    Involment of 5HT1A receptors in the angiogenic-like effects of the Uncaria tomentosa (Willd) DC aqueous extract

    1 p.The aqueous extract of Uncaria tomentosa (EUT) prepared by decoction of the root and stem bark is used in the treatment of several diseases. There is evidence showing the interaction of the extract components with serotonin receptors. Particularly, the 5HT1A receptors strongly involved in the etiology of anxiety and depression disorders. The aim of the present work was to study the involvement of 5HT1A receptors in the anxiogenic-like effect induced by the aqueous extract of Uncaria tomentosa and the influence of estrous cycle. Female albino swiss mice stereotaxically implanted with bilateral stainlesssteel cannuli into lateral ventricles were used. During 20 days, the mice were orally administered with water or EUT. It was used NAN 190 described as 5HT1A autoreceptors partial agonist and 5HT1A postsynaptic receptor antagonist. The animals receiveda 1 μl intracerebroventricular injection of vehicle or NAN 190 (0.10 μg/μl o 0.25 μg/μl). Five minutes later, the mice were tested in the elevated plus maze. The results obtained showed a significant anxiogenic effect induced by EUT and the higher dose of NAN 190 during the estrous although no differences were found when NAN 190 was administered in EUT treated animals. In the diestrous stage, the low dose of NAN 190 induced an anxiolytic effect and this response was blunted in EUT treated animals. Interestingly, an anxiolytic effect was found with the higher dose of NAN 190 in EUT treated animals. Our findings support a differential action of EUT depending on estrous cycle that could be explained as a desensitization of the 5HT1A autoreceptor induced by estrogen. Since the anxiogenic effect of EUT is observed only under desensitization conditions of 5HT1A autoreceptors, it was concluded that EUT could induce anxiety through an indirect stimulation of the raphe nucleus.Fil: Ortiz, María Julia.     Apprehending from our hearts the experience of living in peace

    Este texto presenta una experiencia que busca privilegiar la escuela como escenario para la construcción de identidades diversas, en una comunidad educativa ubicada en la periferia de la ciudad de Bogotá, Colombia. Allí, un grupo de docentes procura transformar los muros de ladrillo por un entretejido poroso e intercultural que permita la circulación y apropiación de referentes de identidad histórica y cotidiana. Es un recorrido a lo largo de 17 años para re-conocernos como docentes y aprendices, desde nuestras historias familiares y trayectorias de vida cotidiana. Describe nuestras transformaciones pedagógicas frente a la arremetida de la urbanización violenta sobre un territorio rural, pero también de asentamiento ancestral indígena. El análisis está centrado sobre nuestro ejercicio de retomar las sabidurías ancestrales como insumo, pero además las enseñanzas de las personas y familias que han llegado a este territorio, desplazadas por la violencia en Colombia.This text presents an experience that seeks to privilege the school as a scenario for the construction of diverse identities, in an educational community located in the periphery of the city of Bogotá, Colombia. There, a group of teachers tries to transform the brick walls by a porous and intercultural interweave that allows the circulation and appropriation of historical and daily identity referents. It is a journey through 17 years to re-know ourselves as teachers and apprentices, from our family histories and trajectories of daily life. It describes our pedagogical transformations in the face of the onslaught of violent urbanization on a rural territory, but also of indigenous ancestral settlement. The analysis is centered on our exercise of retaking the ancestral wisdoms as an input, but also the teachings of the people and families that have arrived in this territory, displaced by the violence in Colombia

    PAI1 is a Marker of Bad Prognosis in Rectal Cancer but Predicts a Better Response to Treatment with PIM Inhibitor AZD1208

    Colorectal cancer (CRC) is the third most common cancer worldwide. The standard treatment in locally advanced rectal cancer is preoperative radiation alone or in combination with chemotherapy, followed by adjuvant chemotherapy. Rectal cancer is highly lethal, with only 20% of patients showing a complete remission (by RECIST) after standard treatment, although they commonly show local or systemic relapse likely due to its late detection and high chemotherapy resistance, among other reasons. Here, we explored the role of PAI1 (Serpin E1) in rectal cancer through the analyses of public patient databases, our own cohort of locally advanced rectal cancer patients and a panel of CRC cell lines. We showed that PAI1 expression is upregulated in rectal tumors, which is associated with decreased overall survival and increased metastasis and invasion in advanced rectal tumors. Accordingly, PAI1 expression is correlated with the expression of (Epithelial-to-Mesenchymal Transition) EMT-associated genes and genes encoding drug targets, including the tyrosine kinases PDGFRb, PDGFRa and FYN, the serine/threonine kinase PIM1 and BRAF. In addition, we demonstrate that cells expressing PAI1 protein are more sensitive to the PIM inhibitor AZD1208, suggesting that PAI1 could be used to predict response to treatment with PIM inhibitors and to complement radiotherapy in rectal tumors.España Consejería de Salud of the Junta de Andalucía (PI-0397-2017)España , Consejeria of Economía, Conocimiento, Empresas y Universidad of the Junta de Andalucia (P18-RT-2501

