    Ineffectiveness in Teacher Bargaining: The Anatomy of a First Strike

    Dans l'étude et la pratique de la négociation collective, on se heurte toujours à la possibilité d'une grève qui peut être potentiellement néfaste. Lorsque, de plus, les parties sont des novices dans l'art de négocier et qu'elles font face à un état de grève pour la première fois, il est probable qu'elles auront un comportement qui laisse à désirer et qui peut conduire à des résultats désastreux.Cette étude traite spécifiquement d'une « première grève » et propose une série de faits qui caractérisent de pareilles situations. Ces facteurs sont ensuite utilisés dans l'analyse des événements qui entourent une grève dans le secteur de l'enseignement où les deux parties, le syndicat et la commission scolaire, en étaient à leur première expérience.Lorsqu'il s'agit d'une première grève, on peut s'attendre à ce que, comparés à des négociateurs plus expérimentés, les responsables de la négociation soient (1) moins bien informés et plus hésitants, ce qui conduit à une perception moins nette des événements et à un comportement irrationnel; (2) qu'ils se montrent méfiants à l'endroit des autres parties; (3) qu'ils s'appuient sur des raisons émotives; (4) qu'ils adoptent une attitude défensive et agressive; (5) qu'ils optent pour des modes de communication inadéquats; (6) qu'ils aient recours à des aides professionnels moins compétents, ce qui a pour effet d'accroître les autres difficultés.L'examen de ce qui s'est passé au cours de cette grève d'enseignants a révélé que, d'une façon générale, le syndicat a subi des préjudices graves et inutiles comme résultat de sa décision de faire la grève, en obtenant qu'un petit peu plus que ce que lui avait offert la commission scolaire immédiatement avant la grève. Des communications réticentes et sélectives, alors que l'une et l'autre parties jouaient à cache-cache, le manque de conseils à la section locale de la part du siège social du syndicat, la négligence des dirigeants syndicaux à renseigner les membres sur des faits importants qui se rapportaient directement au conflit comme les offres salariales, des erreurs dans la stratégie de négociation qui ont placé le syndicat dans une situation désavantageuse, des attitudes de plus en plus émotives et agressives entre les parties au fur et à mesure que les négociations se poursuivaient, autant de faits qui ont caractérisé ces négociations. Le syndicat laissa passer la période la plus propice à la grève au commencement de l'automne au moment de l'ouverture des classes alors que la commission scolaire était moins prête à affronter la grève.La grève elle-même fut assez violente; elle a laissé de profonds ressentiments et une méfiance prolongée entre les parties. Les dirigeants syndicaux ont été emprisonnés une semaine pendant la grève. Les cours furent perturbés et les policiers furent forcés de maîtriser les grévistes. Les membres en général furent surpris par la dureté de la grève et par la fermeté de la commission scolaire à maintenir sa position. Le président de la commission scolaire remet peu après sa démission à cause d'animosité et de pressions sous forme d'appels téléphoniques outranciers, de lettres haineuses et d'altercation publique avec les membres du syndicat.Les événements et les expériences qui ont marqué cette grève diffèrent de la plupart des grèves d'enseignants qui ont été analysées jusqu'ici, ce qui indique que les premières grèves méritent une étude spéciale en tant que forme originale de conflits qui possèdent leurs caractéristiques et leurs dynamisme propres.On peut donc conclure que les parties à une première grève devraient se rendre compte de la probabilité plus forte des dangers dont il vient d'être question et qu'elles devraient mieux se préparer aux réalités de la négociation collective, se doter d'une aide professionnelle compétente, être prêtes à faire des compromis et être conscientes des effets prolongés de comportements néfastes pour les négociations futures.The objective of this paper is to identify characteristics which distinguish first strike situations from other strike actions.     Ecology impacts the decrease of Spirochaetes and Prevotella in the fecal gut microbiota of urban humans

