Representing and Capturing the Experts´ Knowledge in a Design Process

    Get PDF
An object-oriented framework to support the modeling and management of the design process is introduced. It naturally integrates the representation of both the design process itself, and the outcomes that are achieved as the result of the various design activities. The integral view of tracing that was adopted not only captures and manages the products being generated but also the activities that occurred, their associated context and the adopted decisions. The Version Administration System introduced in this paper provides an explicit mechanism to manage the different model versions being generated during the course of a design project as design activities are executed.Fil: Gonnet, Silvio Miguel. 209 p.In the field of regenerative medicine, it has been pointed out the relevance of human adipose derived stem cells (hASCs) for cell therapy. They can be easily obtained, have low immunogenicity, and secrete soluble factors that could regulate neuroinflammation and oxidative stress. However, their transplantation at the site of injury results in a low percentage of survival and engraftment, mainly due to the harsh microenvironment they encounter. To address this issue, it is imperative to pre-adapt cells, so that they resist harmful environmental factors such as oxidative stress. In this study, we preconditioned hASCs (PC-hASCs) with low doses of H2O2 and evaluated their resistance to an oxidative stress insult. PC-hASCs displayed lower levels of ROS, apoptosis and cytotoxicity than hASCs, indicating their increased capacity to resist to oxidative stress. In addition, we analyzed the molecular and bioenergetic mechanisms underlying the survival and adaptation of PC-hASCs under oxidative stress. On these conditions, PC-hASCs i, reduce intracellular ROS levels by overexpressing the transcription factor NRF2 and their related antioxidant enzymes HO-1, SOD-1, GPx-1, and CAT; ii, reduce the secretion of pro-inflammatory molecules COX-2 and IL-1ß by attenuating the expression of NF-kB, and iii, increase the total ATP production rate by adapting their metabolism in order to meet the bioenergetic demand required to survive. Finally, we evaluated the therapeutic role of PC-hASCs to overcome the deleterious effect of oxidative stress in an oligodendroglial cell population, and proved that PC-hASCs restored cell viability and diminished the intracellular ROS levels of these damaged oligodendrocytes by promoting their antioxidant response.Altogether, these findings support that PC-hASCs, given their beneficial advantages, might be considered an important breakthrough in cell¿based therapies

Contribution of green turtles Chelonia mydas to total herbivore biomass in shallow tropical habitats of oceanic islands

Green turtles are megaherbivores with a key role in the dynamics of tropical seagrass meadows, but little is known about their relevance as herbivores in tropical reef habitats. We conducted underwater censuses of green turtles, herbivorous fishes and sea urchins in two distinct tropical regions: Fernando de Noronha (Western Atlantic Ocean) and the Hawaiian Archipelago (Central Pacific Ocean), to assess the contribution of green turtles to the total herbivore biomass in shallow reef habitats of tropical oceanic islands. Juvenile green turtles ranging 40-60 cm were observed at most of the surveyed sites, and hence, could be considered typical components of the shallow reef fauna of tropical oceanic islands. Furthermore, they were usually one of the most abundant species of roving herbivores in many of the sites surveyed. However, the biomass of green turtles was usually much lower than the aggregated biomass of fishes or sea urchins, which usually constituted most of the total herbivore biomass. Green turtles made a major contribution to the total herbivore biomass only in sheltered sites with low rugosity, low coral cover and high algal cover. Further investigation on the trophic redundancy between herbivores is required to assess the actual relevance of green turtles in reef ecosystems of oceanic islands, compared to herbivorous fishes and sea urchins, because different herbivores may target different algal resources and complementarity may be needed to maintain ecosystem functioning across large, naturally varied reefscapes

Hey children! Do you feel listened by your politicians? Evaluating a participative democratic programme

    Get PDF
The objective of this study was to evaluate a participative democracy programme called Ágora infantil, carried out with students between the ages of 10 and 12. As in participatory budgets, the process is based on the research-action model (Lewin, 1946) and consists of the city council opening up decision-making on a part of the municipal budget or a concrete policy to the children of the municipality. This process of diagnosis, prioritization and decision-making was completed over the course of 5 to 7 sessions during school hours and implemented by the Coglobal association. Once the participants make a decision, the city council carries out the project selected in the municipality (the decision made is binding). The main goal of Ágora Infantil is to generate psychological empowerment and wellbeing amongst its participants, and to measure the effects of this programme on children’s psychological empowerment. To this end, we used Zimmerman´s model (Zimmerman, 1995; Zimmerman, 2000). The operationalization of the strengthening was based on: 1) knowledge of local politics (as a component of the interactional dimension of psychological empowerment) and 2) the self-perception of being able to influence the future of the municipality (component of the intrapersonal dimension of psychological empowerment). In addition, wellbeing was evaluated through the perception of being heard (Casas y Bello, 2012) and trust in the city council.

The resting microstate networks (RMN): cortical distributions, dynamics, and frequency specific information flow

A brain microstate is characterized by a unique, fixed spatial distribution of electrically active neurons with time varying amplitude. It is hypothesized that a microstate implements a functional/physiological state of the brain during which specific neural computations are performed. Based on this hypothesis, brain electrical activity is modeled as a time sequence of non-overlapping microstates with variable, finite durations (Lehmann and Skrandies 1980, 1984; Lehmann et al 1987). In this study, EEG recordings from 109 participants during eyes closed resting condition are modeled with four microstates. In a first part, a new confirmatory statistics method is introduced for the determination of the cortical distributions of electric neuronal activity that generate each microstate. All microstates have common posterior cingulate generators, while three microstates additionally include activity in the left occipital/parietal, right occipital/parietal, and anterior cingulate cortices. This appears to be a fragmented version of the metabolically (PET/fMRI) computed default mode network (DMN), supporting the notion that these four regions activate sequentially at high time resolution, and that slow metabolic imaging corresponds to a low-pass filtered version. In the second part of this study, the microstate amplitude time series are used as the basis for estimating the strength, directionality, and spectral characteristics (i.e., which oscillations are preferentially transmitted) of the connections that are mediated by the microstate transitions. The results show that the posterior cingulate is an important hub, sending alpha and beta oscillatory information to all other microstate generator regions. Interestingly, beyond alpha, beta oscillations are essential in the maintenance of the brain during resting state.Comment: pre-print, technical report, The KEY Institute for Brain-Mind Research (Zurich), Kansai Medical University (Osaka
