    Tackling CD147 exosome-based cell-cell signaling by electrochemical biosensing for early colorectal cancer detection

    The great opportunities represented by exosomes in liquid biopsy diagnostics and the relevance of CD147 protein as diagnostic and prognostic cancer biomarker led us to develop the first bio-electroanalytical platform for the determination of exosomal CD147 (exoCD147) by exploiting micro-sized magnetic beads coated with specific anti-CD147 antibodies. The captured exosomal target protein was sandwiched by specific biotin functionalized detector antibodies followed by attaching streptavidin-HRP conjugate to perform the amperometric reading using screen-printed carbon electrodes (SPCEs) as electrode transducers in the presence of hydroquinone (HQ) and H2O2. The analytical and operational characteristics achieved by implementing this simple methodology allowed the sensitive (LOD 29 pg mL-1) and selective determination of CD147 and the analysis of exoCD147 in different but inter-related real clinical scenarios including lysed and entire exosomes previously isolated from CRC cell lines with different metastatic potential.     First bioelectronic immunoplatform for quantitative secretomic analysis of total and metastasis-driven glycosylated haptoglobin

    The glycosylation status of proteins is increasingly used as biomarker to improve the reliability in the diagnosis and prognosis of diseases as relevant as cancer. This feeds the need for tools that allow its simple and reliable analysis and are compatible with applicability in the clinic. With this objective in mind, this work reports the first bioelectronic immunoplatforms described to date for the determination of glycosylated haptoglobin (Hp) and the simultaneous determination of total and glycosylated Hp. The bioelectronic immunoplatform is based on the implementation of non-competitive bioassays using two different antibodies or an antibody and a lectin on the surface of commercial magnetic microcarriers. The resulting bioconjugates are labeled with the horseradish peroxidase (HRP) enzyme, and after their magnetic capture on disposable electroplatforms, the amperometric transduction using the H2O2/hydroquinone (HQ) system allows the single or multiple detection. The developed immunoplatform achieves limits of detection (LODs) of 0.07 and 0.46 ng mL-1 for total and glycosylated Hp in buffer solution, respectively. The immunoplatform allows accurate determination using simple and relatively short protocols (approx. 75 min) of total and glycosylated Hp in the secretomes of in vitro-cultured colorectal cancer (CRC) cells with different metastatic potentials, which is not feasible, due to lack of sensitivity, by means of some commercial ELISA kits and Western blot methodology.Funding Open Access funding provided thanks to the CRUE-CSIC agreement with Springer Nature.     Electrochemical immunoplatform to help managing pancreatic cancer

    Pancreatic ductal adenocarcinoma (PDAC) is the solid tumor with the worst prognosis, representing today the third cause of cancer-related deaths in developed countries and expected to be the second in 2030. Today, CA19-9 remains the only clinically used marker for management of PDAC (FDA-approved as a disease moni- toring marker). This work reports a disposable amperometric immunoplatform for the determination of CA19-9. The immunoplatform skilfully combines the advantages of magnetic microsupports (MBs) for implementation of the immunoassay and amperometric transduction on screen-printed carbon electrodes (SPCEs). The method involves the preparation of sándwich immunocomplexes enzymatically labeled with the enzyme horseradish peroxidase (HRP) on the MBs and uses a detection antibody conjugated to HRP. Once the HRP- labeled sandwich immunocomplexes-bearing MBs were trapped on the SPCE surface, the variation of the catho- dic current was measured in the presence of H2O2 and hydroquinone (HQ), which was directly proportional to the concentration of CA19-9. Under the optimized experimental conditions, the immunoplatform allowed the amperometric determination of CA19-9 standards over the 5.0 to 500 U mL−1 concentration range, with a limit of detection (LOD) value of 1.5 U mL−1 in 1 h. The method exhibits good reproducibility and selectivity and the magnetic immunoconjugates shows a good storage stability. The immunoplatform was applied to the deter- mination of CA19-9 in serum samples of a medium-sized cohort (22 individuals) of healthy subjects and patients diagnosed with PDAC. The obtained results demonstrated the immunoplatform ability to discriminateboth types of individuals within 1 h after sample dilution. The developed immunoplatform represents an improvement in terms of cost, applicability and accessibility compared to the ELISA-based techniques currently used in the clinic

    Electrochemical immunosensing of Growth arrest‐specific 6 in human plasma and tumor cell secretomes

    Growth arrest-specific 6 (GAS6) protein plays a key role in processes related toproliferation,inflammation,angiogenesis,andatheroscleroticplaqueformation.In addition, it has been reported that plasma levels of GAS6 are related to cancerprognosis and other relevant pathologies, such as heart failure or sepsis. Wereport here the first electrochemical immunoplatform for the determination ofGAS6, which has demonstrated to be competitive with other available method-ologies in terms of cost, simplicity, and decentralized application. The developedimmunoplatform involves a sandwich immunoassay using magnetic microparti-cles (MBs) and uses amperometric detection at disposable screen-printed carbonelectrodes (SPCEs). The MBs were modified with an antibody specific to GAS6for its selective capture, which is further recognized by a biotinylated secondaryantibody subsequently labeled with a streptavidin-horseradish peroxidase(Strep-HRP) conjugate. The electrochemical detection was carried out using thehydroquinone (HQ)/H2O2system.     Revealing prevalent cancers by interrogating glycoproteins with sustainable immunoelectrochemical tools

