    Efecto antitrombótico de extractos y compuestos bioactivos de Solanum lycopersicum en un modelo murino de trombosis

    91 p.Las Enfermedades Cardiovasculares (ECV) son la principal causa de muerte en el mundo. En base a esto distintas estrategias se han propuesto para su prevención. Se ha visto que una dieta rica en frutas y hortalizas disminuye el riesgo de padecer ECV. Se conoce bastante el efecto antioxidante de estas, no así su efecto antitrombótico. Por esto, el objetivo de esta tesis fue investigar el efecto antitrombótico de extractos y compuestos bioactivos de S. Lycopersicum, en un modelo murino de trombosis. Se evaluaron ocho extractos y ocho compuestos bioactivos. Para esto se montó un sistema de trombosis fotoquímica in vivo. Se estandarizó con una concentración óptima de rosa de bengala de 50 mg/kg de peso. Las dosis de los anestésicos y relajante muscular se estandarizaron a una concentración de 266,6 mg/kg de peso (2,2,2 tribromoetanol), ketamina a 208,3 mg/kg de peso y xylasina 13,3 mg/kg de peso. La inducción de trombosis se realizó en las arterias mesentéricas utilizando un láser (5 mW, 532 nm). Se determinaron las concentraciones de extractos y compuestos bioactivos de S. Lycopersicum mediante una curva de concentraciones determinando que la concentración a utilizar en los ensayos es de 200 mg/kg de peso. Tanto extractos como compuestos bioactivos, mostraron una inhibición estadísticamente significativa en la formación del trombo. La media de inhibición de la trombosis de extractos y compuestos bioactivos de S. Lycopersicum fue de un ~40% y un ~66%, respectivamente. Esta tesis muestra un nuevo campo en la investigación, enfocándose principalmente a continuar con los estudios de esta hortaliza en la prevención de ECV y de los mecanismos moleculares involucrados en ésta

    Bioinformatics and transcriptomics analyses of the HrpL regulon in Pseudomonas savastanoi pathovars of woody host.

    The species Pseudomonas savastanoi, a member of the Pseudomonas syringae complex, includes four pathovars causing knots or excrescences in woody hosts: P. savastanoi pv. savastanoi (Psv), pv. fraxini (Psf), pv. nerii (Psn) and pv. retacarpa (Psr), comprising isolates from olive, ash, oleander and broom plants, respectively. One of the better characterized pathogenicity factor in this bacterial complex is the type III secretion system (T3SS) and its effector (T3E) repertoire. Transcription of the T3SS structural genes and most of its T3E genes is positively regulated HrpL. We have constructed ∆hrpL mutants in a model strain belonging to each of this four pathovars and analyzed the role of this gene in the pathogenicity of P. savastanoi. As previously reported for Psv and Psn, the hrpL gene is also necessary in Psf and Psr for both symptoms production during a compatible interaction and the induction of the hypersensitive response in a resistant host. To characterize the HrpL regulon, we are currently following two strategies (i) the bioinformatics identification of HrpL-dependent promoters in the genomes of several P. savastanoi pathovars and, (ii) a comparative RNAseq analysis of a wild-type Psv strain and its ∆hrpL mutant. The results obtained will be compared with those published for other P. syringae strains isolated from herbaceous hosts, in order to identify specific HrpL-dependent proteins involved in the interaction with woody hosts.Universidad de Málaga.     We derive by program transformation Pierre Crégut s full-reducing Krivine machine KN from the structural operational semantics of the normal order reduction strategy in a closure-converted pure lambda calculus. We thus establish the correspondence between the strategy and the machine, and showcase our technique for deriving full-reducing abstract machines. Actually, the machine we obtain is a slightly optimised version that can work with open terms and may be used in implementations of proof assistants

    Plasticity coupled with thermo-electric fields: Thermodynamics framework and finite element method computations

    [EN] A consistent thermodynamic-based theoretical framework and three-dimensional finite element formulation is pre- sented, capable of coupling elastic, thermal and electric fields. The complete set of governing equations are obtained from conservation principles for electric charge, energy and momentum. The second principle of thermodynamics is taken into account to introduce the irreversible phenomena, such as plastic dissipation or Joule heating. The constitu- tive relations are derived consistently from the Helmholtz free-energy potential for each corresponding dual variable in terms of the defined set of state variables. We consider the case of linear isotropic hardening model for plasticity, and provide the consistent form of the tangent thermo-electro-elastoplastic modulus through dual variable computa- tions. The latter plays the crucial role in ensuring fast convergence properties of the finite element computations with the proposed coupled plasticity model. The implementation is carried out in a research version of the well-known computer code FEAP.     European Union poverty policies. The case of the statistical policy

    A long-lasting difficulty in dealing with the subject of poverty, both in the scientific and political realms, has been the lack of analytical and methodological instruments that facilitate defining, observing and measuring accurately the social dimensions of the phenomenon. Henceforth, it is crucial to develop further indicators in order to design and implement a consistent statistical policy. After examining these issues, this paper analyses the European Union policy on poverty and, especially, the efforts done in the field of statistical policy.Uma dificuldade de longa data nos estudos sobre a pobreza, quer a nível académico quer a nível político, reside na escasez de instrumentos analíticos e metodológicos para definir, observar e medir as dimensões sociais da pobreza. Portanto, é importante desenvolver ferramentas e indicadores que permitam formular e implementar uma política de estatísticas consistente. Após examinar estas questões, este artigo analisa a política da União Europeia relativamente à pobreza, com destaque para a área das políticas de estatísticas

    Linear elastic mechanical system interacting with coupled thermo-electro-magnetic fields

