    Up until now design and architectural projects without commercial value have been carried out by larger corporations that have had enough resources to expand in these areas with long-term perspectives and become cultural mediators. However, as a result of the growing trend to evolve from philanthropic donations to strategic CSR, intermediary smaller organisations have emerged as cultural consultants, providing tailored solutions aligned with the vision and the needs of a company. Given the author’s internship experience in the cultural consultant experimenta, this thesis aims to define, illustrate and suggests evaluation methods for design and architectural projects as well as provide a practical exemplification with Amorim 2020. Acknowledging the current complex situation in terms of value creation measurement of these kind of projects, but considering its relevance, a broader framework based on the SROI methodology is finally proposed, together with recommendations and potential lines of actions on its application

    Osteological Evidence of Possible Tuberculosis from the Early Medieval Age (6th–11th Century), Northern Italy

    We discuss the probable presence of tuberculosis in non-adults of a medieval rural com- munity in northern Italy with a biocultural perspective. Before birth, mother and child have a closely interconnected relationship, as suggested by the role of microchimerism on maternal health. To better understand maternal–child health and related stress factors, paleopathology has investigated the potential of this relationship in recent years. Diseases with environmental development factors such as infections depend on and are strongly affected by maternal control variables such as, for example, breastfeeding and the growth environment. This article presents ISZ2, the second possible case of childhood tuberculosis identified in northeastern Italy through recent paleopathological criteria. The subject is of primary importance as it not only represents a direct testimony of this infectious disease otherwise known only from historical sources, but also increases our knowledge on the state of health of infants in this area, which to date are scarcely analyze

    Changes in the contents of micro- and trace-elements in wine due to winemaking treatments

    Forty-four mineral elements quantified by ICP-OES and ICP-MS were measured in wines, (a) after wine fining, at three pH levels with 10 different bentonites (1 g·l-1), (b) after addition of yeast hulls from 2 suppliers to wine (180 and 360 mg·1-1). Bentonite fining resulted in statistically significant increases of the large majority of elements, but in significant lower levels of Cu, K, Rb and Zn. The addition of yeast hulls caused a statistically significant depletion of the contents of Ce, Cu, Fe, La, Sb, U, V and Y.

    Age-Associated Decline in Dendritic Cell Function and the Impact of Mediterranean Diet Intervention in Elderly Subjects

    Introduction: Aging is accompanied by increased susceptibility to infection and age-associated chronic diseases. It is also associated with reduced vaccine responses, which is often attributed to immunosenescence and the functional decline of the immune system. Immunosenescence is characterized by a chronic, low-grade, inflammatory state termed inflammaging. Habitants of Mediterranean (MED) regions maintain good health into old age; often attributed to MED diets. Hypothesis: Adoption of a MED-diet by elderly subjects, in Norfolk (UK), may improve immune responses of these individuals and in particular, dendritic cell (DC) function. Experimental approach: A total of 120 elderly subjects (65–79 years old) recruited onto the Nu-AGE study, a multicenter European dietary study specifically addressing the needs of the elderly, across five countries, and were randomized to the control or MED-diet groups, for one year. Blood samples were taken pre- and post-intervention for DC analysis and were compared with each other, and to samples obtained from 45 young (18–40 years old) subjects. MED-diet compliance was assessed using high performance liquid chromatography-with tandem mass spectrometry analysis of urine samples. Immune cell and DC subset numbers and concentrations of secreted proteins were determined by flow cytometric analysis. Results: As expected, reduced myeloid DC numbers were observed in blood samples from elderly subjects compared with young. The elevated secretion of the adipokine, resistin, after ex vivo stimulation of peripheral blood mononuclear cells from elderly subjects, was significantly reduced after MED-diet intervention. Conclusion: This study provides further evidence of numerical and functional effects of aging on DCs. The MED-diet showed potential to impact on the aging immune cells investigated and could provide an economical approach to address problems associated with our aging population

    Cytotoxic targeting of F9 teratocarcinoma tumours with anti-ED-B fibronectin scFv antibody modified liposomes

    We prepared small unilamellar liposomes derivatised with single chain antibody fragments specific for the ED-B domain of B-fibronectin. This extracellular matrix associated protein is expressed around newly forming blood vessels in the vicinity of many types of tumours. The single chain antibody fragments were functionalised by introduction of C-terminal cysteines and linked to liposomes via maleimide groups located at the terminal ends of poly(ethylene glycol) modified phospholipids. The properties of these anti-ED-B single chain antibody fragments-liposomes were analysed in vitro on ED-B fibronectin expressing Caco-2 cells and in vivo by studying their biodistribution and their therapeutic potential in mice bearing subcutanous F9 teratocarcinoma tumours. Radioactively labelled (114mIndium) single chain antibody fragments-liposomes accumulated in the tumours at 2–3-fold higher concentrations during the first 2 h after i.v. injection compared to unmodified liposomes. After 6–24 h both liposome types were found in similar amounts (8–10% injected dose g−1) in the tumours. Animals treated i.v. with single chain antibody fragments-liposomes containing the new cytotoxic agent 2′-deoxy-5-fluorouridylyl-N4-octadecyl-1-β-D-arabinofuranosylcytosine (30 mg kg-1 per dose, five times every 24 h) showed a reduction of tumour growth by 62–90% determined on days 5 and 8, respectively, compared to animals receiving control liposomes. Histological analysis revealed a marked reduction of F9 tumour cells and excessive deposition of fibronectin in the extracellular matrix after treatment with single chain antibody fragments-2-dioxy-5-fluorouridylyl-N4-octadecyl-1-β-D-arabinofuranosylcytosine-liposomes. Single chain antibody fragments-liposomes targeted to ED-B fibronectin positive tumours therefore represent a promising and versatile novel drug delivery system for the treatment of tumours

    Liposomes in Biology and Medicine

    Drug delivery systems (DDS) have become important tools for the specific delivery of a large number of drug molecules. Since their discovery in the 1960s liposomes were recognized as models to study biological membranes and as versatile DDS of both hydrophilic and lipophilic molecules. Liposomes--nanosized unilamellar phospholipid bilayer vesicles--undoubtedly represent the most extensively studied and advanced drug delivery vehicles. After a long period of research and development efforts, liposome-formulated drugs have now entered the clinics to treat cancer and systemic or local fungal infections, mainly because they are biologically inert and biocompatible and practically do not cause unwanted toxic or antigenic reactions. A novel, up-coming and promising therapy approach for the treatment of solid tumors is the depletion of macrophages, particularly tumor associated macrophages with bisphosphonate-containing liposomes. In the advent of the use of genetic material as therapeutic molecules the development of delivery systems to target such novel drug molecules to cells or to target organs becomes increasingly important. Liposomes, in particular lipid-DNA complexes termed lipoplexes, compete successfully with viral gene transfection systems in this field of application. Future DDS will mostly be based on protein, peptide and DNA therapeutics and their next generation analogs and derivatives. Due to their versatility and vast body of known properties liposome-based formulations will continue to occupy a leading role among the large selection of emerging DDS
