    QUANTITATIVE ASPECTS OF COAT COLOR IN OLD KLADRUBER BLACK HORSES

    Base economic characteristics (total revenues, total costs, profit and profitability ratio) of the Slovak Pinzgau breed were calculated in this study. Under the actual production and economic conditions of the breed, production system is operated with loss (-457 € per cow and per year) and with negative profitability ratio (-20%). Optimisation of the production parameters on the level defined in the breed standard (5,200 kg milk per cow and year, 92% for conception rate of cows, 404 days of calving interval and 550 g in daily gain of reared heifers) and improved udder health traits (clinical mastitis incidence and somatic cells score) was of positive impact on the total revenues (+34%), on the effective utilisation of costs (+105%) and balanced profit of dairy systems. Next to the positive profitability of the system, higher quality and security of dairy milk products should be mentioned there. Moreover, direct subsidies as an important factor of positive economic result of dairy cattle systems has to be pointed as well. Subsidies should be provided to compensate the real biological limitation of the local breed farmed in marginal areas. However, improvement of the production parameters of the Slovak Pinzgau breed is recommended with the same attention to reach the economic sustainability of dairy production system. To reach economic sustainability of the breed from practical point of view, the farmer activity should be aimed especially to the enhanced herd management

    The object of this diploma thesis is the evaluation of job satisfaction within the company Nakladatelství Fraus, s. r. o. The aim is to analyse the job satisfaction of the employees of this firm using a questionnaire and idividual interviews and according to the outcomes found to suggest recommendation to make this situation better. The diploma thesis is primary divided into two parts - a theoretical and a practical one. The theoretical part rely on literary sources and explains basic terms such as motivation, its sources and tools or the atributes of job satisfaction. The theoretical part includes the characteristics of the company Nakladatelství Fraus, s. r. o., individual interviews and the evaluation of the questionnaire. The final part pays attention to the suggested recommendations, how to improve the current situation of job satisfaction or dissatisfaction of the employees of the company

    Bakalářská práce se skládá primárně ze dvou částí -- teoretické a praktické. V teoretické části jsou s pomocí literárních zdrojů vysvětleny základní pojmy jako marketing, marketingový mix a jeho nástroje, komunikační mix a jeho složky nebo marketingový výzkum. Praktická část se pak zaměřuje na konkrétní podnik, Nakladatelství Fraus, s. r. o. v Plzni. Jednou ze specializací tohoto nakladatelství jsou učebnice pro základní školy, pro práci je tudíž vhodné. V jednotlivých oddílech praktické části je zahrnuta charakteristika společnosti, popis jednoho úspěšného a jednoho neúspěšného produktu nakladatelství, byl proveden marketingový výzkum a navíc je v této části i podáno doporučení možných zlepšujících opatření do budoucna. Cílem této práce je zhodnocení marketingových aktivit Nakladatelství Fraus a posouzení jednotlivých složek marketingového mixu ve vztahu ke konkrétním vybraným produktům tohoto nakladatelství.Bachelor's Thesis contains primary two parts -- a theoretical and a practical one. In the theoretical part there are explained the basic terms, using the literal sources, such as marketing, marketing mix and its tools, communication mix and its components or marketing research. The practical part is focused on a specific company, Nakladatelství Fraus, s. r. o. with the headquarters in Pilsen. One of the specializations of this publisher are textbooks for primary schools, so it is suitable for the Thesis. Within the particular sections of the practical part there are covered the characteristic of the company, description of one successful and one unsuccessful product, the marketing research was made and, moreover, in this part there is recommendation for the potential future improvements. The aim of this Thesis is the evaluation of the marketing activities of Nakladatelství Fraus and the review of the concrete components of marketing mix in relation to specific products of this publisher

    Interactions of an <it>Arabidopsis</it> RanBPM homologue with LisH-CTLH domain proteins revealed high conservation of CTLH complexes in eukaryotes

    <p>Abstract</p> <p>Background</p> <p>RanBPM (Ran-binding protein in the microtubule-organizing centre) was originally reported as a centrosome-associated protein in human cells. However, RanBPM protein containing highly conserved SPRY, LisH, CTLH and CRA domains is currently considered as a scaffolding protein with multiple cellular functions. A plant homologue of RanBPM has not yet been characterized.</p> <p>Results</p> <p>Based on sequence similarity, we identified a homologue of the human RanBPM in <it>Arabidopsis thaliana.</it> AtRanBPM protein has highly conserved SPRY, LisH, CTLH and CRA domains. Cell fractionation showed that endogenous AtRanBPM or expressed GFP-AtRanBPM are mainly cytoplasmic proteins with only a minor portion detectable in microsomal fractions. AtRanBPM was identified predominantly in the form of soluble cytoplasmic complexes ~230 – 500 kDa in size. Immunopurification of AtRanBPM followed by mass spectrometric analysis identified proteins containing LisH and CRA domains; LisH, CRA, RING-U-box domains and a transducin/WD40 repeats in a complex with AtRanBPM. Homologues of identified proteins are known to be components of the C-terminal to the LisH motif (CTLH) complexes in humans and budding yeast. Microscopic analysis of GFP-AtRanBPM <it>in vivo</it> and immunofluorescence localization of endogenous AtRanBPM protein in cultured cells and seedlings of <it>Arabidopsis</it> showed mainly cytoplasmic and nuclear localization. Absence of colocalization with γ-tubulin was consistent with the biochemical data and suggests another than a centrosomal role of the AtRanBPM protein.</p> <p>Conclusion</p> <p>We showed that as yet uncharacterized <it>Arabidopsis</it> RanBPM protein physically interacts with LisH-CTLH domain-containing proteins. The newly identified high molecular weight cytoplasmic protein complexes of AtRanBPM showed homology with CTLH types of complexes described in mammals and budding yeast. Although the exact functions of the CTLH complexes in scaffolding of protein degradation, in protein interactions and in signalling from the periphery to the cell centre are not yet fully understood, structural conservation of the complexes across eukaryotes suggests their important biological role.</p
