    Multimodal determinants of phase-locked dynamics across deep-superficial hippocampal sublayers during theta oscillations

    Theta oscillations play a major role in temporarily defining the hippocampal rate code by translating behavioral sequences into neuronal representations. However, mechanisms constraining phase timing and cell-type-specific phase preference are unknown. Here, we employ computational models tuned with evolutionary algorithms to evaluate phase preference of individual CA1 pyramidal cells recorded in mice and rats not engaged in any particular memory task. We applied unbiased and hypothesis-free approaches to identify effects of intrinsic and synaptic factors, as well as cell morphology, in determining phase preference. We found that perisomatic inhibition delivered by complementary populations of basket cells interacts with input pathways to shape phase-locked specificity of deep and superficial pyramidal cells. Somatodendritic integration of fluctuating glutamatergic inputs defined cycle-by-cycle by unsupervised methods demonstrated that firing selection is tuneable across sublayers. Our data identify different mechanisms of phase-locking selectivity that are instrumental for flexible dynamical representations of theta sequences

    GREENLION Project: Advanced Manufacturing Processes for Low Cost Greener Li-Ion Batteries

    GREENLION is a Large Scale Collaborative Project within the FP7 (GC.NMP.2011-1) leading to the manufacturing of greener and cheaper Li-Ion batteries for electric vehicle applications via the use of water soluble, fluorine-free, high thermally stable binders, which would eliminate the use of VOCs and reduce the cell assembly cost. The project has 6 key objectives: (i) development of new active and inactive battery materials viable for water processes (green chemistry); (ii) development of innovative processes (coating from aqueous slurries) capable of reducing electrode production cost and avoid environmental pollution; (iii) development of new assembly procedures (including laser cutting and high temperature pre-treatment) capable of substantially reduce the time and the cost of cell fabrication; (iv) lighter battery modules with easier disassembly through eco-designed bonding techniques; (v) waste reduction, which, by making use of the watersolubility of the binder, allows the extensive recovery of the active and inactive battery materials; and (vi) development of automated process and construction of fully integrated battery module for electric vehicle applications with optimized electrodes, cells, and other ancillaries. Achievements during the first 18 months of the project, especially on materials development and water-based electrode fabri cation are reported herein

    This reading list gathers together literature focused on buffalo (also known as bison) to support ongoing efforts to restore this iconic species to its keystone cultural and ecological role. The books and articles compiled here are grounded in a wide range of academic disciplines and approaches, representing many distinct ways of thinking about buffalo within the realm of Western sciences. This is not intended to be an exhaustive account of what is known about buffalo, much of which is held in Indigenous knowledge systems and communities. This academic list is designed as a resource for those working towards buffalo restoration, including conservation practitioners, researchers, parks and government officials, and Tribal communities. It is divided into categories according to either discipline or thematic focus, each containing a subset of pertinent literature. Topics include the paleobiology of bison, histories of buffalo loss, relationships between Indigenous Peoples and buffalo, buffalo restoration and reintroduction, buffalo ecology, buffalo and climate change, governance and human dimensions of buffalo, bison and cattle, bison as livestock, and the genetics of bison. This list is intended as a living document to grow with the increasingly rich scholarship on buffalo, and will be updated regularly

    Aberrant survival of hippocampal Cajal-Retzius cells leads to memory deficits, gamma rhythmopathies and susceptibility to seizures in adult mice

    Cajal-Retzius cells (CRs) are transient neurons, disappearing almost completely in the postnatal neocortex by programmed cell death (PCD), with a percentage surviving up to adulthood in the hippocampus. Here, we evaluate CR’s role in the establishment of adult neuronal and cognitive function using a mouse model preventing Bax-dependent PCD. CRs abnormal survival resulted in impairment of hippocampus-dependent memory, associated in vivo with attenuated theta oscillations and enhanced gamma activity in the dorsal CA1. At the cellular level, we observed transient changes in the number of NPY cells and altered CA1 pyramidal cell spine density. At the synaptic level, these changes translated into enhanced inhibitory currents in hippocampal pyramidal cells. Finally, adult mutants displayed an increased susceptibility to lethal tonic-clonic seizures in a kainate model of epilepsy. Our data reveal that aberrant survival of a small proportion of postnatal hippocampal CRs results in cognitive deficits and epilepsy-prone phenotypes in adulthood.We thank Dr. P.     Differential Temporal and Spatial Progerin Expression during Closure of the Ductus Arteriosus in Neonates

