    Supervivencia de Erwinia amylovora en condiciones de estrés: influencia de la presencia de cobre y la limitación de nutrientes

    La bacteria Erwinia amylovora es el agente causal del fuego bacteriano de las rosáceas, responsable de importantes daños en varios frutales de pepita y plantas ornamentales. Debido a su fácil diseminación y difícil control, está considerada como una enfermedad de cuarentena en la Unión Europea. A pesar de los numerosos estudios realizados sobre E. amylovora, aún no se conoce su ciclo biológico completo, y la información sobre su posible supervivencia en condiciones de estrés y/o en hábitats fuera de hospedadores susceptibles es aún muy escasa. Por ello, en esta memoria se ha planteado estudiar la supervivencia de E. amylovora en un medio mineral con cobre, metal ampliamente usado para el control de bacteriosis de plantas, así como en condiciones de oligotrofia de ambientes naturales como el agua. Los resultados han demostrado que E. amylovora es capaz de sobrevivir frente a dichas situaciones de estrés mediante su entrada en el estado denominado Viable No Cultivable (VNC). Cuando las condiciones vuelven a ser favorables, E. amylovora recupera su cultivabilidad y su poder patógeno, por lo que el estado VNC puede ser considerado como una estrategia de supervivencia para la bacteria. Al observar que E. amylovora en condiciones adversas perdía su capacidad de formar colonias en un medio de cultivo sólido, pero mantenía su viabilidad y patogenicidad, se planteó aumentar la eficiencia de su recuperación en placa. Dado que el cobre es un micronutriente esencial que puede favorecer el crecimiento bacteriano en medios ricos, se optimizó uno de los medios de cultivo más empleados para el aislamiento de E. amylovora añadiendo sulfato de cobre. Así, se consiguió potenciar el crecimiento de este patógeno en condiciones de estrés y retrasar la pérdida de cultivabilidad. También se comprobó que la presencia de cobre en el medio de cultivo producía un incremento acusado en la mucosidad de las colonias de E. amylovoOrdax Ibáñez, M. (2008).     Medfly Ceratitis capitata as Potential Vector for Fire Blight Pathogen Erwinia amylovora: Survival and Transmission

    Monitoring the ability of bacterial plant pathogens to survive in insects is required for elucidating unknown aspects of their epidemiology and for designing appropriate control strategies. Erwinia amylovora is a plant pathogenic bacterium that causes fire blight, a devastating disease in apple and pear commercial orchards. Studies on fire blight spread by insects have mainly focused on pollinating agents, such as honeybees. However, the Mediterranean fruit fly (medfly) Ceratitis capitata (Diptera: Tephritidae), one of the most damaging fruit pests worldwide, is also common in pome fruit orchards. The main objective of the study was to investigate whether E. amylovora can survive and be transmitted by the medfly. Our experimental results show: i) E. amylovora can survive for at least 8 days inside the digestive tract of the medfly and until 28 days on its external surface, and ii) medflies are able to transmit the bacteria from inoculated apples to both detached shoots and pear plants, being the pathogen recovered from lesions in both cases. This is the first report on E. amylovora internalization and survival in/on C. capitata, as well as the experimental transmission of the fire blight pathogen by this insect. Our results suggest that medfly can act as a potential vector for E. amylovora, and expand our knowledge on the possible role of these and other insects in its life cycle

    Metabolic biomarkers in community obese children: effect of obstructive sleep apnea and its treatment

