    Wikification of learning objects using metadata as an alternative context for disambiguation

    We present a methodology to wikify learning objects. Our proposal is focused on two processes: word sense disambiguation and relevant phrase selection. The disambiguation process involves the use of the learning objects metadata as either additional or alternative context. This increases the probability of success when a learning object has a low quality context. The selection of relevant phrases is perf ormed by identifying the highest values of semantic relat edness between the main subject of a learning object and t he phrases. This criterion is useful for achieving the didactic objectives of the learning object

    Fibronectin modulates the endocannabinoid system through the cAMP/PKA pathway during human sperm capacitation

    Fibronectin (Fn) enhances human sperm capacitation via the cAMP/PKA pathway, and the endocannabinoid system participates in this process. Moreover, Fn has been linked to endocannabinoid system components in different cellular models, even though no evidence of such interactions in human sperm is available. Normal semen samples were evaluated over a 4-year period. Our findings suggest that (a) the capacitating effects of Fn were reversed by preincubating the sperm with a cannabinoid receptor 1 (CB1) or transient receptor potential cation channel subfamily V member 1 (TRPV1) antagonist (p < 0.001 and p < 0.05, respectively); (b) cooperation between CB1 and TRPV1 may exist (p < 0.01); (c) the activity of specific fatty acid amide hydroxylase (FAAH) decreased after 1 min (p < 0.01) and increased after 60 min (p < 0.01) of capacitation in the presence of Fn; (d) the effects of Fn on FAAH activity were prevented by preincubating spermatozoa with a protein kinase A (PKA) inhibitor (p < 0.01); (e) Fn modulated both the cyclic adenosine monophosphate concentration and PKA activity (p < 0.05) during early capacitation; and (f) FAAH was a PKA substrate modulated by phosphorylation. These findings indicate that Fn stimulates human sperm capacitation via the cAMP/PKA pathway through modulation of the endocannabinoid system. Understanding the functional competence of human spermatozoa is essential for facilitating clinical advances in infertility treatment and for developing novel contraceptive strategies.Fil: Martínez León, Eduardo Antonio.     Callus Induction and Cellular Suspensions from Murtilla (Ugni molinae Turcz.) for trans-resveratrol Production

    The present study reports for the first time the quantification of resveratrol and the use of biotechnological techniques applied to stilbene productions in species from genus Ugni and calli production from adult explants (fruits). Resveratrol is synthesized by a series of families of higher plants, which has generated much interest in recent years for its antioxidant, anticancer and antitumor properties, which would allow longevity of cells to be prolonged. In this study, leaves and mature fruits were collected from three sites in Southern Chile and correspond to three ecotypes of the Chilean endemic species Ugni molinae Turz. (murtilla). These were established in vitro to prepare the callus and subsequent development of cellular suspensions for trans-resveratrol production. Our results showed that these stilbenes are present in murtilla and that their concentrations vary between ecotypes and tissues, reaching up to 553.5 µg g-1 of t-resveratrol produced in ecotype 3 callus. These values are relatively higher than those found in other plant species. Under optimum culture conditions, extraction of resveratrol from Ugni molinae is scalable to industrial levels, which makes it a viable alternative for obtaining stilbenes.This research was supported by a project of the Innova BíoBío, Corfo-Chile No.12.247

    En cuanto al uso de los conceptos para organizar el conocimiento, Hjorland indica que éstos son concebidos como unidades de pensamiento, y la relación interconceptual es un elemento fundamental ya que los paradigmas, con sus teorías y conceptualizaciones, son por naturaleza instrumentos clasificadores de la realidad con la que interactúa de continuo el individuo. De esta forma, las relaciones semánticas que se establecen entre los conceptos son básicas para sistematizar el conocimiento, pero su significado queda sujeto a la perspectiva teórica desde la cual sean considerados.Según la UNESCO (2005a) la diversidad de las modalidades de acceso al conocimiento constituye una de las características más importantes de la sociedad del aprendizaje y supone también, entre otras cosas, el cuestionamiento de los conceptos de inteligencia en los que prevalecía el carácter estable de los procesos de evaluación y trasmisión de los conocimientos. Así, en el contexto de la realidad actual, la tarea de discriminar información se vuelve un proceso casi imposible, la creciente información actual, principalmente por vía Internet, le resulta al pensamiento humano cada vez más difícil de filtrar, tratar y dominar; lo cual se traduce en barreras (número ilimitado de fuentes, desconocimiento de los mecanismos de filtrado, organización o apropiación cognitiva de la información) que impiden hacer un uso efectivo de ella. Así, el exceso de información no significa mejor comprensión de la realidad, sino que con frecuencia alarmante produce el efecto contrario (UNESCO, 2005a; Dudziacck 2003; Sahagún, 2004, citados por Peña, 2011)

