    Generation of internal solitary waves by frontally forced intrusions in geophysical flows

    International audienceInternal solitary waves are hump-shaped, large-amplitude waves that are physically analogous to surface waves except that they propagate within the fluid, along density steps that typically characterize the layered vertical structure of lakes, oceans and the atmosphere. As do surface waves, internal solitary waves may overturn and break, and the process is thought to provide a globally significant source of turbulent mixing and energy dissipation. Although commonly observed in geophysical fluids, the origins of internal solitary waves remain unclear. Here we report a rarely observed natural case of the birth of internal solitary waves from a frontally forced interfacial gravity current intruding into a two-layer and vertically sheared background environment. The results of the analysis carried out suggest that fronts may represent additional and unexpected sources of internal solitary waves in regions of lakes, oceans and atmospheres that are dynamically similar to the situation examined here in the Saguenay Fjord, Canada

    Interfacial solitary wave run-up in the St. Lawrence Estuary

    Density variations show evidence of interfacial solitary waves (ISW) running up the sloping boundary of an island in the St. Lawrence Estuary, confirming inferences based remote sensing. Further detail is suggested by simulations created with a two-dimensional nonhydrostatic numerical model. The simulations confirm theoretical predictions of the location of wave breaking, something that is difficult to observe in the field. Two other results of the simulations match laboratory findings: the creation of turbulent boluses that propagate upslope of the breaking zone, and the creation of an intermediate layer that transports mixed water away from the mixing site. Although our sampling could not resolve the intermediate mixing layer, it did provide evidence of boluses. In addition to ISW breaking the bolus and intrusion effects may also be important in coastal regions

    In a manner similar to that of surface waves on beaches, high-frequency interfacial waves (IWs) may break when approaching the \u27internal coastline,\u27 where the undisturbed pycnocline intersects the shoaling bottom. This process has been studied previously in idealized laboratory and numerical experiments but there are few field observations to document the properties of IWs shoaling on natural internal beaches. This paper presents observations of currents, density and turbulence collected inshore of the break point of an internal beach of the St. Lawrence Estuary. A series of large- and small-amplitude complex-shaped and unorganized internal boluses was observed. Including such effects in numerical models demands improvements in the understanding of several aspects of the energetics, especially those relating to turbulence generation, and observations are needed to build this understanding. To address some of these issues in the estuarine context, we undertook an intensive field program for 10 days in the summer of 2008 in the St. Lawrence Estuary. The sampling involved shore-based photogrammetry, ship-based surveys, and an array of moorings in the shoaling region that held both conventional and turbulence-resolving sensors. The measurements shed light on many aspects of the wave shoaling process. Wave arrivals were generally phase-locked with the M2 tide, providing hints about far-field forcing. In the deeper part of the study domain, the waves propagated according to the predictions of linear theory. In intermediate-depth waters, the waves traversed the field site perpendicularly to isobaths, a pattern that continued as the waves transformed nonlinearly. Acoustic Doppler velocimeters permitted inference of the turbulent energetics, and two main features were studied. First, during a period of shoaling internal waves, turbulence dissipation rates exceeded values associated with tidal shear by an order of magnitude. Second, the evolving spectral signatures associated with a particular wave-shoaling event suggest that the turbulence is at least partly locally generated. Overall, the results of this study suggest that parameterizations of wave-induced mixing could employ relatively simple dynamics in deep water, but may have to handle a wide suite of turbulence generation and transport mechanisms in inshore regions.The work was supported by the Killam Foundation, the Natural Sciences and Engineering Research Council of Canada, the Canadian Foundation for Innovation, the Canadian Foundation for Climate and Atmospheric Sciences, and the Canadian Department of Fisheries and Oceans.2013-07-3

    A Novel Device for the Measurement of the Mechanical and Magnetic Axes of Superconducting Magnet Assemblies for Accelerators

    In the context of the LHC superconducting magnet production, especially for dipoles and quadrupoles due to their complexity, it is foreseen to perform acceptance tests, at an early production stage, to detect possible significant deviations from the design values. The knowledge of the magnetic field geometry is very important, especially for the main magnets. In order to get this information a new device has been conceived that measures the magnets at room temperature during different stages of construction. This device incorporates a sensitive measuring probe and an efficient data acquisition system because the coils are only powered at about 10-5 of the nominal D.C. current. It is dedicated to Quadrupole and Dipole (by using Quadrupole-Configured Dipole (QCD) transformation) magnets, but is also easily adaptable to higher order magnets (n = 3, 4 and 5) by specific orientation of the search coils. It is equipped with magnetic sensors (4 fixed tangential coils and AC excitation current for the magnet) and position sensors (3D-laser tracker and light reflector) that allow the simultaneous detection of the magnetic field axis and the cold bore axis. It is equipped as well with a set of 4 LEDs and associated with a CCD camera that allows both the measurement of the cold bore diameter and its position with respect to the mole.     Regional variations in early life stages response to a temperature gradient in the northern shrimp Pandalus borealis and vulnerability of the populations to ocean warming

