    Enhanced phenylpyruvic acid production with Proteus Vulgaris by optimizing of the fermentation medium

    Alpha keto acids are important food additives, which commonly produced by microbial deamination of amino acids. In this study, production of phenylpyruvic acid (PPA), which is the alpha keto acid of phenylalanine was enhanced in 2-l bench scale bioreactors by optimizing of fermentation medium composition using the Box-Behnken Response Surface Methodology (RSM). Optimum glucose, yeast extract, and phenylalanine concentrations were determined to be 119.4 g 1−1, 3.7 g 1−1, and 14.8 g 1−1, respectively, for PPA production, and 163.8 g 1−1, 10.8 g 1−1, and 9.8 g 1−1, respectively, for biomass production. Under these optimum conditions, PPA concentration was enhanced to 1349 mg 1−1, which was 28% and 276% higher than the unoptimized bioreactor and shake-flask fermentations, respectively. Moreover, P. vulgaris biomass concentration was optimized at 4.36 g 1−1, which was 34% higher than under the unoptimized bioreactor condition. Overall, this study demonstrated that optimization of the fermentation media improved PPA concentration and biomass production in bench scale bioreactors compared to previous studies in the literature and sets the stage for scale up to industrial production

    Heterometallic cobalt(ii) calix[6 and 8]arenes: synthesis, structure and electrochemical activity

    Heterometallic cobalt p-tert-butylcalix[6 and 8]arenes have been generated from the in situ reaction of lithium reagents (n-BuLi or t-BuOLi) or NaH with the parent calix[n]arene and subsequent reaction with CoBr2. The reverse route, involving the addition of in situ generated Li[Co(Ot-Bu)3] to p-tert-butylcalix[6 and 8]arene, has also been investigated. X-ray crystallography reveals the formation of complicated products incorporating differing numbers of cobalt and lithium or sodium centers, often with positional disorder, as well as, in some cases, the retention of halide. The electrochemical analysis revealed several oxidation events related to the subsequent oxidation of Co(ii) centers and the reduction of the metal cation at negative potentials. Moreover, the electrochemical activity of the phenol moieties of the parent calix[n]arenes resulted in dimerized products or quinone derivatives, leading to insoluble oligomeric products that deposit and passivate the electrode. Preliminary screening for electrochemical proton reduction revealed good activity for a number of these systems. Results suggest that [Co6Na(NCMe)6(μ-O)(p-tert-butylcalix[6]areneH)2Br]·7MeCN (6·7MeCN) is a promising molecular catalyst for electrochemical proton reduction, with a mass transport coefficient, catalytic charge transfer resistance and current magnitude at the catalytic turnover region that are comparable to those of the reference electrocatalyst (Co(ii)Cl2)

    Short-term post-harvest stress that affects profiles of volatile organic compounds and gene expression in rocket salad during early post-harvest senescence

    Once harvested, leaves undergo a process of senescence which shares some features with developmental senescence. These include changes in gene expression, metabolites, and loss of photosynthetic capacity. Of particular interest in fresh produce are changes in nutrient content and the aroma, which is dependent on the profile of volatile organic compounds (VOCs). Leafy salads are subjected to multiple stresses during and shortly after harvest, including mechanical damage, storage or transport under different temperature regimes, and low light. These are thought to impact on later shelf life performance by altering the progress of post-harvest senescence. Short term stresses in the first 24 h after harvest were simulated in wild rocket (Diplotaxis tenuifolia). These included dark (ambient temperature), dark and wounding (ambient temperature), and storage at 4 \ub0C in darkness. The effects of stresses were monitored immediately afterwards and after one week of storage at 10 \ub0C. Expression changes in two NAC transcription factors (orthologues of ANAC059 and ANAC019), and a gene involved in isothiocyanate production (thiocyanate methyltransferase, TMT) were evident immediately after stress treatments with some expression changes persisting following storage. Vitamin C loss and microbial growth on leaves were also affected by stress treatments. VOC profiles were differentially affected by stress treatments and the storage period. Overall, short term post-harvest stresses affected multiple aspects of rocket leaf senescence during chilled storage even after a week. However, different stress combinations elicited different responses

    Exchange coupling in CaMnO3_3 and LaMnO3_3: configuration interaction and the coupling mechanism

    The equilibrium structure and exchange constants of CaMnO3_3 and LaMnO3_3 have been investigated using total energy unrestricted Hartree-Fock (UHF) and localised orbital configuration interaction (CI) calculations on the bulk compounds and Mn2_2O1114_{11}^{14-} and Mn2_2O1116_{11}^{16-} clusters. The predicted structure and exchange constants for CaMnO3_3 are in reasonable agreement with estimates based on its N\'eel temperature. A series of calculations on LaMnO3_3 in the cubic perovskite structure shows that a Hamiltonian with independent orbital ordering and exchange terms accounts for the total energies of cubic LaMnO3_3 with various spin and orbital orderings. Computed exchange constants depend on orbital ordering. UHF calculations tend to underestimate exchange constants in LaMnO3_3, but have the correct sign when compared with values obtained by neutron scattering; exchange constants obtained from CI calculations are in good agreement with neutron scattering data provided the Madelung potential of the cluster is appropriate. Cluster CI calculations reveal a strong dependence of exchange constants on Mn d eg_g orbital populations in both compounds.     Pharmacological Inhibition of polysialyltransferase ST8SiaII Modulates Tumour Cell Migration

