    3D Bioprinted Structures from Cells of Non-Epithelial Mesodermal and Endodermal Lineage Using a Custom Accessible 3D Bioprinting Platform

    Prior work within our lab has demonstrated the ability to print both murine and human mammary organoids and tumoroids in vitro that can also be reliably transplanted into a murine host for translational studies. Peripherally, this bioprinting system has also been used for 3D printing neurons, stem cells, cancer cells, and a primary cell line rich with fibroblasts, but each of these efforts were with cells of ectodermal lineage. Thus, the system\u27s capacity for use on cells of other origins had been untested. To address this, we have now developed protocols for cells of endodermal and non-epithelial mesodermal/mesenchymal lineage. In this work, we find that we can produce reliable organoids, tumoroids, and other in vitro structures from them, thus expanding the functional range of our open 3D bioprinting platform.     The Purpose of this Booklet This booklet is intended to provide succinct guidance to teachers, leaders and other practitioners in schools on current research and methodologies related to coaching. Coaching is increasingly seen as an important, key element in practitioner professional learning. The Welsh Government’s recent publication ‘Investing in excellence: Our national workforce development plan 2019–21’ (Welsh Government, 2019) includes commitments to: • a dedicated programme of coaching and mentoring and a set of standards for mentor support along with a programme of common professional learning for mentors. (designed for) practitioners in the early career stages • the introduction of an enhanced programme that includes coaching, mentoring and a high-level development programme (to support leadership and succession planning) The national professional learning model (NAPL) is the government’s vehicle for the design and delivery of professional learning (https://gov.wales/national-approach-professional-learning-napl ). In the context of practitioner coaching and mentoring, the following design features are particularly pertinent: The professional learner is near the centre of our national approach. Professional Learning should be intended and designed to be a personalised response to individual professional learners’ needs, taking into account their experience, expertise and aspirations. (Welsh Government, 2019) There are clearly links to effective teaching and learning also, and hence to the Welsh Government’s professional standards for teaching and leadership (Welsh Government, 2018). Whilst the focus in this booklet is on the teaching profession, coaching has already been shown to be truly beneficial for the development of a wide range of professions, and for activities both inside and outside work (Passmore & Fillery-Travis, 2011).     The significance and challenges of turnover and retention of millennial professionals

    Purpose – This paper aims to provide readers with a comprehensive overview of the current state of the millennial literature, highlighting the significance and challenges of millennial professionals, their reported high turnover and the various recommendations designed to engage and retain them. Design/methodology/approach – An integrated review approach was applied to synthesise contemporary peer-reviewed articles, supplemented by legacy and grey literature and relevant book chapters, to comprehensively explore and construct a cohesive overview of the current research on the millennial workforce. Findings – Within the wealth of available information, examining the various studies on millennial turnover reveals diverse theories, evidence and opportunities for advancement, underscoring the necessity for more robust empirical studies. The investigation identified three overarching retention strategy themes: (1) intergenerational conflict management, (2) workplace adaptations and (3) solutions rooted in a protean career orientation. In alignment with protean career concepts, coaching shows promise as an underexplored option. Practical implications – This article holds practical significance by offering researchers a comprehensive and cohesive overview of the millennial literature. Additionally, it gives organisations a novel perspective on the crucial role coaching can play in engaging and retaining millennial employees. Originality/value – The increased focus on retaining millennial workers in recent decades has spurred a proliferation of articles and books on this subject. However, this body of research remains fragmented, lacking an overview that provides a clear picture of its current state. This review aims to bridge this gap.     LIS Programs and Data Centers: Integrating Expertise

    Scientific data centers have provided data services to research communities for decades and are invaluable educational partners for iSchools developing academic programs in data curation. This paper presents analyses from three years of internship placements at the National Center for Atmospheric Research and interviews with managers at prominent data centers across the country, as part of the Data Curation Education in Research Centers project. Key benefits of the internship program are identified, from the perspective of student learning and the contributions made by iSchool students to data center operations. The interviews extend the case results, providing evidence for potential data curation internship programs at data centers. The DCERC education model fosters integration of expertise across the iSchool and data center communities, enriching academic preparation with state-of-the-art practical experience in ways that are vital to the emerging data profession and its ability to meet the future demands of data-intensive research.ye

    Host phenotype characteristics and MC1R in relation to early-onset basal cell carcinoma.

