Benzo[a]Pyrene Diol Epoxide Perturbation of Cell Cycle Kinetics of Synchronized Mouse Liver Epithelial Cells
A cell cycle synchronization system is described for the analysis of the perturbation of cell cycle kinetics and the cycle-phase specificity of chemicals and other agents. We used the system to study the effects of ({+-})r-7, t-8-dihydroxy-t-9, 10-oxy-7,8,9,10-tetrahydrobenzo[a]pyrene (BaP diol epoxide) upon the cell cycle of mouse liver epithelial cells(NMuLi). BaP diol epoxide(0.6 uM) was added to replated cultures of NMuLi cells that had been synchronized in various stages of the cell cycle by centrifugal elutriation. DNA histograms were obtained by flow cytometry as a function of time after replating. The data were analyzed by a computer modeling routine and reduced to a few graphs illustrating the 'net effects' of the BaP diol epoxide relative to controls. BaP diol epoxide slowed S-phase traversal in all samples relative to their respective control. Traversal through G{sub 2}M was also slowed by at least 50%. BaP diol epoxide had no apparent effect upon G{sub 1} traversal by cycling cells, but delayed the recruitment of quiescent G{sub 0} cells by about 2 hrs. The methods described constitute a powerful new approach for probing the cell cycle effects of a wide variety of agents. The present system appears to be extremely sensitive and capable of characterizing the action of agents on each phase of the cell cycle. The methods are automatable and would allow for the assay and possible differential characterization of mutagens and carcinogens
Conservative evaluation of the uncertainty in the LAGEOS-LAGEOS II Lense-Thirring test
We deal with the test of the general relativistic gravitomagnetic
Lense-Thirring effect currently ongoing in the Earth's gravitational field with
the combined nodes \Omega of the laser-ranged geodetic satellites LAGEOS and
LAGEOS II.
One of the most important source of systematic uncertainty on the orbits of
the LAGEOS satellites, with respect to the Lense-Thirring signature, is the
bias due to the even zonal harmonic coefficients J_L of the multipolar
expansion of the Earth's geopotential which account for the departures from
sphericity of the terrestrial gravitational potential induced by the
centrifugal effects of its diurnal rotation. The issue addressed here is: are
the so far published evaluations of such a systematic error reliable and
realistic? The answer is negative. Indeed, if the difference \Delta J_L among
the even zonals estimated in different global solutions (EIGEN-GRACE02S,
EIGEN-CG03C, GGM02S, GGM03S, ITG-Grace02, ITG-Grace03s, JEM01-RL03B, EGM2008,
AIUB-GRACE01S) is assumed for the uncertainties \delta J_L instead of using
their more or less calibrated covariance sigmas \sigma_{J_L}, it turns out that
the systematic error \delta\mu in the Lense-Thirring measurement is about 3 to
4 times larger than in the evaluations so far published based on the use of the
sigmas of one model at a time separately, amounting up to 37% for the pair
EIGEN-GRACE02S/ITG-Grace03s. The comparison among the other recent GRACE-based
models yields bias as large as about 25-30%. The major discrepancies still
occur for J_4, J_6 and J_8, which are just the zonals the combined

Beings in their own right? Exploring Children and young people's sibling and twin relationships in the Minority World
This paper examines the contributions that the sociological study of sibship and twinship in the Minority World can make to childhood studies. It argues that, in providing one forum within which to explore children and young people's social relationships, we can add to our understanding of children and young people's interdependence and develop a more nuanced understanding of agency. As emergent subjects, children, young people and adults are in a process of ‘becoming’. However, this does not mean that they can ‘become’ anything they choose to. The notion of negotiated interdependence (Punch 2002) is useful in helping us to grasp the contingent nature of children and young people's agency
Compassion Stress and the Qualitative Researcher
Human subjects are carefully protected in the research process. However, the same consideration is not currently being given to the qualitative researcher, even those investigating topics that are likely to elicit powerful emotions. The role of researcher’s emotional responses and the self-care strategies that, in some circumstances, are appropriate for the researcher and other research support personnel have not received the attention they deserve in qualitative research literature. Based on experience in conducting research on the topic of self-directed learning and breast cancer, and on the limited literature available, the author makes the case for the use of strategies such as counseling, peer debriefing, and journal writing as means of dealing with the potential for “compassion stress” as experienced by the researcher and other research support personnel. Astrobites as a Community-led Model for Education, Science Communication, and Accessibility in Astrophysics
Support for early career astronomers who are just beginning to explore
astronomy research is imperative to increase retention of diverse practitioners
in the field. Since 2010, Astrobites has played an instrumental role in
engaging members of the community -- particularly undergraduate and graduate
students -- in research. In this white paper, the Astrobites collaboration
outlines our multi-faceted online education platform that both eases the
transition into astronomy research and promotes inclusive professional
development opportunities. We additionally offer recommendations for how the
astronomy community can reduce barriers to entry to astronomy research in the
coming decade
Constraints from orbital motions around the Earth of the environmental fifth-force hypothesis for the OPERA superluminal neutrino phenomenology
It has been recently suggested by Dvali and Vikman that the superluminal
neutrino phenomenology of the OPERA experiment may be due to an environmental
feature of the Earth, naturally yielding a long-range fifth force of
gravitational origin whose coupling with the neutrino is set by the scale M_*,
in units of reduced Planck mass. Its characteristic length lambda should not be
smaller than one Earth's radius R_e, while its upper bound is expected to be
slightly smaller than the Earth-Moon distance (60 R_e). We analytically work
out some orbital effects of a Yukawa-type fifth force for a test particle
moving in the modified field of a central body. Our results are quite general
since they are not restricted to any particular size of lambda; moreover, they
are valid for an arbitrary orbital configuration of the particle, i.e. for any
value of its eccentricity . We find that the dimensionless strength coupling
parameter alpha is constrained to |alpha| <= 1 10^-10-4 10^-9 for 1 R_e <=
lambda <= 10 R_e by the laser data of the Earth's artificial satellite LAGEOS
II, corresponding to M_* >= 4 10^9 -1.6 10^10. The Moon perigee allows to
obtain |alpha| <= 3 10^-11 for the Earth-Moon pair in the range 15 R_e <=
lambda = 3 10^10 - 4.5 10^10. Our results
are neither necessarily limited to the superluminal OPERA scenario nor to the
Dvali-Vikman model, in which it is M_* = 10^-6 at lambda = 1 R_e, in contrast
with our bounds: they generally extend to any theoretical scenario implying a

