    BIOMEX (Biology and Mars Experiment): Preliminary results on Antarctic black cryptoendolithic fungi in ground based experiments

    The main goal for astrobiologists is to find traces of present or past life in extraterrestrial environment or in meteorites. Biomolecules, such as lipids, pigments or polysaccharides, may be useful to establish the presence of extant or extinct life (Simoneit, B et al., 1998). BIOMEX (Biology and Mars Experiment) aims to measure to what extent biomolecules, such as pigments and cellular components, preserve their stability under space and Mars-like conditions. The experiment has just been launched in the space and will be exposed on EXPOSE-R payload to the outside of the International Space Station (ISS) for about 2 years. Among a number of extremophilic microorganisms tested, the Antarctic cryptoendolithic black fungus Cryomyces antarcticus CCFEE 515 was included in the experiment. The fungus, living in the airspaces of porous rocks, was already chosen in previous astrobiological investigation for studying the interplanetary transfer of life via meteorites. In that context, the fungus survived 18 months of exposure outside of the ISS (Onofri al., 2012); for all these reasons it is considered an optimal eukaryotic model for astrobiological exploration. Before launch dried samples were exposed, in ground based experiments, to extreme conditions, including vacuum, irradiation and temperature cycles.Upon sample re-hydration and survival analysis, including colony forming ability, Propidium MonoAzide (PMA) assay-coupled quantitative PCR (Mohapatra and La Duc, 2012) all the test systems survived, neither any DNA damage was detectable. Our analyses focused also on mineral-microorganisms interactions and stability/degradation of typical fungal macromolecules, in particular melanin, when exposed to space and simulated Martian conditions, contributing to the development of libraries of biosignatures in rocks, supporting future exploration missions

    A numerical study of the RG equation for the deformed O(3)O(3) nonlinear sigma model

    The Renormalization Group equation describing the evolution of the metric of the nonlinear sigma model poses some nice mathematical problems involving functional analysis, differential geometry and numerical analysis. In this article we briefly report some results obtained from the numerical study of the solutions in the case of a two dimensional target space (deformation of the O(3)O(3) sigma model). In particular, our analysis shows that the so-called sausages define an attracting manifold in the U(1)U(1)-symmetric case, at one-loop level. Moreover, data from two-loop evolution are used to test the association put forward in Nucl.     We define the "sum of squares of the wavelengths" of a Riemannian surface (M,g) to be the regularized trace of the inverse of the Laplacian. We normalize by scaling and adding a constant, to obtain a "mass", which is scale invariant and vanishes at the round sphere. This is an anlaog for closed surfaces of the ADM mass from general relativity. We show that if M has positive genus then on each conformal class, the mass attains a negative minimum. For the minimizing metric, there is a sharp logarithmic Hardy-Littlewood-Sobolev inequality and a Moser-Trudinger-Onofri type inequality.Comment: 8 page

    The amyloidogenic potential and behavioral correlates of stress

    Observations of elevated basal cortisol levels in Alzheimer's disease (AD) patients prompted the hypothesis that stress and glucocorticoids (GC) may contribute to the development and/or maintenance of AD. Consistent with that hypothesis, we show that stress and GC provoke misprocessing of amyloid precursor peptide in the rat hippocampus and prefrontal cortex, resulting in increased levels of the peptide C-terminal fragment 99 (C99), whose further proteolytic cleavage results in the generation of amyloid-beta (Abeta). We also show that exogenous Abeta can reproduce the effects of stress and GC on C99 production and that a history of stress strikingly potentiates the C99-inducing effects of Abeta and GC. Previous work has indicated a role for Abeta in disruption of synaptic function and cognitive behaviors, and AD patients reportedly show signs of heightened anxiety. Here, behavioral analysis revealed that like stress and GC, Abeta administration causes spatial memory deficits that are exacerbated by stress and GC; additionally, Abeta, stress and GC induced a state of hyperanxiety. Given that the intrinsic properties of C99 and Abeta include neuroendangerment and behavioral impairment, our findings suggest a causal role for stress and GC in the etiopathogenesis of AD, and demonstrate that stressful life events and GC therapy can have a cumulative impact on the course of AD development and progression.CC and IS were supported by stipends from the Max Planck Society and EU Marie Curie Training Fellowships (at University College London, UK).     REST/NRSF drives homeostatic plasticity of inhibitory synapses in a target-dependent fashion

    The repressor-element 1-silencing transcription/neuron-restrictive silencer factor (REST/NRSF) controls hundreds of neuron-specific genes. We showed that REST/NRSF downregulates glutamatergic transmission in response to hyperactivity, thus contributing to neuronal homeostasis. However, whether GABAergic transmission is also implicated in the homeostatic action of REST/NRSF is unknown. Here, we show that hyperactivity-induced REST/NRSF activation, triggers a homeostatic rearrangement of GABAergic inhibition, with increased frequency of miniature inhibitory postsynaptic currents (IPSCs) and amplitude of evoked IPSCs in mouse cultured hippocampal neurons. Notably, this effect is limited to inhibitory-onto-excitatory neuron synapses, whose density increases at somatic level and decreases in dendritic regions, demonstrating a complex target- and area-selectivity. The upscaling of perisomatic inhibition was occluded by TrkB receptor inhibition and resulted from a coordinated and sequential activation of the Npas4 and Bdnf gene programs. On the opposite, the downscaling of dendritic inhibition was REST-dependent, but BDNF-independent. The findings highlight the central role of REST/NRSF in the complex transcriptional responses aimed at rescuing physiological levels of network activity in front of the ever-changing environment

    Synergistic interaction between the type III secretion system of the endophytic bacterium <i>Pantoea agglomerans</i> DAPP-PG 734 and the virulence of the causal agent of olive knot <i>Pseudomonas savastanoi pv. savastanoi</i> DAPP-PG 722

    The endophytic bacterium Pantoea agglomerans DAPP-PG 734 was previously isolated from olive knots caused by infection with Pseudomonas savastanoi pv. savastanoi DAPP-PG 722. Whole-genome analysis of this P. agglomerans strain revealed the presence of a Hypersensitive response and pathogenicity (Hrp) type III secretion system (T3SS). To assess the role of the P. agglomerans T3SS in the interaction with P. savastanoi pv. savastanoi, we generated independent knockout mutants in three Hrp genes of the P. agglomerans DAPP-PG 734 T3SS (hrpJ, hrpN, and hrpY). In contrast to the wildtype control, all three mutants failed to cause a hypersensitive response when infiltrated in tobacco leaves, suggesting that P. agglomerans T3SS is functional and injects effector proteins in plant cells. In contrast to P. savastanoi pv. savastanoi DAPP-PG 722, the wildtype strain P. agglomerans DAPP-PG 734 and its Hrp T3SS mutants did not cause olive knot disease in 1-year-old olive plants. Coinoculation of P. savastanoi pv. savastanoi with P. agglomerans wildtype strains did not significantly change the knot size, while the DAPP-PG 734 hrpY mutant induced a significant decrease in knot size, which could be complemented by providing hrpY on a plasmid. By epifluorescence microscopy and confocal laser scanning microscopy, we found that the localization patterns in knots were nonoverlapping for P. savastanoi pv. savastanoi and P. agglomerans when coinoculated. Our results suggest that suppression of olive plant defences mediated by the Hrp T3SS of P. agglomerans DAPP-PG 734 positively impacts the virulence of P. savastanoi pv. savastanoi DAPP-PG 722