    Objective: Obesity and obstructive sleep apnea in children have been associated with metabolic morbidities. The present study aimed to evaluate the presence of metabolic alterations among obese children recruited from the community, with and without obstructive sleep apnea syndrome (OSAS), and the impact of treatment of OSAS on metabolic profiles. Methods: A cross-sectional, prospective, multicenter study of Spanish children aged 3-14 years with a body mass index (BMI) ?95th percentile for age and sex were randomly selected in the first phase. Four groups emerged for follow-up: (1) no treatment; (2) dietary intervention; (3) surgical treatment of OSA; and (4) continuous positive airway pressure (CPAP) treatment of OSA. Fasting blood tests were performed at baseline (T0) and approximately one year after the intervention (T1). Results: A total of 113 obese children with a mean age of 11.3 ± 2.9 years completed T0 and T1 assessments. Their mean BMI z-score at T1 was 1.34 ± 0.59, and mean Respiratory Disturbance Index was 8.6 ± 13.0 at T0 and 3.3 ± 4.0/hour total sleep time at T1. Only glucose fasting levels differed among metabolic parameters in obese children with OSAS and without OSAS at baseline (T0) (p = 0.018). There were statistically significant differences between surgically treated OSAS (p = 0.002), and CPAP-treated OSAS (p = 0.024) versus the non-OSAS group in the glucose levels between baseline (T0) and follow-up (T1) after controlling for age and change in BMI. Significant univariate associations between BMI and C-reactive protein, insulin, and homeostasis model assessment of insulin resistance emerged at both T0 and T1. Conclusions: Concurrent obesity and OSAS could promote metabolic and inflammatory alterations, and the latter appeared to be sensitive to OSAS treatment outcomes.     El género Erwinia, que debe su nombre a la memoria del fitopatólogo Erwin F. Smith, se creó inicialmente para agrupar a las enterobacterias asociadas a las plantas, Gram negativas, bacilares, no formadoras de esporas y móviles (Winslow et al., 1920). Por ello, los miembros de este género incluían además enterobacterias saprofitas ecológicamente asociadas a plantas, así como patógenos oportunistas del hombre y los animales (Brenner, 1984). Esta heterogeneidad de especies fue la causa de que el género Erwinia fuera objeto de varias reclasificaciones. Finalmente, gracias al avance de las técnicas moleculares, las especies del género Erwinia se clasificaron en cuatro grupos filogenéticos basados en la comparación de secuencias del ADN ribosómico 16S (Hauben et al., 1998). El grupo I (género Erwinia) representa a las verdaderas erwinias e incluye diversas especies, que producen necrosis o marchitamientos en plantas, o que pueden ser epifitas. Erwinia amylovora es la especie tipo de este género. El grupo II (actuales géneros Pectobacterium y Dickeya) agrupa especies que originan podredumbres blandas en un amplio rango de hospedadores debido a su gran actividad pectolítica. El grupo III (actual género Brenneria y la especie Dickeya paradisiaca) incluye varias especies que afectan a plantas leñosas produciendo generalmente chancros y exudados. El grupo IV (género Pantoea) contiene especies que son saprofitas o patógenos más o menos frecuentemente oportunistas de plantas, animales y del hombre. Entre ellas destaca la antigua Erwinia herbicola, actualmente denominada Pantoea agglomerans, frecuentemente asociada en rosáceas a E. amylovora

    Echocardiographic Changes with Positive Airway Pressure Therapy in Obesity Hypoventilation Syndrome. Long-Term Pickwick Randomized Controlled Clinical Trial

    Spanish Sleep Network.[Rationale] Obesity hypoventilation syndrome (OHS) has been associated with cardiac dysfunction. However, randomized trials assessing the impact of long-term noninvasive ventilation (NIV) or continuous positive airway pressure (CPAP) on cardiac structure and function assessed by echocardiography are lacking. Rationale: Obesity hypoventilation syndrome (OHS) has been associated with cardiac dysfunction. However, randomized trials assessing the impact of long-term noninvasive ventilation (NIV) or continuous positive airway pressure (CPAP) on cardiac structure and function assessed by echocardiography are lacking.[Objectives] In a prespecified secondary analysis of the largest multicenter randomized controlled trial of OHS (Pickwick Project; N = 221 patients with OHS and coexistent severe obstructive sleep apnea), we compared the effectiveness of three years of NIV and CPAP on structural and functional echocardiographic changes.[Methods] At baseline and annually during three sequential years, patients underwent transthoracic two-dimensional and Doppler echocardiography. Echocardiographers at each site were blinded to the treatment allocation. Statistical analysis was performed using a linear mixed-effects model with a treatment group and repeated measures interaction to determine the differential effect between CPAP and NIV. Measurements and Main Results: A total of 196 patients were analyzed: 102 were treated with CPAP and 94 were treated with NIV. Systolic pulmonary artery pressure decreased from 40.5 ± 1.47 mm Hg at baseline to 35.3 ± 1.33 mm Hg at three years with CPAP, and from 41.5 ± 1.56 mm Hg to 35.5 ± 1.42 with NIV (P < 0.0001 for longitudinal intragroup changes for both treatment arms). However, there were no significant differences between groups. NIV and CPAP therapies similarly improved left ventricular diastolic dysfunction and reduced left atrial diameter. Both NIV and CPAP improved respiratory function and dyspnea.[Conclusions] In patients with OHS who have concomitant severe obstructive sleep apnea, long-term treatment with NIV and CPAP led to similar degrees of improvement in pulmonary hypertension and left ventricular diastolic dysfunction