Representations of sport in the revolutionary socialist press in Britain, 1988–2012
This paper considers how sport presents a dualism to those on the far left of the political spectrum. A long-standing, passionate debate has existed on the contradictory role played by sport, polarised between those who reject it as a bourgeois capitalist plague and those who argue for its reclamation and reformation. A case study is offered of a political party that has consistently used revolutionary Marxism as the basis for its activity and how this party, the largest in Britain, addresses sport in its publications. The study draws on empirical data to illustrate this debate by reporting findings from three socialist publications. When sport did feature it was often in relation to high profile sporting events with a critical tone adopted and typically focused on issues of commodification, exploitation and alienation of athletes and supporters. However, readers’ letters, printed in the same publications, revealed how this interpretation was not universally accepted, thus illustrating the contradictory nature of sport for those on the far left
An exact expression to calculate the derivatives of position-dependent observables in molecular simulations with flexible constraints
In this work, we introduce an algorithm to compute the derivatives of
physical observables along the constrained subspace when flexible constraints
are imposed on the system (i.e., constraints in which the hard coordinates are
fixed to configuration-dependent values). The presented scheme is exact, it
does not contain any tunable parameter, and it only requires the calculation
and inversion of a sub-block of the Hessian matrix of second derivatives of the
function through which the constraints are defined. We also present a practical
application to the case in which the sought observables are the Euclidean
coordinates of complex molecular systems, and the function whose minimization
defines the constraints is the potential energy. Finally, and in order to
validate the method, which, as far as we are aware, is the first of its kind in
the literature, we compare it to the natural and straightforward

Eosinophils Are Important for Protection, Immunoregulation and Pathology during Infection with Nematode Microfilariae
Eosinophil responses typify both allergic and parasitic helminth disease. In helminthic disease, the role of eosinophils can be both protective in immune responses and destructive in pathological responses. To investigate whether eosinophils are involved in both protection and pathology during filarial nematode infection, we explored the role of eosinophils and their granule proteins, eosinophil peroxidase (EPO) and major basic protein-1 (MBP-1), during infection with Brugia malayi microfilariae. Using eosinophil-deficient mice (PHIL), we further clarify the role of eosinophils in clearance of microfilariae during primary, but not challenge infection in vivo. Deletion of EPO or MBP-1 alone was insufficient to abrogate parasite clearance suggesting that either these molecules are redundant or eosinophils act indirectly in parasite clearance via augmentation of other protective responses. Absence of eosinophils increased mast cell recruitment, but not other cell types, into the broncho-alveolar lavage fluid during challenge infection. In addition absence of eosinophils or EPO alone, augmented parasite-induced IgE responses, as measured by ELISA, demonstrating that eosinophils are involved in regulation of IgE. Whole body plethysmography indicated that nematode-induced changes in airway physiology were reduced in challenge infection in the absence of eosinophils and also during primary infection in the absence of EPO alone. However lack of eosinophils or MBP-1 actually increased goblet cell mucus production. We did not find any major differences in cytokine responses in the absence of eosinophils, EPO or MBP-1. These results reveal that eosinophils actively participate in regulation of IgE and goblet cell mucus production via granule secretion during nematode-induced pathology and highlight their importance both as effector cells, as damage-inducing cells and as supervisory cells that shape both innate and adaptive immunity
