    Meglumine antimoniate and miltefosine combined with allopurinol sustain pro-inflammatory immune environments during canine leishmaniosis treatment

    Canine leishmaniosis (CanL) caused by Leishmania infantum is a zoonotic disease of global concern. Antileishmanial drug therapies commonly used to treat sick dogs improve their clinical condition, although when discontinued relapses can occur. Thus, the current study aims to evaluate the effect of CanL treatments in peripheral blood, lymph node, and bone marrow cytokine profile associated with clinical recovery. Two groups of six dogs diagnosed with CanL were treated with miltefosine combined with allopurinol and meglumine antimoniate combined with allopurinol (MT+A and MG+A), respectively. At diagnosis and after treatment, during a 3-month follow-up, clinical signs, hematological and biochemical parameters, urinalysis results and antileishmanial antibody titers were registered. Furthermore, peripheral blood, popliteal lymph node, and bone marrow samples were collected to assess the gene expression of IL-2, IL-4, IL-5, IL-10, IL-12, TNF-α, TGF-β, and IFN-γ by qPCR. In parallel, were also evaluated samples obtained from five healthy dogs. Both treatment protocols promoted the remission of clinical signs as well as normalization of hematological and biochemical parameters and urinalysis values. Antileishmanial antibodies returned to non-significant titers in all dogs. Sick dogs showed a generalized upregulation of IFN-γ and downregulation of IL-2, IL-4, and TGF-β, while gene expression of IL-12, TNF-α, IL-5, and IL-10 varied between groups and according to evaluated tissue. A trend to the normalization of cytokine gene expression was induced by both miltefosine and meglumine antimoniate combined therapies. However, IFN-γ gene expression was still up-regulated in the three evaluated tissues. Furthermore, the effect of treatment in the gene expression of cytokines that were not significantly changed by infection, indicates that miltefosine and meglumine antimoniate combined therapy directly affects cytokine generation. Both combined therapies are effective in CanL treatment, leading to sustained pro-inflammatory immune environments that can compromise parasite survival and favor dogs' clinical cure. In the current study, anti-inflammatory and regulatory cytokines do not seem to play a prominent role in CanL or during clinical recovery.publishersversionpublishe

    The understanding of information about oral chemotherapy treatment among outpatients at a University Hospital in Ceará, Brazil

    This study assessed the understanding of information about oral chemotherapy treatment, among patients attending the hematology outpatients' ward of a University hospital. A questionnaire with patient data and details of the treatment was applied to 147 outpatients. The analysis of variables and classification of the level of understanding about the treatment followed methods established in the literature with some adjustments. The study found a significant association between the level of understanding about treatment with the level of education and the occupation of the respondents. Regarding the correct acquisition and administration of the medicine-its name, dose and frequency of administration-there was agreement between the medical prescriptions and the responses for 90.7, 88.0 and 99.3 % of patients, respectively. Patients had less knowledge about the duration and side-effects of the treatment. The level of treatment understanding was classified as good for 74.1 % of those interviewed, suggesting a rational use of oral chemotherapy by the majority of patients.Colegio de Farmacéuticos de la Provincia de Buenos Aire

    Estado e agricultores familiares: uma análise interpretativa sobre o desenvolvimento rural no Sul de Minas Gerais.

    Este trabalho procurou identificar como os atores sociais vinculados à agricultura familiar (lideranças de produtores familiares, profissionais de ciências agrárias, autoridades municipais, pesquisadores e professores universitários) interpretam o papel do Estado para o desenvolvimento dessa categoria na região sul de Minas Gerais. Com relação à coleta de dados, utilizou-se a técnica "focused interview". Os entrevistados foram seledonados pelo método não-probabilístico de amostragem por julgamento. Os resultados revelaram que a interpretação feita pelos entrevistados aproxima-se mais da perspectiva do Estado patrimonialistaburocrático autoritário, tendo em vista que os depoimentos dos entrevistados corroboram com a reprodução das relações de dependência dos agricultores familiares em relação ao Estado. Por outro lado, alguns atores reconhecem a necessidade de mecanismos que incentivem a participação social, no entanto, salientaram que a mobilização dos produtores familiares na região ainda é incipiente e desarticulada

    Chemical Composition and Evaluation of Antitumoral Activity of Leaf and Root Essential Oils of Conyza canadensis (Asteraceae)

