    Development of sandwich and competitive ELISA formats to determine ß-conglycinin: Evaluation of their performance to detect soy in processed food

    Two ELISA formats (sandwich and indirect competitive) were developed to quantify ß-conglycinin, a major soy allergen. Their performance was evaluated using three model foods incurred with soy proteins. The sandwich format detects the addition of 0.005% and 0.05% soy proteins in pasteurized sausages and baked bread. However, the competitive format detects only 0.1 and 0.5%, respectively. ß-conglycinin was not detected in sterilized pâtè with any format. An industrial prototype of the sandwich ELISA was in-house validated, showing acceptable results of repeatability, reproducibility and robustness. Model foods spiked with ß-conglycinin after processing showed recoveries between 93.3 and 138.7%. However, in model foods incurred with soy proteins before processing the recovery decreased with the increase of the severity of heat treatment applied. The sandwich format could differentiate most of the retail foods with soy declared or not as ingredient.     Depositional sequences and ammonoid assemblages in the upper Cenomanian-lower Santonian of the Iberian Peninsula (Spain and Portugal)

    A clear relationship exists between eustatic sea-level rises and falls recorded as cyclical depositional sequences and ammonite faunas during the Cenomanian-Santonian in the Iberian and West Portuguese basins. Most of the faunal turnovers correlate with stratigraphic intervals related to marine transgressions, maximum flooding of the shelf (locally associated to anoxic events), and marine regressions. Specifically, within each depositional sequence, three distinct and identical events of morphological change occur, involving ammonoids belonging to different groups. Transgressive sediments are characterized by moderately ornamented, inflated and evolute morphologies, which are replaced by smooth, involute and compressed oxycones (the most hydrodynamic shells) during maximum flooding (and to a lesser extent at the early highstand) of the sequences. The latter morphologies in turn are replaced by coarsely ornamented and evolute shells during late highstands. We conclude that ammonoid faunal analysis can be used to trace sea-level changes and provides an additional tool for sequence stratigraphy

    Development of sandwich ELISA and lateral flow immunoassay to detect almond in processed food

    Almond (Prunus dulcis) represents a potential allergenic hazard that should be included in Allergen Control Plans. In this study, sandwich ELISA and lateral flow immunoassay (LFIA), using amandin (Pru du 6) as the target protein, were developed to detect almond in processed food and validated according to international guides. ELISA could detect 2 ng/mL and LFIA 30 ng/mL of pure amandin. No cross-reactivity was found on a panel of 50 food commodities with the exception of Pecan nut, Brazil nut and chestnut for which the cross-reactivity was lower than 0.02%. Furthermore, ELISA and LFIA were able to detect 0.12 and 0.70 ppm of almond protein in foods spiked with almond extract whereas 0.20 and 2.0 ppm could be detected in baked cookies incurred with almond, respectively. Both techniques could be applied for food manufacturers and control agencies for monitoring the presence of almond traces in food and working surfaces. © 2021 The Author(s

    Non-iterative computation of Gauss-Jacobi quadrature

    Asymptotic approximations to the zeros of Jacobi polynomials are given, with methods to obtain the coefficients in the expansions. These approximations can be used as standalone methods for the noniterative computation of the nodes of Gauss--Jacobi quadratures of high degree (n≥100n\ge 100). We also provide asymptotic approximations for functions related to the first-order derivative of Jacobi polynomials which are used for computing the weights of the Gauss--Jacobi quadrature. The performance of the asymptotic approximations is illustrated with numerical examples, and it is shown that nearly double precision relative accuracy is obtained for both the nodes and the weights when n≥100n\ge 100 and −1<α,β≤5-1< \alpha, \beta\le 5. For smaller degrees the approximations are also useful as they provide 10−1210^{-12} relative accuracy for the nodes when n≥20n\ge 20, and just one Newton step would be sufficient to guarantee double precision accuracy in that cases

    Influence of Age on Stress Responses to Metabolic Cage Housing in Rats

    SUMMARY 1. We studied the effect of isolation stress in 3-and 12-month-old rats individually housed in metabolic cages for 7 days. Urine (24 hr) was collected daily from one group of animals of each age. The other group was tested in an open field and on a hot plate on days 1 and 7. 2. Total deambulation in the open-field test was lower in young than in older rats both on day 1 (54.7 Ϯ 9.9 vs 80 Ϯ 8.9 crossings/session; P Ͻ 0.04) and on day 7 (21 Ϯ 9 vs 48 Ϯ 7 crossings per session; P Ͻ 0.04) and decreased significantly in the two groups when tested on day 7 (P Ͻ 0.03). Latency to paw-licking in the hot-plate test was longer in young than in older animals on day 1 (14 Ϯ 2 vs 8 Ϯ 4 sec; P Ͻ 0.05) but was similar in the two groups on day 7. 3. Urinary excretions of norepinephrine (NE) and epinephrine (E) were determined by HPLC with electrochemical detection. Urinary NE in day 1 was similar in young and older animals (2627 Ϯ 828 vs 3069 Ϯ 598 ng/24 hr). In young animals NE excretion decreased along the study and was significantly (P Ͻ 0.02) lower than on day 1 during the last 3 days of the study. Conversely, in older animals urinary excretion of NE remained similar throughout the study. On day 7 urinary excretion of NE in older animals was about two fold that in young rats. Urinary E was similar in young and older rats (341 Ϯ 127 vs 532 Ϯ 256 ng/24 hr) on day 1 and showed a tendency to increase throughout the study. 4. Urinary monoamine oxidase inhibitory (IMAO) activity was determined by testing the ability of urine extracts to inhibit rat liver MAO activity in vitro and was higher in young than in older animals throughout the study (day 1, 54.8 Ϯ 4.2 vs 25.1 Ϯ 5.1%; P Ͻ 0.02). In young rats excretion of IMAO was significantly higher during the last 3 days of the study than on day 1 (P Ͻ 0.05). In older animals urinary IMAO showed a tendency to increase at the end of the study. 5. Isolation stress caused by housing rats in metabolic cages results in different behavioral and metabolic responses in young and older animals. Young animals exhibit a lower locomotor and analgesic response and excrete lower amounts of NE and higher IMAO activity in the urine than older rats. The metabolic and behavioral responses to isolation stress are highly dependent on the age of the animals tested. These results should be taken into consideration when designing experiments requiring the use of metabolic cages