    Basal cell carcinoma (BCC) incidence is increasing, particularly among adults under the age of 40 years. Pigment-related characteristics are associated with BCC in older populations, but epidemiologic studies among younger individuals and analyses of phenotype-genotype interactions are limited. We examined self-reported phenotypes and melanocortin 1 receptor gene (MC1R) variants in relation to early-onset BCC. BCC cases (n=377) and controls with benign skin conditions (n=390) under the age of 40 years were identified through Yale's Dermatopathology database. Factors most strongly associated with early-onset BCC were skin reaction to first summer sun for 1 hour (severe sunburn vs. tan odds ratio (OR)=12.27, 95% confidence interval (CI)=4.08-36.94) and skin color (very fair vs. olive OR=11.06, 95% CI=5.90-20.74). Individuals with two or more MC1R non-synonymous variants were 3.59 times (95% CI=2.37-5.43) more likely to have BCC than those without non-synonymous variants. All host characteristics and MC1R were more strongly associated with multiple BCC case status (37% of cases) than a single BCC case status. MC1R, number of moles, skin reaction to first summer sun for 1 hour, and hair and skin color were independently associated with BCC. BCC risk conferred by MC1R tended to be stronger among those with darker pigment phenotypes, traditionally considered to be at low risk of skin cancer

    Gammaretrovirus-mediated correction of SCID-X1 is associated with skewed vector integration site distribution in vivo

    We treated 10 children with X-linked SCID (SCID-X1) using gammaretrovirus-mediated gene transfer. Those with sufficient follow-up were found to have recovered substantial immunity in the absence of any serious adverse events up to 5 years after treatment. To determine the influence of vector integration on lymphoid reconstitution, we compared retroviral integration sites (RISs) from peripheral blood CD3(+) T lymphocytes of 5 patients taken between 9 and 30 months after transplantation with transduced CD34(+) progenitor cells derived from 1 further patient and I healthy donor. Integration occurred preferentially in gene regions on either side of transcription start sites, was clustered, and correlated with the expression level in CD34(+) progenitors during transduction. In contrast to those in CD34(+) cells, RISs recovered from engrafted CD3(+)T cells were significantly overrepresented within or near genes encoding proteins with kinase or transferase activity or involved in phosphorus metabolism. Although gross patterns of gene expression were unchanged in transduced cells, the divergence of RIS target frequency between transduced progenitor cells and post-thymic T lymphocytes indicates that vector integration influences cell survival, engraftment, or proliferation

    Pressure promoted low-temperature melting of metal–organic frameworks

    International audienceMetal–organic frameworks (MOFs) are microporous materials with huge potential for chemical processes. Structural collapse at high pressure, and transitions to liquid states at high temperature, have recently been observed in the zeolitic imidazolate framework (ZIF) family of MOFs. Here, we show that simultaneous high-pressure and high-temperature conditions result in complex behaviour in ZIF-62 and ZIF-4, with distinct high- and low-density amorphous phases occurring over different regions of the pressure–temperature phase diagram. In situ powder X-ray diffraction, Raman spectroscopy and optical microscopy reveal that the stability of the liquid MOF state expands substantially towards lower temperatures at intermediate, industrially achievable pressures and first-principles molecular dynamics show that softening of the framework coordination with pressure makes melting thermodynamically easier. Furthermore, the MOF glass formed by melt quenching the high-temperature liquid possesses permanent, accessible porosity. Our results thus imply a route to the synthesis of functional MOF glasses at low temperatures, avoiding decomposition on heating at ambient pressure
