    Sustainability in Nutrition & Dietetics: Where are we now?

    With the topic of sustainability becoming a regular feature in all aspects of our lives, this year, the CN Team launched a new CN Award category focusing on sustainability initiatives. In turn, this got us thinking further about the nutrition and dietetic arena as a whole – not just about those of you on the frontline, but also organisations, groups, associations and industry – what action is taking place, and what roles are being developed to actively make nutrition and dietetics more sustainable? So, with this in mind, CN Team members Faye and Jodie sought to do a bit of investigating, in order to discover how the world of nutrition and dietetics is becoming more sustainable! As a result, within this article, we provide a snapshot of initiatives and developments that have, or are, taking place (this is by no means exhaustive), which we hope will further encourage and inspire us all to think more sustainably, and positively acknowledge and recognise what is being done to make an all important difference to the world we live in

    A spatial analytical framework of Toronto using machine learning and spatial regressions

In Canada, injuries are the leading cause of potential years of life lost before the age of 65. The geographical patterns of injury, however, are evident both over space and time, suggesting the possibility of spatial optimization of policies at the neighborhood scale to mitigate injury risk, foster prevention, and control within metropolitan regions. In this paper, Canada’s National Ambulatory Care Reporting System is used to assess unintentional and intentional injuries for Toronto between 2004 and 2010, exploring the spatial relations of injury throughout the city, together with Wellbeing Toronto data. Corroborating with these findings, spatial autocorrelations at global and local levels are performed for the reported over 1.7 million injuries. The sub-categorization for Toronto’s neighborhood further distills the most vulnerable communities throughout the city, registering a robust spatial profile throughout. Individual neighborhoods pave the need for distinct policy profiles for injury prevention. This brings one of the main novelties of this contribution. A comparison of the three regression models is carried out. The findings suggest that the performance of spatial regression models is significantly stronger, showing evidence that spatial regressions should be used for injury research. Wellbeing Toronto data performs reasonably well in assessing unintentional injuries, morbidity, and falls. Less so to understand the dynamics of intentional injuries. The results enable a framework to allow tailor-made injury prevention initiatives at the neighborhood level as a vital source for planning and participatory decision making in the medical field in developed cities such as Toronto.publishersversionpublishe

Its industry focuses predominantly on the service sector and immediate economic growth seems to steer India into the economic superpower. India's unique business landscape is felt at a regional level, where massive urbanization has become an unavoidable consequence of population growth and spatial allocation to the economic hubs of metropolitan cities. Mumbai, one of the world's largest cities, represents a unique combination of a diverse economic landscape and the growth of a megacity. The role of Mumbai in India's growth is of crucial importance for India's business landscape. This paper explores the massive urbanization processes of Mumbai's peri-urban areas and compares urban sprawl with the location of its business landscape. A spatial accounting methodology based on the proximity of Mumbai's different economic hubs will be used to measure the underlying pattern of the Mumbai region, concerning past and present urbanization, and the effect of this urbanization process has on the possible location of businesses. This business-urban ecosystem perspective will be implemented by a spatial analysis on the correlation between urban compactness and urban footprints, in relation to business concentration and its spatiotemporal evolution over the last hundred years.publishersversionpublishe

    Putative regulatory sites unraveled by network-embedded thermodynamic analysis of metabolome data

    As one of the most recent members of the omics family, large-scale quantitative metabolomics data are currently complementing our systems biology data pool and offer the chance to integrate the metabolite level into the functional analysis of cellular networks. Network-embedded thermodynamic analysis (NET analysis) is presented as a framework for mechanistic and model-based analysis of these data. By coupling the data to an operating metabolic network via the second law of thermodynamics and the metabolites' Gibbs energies of formation, NET analysis allows inferring functional principles from quantitative metabolite data; for example it identifies reactions that are subject to active allosteric or genetic regulation as exemplified with quantitative metabolite data from Escherichia coli and Saccharomyces cerevisiae. Moreover, the optimization framework of NET analysis was demonstrated to be a valuable tool to systematically investigate data sets for consistency, for the extension of sub-omic metabolome data sets and for resolving intracompartmental concentrations from cell-averaged metabolome data. Without requiring any kind of kinetic modeling, NET analysis represents a perfectly scalable and unbiased approach to uncover insights from quantitative metabolome data

    Transcriptional regulation of NAD metabolism in bacteria: genomic reconstruction of NiaR (YrxA) regulon

    A comparative genomic approach was used to reconstruct transcriptional regulation of NAD biosynthesis in bacteria containing orthologs of Bacillus subtilis gene yrxA, a previously identified niacin-responsive repressor of NAD de novo synthesis. Members of YrxA family (re-named here NiaR) are broadly conserved in the Bacillus/Clostridium group and in the deeply branching Fusobacteria and Thermotogales lineages. We analyzed upstream regions of genes associated with NAD biosynthesis to identify candidate NiaR-binding DNA motifs and assess the NiaR regulon content in these species. Representatives of the two distinct types of candidate NiaR-binding sites, characteristic of the Firmicutes and Thermotogales, were verified by an electrophoretic mobility shift assay. In addition to transcriptional control of the nadABC genes, the NiaR regulon in some species extends to niacin salvage (the pncAB genes) and includes uncharacterized membrane proteins possibly involved in niacin transport. The involvement in niacin uptake proposed for one of these proteins (re-named NiaP), encoded by the B. subtilis gene yceI, was experimentally verified. In addition to bacteria, members of the NiaP family are conserved in multicellular eukaryotes, including human, pointing to possible NaiP involvement in niacin utilization in these organisms. Overall, the analysis of the NiaR and NrtR regulons (described in the accompanying paper) revealed mechanisms of transcriptional regulation of NAD metabolism in nearly a hundred diverse bacteria