    In order to define the relative vulnerability of northern shrimp (Pandalus borealis) populations to the ongoing global warming, we compared the thermal performance curves for survival and growth in the first three pelagic larval stages from three populations of the Northwest Atlantic. Egg carrying females were obtained from different regions characterized by distinct sea surface temperature (SST) conditions for larval development in spring. Two independent experiments were conducted in two different years. In spring 2012, larvae from females captured in the Lower St Lawrence Estuary (LE) and in the Northeast Gulf of St Lawrence (GSL) were compared. In spring 2014, larvae from females captured in the LE and on the Labrador–Newfoundland Shelf (Northwest Atlantic, NWA) were used. The LE larvae were used both years and served as the reference population for comparisons. In 2012 and 2014, groups of 25 newly hatched northern shrimp larvae from each source population were incubated at six temperatures (0, 3, 6, 9, 12, and 15 °C) to monitor and compare survival and growth at moult. Northern shrimp larvae from the LE (warmer May–June SST) had a higher optimal temperature range for survival compared to larvae from the GSL and the NWA (colder May–June SST) populations. However, in 2012 growth performance at moult was reduced at higher temperatures for the LE population compared to the GSL population. The differences in thermal performance curves observed may suggest the presence of a certain level of local adaptation in response to the different regional SST regimes in spring–early summer. Northern shrimp larvae in the Northeast Gulf of St Lawrence and Northwest Atlantic shelf could benefit from warmer early-spring temperatures; however, larvae from the Lower Estuary may be closer to their upper tolerance limits and thus more likely at risk of negative impact of future warming of surface water masses. -- Keywords : Northern shrimp ; Larval survival ; Larval growth ; Macrophysiology ; Conservation physiology ; Climate change

    Middle and Late Pleistocene environmental history of the Marsworth area, south-central England

    To elucidate the Middle and Late Pleistocene environmental history of south-central England, we report the stratigraphy, sedimentology, palaeoecology and geochronology of some deposits near the foot of the Chiltern Hills scarp at Marsworth, Buckinghamshire. The Marsworth site is important because its sedimentary sequences contain a rich record of warm stages and cold stages, and it lies close to the Anglian glacial limit. Critical to its history are the origin and age of a brown pebbly silty clay (diamicton) previously interpreted as weathered till. The deposits described infill a river channel incised into chalk bedrock. They comprise clayey, silty and gravelly sediments, many containing locally derived chalk and some with molluscan, ostracod and vertebrate remains. Most of the deposits are readily attributed to periglacial and fluvial processes, and some are dated by optically stimulated luminescence to Marine Isotope Stage (MIS) 6. Although our sedimentological data do not discriminate between a glacial or periglacial interpretation of the diamicton, amino-acid dating of three molluscan taxa from beneath it indicates that it is younger than MIS 9 and older than MIS 5e. This makes a glacial interpretation unlikely, and we interpret the diamicton as a periglacial slope deposit. The Pleistocene history reconstructed for Marsworth identifies four key elements: (1) Anglian glaciation during MIS 12 closely approached Marsworth, introducing far-travelled pebbles such as Rhaxella chert and possibly some fine sand minerals into the area. (2) Interglacial environments inferred from fluvial sediments during MIS 7 varied from fully interglacial conditions during sub-stages 7e and 7c, cool temperate conditions during sub-stage 7b or 7a, temperate conditions similar to those today in central England towards the end of the interglacial, and cool temperate conditions during sub-stage 7a. (3) Periglacial activity during MIS 6 involved thermal contraction cracking, permafrost development, fracturing of chalk bedrock, fluvial activity, slopewash, mass movement and deposition of loess and coversand. (4) Fully interglacial conditions during sub-stage 5e led to renewed fluvial activity, soil formation and acidic weathering

    Lack of Knowledge of HIV Status a Major Barrier to HIV Prevention, Care and Treatment Efforts in Kenya: Results from a Nationally Representative Study