    The leaf and root oils of Conyza canadensis were studied for chemical composition and antitumor activity. The results showed that there is great variation in the composition of the oils obtained from different parts. The main components in the leaf oil were limonene, caryophyllene oxide and espatulenol. In the root oil the major component was the acetylenic ester lachnophyllum methyl ester. It was observed that according to the collection time (6 and 16 hours), significant variations in the content of the main components of this essential oil of leaves can occur. Limonene, spatulenol and caryophyllene oxide presented a distribution of 61% / 5.4% / 12.5% ​​and 38% / 10.7% / 22.3% in oils obtained from plants collected at 6 and 16 hours, respectively. The antitumor activity of the oils showed that leaf oil had a greater potential for inhibition, and this oil was distinguished by the activity against neoplastic cell lines K562 (leukemia) and NCI-ADR / RES (ovary with multidrug resistance phenotype ) with TGI values ​​of 16.8 and 19.0 mg.mL-1, respectively. Comparing the leaf oils and their tumor cell inhibition potentials, it was noted that this activity is higher in the oil with higher contents of monoterpene limonene.     It is an unequivocal fact that Amazonian tropical forest is the largest remaining primary forest in the world. The ecosystem in the region is e tremely comple with high biodiversity (Peres et al. 2010). Conservation and protection of the dynamic forest and river regions is e tremely important not only for the natural environments, but also for the economy and social dependence of benefits from such abundant natural environments. Important natural parameters that affect status of the natural environments include light (natural sunlight), soil, and water, which abundantly e ist in the Amazon region. Solar energy is the primary energy source for the majority of living organisms in both terrestrial and aquatic ecosystems, and drives the diurnal and seasonal cycles of biogeochemical processes (Monteith & Unsworth 2013). In particular, in situ light data remains one of the most underappreciated data measurements although having a significant impact on the physical, chemical and biological processes in the ecosystem (Johnsen 2012). Soil provides the fundamental basis for all terrestrial living organisms including the Amazonian forests as well as life-sustaining infrastructure for human society. Water is the most essential single entity to constitute all organisms from a single cell to the earth. Understanding of importance and roles of each factor and interaction of such comple dynamics in the natural environments can serve as fundamental platform for natural scientists, particularly for young scientists such as university students. The objective of this workshop was to provide hand- on scientific and environmental education for university students in Manaus, Amazonas, Brazil through practical field measurements using the three most important parameters in the natural ecosystem composed of natural sunlight, soil, and water.     Construction and validation of an developmental profile assessment tool for children with autistic spectrum disorder

    Nos últimos anos a investigação tem dado particular relevância às alterações do Processamento Sensorial nas crianças com perturbações do espectro do autismo (PEA) e a literatura refere que entre 42% a 88% das crianças com PEA apresentam este tipo de disfunção. Nesta linha foi definido um projeto de investigação centrado na construção de uma escala que avalie a tradicional tríade que caracteriza as crianças com PEA (Interação, Comunicação e Comportamento e interesses repetitivos e estereotipados), enriquecida pela inclusão de um novo domínio: o Processamento Sensorial. Com a construção e validação desta escala pretendemos que pais e profissionais utilizem colaborativamente um instrumento de avaliação da intervenção que lhes permita monitorizar o processo de apoio e adequar as suas práticas. Neste artigo descrevemos os procedimentos e os resultados das sucessivas fases de construção do instrumento, desde as análises iniciais mais qualitativas até aos estudos centrados na análise quantitativa dos itens.During the last few years, research has focused on changes in Sensory Processing in children with Autistic Spectrum Disorder (ASD). As a result, literature has shown that between 42% and 88% of children with ASD present this type of disorder. Based on these findings, a research project was designed centring on the construction of a tool to assess the traditional triad that characterizes children with ASD (Interaction, Communication and Behaviour and Repetitive and Stereotyped Interests), to which was added a new domain: Sensory Processing. By constructing and validating this assessment tool, the intention is for parents and professionals to collaboratively apply this intervention assessment instrument in order to monitor the support process and adapt their practices.     Spatial patterns and temporal variability of drought in Western Iran

    An analysis of drought in western Iran from 1966 to 2000 is presented using monthly precipitation data observed at 140 gauges uniformly distributed over the area. Drought conditions have been assessed by means of the Standardized Precipitation Index (SPI). To study the long-term drought variability the principal component analysis was applied to the SPI field computed on 12-month time scale. The analysis shows that applying an orthogonal rotation to the first two principal component patterns, two distinct sub-regions having different climatic variability may be identified. Results have been compared to those obtained for the largescale using re-analysis data suggesting a satisfactory agreement. Furthermore, the extension of the large-scale analysis to a longer period (1948–2007) shows that the spatial patterns and the associated time variability of drought are subjected to noticeable changes. Finally, the relationship between hydrological droughts in the two sub-regions and El Niño Southern Oscillation events has been investigated finding that there is not clear evidence for a link between the two phenomen