    Respiratory Polygraphy Patterns and Risk of Recurrent Cardiovascular Events in Patients With Acute Coronary Syndrome

    Introduction: Obstructive sleep apnea (OSA) severity is based on the apnea-hypopnea index (AHI). The AHI is a simplistic measure that is inadequate for capturing disease severity and its consequences in cardiovascular diseases (CVDs). Deleterious effects of OSA have been suggested to influence the prognosis of specific endotypes of patients with acute coronary syndrome (ACS). We aim to identify respiratory polygraphy (RP) patterns that contribute to identifying the risk of recurrent cardiovascular events in patients with ACS. Methods: Post hoc analysis of the ISAACC study, including 723 patients admitted for a first ACS (NCT01335087) in which RP was performed. To identify specific RP patterns, a principal component analysis (PCA) was performed using six RP parameters: AHI, oxygen desaturation index, mean and minimum oxygen saturation (SaO2), average duration of events and percentage of time with SaO2 < 90%. An independent HypnoLaus population-based cohort was used to validate the RP components. Results: From the ISAACC study, PCA showed that two RP components accounted for 70% of the variance in the RP data. These components were validated in the HypnoLaus cohort, with two similar RP components that explained 71.3% of the variance in the RP data. The first component (component 1) was mainly characterized by low mean SaO2 and obstructive respiratory events with severe desaturation, and the second component (component 2) was characterized by high mean SaO2 and long-duration obstructive respiratory events without severe desaturation. In the ISAACC cohort, component 2 was associated with an increased risk of recurrent cardiovascular events in the third tertile with an adjusted hazard ratio (95% CI) of 2.44 (1.07 to 5.56; p-value = 0.03) compared to first tertile. For component 1, no significant association was found for the risk of recurrent cardiovascular events.     Survival Strategy of Erwinia amylovora against Copper: Induction of the Viable-but-Nonculturable State

    Copper compounds, widely used to control plant-pathogenic bacteria, have traditionally been employed against fire blight, caused by Erwinia amylovora. However, recent studies have shown that some phytopathogenic bacteria enter into the viable-but-nonculturable (VBNC) state in the presence of copper. To determine whether copper kills E. amylovora or induces the VBNC state, a mineral medium without copper or supplemented with 0.005, 0.01, or 0.05 mM Cu(2+) was inoculated with 10(7) CFU/ml of this bacterium and monitored over 9 months. Total and viable cell counts were determined by epifluorescence microscopy using the LIVE/DEAD kit and by flow cytometry with 5-cyano-2,3-ditolyl tetrazolium chloride and SYTO 13. Culturable cells were counted on King's B nonselective solid medium. Changes in the bacterial morphology in the presence of copper were observed by scanning electron microscopy. E. amylovora entered into the VBNC state at all three copper concentrations assayed, much faster when the copper concentration increased. The addition of different agents which complex copper allowed the resuscitation (restoration of culturability) of copper-induced VBNC cells. Finally, copper-induced VBNC cells were virulent only for the first 5 days, while resuscitated cells always regained their pathogenicity on immature fruits over 9 months. These results have shown, for the first time, the induction of the VBNC state in E. amylovora as a survival strategy against copper

    Exopolysaccharides favor the survival of Erwinia amylovora under copper stress through different strategies

    Erwinia amylovora causes fire blight, a destructive disease of rosaceous plants very difficult to control. We demonstrated that copper, employed to control plant diseases, induces the &quot;viable-but-nonculturable&quot; (VBNC) state in E. amylovora. Moreover, it was previously reported that copper increases production of its main exopolysaccharide (EPS), amylovoran. In this work, the copper-complexing ability of amylovoran and levan, other major EPS of E. amylovora, was demonstrated. Following this, EPS-deficient mutants were used to determine the role of these EPSs in survival of this bacterium in AB mineral medium with copper, compared to their wild type strain and AB without copper. Total, viable and culturable counts of all strains were monitored for six months. With copper, a larger fraction of the viable population of EPS mutants entered into the VBNC state, and earlier than their wild type strain, showing the contribution of both EPSs to long-term survival in a culturable state. Further, we demonstrated that both EPSs can be used as carbon source by E. amylovora under deprivation conditions. Overall, these previously unreported functions of amylovoran and levan provide survival advantages for E. amylovora, which could contribute to its enhanced persistence in nature.