    BACKGROUND: We analyzed HIV testing rates, prevalence of undiagnosed HIV, and predictors of testing in the Kenya AIDS Indicator Survey (KAIS) 2007. METHODS: KAIS was a nationally representative sero-survey that included demographic and behavioral indicators and testing for HIV, HSV-2, syphilis, and CD4 cell counts in the population aged 15-64 years. We used gender-specific multivariable regression models to identify factors independently associated with HIV testing in sexually active persons. RESULTS: Of 19,840 eligible persons, 80% consented to interviews and blood specimen collection. National HIV prevalence was 7.1% (95% CI 6.5-7.7). Among ever sexually active persons, 27.4% (95% CI 25.6-29.2) of men and 44.2% (95% CI 42.5-46.0) of women reported previous HIV testing. Among HIV-infected persons, 83.6% (95% CI 76.2-91.0) were unaware of their HIV infection. Among sexually active women aged 15-49 years, 48.7% (95% CI 46.8-50.6) had their last HIV test during antenatal care (ANC). In multivariable analyses, the adjusted odds ratio (AOR) for ever HIV testing in women ≥35 versus 15-19 years was 0.2 (95% CI: 0.1-0.3; p<0.0001). Other independent associations with ever HIV testing included urban residence (AOR 1.6, 95% CI: 1.2-2.0; p = 0.0005, women only), highest wealth index versus the four lower quintiles combined (AOR 1.8, 95% CI: 1.3-2.5; p = 0.0006, men only), and an increasing testing trend with higher levels of education. Missed opportunities for testing were identified during general or pregnancy-specific contacts with health facilities; 89% of adults said they would participate in home-based HIV testing. CONCLUSIONS: The vast majority of HIV-infected persons in Kenya are unaware of their HIV status, posing a major barrier to HIV prevention, care and treatment efforts. New approaches to HIV testing provision and education, including home-based testing, may increase coverage. Targeted interventions should involve sexually active men, sexually active women without access to ANC, and rural and disadvantaged populations

    Vinculin–actin interaction couples actin retrograde flow to focal adhesions, but is dispensable for focal adhesion growth

    In migrating cells, integrin-based focal adhesions (FAs) assemble in protruding lamellipodia in association with rapid filamentous actin (F-actin) assembly and retrograde flow. How dynamic F-actin is coupled to FA is not known. We analyzed the role of vinculin in integrating F-actin and FA dynamics by vinculin gene disruption in primary fibroblasts. Vinculin slowed F-actin flow in maturing FA to establish a lamellipodium–lamellum border and generate high extracellular matrix (ECM) traction forces. In addition, vinculin promoted nascent FA formation and turnover in lamellipodia and inhibited the frequency and rate of FA maturation. Characterization of a vinculin point mutant that specifically disrupts F-actin binding showed that vinculin–F-actin interaction is critical for these functions. However, FA growth rate correlated with F-actin flow speed independently of vinculin. Thus, vinculin functions as a molecular clutch, organizing leading edge F-actin, generating ECM traction, and promoting FA formation and turnover, but vinculin is dispensible for FA growth

    Quantum Dots for Multiplexed Detection and Characterisation of Prostate Cancer Cells Using a Scanning Near-Field Optical Microscope

    In this study scanning near-field optical microscopy (SNOM) has been utilised in conjunction with quantum dot labelling to interrogate the biomolecular composition of cell membranes. The technique overcomes the limits of optical diffraction found in standard fluorescence microscopy and also yields vital topographic information. The technique has been applied to investigate cell-cell adhesion in human epithelial cells. This has been realised through immunofluorescence labelling of the cell-cell adhesion protein E-cadherin. Moreover, a dual labelling protocol has been optimised to facilitate a comparative study of the adhesion mechanisms and the effect of aberrant adhesion protein expression in both healthy and cancerous epithelial cells. This study reports clear differences in the morphology and phenotype of healthy and cancerous cells. In healthy prostate epithelial cells (PNT2), E-cadherin was predominantly located around the cell periphery and within filopodial extensions. The presence of E-cadherin appeared to be enhanced when cell-cell contact was established. In contrast, examination of metastatic prostate adenocarcinoma cells (PC-3) revealed no E-cadherin labelling around the periphery of the cells. This lack of functional E-cadherin in PC-3 cells coincided with a markedly different morphology and PC-3 cells were not found to form close cell-cell associations with their neighbours. We have demonstrated that with a fully optimised sample preparation methodology, multiplexed quantum dot labelling in conjunction with SNOM imaging can be successfully applied to interrogate biomolecular localisation within delicate cellular membranes