The worldwide incidence and death toll of colorectal and pancreatic cancers (CRC and PDAC) have increased considerably since 1990. For this reason, both early detection and regular follow-up are considered key factors in improving patient prognosis. In this sense, the determination of the total content of specific proteins and their aberrantly glycosylated fraction in oncologic processes could help to achieve the proposed goals. Results and Discussion. In this work, two simple but highly competitive electrochemical immunoplatforms for the determination of total and glycosylated post-translational modified haptoglobin (Hp) [1], and CA19-9 [2] (candidate biomarkers associated with colorectal and pancreatic cancer, respectively) are presented. As seen in Figure 1, these biotools are uplifted in the use of magnetic immunocaptors and another antibody or a lectin as detector elements lastly labeled with HRP, which enables subsequent amperometric detection. The presented bioplatforms exhibit attractive characteristics in terms of simplicity, affordability, and point-of-care application compared to the conventional available methodologies, highlighting low detection limits (0.07 and 0.46 ng mL¿1 for total and glycosylated Hp, respectively, and 1.5 U mL¿1 for CA19-9), and short assay times (< 2 h). The workability of these quantitative bioplatforms for the analysis of secretomes from cultured CRC cells with the distinct potential to metastasize (Hp) or serum samples from healthy and PDAC-diagnosed subjects (CA19-9) was assessed to definitely confirm full exploitation of all the above exposed enticing attributes. Conclusions. Our findings clearly revealed the unquestionable ability of these modern electrochemical immunoplatforms to discriminate between healthy and cancer-diagnosed subjects, as well as to assess disease progression, positioning these simple but effective methodologies as advanced electroanalytical tools with proven real biomedical applications, and the hope of aiding in the accurate diagnosis of prevalent and high mortality cancers

    Individual profiles and wine consumption patterns in Tenerife.: A multinomial logit model

    The aim of this study is to analyze the decisions on the frequency of wine consumption to detect the individual characteristics of the potential consumer that influence the propensity towards each decision. Discrete choice models, and in particular, logit models have been seen as tools which are able to identify the way in which a specific combination of characteristics that define the residents of Tenerife conditions his decision on consumption frequency. In general, a regular consumption is not usual for women, which only do so occasionally. On the other hand, the ratio of occasional consumption probability to high frequency consumption probability falls as the age increases. In this sense, the identification of the combinations of individual characteristics associated with patterns of extreme consumption clearly indicates that women and young people are the two consumer segments which should be objects of specific marketing strategies. Any case, the design of such strategies requires investigating about individual preferences and goes beyond the scope of this paper.En este trabajo se analizan las decisiones sobre la frecuencia de consumo de vino de los residentes en Tenerife con objeto de detectar las características individuales del potencial consumidor que determinan una mayor propensión hacia cada una de estas decisiones. Los modelos de elección discreta y en particular los modelos logit se han mostrado como una herramienta capaz de desentrañar el modo en que ciertas combinaciones de atributos que definen al individuo en cuestión condicionan su decisión sobre la frecuencia de consumo. En general, las mujeres no pueden incluirse en el grupo de consumidores de alta frecuencia y si consumen, sólo lo hacen ocasionalmente. Por otro lado, el ratio entre la probabilidad de consumo ocasional y la probabilidad de alta frecuencia disminuye a medida que aumenta la edad. En este sentido, la identificación de combinaciones de características individuales asociadas a patrones de consumo extremos muestra con claridad que las mujeres y los jóvenes constituyen dos segmentos de consumidores a los que convendría dirigir estrategias comerciales específicas. En cualquier caso, el diseño de estas estrategias requiere conocer las preferencias individuales y queda fuera del alcance de este trabajo

    The present paper tries to assess the way that individual socio-economic features have an influence on the frequency of consumption of different kinds of wine and, on the other hand, on the number of different kinds of wine that are consumed, so that distinct consumer profiles can be identified. Discrete choice models are used for this purpose: binomial logit models and Poisson models, respectively. We use data from a survey on wine consumption in Tenerife that took place between April and May of 2001.Este artículo pretende evaluar en qué medida los rasgos socioeconómicos de los individuos influyen, por un lado, en la frecuencia de consumo de los diferentes tipos de vino y por otro lado, en el número de tipos de vino diferentes que se consumen, con el propósito de identificar distintos perfiles de consumidores de vino. Las herramientas estadísticas utilizadas en ambos casos se encuadran dentro de los modelos de elección discreta: modelo logit binomial y modelos de Poisson, respectivamente y los datos empleados proceden de una encuesta sobre consumo de vino en Tenerife realizada en abril y mayo de 2001