    Xanthinuria: a rare cause of urolithiasis in the cat

    Xanthinuria is a very rare disease in cats. Its etiology may have a genetic origin or may be due to an iatrogenic xan- thine-dehydrogenase inhibition that nally results in urolithiasis. The present work reports two cases of xanthine uro- lithiasis in European Shorthair unrelated male and female cats. Both uroliths were analyzed by stereoscopic microsco- py, infrared spectroscopy and scanning electron microscopy. Besides the report of these two clinical cases, a detailed pathophysiologic review and some updated recommendations for diagnosis and treatment for this condition were done.La xantinuria es una patología que se presenta raramente en los gatos. Su etiología puede tener origen genético o de- berse a una inhibición yatrogénica de la enzima xantina deshidrogenasa, que generalmente se mani esta con urolitiasis. En este trabajo se informa el hallazgo de dos urolitos de xantina en dos gatos, un macho y una hembra, de raza Euro- pea de pelo corto, no emparentados. Los urolitos fueron analizados mediante microscopía estereoscópica, espectrosco- pía infrarroja y microscopía electrónica de barrido.     Personalized Modeling of Atrial Activation and P-waves: a Comparison Between Invasive and Non-Invasive Cardiac Mapping

    Biatrial personalized models incorporating functional and anatomical features are becoming a promising tool for planning therapy for patients with atrial fibrillation (AF). Conduction velocity (CV) is one of the main features to be matched during the process of functional personalization, as it can identify electrical abnormalities in the cardiac tissue. The spatial distribution of CV can be estimated from local activation times (LAT) maps from non-invasive electrocardiographic imaging (ECGI) or invasive electroanatomical mapping systems (EAMS). We investigated the effect of using either invasive LAT maps from EAMS or non-invasive LAT maps from ECGI to personalize two biatrial models by comparing the virtual P-waves obtained from these LAT maps with the measured P-waves from the surface electrocardiogram (ECG). For both modalities – ECGI and EAMS – we found a qualitative match between simulated and measured P-waves but observed quantitative differences. The root-mean-square error (RMSE) between measured and simulated signals for patient A was 0.26±0.11 mV and 0.38±0.31 mV, while for patient B it was 0.21±0.09 mV and 0.14±0.05 mV for EAMS and ECGI, respectively. The correlation between measured and simulated signals from ECGI and EAMS was 0.69±0.34 and 0.71±0.26 for patient A and 0.71±0.18 and 0.72±0.18 for patient B. Our results suggest that LAT maps from ECGI and EAMS show differences, which are also reflected in the computed P-wave on the body surface

    Human fluids alter DNA-acquisition in Acinetobacter baumannii

    Transformation is one of the mechanisms of acquisition of foreign genetic material leading to the emergence of multidrug resistant (MDR) bacteria. Recently, human serum albumin (HSA) was shown to specifically increase transformation frequency in the nosocomial pathogen Acinetobacter baumannii. To further assess the relevance of HSA as a possible modulator of A. baumannii transformation in host-pathogen interactions, in this work we examined the effect of different human fluids. We observed a significant increase in transformation frequencies in the presence of pleural fluid, whole blood cells and liquid ascites, and to a lesser extent with urine. The observed effects correlate with both HSA and bacterial content found in the assayed patient fluids. Taken together, these results are in agreement with our previous findings that highlight HSA as a possible host signal with the ability to trigger natural transformation in A. baumannii.Fil: Martinez, Jasmine.     Cyclic AMP efflux, via MRPs and A1 adenosine receptors, is critical for bovine sperm capacitation