    [EN] A fully-coupled thermodynamic-based transient finite element formulation is proposed in this article for electric, magnetic, thermal and mechanic fields interactions limited to the linear case. The governing equations are obtained from conservation principles for both electric and magnetic flux, momentum and energy. A full-interaction among different fields is defined through Helmholtz free-energy potential, which provides that the constitutive equations for corresponding dual variables can be derived consistently. Although the behavior of the material is linear, the coupled interactions with the other fields are not considered limited to the linear case. The implementation is carried out in a research version of the research computer code FEAP by using 8-node isoparametric 3D solid elements. A range of numerical examples are run with the proposed element, from the relatively simple cases of piezoelectric, piezomagnetic, thermoelastic to more complicated combined coupled cases such as piezo-pyro-electric, or piezo-electro-magnetic.     Comparative Analysis of the Type III Secretion System Effector Repertoires of Pseudomonas savastanoi Pathovars Pathogenic on Woody Hosts

    Comunicación de tipo pósterThe species Pseudomonas savastanoi, a member of the Pseudomonas syringae complex, includes four pathovars causing knots or excrescences in woody hosts: P. savastanoi pv. savastanoi (Psv), pv. fraxini (Psf), pv. nerii (Psn) and pv. retacarpa (Psr), comprising isolates from olive, ash, oleander and broom plants, respectively. Pathogenicity of P. savastanoi is dependent, among other factors, on the type III secretion system (T3SS) and its effector (T3E) repertoire. Furthermore, a putative role in the interaction with woody hosts has been suggested for several of these T3E. The recent availability of the genome sequences of several P. savastanoi strains isolated from different hosts has facilitated bioinformatics predictions of their T3SS genes and T3E pools, the study of their distribution in other strains of the P. syringae complex isolated from woody hosts and the functional analysis of several of these secreted proteins. As previously reported for Psv, Psn and Psf, here we show that pathogenicity of Psr ICMP16945, is also dependent on the T3SS. Psv strains NCPPB 3335, ICMP4352 and PseNe107 share a core set of at least 22 T3E, 18 of which are also encoded in Psn ICMP16943, Psf ICMP7711 and Psr ICMP16945. However, these three strains encode truncated versions of 1-2 of these 18 T3E and, Psr ICMP16945 contains three pathovarspecific T3E. Our results also show that several T3E, including HopAO1, are phylogenetically clustered across the P. syringae complex according to the woody/herbaceous nature of their host of isolation, suggesting host specialization of these effectors in this complex.Universidad de Málaga.     En este trabajo se desarrolla un nuevo indicador mensual de cuellos de botella en las cadenas de suministro utilizando artículos de prensa. El índice de cuellos de botella de oferta (SBI, por sus siglas en inglés) proporciona una narración coherente de los problemas de suministro relacionados con las guerras, las catástrofes naturales, las huelgas y, más recientemente, la pandemia ocasionada por el COVID-19. Las innovaciones en el SBI tienen importantes implicaciones macroeconómicas: un aumento del SBI actúa como un shock de costes, disminuyendo la producción industrial y el empleo, y presionando al alza los precios, lo que dificulta la actuación de la política monetaria.We develop a new monthly indicator of supply bottlenecks using newspaper articles. The supply bottlenecks index (SBI) provides a consistent narrative of supply issues related to wars, natural disasters, strikes and, most recently, the COVID-19 pandemic. Innovations in the SBI have important macroeconomic implications: an increase in the SBI functions as a cost-push shock, decreasing industrial production and employment, and pushing prices up, so that monetary policy faces important trade-offs

    Análisis genómico y transcriptómico de los efectores del sistema de secreción tipo III en Pseudomonas savastanoi pv. savastanoi NCPPB 3335

    Presentación oralLa especie Pseudomonas savastanoi pertenece al complejo Pseudomonas syringae, el cual está constituido por un conjunto de bacterias fitopatógenas Gram (-) de alto interés agrícola y económico. Dentro de esta especie se han descrito hasta la fecha cuatro patovares capaces de infectar huéspedes leñosos: pv. savastanoi (aislados de olivo), pv. nerii (aislados de adelfa), pv. fraxini (aislados de fresno) y pv. retacarpa (aislados de retama). La cepa NCPPB 3335 del patovar savastanoi (Psv), establecida como modelo para el estudio de la interacción de Pseudomonas patógenas con plantas leñosas, es el agente causal de la tuberculosis del olivo, enfermedad caracterizada por la aparición de tumores en las partes aéreas de la planta. Uno de los factores de patogenicidad de Psv más relevante es el sistema de secreción tipo III (T3SS) y su repertorio de efectores (T3E). La disponibilidad de la secuencia de Psv NCPPB 3335 nos ha permitido la predicción bioinformática de los genes que codifican su T3SS y su repertorio de T3E en base a la homología con otros efectores previamente descritos. Esta cepa presenta un repertorio de efectores constituido por 28 T3E. Con el objetivo de analizar la expresión del repertorio de T3E en Psv, se ha llevado a cabo un análisis RNAseq comparativo entre la cepa silvestre Psv NCPPB 3335 y un mutante de la misma del gen hrpL, el cual codifica un activador transcripcional de los genes del T3SS y de la mayoría de sus T3E. Las secuencias procedentes de este RNAseq se están analizando actualmente y los resultados que se obtengan del mismo nos permitirá no sólo analizar la expresión de los T3E que hemos identificado en esta cepa, sino también caracterizar el regulón HrpL completo en la misma y quizás identificar nuevos posibles T3E. Los resultados obtenidos, se compararán con una predicción de promotores regulados por HrpL (hrp-box), que se llevará a cabo utilizando una herramienta bioinformática generada por nuestro equipo.Universidad de Málaga.