    Streptococcus pyogenes is an important global pathogen, causing considerable morbidity and mortality, especially in low and middle income countries where rheumatic heart disease and invasive infections are common. There is a number of promising vaccine candidates, most notably those based on the M protein, the key virulence factor for the bacterium. Vaccines against Streptococcus pyogenes are considered as impeded vaccines because of a number of crucial barriers to development. Considerable effort is needed by key players to bring current vaccine candidates through phase III clinical trials and there is a clear need to develop a roadmap for future development of current and new candidates

    Relationship between Expression of the Family of M Proteins and Lipoteichoic Acid to Hydrophobicity and Biofilm Formation in Streptococcus pyogenes

    Background: Hydrophobicity is an important attribute of bacteria that contributes to adhesion and biofilm formation. Hydrophobicity of Streptococcus pyogenes is primarily due to lipoteichoic acid (LTA) on the streptococcal surface but the mechanism(s) whereby LTA is retained on the surface is poorly understood. In this study, we sought to determine whether members of the M protein family consisting of Emm (M protein), Mrp (M-related protein), Enn (an M-like protein), and the streptococcal protective antigen (Spa) are involved in anchoring LTA in a manner that contributes to hydrophobicity of the streptococci and its ability to form biofilms. Methodology/Principal Findings: Isogenic mutants defective in expression of emm, mrp, enn, and/or spa genes of eight different serotypes and their parental strains were tested for differences in LTA bound to surface proteins, LTA released into the culture media, and membrane-bound LTA. The effect of these mutations on the ability of streptococci to form a hydrophobic surface and to generate biofilms was also investigated. A recombinant strain overexpressing Emm1 was also engineered and similarly tested. The serotypes tested ranged from those that express only a single M protein gene to those that express two or three members of the M protein family.     Analysis of Wetland Landcover Change in Great Lakes Urban Areas Using Self-Organizing Maps

    Wetland loss and subsequent reduction of wetland ecosystem services in the Great Lakes region has been driven, in part, by changing landcover and increasing urbanization. With landcover change data, digital elevation models (DEM), and self-organizing maps (SOM), this study explores changing landcover and the flood mitigation attributes of wetland areas over a 15-year period in Toronto and Chicago. The results of this analysis show that (1) in the city of Toronto SOM clusters, the landcover change correlations with wetland volume and wetland area range between −0.1 to −0.5, indicating that a more intense landcover change tends to be correlated with small shallow wetlands, (2) in the city of Chicago SOM clusters, the landcover change correlations with wetland area range between −0.1 to −0.7, the landcover change correlations with wetland volume per area range between −0.1 to 0.8, and the landcover change correlations with elevation range between −0.2 to −0.6, indicating that more intense landcover change tends to be correlated with spatially small wetlands that have a relatively high water-storage capacity per area and are located at lower elevations. In both cities, the smallest SOM clusters represent wetland areas where increased landcover change is correlated with wetland areas that have high flood mitigation potential.     Comparative studies of courtship behaviour in sympatric sibling species of Zaprionus (Diptera - Drosophilidae)

    Zaprionus tuberculatus and Z. sepsoides are two sibling species of drosophilid fly which are sympatric throughout tropical Africa. Their sexual behaviour has previously received little study. In this thesis, the courtship behaviours of the two species are described, and the descriptions compared with previously published descriptions by other authors. Three different laboratory-bred strains of Z. sepsoides, and two of Z. tuberculatus, were studied, and their male courtship "songs" (produced by male wing vibration) were compared. The main objects of the project were to study the mechanisms by which members of these two species identify the sex of other individuals (sexual dimorphism being slight); and the isolating mechanisms operating between the two species. Preliminary observations suggested that males could probably identify females by chemosensory cues. However, males would sometimes also court each other. Further experiments on the males' ability to distinguish sex-specific pheromones gave ambiguous results. There were no grounds to suspect that females could distinguish between the sexes by chemosensory means, and further experiments gave no evidence for such an ability. Preliminary observations indicated that that there was an effective pre-mating reproductive barrier between male Z. sepsoides and female Z. tuberculatus, but male Z. tuberculatus were sometimes able to copulate with female Z. sepsoides. These interspecific matings resulted in infertile eggs. Further experiments seemed to confirm that the isolating mechanism between the two species is asymmetric, the onus of species recognition lying largely on Z. sepsoides. Male Z. sepsoides apparently recognise a female's species by chemosensory means, while female Z. sepsoides recognize a male's species by his "Type-2" courtship "song". There was also some evidence that female Z. sepsoides could distinguish between males of the two species by olfactory means. Z. tuberculatus were not shown to be able to distinguish reliably between the two species. The relevance of these findings to current theories about mechanisms of speciation is discussed.</p