    Compared to the huge microbial diversity in most mammals, human gut microbiomes have lost diversity while becoming specialized for animal-based diets - especially compared to chimps, their genetically closest ancestors. The lowered microbial diversity within the gut of westernized populations has also been associated with different kinds of chronic inflammatory diseases in humans. To further deepen our knowledge on phylogenetic and ecologic impacts on human health and fitness, we established the herein presented biobank as well as its comprehensive microbiota analysis. In total, 368 stool samples from 38 different animal species, including Homo sapiens, belonging to four diverse mammalian orders were collected at seven different locations and analyzed by 16S rRNA gene amplicon sequencing. Comprehensive data analysis was performed to (i) determine the overall impact of host phylogeny vs. diet, location, and ecology and to (ii) examine the general pattern of fecal bacterial diversity across captive mammals and humans.By using a controlled study design with captive mammals we could verify that host phylogeny is the most dominant driver of mammalian gut microbiota composition. However, the effect of ecology appears to be able to overcome host phylogeny and should therefore be studied in more detail in future studies. Most importantly, our study could observe a remarkable decrease of Spirochaetes and Prevotella in westernized humans and platyrrhines, which is probably not only due to diet, but also to the social behavior and structure in these communities.Our study highlights the importance of phylogenetic relationship and ecology within the evolution of mammalian fecal microbiota composition. Particularly, the observed decrease of Spirochaetes and Prevotella in westernized communities might be associated to lifestyle dependent rapid evolutionary changes, potentially involved in the establishment of dysbiotic microbiomes, which promote the etiology of chronic diseases

    Reliability of an adapted core strength endurance test battery in individuals with axial spondylarthritis

    Objectives To adapt the core strength endurance test battery (aCSE), previously used for testing athletes, to a target group of patients with axial spondylarthritis (axSpA), to evaluate its intra-tester reliability and its associationswith disease-specific factors. Methods A cross-sectional study was conducted at axSpA exercise therapy groups, including both axSpA patients and the physiotherapist group leaders (PTs). The aCSE was used to measure the isometric strength endurance of the ventral, lateral, and dorsal core muscle chains (measured in seconds), as well as to assess the disease-specific factors of functional status, selfreported pain, and perceived strength performance. The aCSE was repeated after 7–14 days to measure intra-tester reliability for the same rater (PT group leader). Reliability was calculated as an intra-class correlation coefficient (ICC) using a nested design. The associations between ventral, lateral, and dorsal strength endurance and the disease-specific factors were calculated using Pearson correlation coefficients. Results Study participants were 13 PT group leaders and 62 axSpA patients. The latter were all capable of performing the aCSE, with the exception of one individual. A moderate to substantial intra-rater reliability (ICCs (95%CI)) was found for the ventral (0.54 (0.35, 0.74)), lateral (0.52 (0.33, 0.70)), and dorsal (0.71 (0.58, 0.86)) core muscle chains. None of the aCSE measures correlated with the disease-specific factors. Conclusion The aCSE was found to be a reliable test battery for assessing core strength endurance in axSpA patients. Interestingly, aCSE performance was not associated with any disease-specific factors

    Short-term physical exercise impacts on the human holobiont obtained by a randomised intervention study

    Background Human well-being has been linked to the composition and functional capacity of the intestinal microbiota. As regular exercise is known to improve human health, it is not surprising that exercise was previously described to positively modulate the gut microbiota, too. However, most previous studies mainly focused on either elite athletes or animal models. Thus, we conducted a randomised intervention study that focused on the effects of different types of training (endurance and strength) in previously physically inactive, healthy adults in comparison to controls that did not perform regular exercise. Overall study duration was ten weeks including six weeks of intervention period. In addition to 16S rRNA gene amplicon sequencing of longitudinally sampled faecal material of participants (six time points), detailed body composition measurements and analysis of blood samples (at baseline and after the intervention) were performed to obtain overall physiological changes within the intervention period. Activity tracker devices (wrist-band wearables) provided activity status and sleeping patterns of participants as well as exercise intensity and heart measurements. Conclusions We could show that different types of exercise have distinct but moderate effects on the overall physiology of humans and very distinct microbial changes in the gut. The observed overall changes during the intervention highlight the importance of physical activity on well-being. Future studies should investigate the effect of exercise on a longer timescale, investigate different training intensities and consider high-resolution shotgun metagenomics technology.     Boolean analysis reveals systematic interactions among low-abundance species in the human gut microbiome