    Closure of the ductus arteriosus (DA) at birth is essential for the transition from fetal to postnatal life. Before birth the DA bypasses the uninflated lungs by shunting blood from the pulmonary trunk into the systemic circulation. The molecular mechanism underlying DA closure and degeneration has not been fully elucidated, but is associated with apoptosis and cytolytic necrosis in the inner media and intima. We detected features of histology during DA degeneration that are comparable to Hutchinson Gilford Progeria syndrome and ageing. Immunohistochemistry on human fetal and neonatal DA, and aorta showed that lamin A/C was expressed in all layers of the vessel wall. As a novel finding we report that progerin, a splicing variant of lamin A/C was expressed almost selectively in the normal closing neonatal DA, from which we hypothesized that progerin is involved in DA closure. Progerin was detected in 16.2%±7.2 cells of the DA. Progerin-expressing cells were predominantly located in intima and inner media where cytolytic necrosis accompanied by apoptosis will develop. Concomitantly we found loss of α-smooth muscle actin as well as reduced lamin A/C expression compared to the fetal and non-closing DA. In cells of the adjacent aorta, that remains patent, progerin expression was only sporadically detected in 2.5%±1.5 of the cells. Data were substantiated by the detection of mRNA of progerin in the neonatal DA but not in the aorta, by PCR and sequencing analysis. The fetal DA and the non-closing persistent DA did not present with progerin expressing cells. Our analysis revealed that the spatiotemporal expression of lamin A/C and progerin in the neonatal DA was mutually exclusive. We suggest that activation of LMNA alternative splicing is involved in vascular remodeling in the circulatory system during normal neonatal DA closure

    A Prospective Cohort of SARS-CoV-2-Infected Health Care Workers: Clinical Characteristics, Outcomes, and Follow-up Strategy

    Background. During the coronavirus disease 2019 (COVID-19) outbreaks, health care workers (HCWs) are at a high risk of infection. Strategies to reduce in-hospital transmission between HCWs and to safely manage infected HCWs are lacking. Our aim was to describe an active strategy for the management of COVID-19 in severe acute respiratory syndrome coronavirus 2 (SARSCoV-2)-infected HCWs and investigate its outcomes. Methods. A prospective cohort study of SARS-CoV-2-infected health care workers in a tertiary teaching hospital in Barcelona, Spain, was performed. An active strategy of weekly polymerase chain reaction screening of HCWs for SARS-CoV-2 was established by the Occupational Health department. Every positive HCW was admitted to the Hospital at Home Unit with daily assessment online and in-person discretionary visits. Clinical and epidemiological data were recorded. Results. Of the 590 HCWs included in the cohort, 134 (22%) were asymptomatic at diagnosis, and 15% (89 patients) remained asymptomatic during follow-up. A third of positive cases were detected during routine screening. The most frequent symptoms were cough (68%), hyposmia/anosmia (49%), and fever (41%). Ten percent of the patients required specific treatment at home, while only 4% of the patients developed pneumonia. Seventeen patients required a visit to the outpatient clinic for further evaluation, and 6 of these (1%) required hospital admission. None of the HCWs included in this cohort required intensive care unit admission or died. Conclusions. Active screening for SARS-CoV-2 among HCWs for early diagnosis and stopping in-hospital transmission chains proved efficacious in our institution, particularly due to the high percentage of asymptomatic HCWs. Follow-up of HCWs in Hospital at Home units is safe and effective, with low rates of severe infection and readmission.     A draft physical map of a D-genome cotton species (Gossypium raimondii)

    <p>Abstract</p> <p>Background</p> <p>Genetically anchored physical maps of large eukaryotic genomes have proven useful both for their intrinsic merit and as an adjunct to genome sequencing. Cultivated tetraploid cottons, <it>Gossypium hirsutum </it>and <it>G. barbadense</it>, share a common ancestor formed by a merger of the A and D genomes about 1-2 million years ago. Toward the long-term goal of characterizing the spectrum of diversity among cotton genomes, the worldwide cotton community has prioritized the D genome progenitor <it>Gossypium raimondii </it>for complete sequencing.</p> <p>Results</p> <p>A whole genome physical map of <it>G. raimondii</it>, the putative D genome ancestral species of tetraploid cottons was assembled, integrating genetically-anchored overgo hybridization probes, agarose based fingerprints and 'high information content fingerprinting' (HICF). A total of 13,662 BAC-end sequences and 2,828 DNA probes were used in genetically anchoring 1585 contigs to a cotton consensus genetic map, and 370 and 438 contigs, respectively to <it>Arabidopsis thaliana </it>(AT) and <it>Vitis vinifera </it>(VV) whole genome sequences.</p> <p>Conclusion</p> <p>Several lines of evidence suggest that the <it>G. raimondii </it>genome is comprised of two qualitatively different components. Much of the gene rich component is aligned to the <it>Arabidopsis </it>and <it>Vitis vinifera </it>genomes and shows promise for utilizing translational genomic approaches in understanding this important genome and its resident genes. The integrated genetic-physical map is of value both in assembling and validating a planned reference sequence.</p