    YesPolysialic acid (polySia), an α-2,8-glycosidically linked polymer of sialic acid, is a developmentally regulated posttranslational modification predominantly found on NCAM (neuronal cell adhesion molecule). Whilst high levels are expressed during development, peripheral adult organs do not express polySia-NCAM. However, tumours of neural crest-origin re-express polySia-NCAM: its occurrence correlates with aggressive and invasive disease and poor clinical prognosis in different cancer types, notably including small cell lung cancer (SCLC), pancreatic cancer and neuroblastoma. In neuronal development, polySia-NCAM biosynthesis is catalysed by two polysialyltransferases, ST8SiaII and ST8SiaIV, but it is ST8SiaII that is the prominent enzyme in tumours. The aim of this study was to determine the effect of ST8SiaII inhibition by a small molecule on tumour cell migration, utilising cytidine monophosphate (CMP) as a tool compound. Using immunoblotting we showed that CMP reduced ST8iaII-mediated polysialylation of NCAM. Utilizing a novel HPLC-based assay to quantify polysialylation of a fluorescent acceptor (DMB-DP3), we demonstrated that CMP is a competitive inhibitor of ST8SiaII (Ki = 10 μM). Importantly, we have shown that CMP causes a concentration-dependent reduction in tumour cell-surface polySia expression, with an absence of toxicity. When ST8SiaII-expressing tumour cells (SH-SY5Y and C6-STX) were evaluated in 2D cell migration assays, ST8SiaII inhibition led to significant reductions in migration, while CMP had no effect on cells not expressing ST8SiaII (DLD-1 and C6-WT). The study demonstrates for the first time that a polysialyltransferase inhibitor can modulate migration in ST8SiaII-expressing tumour cells.     Search for VHE gamma rays from SS433/W50 with the CANGAROO-II telescope

    SS433, located at the center of the supernova remnant W50, is a close proximity binary system consisting of a compact star and a normal star. Jets of material are directed outwards from the vicinity of the compact star symmetrically to the east and west. Non-thermal hard X-ray emission is detected from lobes lying on both sides. Shock accelerated electrons are expected to generate sub-TeV gamma rays through the inverse-Compton process in the lobes. Observations of the western X-ray lobe region of SS433/W50 system have been performed to detect sub-TeV gamma-rays using the 10m CANGAROO-II telescope in August and September, 2001, and July and September, 2002. The total observation times are 85.2 hours for ON source, and 80.8 hours for OFF source data. No significant excess of sub-TeV gamma rays has been found at 3 regions of the western X-ray lobe of SS433/W50 system. We have derived 99% confidence level upper limits to the fluxes of gamma rays and have set constraints on the strengths of the magnetic fields assuming the synchrotron/inverse-Compton model for the wide energy range of photon spectrum from radio to TeV. The derived lower limits are 4.3 microgauss for the center of the brightest X-ray emission region and 6.3 microgauss for the far end from SS433 in the western X-ray lobe. In addition, we suggest that the spot-like X-ray emission may provide a major contribution to the hardest X-ray spectrum in the lobe.Comment: 7 pages, 8 figures, to be published in Astroparticle Physic

    Impact of percutaneous coronary intervention timing on 5-year outcome in patients with non-ST-segment elevation acute coronary syndromes. The ‘wait a day’ approach might be safer

    Background The OPTIMA trial was a randomised multicentre trial exploring the influence of the timing of percutaneous coronary intervention (PCI) on patient outcomes in an intermediate to high risk non-ST-elevation acute coronary syndrome (NSTE-ACS) population. In order to decide the best treatment strategy for patients presenting with NSTEACS, long-term outcomes are essential. Methods Five-year follow-up data from 133 of the 142 patients could be retrieved (94 %). The primary endpoint was a composite of death and spontaneous myocardial infarction (MI). Spontaneous MI was defined as MI occurring more than 30 days after randomisation. Secondary endpoints were the individual outcomes of death, spontaneous MI or re-PCI. Results No significant difference with respect to the primary endpoint was observed (17.8 vs. 10.1 %; HR 1.55, 95 % CI: 0.73–4.22, p = 0.21). There was no significant difference in mortality rate. However, spontaneous MI was significantly more common in the group receiving immediate PCI (11.0 vs. 1.4 %; HR 4.46, 95 % CI: 1.21–16.50, p = 0.02). We did not find a significant difference between the groups with respect to re-PCI rate. Conclusion There was no difference in the composite of death and spontaneous MI. The trial suggests an increased long-term risk of spontaneous MI for patients treated with immediate PCI

    Frame Theory for Signal Processing in Psychoacoustics

    This review chapter aims to strengthen the link between frame theory and signal processing tasks in psychoacoustics. On the one side, the basic concepts of frame theory are presented and some proofs are provided to explain those concepts in some detail. The goal is to reveal to hearing scientists how this mathematical theory could be relevant for their research. In particular, we focus on frame theory in a filter bank approach, which is probably the most relevant view-point for audio signal processing. On the other side, basic psychoacoustic concepts are presented to stimulate mathematicians to apply their knowledge in this field
