (PLA) solution. Following mouth rinse administration, participants completed a 30 second sprint on a cycle ergometer against a 0.075 g·kg-1 BM resistance. Eight participants achieved a greater peak power output (PPO) in the CHO trial, resulting in a significantly greater PPO compared with PLA (13.51 ± 2.19 vs. 13.20 ± 2. 14 W·kg-1, p < 0.05). Magnitude inference analysis reported a likely benefit (81% likelihood) of the CHO mouth rinse on PPO. In the CHO trial, mean power output (MPO) showed a trend for being greater in the first 5 seconds of the sprint and lower for the remainder of the sprint compared with the PLA trial (p > 0.05). No significant between-trials difference was reported for fatigue index, perceived exertion, arousal and nausea levels, or blood lactate and glucose concentrations. Serial administration of a CHO mouth rinse may significantly improve PPO during a cycle sprint. This improvement appears confined to the first 5 seconds of the sprint, and may come at a greater relative cost for the remainder of the sprint

    Changes in perceived mental fatigue, physical fatigue and mood state during a 4-day national junior orienteering competition preparation camp

    Mental fatigue (MF) has been shown to acutely impair the psychological responses and endurance running performance of orienteers. This study aimed to explore MF levels experienced by orienteers during a 4-day competition preparation camp that consisted of simulated sprint, middle-distance, long-distance, relay and night races. Eleven national junior orienteers participated in the study (age: 15–17 years, height: 1.69 ± 0.07 m and body mass: 59.9 ± 5.22 kg). Subjective ratings of MF, motivation, stress, physical fatigue (PF) and tiredness were measured using a 100-mm visual analogue scale. The Brunel Mood Scale (BRUMS) was utilized to assess the mood state of the orienteers. The self-report measures were taken within 30 min of waking, immediately after the post-training session, and after 24 and 48 h following the final training session. The pre–post orienteering training combined analysis showed that there was a moderate increase in perceived MF (ES = 1.06 [0.66, 1.45]), PF (ES = 1.07 [0.69, 1.45]) and BRUMS fatigue (ES = 0.74 [0.4, 1.1]) after orienteering training. At 48 h post the final training session, MF remained moderately elevated (ES = 0.86 [−0.07, 1.75]), while PF also remained elevated to a small extent (ES = 0.46 [−0.46, 1.39]) compared to the pre-training values. A moderate impairment was still observed in BRUMS vigor (ES = −1.02 [−1.65, −0.36]), but BRUMS confusion scores were moderately lower (ES = −0.85 [−1.71, 0.04]) than pre-training values. This study found that orienteering training induced acute MF, persisting for at least 48 h after the final session

    The effect of RaceRunning on cardiometabolic disease risk factors and functional mobility in young people with moderate-to-severe cerebral palsy:A feasibility study

    Introduction: There is consistent evidence that people with cerebral (CP) do not engage in the recommended physical activity guidelines for the general population from a young age. Participation in moderate-to-vigorous physical activity is particularly reduced in people with CP who have moderate-to-severe disability. RaceRunning is a growing disability sport that provides an opportunity for people with moderate-to-severe disability to participate in physical activity in the community. It allows those who are unable to walk independently, to propel themselves using a RaceRunning bike, which has a breastplate for support but no pedals. The aim of this study is to examine the feasibility and acceptability of RaceRunning for young people with moderate-to-severe CP and the feasibility of conducting a definitive study of the effect of RaceRunning on cardiometabolic disease risk factors and functional mobility. Methods and analysis: Twenty-five young people (age 5-21 yr) with CP or acquired brain injury affecting co-ordination will be included in this single arm intervention study. Participants will take part in one RaceRunning session each week for 24 weeks. Outcomes assessed at baseline, 12 and 24 weeks include body mass index, waist circumference, blood pressure, muscle strength, cardiorespiratory fitness, physical activity and sedentary behaviour, functional mobility, activity competence and psychosocial impact. Adverse events will be systematically recorded throughout the 24 weeks. Focus groups will be conducted with participants and/or parents to explore their views and experiences of taking part in RaceRunning. Ethics and dissemination: Approval has been granted by Queen Margaret University Research Ethics Committee (REC) and the South East of Scotland REC. Results will be disseminated through peer-reviewed journals and distributed to people with CP and their families through RaceRunning and Athletic Clubs, NHS trusts, and organisations for people with disabilities

    MPs’ principals and the substantive representation of disadvantaged immigrant groups