    The analysis of microbiome compositions in the human gut has gained increasing interest due to the broader availability of data and functional databases and substantial progress in data analysis methods, but also due to the high relevance of the microbiome in human health and disease. While most analyses infer interactions among highly abundant species, the large number of low-abundance species has received less attention. Here we present a novel analysis method based on Boolean operations applied to microbial co-occurrence patterns. We calibrate our approach with simulated data based on a dynamical Boolean network model from which we interpret the statistics of attractor states as a theoretical proxy for microbiome composition. We show that for given fractions of synergistic and competitive interactions in the model our Boolean abundance analysis can reliably detect these interactions. Analyzing a novel data set of 822 microbiome compositions of the human gut, we find a large number of highly significant synergistic interactions among these low-abundance species, forming a connected network, and a few isolated competitive interactions

    Extended analysis of a genome-wide association study in primary sclerosing cholangitis detects multiple novel risk loci.

    A limited number of genetic risk factors have been reported in primary sclerosing cholangitis (PSC). To discover further genetic susceptibility factors for PSC, we followed up on a second tier of single nucleotide polymorphisms (SNPs) from a genome-wide association study (GWAS). We analyzed 45 SNPs in 1221 PSC cases and 3508 controls. The association results from the replication analysis and the original GWAS (715 PSC cases and 2962 controls) were combined in a meta-analysis comprising 1936 PSC cases and 6470 controls. We performed an analysis of bile microbial community composition in 39 PSC patients by 16S rRNA sequencing. Seventeen SNPs representing 12 distinct genetic loci achieved nominal significance (p(replication) <0.05) in the replication. The most robust novel association was detected at chromosome 1p36 (rs3748816; p(combined)=2.1 × 10(-8)) where the MMEL1 and TNFRSF14 genes represent potential disease genes. Eight additional novel loci showed suggestive evidence of association (p(repl) <0.05). FUT2 at chromosome 19q13 (rs602662; p(comb)=1.9 × 10(-6), rs281377; p(comb)=2.1 × 10(-6) and rs601338; p(comb)=2.7 × 10(-6)) is notable due to its implication in altered susceptibility to infectious agents. We found that FUT2 secretor status and genotype defined by rs601338 significantly influence biliary microbial community composition in PSC patients. We identify multiple new PSC risk loci by extended analysis of a PSC GWAS.     BackgroundTreg cells have been shown to be an important part of immune-homeostasis and IL-2 which is produced upon T cell receptor (TCR)-dependent activation of T lymphocytes has been demonstrated to critically participate in Treg development.ObjectiveTo evaluate small molecule inhibitors (SMI) for the identification of novel IL-2/Treg enhancing compounds.Materials and methodsWe used TCR-dependent and allergen-specific cytokine secretion of human and mouse T cells, next generation messenger ribonucleic acid sequencing (RNA-Seq) and two different models of allergic airway inflammation to examine lead SMI-compounds.ResultsWe show here that the reported 3-phosphoinositide dependent kinase-1 (PDK1) SMI BX-795 increased IL-2 in culture supernatants of Jurkat E6-1 T cells, human peripheral blood mononuclear cells (hPBMC) and allergen-specific mouse T cells upon TCR-dependent and allergen-specific stimulation while concomitantly inhibiting Th2 cytokine secretion. RNA-Seq revealed that the presence of BX-795 during allergen-specific activation of T cells induces a bona fide Treg cell type highly similar to iTreg but lacking Foxp3 expression. When applied in mugwort pollen and house dust mite extract-based models of airway inflammation, BX-795 significantly inhibited Th2 inflammation including expression of Th2 signature transcription factors and cytokines and influx into the lungs of type 2-associated inflammatory cells such as eosinophils.ConclusionsBX-795 potently uncouples IL-2 production from Th2 inflammation and induces Th-IL-2 cells, which highly resemble induced (i)Tregs. Thus, BX-795 may be a useful new compound for the treatment of allergic diseases
