    Rheology of living cells

    The mechanical behavior of living cells, during planktonic growth, has been thoroughly explored combining common biological techniques with rheology and rheo-imaging measurements. Under a shear flow, bacterial cultures of Staphylococcus aureus revealed a complex and rich rheological behavior not usually accessed in biological studies. In particular, in stationary shear flow, the viscosity increased during the exponential phase and returned close to its initial value at the late phase of growth, accompanied by the stabilization of the bacterial population. In oscillatory flow, the elastic and viscous moduli exhibited power-law behaviors whose exponents are dependent on the bacteria growth stage, and can be associated to a Soft Glassy Material behavior. These behaviors were framed in a microscopic model that suggests the formation of a dynamic web-like structure, where specific aggregation phenomena may occur, depending on growth stage and cell density. Furthermore, systematic measurements combining optical density and dry weight techniques presented new evidences, which confirmed that the observed cell aggregation patterns developed during growth, under shear, can not only be cell density dependent.info:eu-repo/semantics/publishedVersio

    Scaling of the elastic contribution to the surface free energy of a nematic on a sawtoothed substrate

    We characterize the elastic contribution to the surface free energy of a nematic in presence of a sawtooth substrate. Our findings are based on numerical minimization of the Landau-de Gennes model and analytical calculations on the Frank-Oseen theory. The nucleation of disclination lines (characterized by non-half-integer winding numbers) in the wedges and apexes of the substrate induces a leading order proportional to qlnq to the elastic contribution to the surface free energy density, q being the wavenumber associated with the substrate periodicity.Comment: 7 pages, 6 figures, accepted for publication in Physical Review

    Interspecific interactions among functionally diverse frugivores and their outcomes for plant reproduction: a new approach based on camera-trap data and tailored null models

    Research ArticleThe study of plant-frugivore interactions is essential to understand the ecology and evolution of many plant communities. However, very little is known about how interactions among frugivores indirectly affect plant reproductive success. In this study, we examined direct interactions among vertebrate frugivores sharing the same fruit resources. Then, we inferred how the revealed direct interspecific interactions could lead to indirect (positive or negative) effects on reproductive success of fleshy fruited plants. To do so, we developed a new analytical approach that combines camera trap data (spatial location, visitor species, date and time, activity) and tailored null models that allowed us to infer spatial-temporal interactions (attraction, avoidance or indifference) between pairs of frugivore species. To illustrate our approach, we chose to study the system composed by the Mediterranean dwarf palm, Chamaerops humilis, the Iberian pear tree, Pyrus bourgaeana, and their shared functionally diverse assemblages of vertebrate frugivores in a Mediterranean area of SW Spain. We first assessed the extent to which different pairs of frugivore species tend to visit the same or different fruiting individual plants. Then, for pairs of species that used the same individual plants, we evaluated their spatial-temporal relationship. Our first step showed, for instance, that some prey frugivore species (e.g. lagomorphs) tend to avoid those C. humilis individuals that were most visited by their predators (e.g. red foxes). Also, the second step revealed temporal attraction between large wild and domestic frugivore ungulates (e.g. red deer, cows) and medium-sized frugivores (e.g. red foxes) suggesting that large mammals could facilitate the C. humilis and P. bourgaeana exploitation to other smaller frugivores by making fruits more easily accessible. Finally, our results allowed us to identify direct interaction pathways, that revealed how the mutualistic and antagonistic relations between animal associates derived into indirect effects on both plants seed dispersal success. For instance, we found that large-sized seed predators (e.g. ungulates) had a direct positive effect on the likelihood of visits by legitimate seed dispersers (e.g. red foxes) to both fleshy fruited plants.     Balance training reduces fear of falling and improves dynamic balance and isometric strength in institutionalised older people: A randomised trial

    Question What is the effect of a balance training protocol with the Biodex Balance System in institutionalised older people with fear of falling? Design Randomised controlled trial with concealed allocation and assessor blinding. Participants Forty older people who lived in a nursing home and had fear of falling. Intervention The experimental group completed a 12-week balance training protocol based on balancing/rebalancing training with the Biodex Balance System, with two sessions per week. During the training period, participants in both groups received the same multidisciplinary care (such as physiotherapy, occupational therapy and nursing) that they usually received in the nursing home. Outcome measures The primary outcome was fear of falling (Falls Efficacy Scale International questionnaire). Secondary outcomes were dynamic balance (Fall Risk Test) and isometric strength (torque of knee flexor and extensor isometric strength measured with an isokinetic dynamometer). Outcome measures were taken before and after the training program protocol. Results Compared to the control group, the exercise group had significantly greater improvements at 12 weeks in fear of falling (by 8 points, 95% CI 4 to 12), in dynamic balance (by 2 degrees, 95% CI 1 to 3), and in isometric strength of the knee flexors (by 7 Nm, 95% CI 3 to 11) and knee extensors (by 7 Nm, 95% CI 1 to 13). Conclusion The training program was feasible and effective in reducing fear of falling and improving dynamic balance and isometric strength in institutionalised older people with fear of falling.     The reaction mechanism of metallo-beta-lactamases is tuned by the conformation of an active site mobile loop

    Carbapenems are "last resort" β-lactam antibiotics used to treat serious and life-threatening health care-associated infections caused by multidrug-resistant Gram-negative bacteria. Unfortunately, the worldwide spread of genes coding for carbapenemases among these bacteria is threatening these life-saving drugs. Metallo-β-lactamases (MβLs) are the largest family of carbapenemases. These are Zn(II)-dependent hydrolases that are active against almost all β-lactam antibiotics. Their catalytic mechanism and the features driving substrate specificity have been matter of intense debate. The active sites of MβLs are flanked by two loops, one of which, loop L3, was shown to adopt different conformations upon substrate or inhibitor binding, and thus are expected to play a role in substrate recognition. However, the sequence heterogeneity observed in this loop in different MβLs has limited the generalizations about its role. Here, we report the engineering of different loops within the scaffold of the clinically relevant carbapenemase NDM-1. We found that the loop sequence dictates its conformation in the unbound form of the enzyme, eliciting different degrees of active-site exposure. However, these structural changes have a minor impact on the substrate profile. Instead, we report that the loop conformation determines the protonation rate of key reaction intermediates accumulated during the hydrolysis of different β-lactams in all MβLs. This study demonstrates the existence of a direct link between the conformation of this loop and the mechanistic features of the enzyme, bringing to light an unexplored function of active-site loops on MβLs.Fil: Palacios, Antonela Rocio.     Nephron segment-specific gene expression using AAV vectors.

    AAV9 vector provides efficient gene transfer in all segments of the renal nephron, with minimum expression in non-renal cells, when administered retrogradely via the ureter. It is important to restrict the transgene expression to the desired cell type within the kidney, so that the physiological endpoints represent the function of the transgene expressed in that specific cell type within kidney. We hypothesized that segment-specific gene expression within the kidney can be accomplished using the highly efficient AAV9 vectors carrying the promoters of genes that are expressed exclusively in the desired segment of the nephron in combination with administration by retrograde infusion into the kidney via the ureter.