    Sperm capacitation has been largely associated with an increase in cAMP, although its relevance in the underlying mechanisms of this maturation process remains elusive. Increasing evidence shows that the extrusion of cAMP through multidrug resistance associated protein 4 (MRP4) regulates cell homeostasis not only in physiological but also in pathophysiological situations and studies from our laboratory strongly support this assumption. In the present work we sought to establish the role of cAMP efflux in the regulation of sperm capacitation. Sperm capacitation was performed in vitro by exposing bovine spermatozoa to bicarbonate 40 and 70 mM; cAMP; probenecid (a MRPs general inhibitor) and an adenosine type 1 receptor (A1 adenosine receptor) selective antagonist (DPCPX). Capacitation was assessed by chlortetracycline assay and lysophosphatidylcholine-induced acrosome reaction assessed by PSA-FITC staining. Intracellular and extracellular cAMP was measured by radiobinding the regulatory subunit of PKA under the same experimental conditions. MRP4 was detected by western blot and immunohistochemistry assays. Results showed that the inhibition of soluble adenylyl cyclase significantly inhibited bicarbonate-induced sperm capacitation. Furthermore, in the presence of 40 and 70 mM bicarbonate bovine spermatozoa synthesized and extruded cAMP. Interestingly, in the absence of IBMX (a PDEs inhibitor) cAMP efflux still operated in sperm cells, suggesting that cAMP extrusion would be a physiological process in the spermatozoa complementary to the action of PDE. Blockade of MRPs by probenecid abolished the efflux of the cyclic nucleotide resulting not only in the accumulation of intracellular cAMP but also in the inhibition of bicarbonate-induced sperm capacitation. The effect of probenecid was abolished by exposing sperm cells to cAMP. The high-affinity efflux pump for cAMP, MRP4 was expressed in bovine spermatozoa and localized to the midpiece of the tail as previously reported for soluble adenylyl cyclase and A1 adenosine receptor. Additionally, blockade of A1 adenosine receptor abolished not only bicarbonate-induced sperm capacitation but also that stimulated by cAMP. Present findings strongly support that cAMP efflux, presumably through MRP4, and the activation of A1 adenosine receptor regulate some events associated with bicarbonate-induced sperm capacitation, and further suggest a paracrine and/or autocrine role for cAMP.Fil: Osycka Salut, Claudia Elena.     Progress towards Elimination of HIV Mother-to-Child Transmission in the Dominican Republic from 1999 to 2011

    In 1999, prevention of mother-to-child transmission (pMTCT) using antiretrovirals was introduced in the Dominican Republic (DR). Highly active antiretroviral therapy (HAART) was introduced for immunosuppressed persons in 2004 and for pMTCT in 2008. To assess progress towards MTCT elimination, data from requisitions for HIV nucleic acid amplification tests for diagnosis of HIV infection in perinatally exposed infants born in the DR from 1999 to 2011 were analyzed. The MTCT rate was 142/1,274 (11.1%) in 1999?2008 and 12/302 (4.0%) in 2009?2011 (P \u3c .001), with a rate of 154/1,576 (9.8%) for both periods combined. This decline was associated with significant increases in the proportions of women who received prenatal HAART (from 12.3% to 67.9%) and infants who received exclusive formula feeding (from 76.3% to 86.1%) and declines in proportions of women who received no prenatal antiretrovirals (from 31.9% to 12.2%) or received only single-dose nevirapine (from 39.5% to 19.5%). In 2007, over 95% of DR pregnant women received prenatal care, HIV testing, and professionally attended delivery. However, only 58% of women in underserved sugarcane plantation communities (2007) and 76% in HIV sentinel surveillance hospitals (2003?2005) received their HIV test results. HIV-MTCT elimination is feasible but persistent lack of access to critical pMTCT measures must be addressed

    Re-thinking the Etiological Framework of Neurodegeneration

    Neurodegenerative diseases are among the leading causes of disability and death worldwide. The disease-related socioeconomic burden is expected to increase with the steadily increasing life expectancy. In spite of decades of clinical and basic research, most strategies designed to manage degenerative brain diseases are palliative. This is not surprising as neurodegeneration progresses "silently" for decades before symptoms are noticed. Importantly, conceptual models with heuristic value used to study neurodegeneration have been constructed retrospectively, based on signs and symptoms already present in affected patients;a circumstance that may confound causes and consequences. Hence, innovative, paradigm-shifting views of the etiology of these diseases are necessary to enable their timely prevention and treatment. Here, we outline four alternative views, not mutually exclusive, on different etiological paths toward neurodegeneration. First, we propose neurodegeneration as being a secondary outcome of a primary cardiovascular cause with vascular pathology disrupting the vital homeostatic interactions between the vasculature and the brain, resulting in cognitive impairment, dementia, and cerebrovascular events such as stroke. Second, we suggest that the persistence of senescent cells in neuronal circuits may favor, together with systemic metabolic diseases, neurodegeneration to occur. Third, we argue that neurodegeneration may start in response to altered body and brain trophic interactions established via the hardwire that connects peripheral targets with central neuronal structures or by means of extracellular vesicle (E\-mediated communication. Lastly, we elaborate on how lifespan body dysbiosis may be linked to the origin of neurodegeneration. We highlight the existence of bacterial products that modulate the gut-brain axis causing neuroinflammation and neuronal dysfunction. As a concluding section, we end by recommending research avenues to investigate these etiological paths in the future. We think that this requires an integrated, interdisciplinary conceptual research approach based on the investigation of the multimodal aspects of physiology and pathophysiology. It involves utilizing proper conceptual models, experimental animal units, and identifying currently unused opportunities derived from human data. Overall, the proposed etiological paths and experimental recommendations will be important guidelines for future cross-discipline research to overcome the translational roadblock and to develop causative treatments for neurodegenerative diseases