    This article provides an alternative understanding of the substantive representation of immigrant-origin citizens compared to previous work in the ‘politics of presence’ tradition. Rather than assuming that the representational activities of members of parliaments (MPs) are underpinned by intrinsic motivations, it highlights extrinsic motives. Drawing on principal–agent theory, the article conceptualises MPs as delegates who are to act on behalf of their main principals, constituents and party bodies. This approach permits the rigorous analysis of the impact of electoral rules, candidate selection methods and legislative organisation on substantive representation. Based on an analysis of more than 20,000 written parliamentary questions tabled in the 17th German Bundestag (2009–2013), empirical findings suggest that electoral rules do not influence the relationship between MPs and their principals in relation to the substantive representation of disadvantaged immigrant groups; however, results indicate that candidate selection methods as well as powerful parliamentary party group leaderships do

    A comprehensive framework for assessing the accuracy and uncertainty of global above-ground biomass maps

    Over the past decade, several global maps of above-ground biomass (AGB) have been produced, but they exhibit significant differences that reduce their value for climate and carbon cycle modelling, and also for national estimates of forest carbon stocks and their changes. The number of such maps is anticipated to increase because of new satellite missions dedicated to measuring AGB. Objective and consistent methods to estimate the accuracy and uncertainty of AGB maps are therefore urgently needed. This paper develops and demonstrates a framework aimed at achieving this. The framework provides a means to compare AGB maps with AGB estimates from a global collection of National Forest Inventories and research plots that accounts for the uncertainty of plot AGB errors. This uncertainty depends strongly on plot size, and is dominated by the combined errors from tree measurements and allometric models (inter-quartile range of their standard deviation (SD) = 30–151 Mg ha−1). Estimates of sampling errors are also important, especially in the most common case where plots are smaller than map pixels (SD = 16–44 Mg ha−1). Plot uncertainty estimates are used to calculate the minimum-variance linear unbiased estimates of the mean forest AGB when averaged to 0.1∘. These are used to assess four AGB maps: Baccini (2000), GEOCARBON (2008), GlobBiomass (2010) and CCI Biomass (2017). Map bias, estimated using the differences between the plot and 0.1∘ map averages, is modelled using random forest regression driven by variables shown to affect the map estimates. The bias model is particularly sensitive to the map estimate of AGB and tree cover, and exhibits strong regional biases. Variograms indicate that AGB map errors have map-specific spatial correlation up to a range of 50–104 km, which increases the variance of spatially aggregated AGB map estimates compared to when pixel errors are independent. After bias adjustment, total pantropical AGB and its associated SD are derived for the four map epochs. This total becomes closer to the value estimated by the Forest Resources Assessment after every epoch and shows a similar decrease. The framework is applicable to both local and global-scale analysis, and is available at https://github.com/arnanaraza/PlotToMap. Our study therefore constitutes a major step towards improved AGB map validation and improvement

    DISC1 regulates N-methyl-D-aspartate receptor dynamics:abnormalities induced by a Disc1 mutation modelling a translocation linked to major mental illness

    Abstract The neuromodulatory gene DISC1 is disrupted by a t(1;11) translocation that is highly penetrant for schizophrenia and affective disorders, but how this translocation affects DISC1 function is incompletely understood. N-methyl-D-aspartate receptors (NMDAR) play a central role in synaptic plasticity and cognition, and are implicated in the pathophysiology of schizophrenia through genetic and functional studies. We show that the NMDAR subunit GluN2B complexes with DISC1-associated trafficking factor TRAK1, while DISC1 interacts with the GluN1 subunit and regulates dendritic NMDAR motility in cultured mouse neurons. Moreover, in the first mutant mouse that models DISC1 disruption by the translocation, the pool of NMDAR transport vesicles and surface/synaptic NMDAR expression are increased. Since NMDAR cell surface/synaptic expression is tightly regulated to ensure correct function, these changes in the mutant mouse are likely to affect NMDAR signalling and synaptic plasticity. Consistent with these observations, RNASeq analysis of the translocation carrier-derived human neurons indicates abnormalities of excitatory synapses and vesicle dynamics. RNASeq analysis of the human neurons also identifies many differentially expressed genes previously highlighted as putative schizophrenia and/or depression risk factors through large-scale genome-wide association and copy number variant studies, indicating that the translocation triggers common disease pathways that are shared with unrelated psychiatric patients. Altogether, our findings suggest that translocation-induced disease mechanisms are likely to be relevant to mental illness in general, and that such disease mechanisms include altered NMDAR dynamics and excitatory synapse function. This could contribute to the cognitive disorders displayed by translocation carriers