    Integrative immune transcriptomic classification improves patient selection for precision immunotherapy in advanced gastro-oesophageal adenocarcinoma

    BACKGROUND: Advanced gastro-oesophageal cancer (GEA) treatment has been improved by the introduction of immune checkpoint inhibitors (CPIs), yet identifying predictive biomarkers remains a priority, particularly in patients with a combined positive score (CPS) < 5, where the benefit is less clear. Our study assesses certain immune microenvironment features related to sensitivity or resistance to CPIs with the aim of implementing a personalised approach across CPS < 5 GEA. DESIGN: Through integrative transcriptomic and clinicopathological analyses, we studied in both a retrospective and a prospective cohort, the immune tumour microenvironment features. We analysed the cell types composing the immune infiltrate highlighting their functional activity. RESULTS: This integrative study allowed the identification of four different groups across our patients. Among them, we identified a cluster whose tumours expressed the most gene signatures related to immunomodulatory pathways and immunotherapy response. These tumours presented an enriched immune infiltrate showing high immune function activity that could potentially achieve the best benefit from CPIs. Finally, our findings were proven in an external CPI-exposed population, where the use of our transcriptomic results combined with CPS helped better identify those patients who could benefit from immunotherapy than using CPS alone (p = 0.043). CONCLUSIONS: This transcriptomic classification could improve precision immunotherapy for GEA

    Extra-uterine (abdominal) full term foetus in a 15-day pregnant rabbit

    [EN] Background: While ectopic pregnancies account for 1-2% of all pregnancies, abdominal pregnancy is extremely rare, accounting for approximately 1% of ectopic pregnancies. Extrauterine abdominal pregnancy is defined as the implantation and development of an embryo in the peritoneal cavity. The present report is the first of an incidental case of abdominal pregnancy within four full-term foetus simultaneously with 2 weeks of physiological gestation in a healthy doe rabbit. Case presentation: The doe was born on November 3, 2014 and the first partum took place on May 18, 2015. The doe had previously delivered and weaned an average of 12.0 +/- 1.41 live kits at birth (no stillbirths were recorded) during 5 consecutive pregnancies. The last mating was on December 18, 2015 and the detection of pregnancy failure post breeding (by abdominal palpation) on December 31, 2015. Then, the doe was artificially inseminated on January 27, 2016, diagnosed pregnant on February 11, 2016 and subsequently euthanized to recover the foetus. A ventral midline incision revealed a reproductive tract with 12 implantation sites with 15 days old foetus and 4 term foetus in abdominal cavity. There were two foetus floating on either side of the abdominal cavity and two suspended near the greater curvature of the stomach. They were attached to internal organs by means of one or 2 thread-like blood vessels that linked them to the abdominal surfaces.     Bow shocks, nova shells, disc winds and tilted discs: the nova-like V341 Ara has it all

    V341 Ara was recently recognized as one of the closest (d ≃ 150 pc) and brightest (V ≃ 10) nova-like cataclysmic variables. This unique system is surrounded by a bright emission nebula, likely to be the remnant of a recent nova eruption. Embedded within this nebula is a prominent bow shock, where the system’s accretion disc wind runs into its own nova shell. In order to establish its fundamental properties, we present the first comprehensive multiwavelength study of the system. Long-term photometry reveals quasi-periodic, super-orbital variations with a characteristic time-scale of 10–16 d and typical amplitude of ≃1 mag. High-cadence photometry from theTransiting Exoplanet Survey Satellite (TESS) reveals for the first time both the orbital period and a ‘negative superhump’ period. The latter is usually interpreted as the signature of a tilted accretion disc. We propose a recently developed disc instability model as a plausible explanation for the photometric behaviour. In our spectroscopic data, we clearly detect antiphased absorption and emission-line components. Their radial velocities suggest a high mass ratio, which in turn implies an unusually low white-dwarf mass. We also constrain the wind mass-loss rate of the system from the spatially resolved [O III] emission produced in the bow shock; this can be used to test and calibrate accretion disc wind models. We suggest a possible association between V341 Ara and a ‘guest star’ mentioned in Chinese historical records in AD 1240. If this marks the date of the system’s nova eruption, V341 Ara would be the oldest recovered nova of its class and an excellent laboratory for testing nova theory
