Session E8: Fish Migration Possibilities in the Netherlands; State of the Art (Barriers, Solutions, Monitoring)
Abstract:
Measures to restore fish migration is gaining a major boost in the Netherlands since the Water Framework Directive, Benelux Regulation and Eel Directive are operational. Water managers, responsible for a good ecological status, are working hard to reach free fish migration in their waters.
Currently, 1.124 measures are realized at barriers like weirs, pumping stations, sluices and culverts. 2.745 barriers are identified. Since 2000, there is a strong development of measures and new techniques.
These results are based on a study “The Netherlands living with Fish Migration”, that was started in 2008. Target of this study was the evaluation of the progress in approach of measures and to gather information on improvement of fish migration in the Netherlands. Within this project a database was created with the prioritized barriers and measures for fish migration. This data is supplied by the water managers. Every four years water managers are asked for an update and an evaluation is carried out.
In this presentation the results are discussed, with focus on type of barrier, solutions and intensity of monitoring carried out by the water managers. Also an overview of planning, with respect to WFD targets, is given. A map is created of for migratory fish accessible inland waters in up- and downstream direction. Database and maps are good medium for exchanging knowledge between water managers and other stakeholders
Using the lost letter technique to measure real-life behavioral effects of alcohol use
Introduction and aims: People’s behaviors after alcohol use are more extreme and alcohol has a narrowing effect on a person’s cognitive capacity. We tested this effect, making use of the Lost-Letter Technique (LLT), in which the return rate of “lost letters” by public mail is used to measure altruistic behavior. We hypothesized that return rates would be lower when people are under the influence of alcohol, and that an anti-alcohol prime in the address would even further decrease return rates. Design and methods: 768 letters were dropped, half of those at times when some degree of intoxication was likely versus when sober passers-by were common. Three different addressees were used to convey an anti-alcohol cue, a charitable cue, and a neutral cue. Results: The average response rate was 33.1%, and was significantly lower during late bar hours than during lunch hours (20.3% versus 45.8%). Also, when envelopes were found by people who were more likely to have consumed alcohol, anti-alcohol cues gained significantly lower response rates (12.7% versus 23.1%). Discussion and conclusions: This study confirms that alcohol use causes less altruistic behavior and different responses to alcohol-related cues, suggesting that LLT is a promising method to study people’s behavior. Future research could also focus on other substances, (mood-)states, and technology as a means of data collection

The parental overprotection scale: associations with child and parental anxiety
Background: Parental overprotection has commonly been implicated in the development and maintenance of childhood anxiety disorders. Overprotection has been assessed using questionnaire and observational methods interchangeably; however, the extent to which these methods access the same construct has received little attention. Edwards, 2008 and Edwards et al., 2010 developed a promising parent-report measure of overprotection (OP) and reported that, with parents of pre-school children, the measure correlated with observational assessments and predicted changes in child anxiety symptoms. We aimed to validate the use of the OP measure with mothers of children in middle childhood, and examine its association with child and parental anxiety.
Methods: Mothers of 90 children (60 clinically anxious, 30 non-anxious) aged 7–12 years completed the measure and engaged in a series of mildly stressful tasks with their child.
Results: The internal reliability of the measure was good and scores correlated significantly with observations of maternal overprotection in a challenging puzzle task. Contrary to expectations, OP was not significantly associated with child anxiety status or symptoms, but was significantly associated with maternal anxiety symptoms.
Limitations: Participants were predominantly from affluent social groups and of non-minority status. Overprotection is a broad construct, the use of specific sub-dimensions of behavioural constructs may be preferable.
Conclusions: The findings support the use of the OP measure to assess parental overprotection among 7–12 year-old children; however, they suggest that parental responses may be more closely related to the degree of parental rather than child anxiety
Comprehensive diagnostics of acute myeloid leukemia by whole transcriptome RNA sequencing
Acute myeloid leukemia (AML) is caused by genetic aberrations that also govern the prognosis of patients and guide risk-adapted and targeted therapy. Genetic aberrations in AML are structurally diverse and currently detected by different diagnostic assays. This study sought to establish whole transcriptome RNA sequencing as single, comprehensive, and flexible platform for AML diagnostics. We developed HAMLET (Human AML Expedited Transcriptomics) as bioinformatics pipeline for simultaneous detection of fusion genes, small variants, tandem duplications, and gene expression with all information assembled in an annotated, user-friendly output file. Whole transcriptome RNA sequencing was performed on 100 AML cases and HAMLET results were validated by reference assays and targeted resequencing. The data showed that HAMLET accurately detected all fusion genes and overexpression of EVI1 irrespective of 3q26 aberrations. In addition, small variants in 13 genes that are often mutated in AML were called with 99.2% sensitivity and 100% specificity, and tandem duplications in FLT3 and KMT2A were detected by a novel algorithm based on soft-clipped reads with 100% sensitivity and 97.1% specificity. In conclusion, HAMLET has the potential to provide accurate comprehensive diagnostic information relevant for AML classification, risk assessment and targeted therapy on a single technology platform
Recommendations for the introduction of metagenomic high-throughput sequencing in clinical virology, part I:Wet lab procedure
Metagenomic high-throughput sequencing (mHTS) is a hypothesis-free, universal pathogen detection technique for determination of the DNA/RNA sequences in a variety of sample types and infectious syndromes. mHTS is still in its early stages of translating into clinical application. To support the development, implementation and standardization of mHTS procedures for virus diagnostics, the European Society for Clinical Virology (ESCV) Network on Next-Generation Sequencing (ENNGS) has been established. The aim of ENNGS is to bring together professionals involved in mHTS for viral diagnostics to share methodologies and experiences, and to develop application recommendations. This manuscript aims to provide practical recommendations for the wet lab procedures necessary for implementation of mHTS for virus diagnostics and to give recommendations for development and validation of laboratory methods, including mHTS quality assurance, control and quality assessment protocols
The human DEK oncogene regulates DNA damage response signaling and repair
The human DEK gene is frequently overexpressed and sometimes amplified in human cancer. Consistent with oncogenic functions, Dek knockout mice are partially resistant to chemically induced papilloma formation. Additionally, DEK knockdown in vitro sensitizes cancer cells to DNA damaging agents and induces cell death via p53-dependent and -independent mechanisms. Here we report that DEK is important for DNA double-strand break repair. DEK depletion in human cancer cell lines and xenografts was sufficient to induce a DNA damage response as assessed by detection of γH2AX and FANCD2. Phosphorylation of H2AX was accompanied by contrasting activation and suppression, respectively, of the ATM and DNA-PK pathways. Similar DNA damage responses were observed in primary Dek knockout mouse embryonic fibroblasts (MEFs), along with increased levels of DNA damage and exaggerated induction of senescence in response to genotoxic stress. Importantly, Dek knockout MEFs exhibited distinct defects in non-homologous end joining (NHEJ) when compared to their wild-type counterparts. Taken together, the data demonstrate new molecular links between DEK and DNA damage response signaling pathways, and suggest that DEK contributes to DNA repair