    El radioteatro en España: marco de referencia para una aproximación diacrónica

    The main objective of this article is to give a contextualization of radio drama in Spain, over a period of three decades in the last century, between the 1940s and the 1970s. Through retrospective analysis on its history some of the keys related to narrative and drama genres implementation are assessed. The research methodology used can be classified as theoretical-conceptual, since it has been based on the search, compilation and analysis of documentation that have allowed radio theater to be contextualized in those three decades. As a result of the research, the relationship between radio plays and social context is revealed and the contributions of the main Spanish authors and directors of radio drama in those decades are referenced. The final conclusion points to the fact that radio drama throughout radio history has been appearing and disappearing conditioned by programmers and public demand. Despite this, even by the end of the seventies, when radio programmers stood up for informative content, some radio dramatic serials would continue in the radio programming: La Saga de los Porretas (Cadena SER) or Sobrenatural and Historias (RNE).El objetivo del presente artículo es ofrecer una contextualización del radioteatro en España entre las décadas de los años cuarenta a los años setenta del siglo pasado. A través de un análisis retrospectivo de su historia, se hace hincapié en algunas de las claves de su implantación como género narrativo y dramático. Se utiliza una metodología teórico-conceptual, basada en el análisis documental, que ha permitido circunscribir y referenciar temporalmente el radioteatro en el periodo señalado. El resultado del análisis pone de manifiesto la relación de los radioteatros con la situación social y la importancia de su legado literario y cultural. Igualmente, se referencian las aportaciones de los autores y realizadores españoles de radioteatro en esas décadas. La conclusión pone de manifiesto que, a lo largo de la historia de la radio, el radiodrama aparece y desaparece condicionado por sus programadores y por la demanda del público. A pesar de la apuesta de la radio por los contenidos informativos en su programación, a partir de finales de los años setenta, se mantuvieron en antena algunos seriales, como La saga de los Porretas (1976-1988), en la Cadena SER, y Sobrenatural (1994-1996), o Historias (1997-2003), en Radio Nacional de España

    A high magnesium concentration in citrate dialysate prevents oxidative stress and damage in human monocytes in vitro

    Background. The use of dialysis fluids (DFs) during haemodialysis has been associated with increased oxidative stress and reduced serum magnesium (Mg) levels, contributing to chronic inflammation. Since the role of Mg in modulating immune function and reducing oxidative stress has been demonstrated, the aim of this study was to characterize in vitro whether increasing the Mg concentration in DFs could protect immune cells from oxidative stress and damage. Methods. The effect of citrate [citrate dialysis fluid (CDF), 1 mM] or acetate [acetate dialysis fluid (ADF), 3 mM] dialysates with low (0.5 mM; routinely used) or high (1 mM, 1.25 mM and 2 mM) Mg concentrations was assessed in THP-1 human monocytes. The levels of reactive oxygen species (ROS), malondialdehyde (MDA) and oxidized/reduced (GSSG/GSH) glutathione were quantified under basal and inflammatory conditions (stimulation with lipopolysaccharide, LPS). Results. The increase of Mg in CDF resulted in a significant reduction of ROS production under basal and inflammatory conditions (extremely marked in 2 mM Mg; P< 0.001). These effects were not observed in ADF. Interestingly, in a dose-dependent manner, high Mg doses in CDF reduced oxidative stress in monocytes under both basal and inflammatory conditions. In fact, 2 mM Mg significantly decreased the levels of GSH, GSSG and MDA and the GSSG/GSH ratio in relation to 0.5 mM Mg. Conclusions. CDF produces lower oxidative stress than ADF.     Increasing the Magnesium Concentration in Various Dialysate Solutions Differentially Modulates Oxidative Stress in a Human Monocyte Cell Line

    Oxidative stress is exacerbated in hemodialysis patients by several factors, including the uremic environment and the use of dialysis fluids (DFs). Since magnesium (Mg) plays a key role in modulating immune function and in reducing oxidative stress, we aimed to evaluate whether increasing the Mg concentration in different DFs could protect against oxidative stress in immunocompetent cells in vitro. Effect of ADF (acetate 3 mM), CDF (citrate 1 mM), and ACDF (citrate 0.8 mM + acetate 0.3 mM) dialysates with Mg at standard (0.5 mM) or higher (1, 1.25, and 2 mM) concentrations were assessed in THP-1 monocyte cultures. Reactive oxygen species (ROS) and malondialdehyde (MDA) levels were quantified under basal and uremic conditions (indoxyl sulfate (IS) treatment). Under uremic conditions, the three DFs with 0.5 mM Mg promoted higher ROS production and lipid damage than the control solution. However, CDF and ACDF induced lower levels of ROS and MDA, compared to that induced by ADF. High Mg concentration (1.25 and/or 2 mM) in CDF and ACDF protected against oxidative stress, indicated by reduced ROS and MDA levels compared to respective DFs with standard concentration of Mg. Increasing Mg concentrations in ADF promoted high ROS production and MDA content. Thus, an increase in Mg content in DFs has differential effects on the oxidative stress in IS-treated THP-1 cells depending on the dialysate used.Instituto de Salud Carlos IIISociedad Española de Nefrologí
