    Prevalence and diversity of Chlamydiales in Swiss ruminant farms.

    Chlamydia and Chlamydia-related bacteria are known to infect various organisms and may cause a wide range of diseases, especially in ruminants. To gain insight into the prevalence of these bacteria in the ruminant environment, we applied a pan-Chlamydiales PCR followed by sequencing to 72 ruminant environmental samples from water, feed bunks and floors. Chlamydiales from four family-level lineages were detected indicating a high biodiversity of Chlamydiales in ruminant farms. Parachlamydiaceae were detected in all three types of environmental samples and was the most abundant family-level taxon (60%). In contrast, only one bacterium from each of the following family-level lineages was identified: Chlamydiaceae, Criblamydiaceae and Simkaniaceae. The observed high prevalence of Parachlamydiaceae in water samples may suggest water as the main source of contamination for ruminants as well as their environment due to spoilage. The absence of reported infections in the investigated ruminant farms might indicate that either detected Chlamydiales are of reduced pathogenicity or infective doses have not been reached

    Modeling and numerical study of primary breakup under diesel conditions

    A recently introduced stochastic model for reduced numerical simulation of primary jet breakup is evaluated by comparing model predictions to DNS results for primary jet breakup under diesel conditions. The model uses one-dimensional turbulence (ODT) to simulate liquid and gas time advancement along a lateral line of sight. This one-dimensional domain is interpreted as a Lagrangian object that is advected downstream at the jet bulk velocity, thus producing a flow state expressed as a function of streamwise and lateral location. Multiple realizations are run to gather ensemble statistics that are compared to DNS results. The model incorporates several empirical extensions of the original ODT model that represent the phenomenology governing the Weber number dependence of global jet structure. The model as previously formulated, including the assigned values of tunable parameters, is used here without modification in order to test its capability to predict various statistics of droplets generated by primary breakup. This test is enabled by the availability of DNS results that are suitable for model validation. Properties that are examined are the rate of bulk liquid mass conversion into droplets, the droplet size distribution, and the dependence of droplet velocities on droplet diameter. Quantities of greatest importance for engine modeling are found to be predicted with useful accuracy, thereby demonstrating a more detailed predictive capability by a highly reduced numerical model of primary jet breakup than has previously been achieved

    Architects in Europe: Models of Professionalisation and Potential Implications for the Planning Profession

    Data from the Architects’ Council of Europe (ACE) show a very inhomogeneous distribution of architects among European countries, for example, high numbers of architects in Italy and Germany versus low numbers in the UK and France. These discrepancies cannot be explained by differences in the domestic construction markets. This paper reviews models of professionalisation to explain the heterogeneity among European architects and shows potential implications for the planning profession in Europe. We analyse the 2018 ACE data for the five European countries with the strongest labour markets for architects. Our findings led to three main conclusions: Firstly, such models of professionalisation add to an understanding of statistical data on architects in Europe. Secondly, a better understanding of the statistical data requires more than a single theory. Thirdly, as to the planning profession, the standard process model of professionalisation may work well for the UK, but not for the other European countries.Peer Reviewe

    Canine NAPEPLD-associated models of human myelin disorders

    Canine leukoencephalomyelopathy (LEMP) is a juvenile-onset neurodegenerative disorder of the CNS white matter currently described in Rottweiler and Leonberger dogs. Genome-wide association study (GWAS) allowed us to map LEMP in a Leonberger cohort to dog chromosome 18. Subsequent whole genome re-sequencing of a Leonberger case enabled the identification of a single private homozygous non-synonymous missense variant located in the highly conserved metallo-beta-lactamase domain of the N-acyl phosphatidylethanolamine phospholipase D (NAPEPLD) gene, encoding an enzyme of the endocannabinoid system. We then sequenced this gene in LEMP-affected Rottweilers and identified a different frameshift variant, which is predicted to replace the C-terminal metallo-beta-lactamase domain of the wild type protein. Haplotype analysis of SNP array genotypes revealed that the frameshift variant was present in diverse haplotypes in Rottweilers, and also in Great Danes, indicating an old origin of this second NAPEPLD variant. The identification of different NAPEPLD variants in dog breeds affected by leukoencephalopathies with heterogeneous pathological features, implicates the NAPEPLD enzyme as important in myelin homeostasis, and suggests a novel candidate gene for myelination disorders in people

    Good Management of European UNESCO Industrial World Heritage Sites in the Context of Urban and Regional Planning

    This paper addresses the general theme of conservation and/or change of heritage sites in urban and regional planning. The particular focus is on the management of UNESCO industrial World Heritage sites in Europe. Industrial heritage refers back to historic innovation and continues to be embedded today in innovation-orientated urban and regional development. The question that our study examines is: In which aspects does the management of an industrial World Heritage site coincide with (or differ from) that of areas of innovation? We identified criteria both for the management of industrial heritage sites (“Good Practice Wheel”) and areas of innovation (success factors for European Science and Technology Parks). In an online survey, we asked managers of heritage sites to evaluate management criteria. Completed survey responses were obtained for 22 of 38 European sites (58%) in 12 of 15 countries concerned (80%). Our study clearly shows an overlap of conservation and innovation priorities even in the management of UNESCO industrial World Heritage sites, and underlines the importance of integrative, “good” heritage management.Peer Reviewe

    Assessment of a Multiphase Formulation of One-Dimensional Turbulence using Direct Numerical Simulation of a Decaying Turbulent Interfacial Flow

    The interaction between turbulence and surface tension is studied numerically using the one-dimensional-turbulence (ODT) model. ODT is a stochastic model simulating turbulent flow evolution along a notional one-dimensional line of sight by applying instantaneous maps that represent the effects of individual turbulent eddies on property fields. It provides affordable high resolution of interface creation and property gradients within each phase, which are key for capturing the local behavior as well as overall trends, and has been shown to reproduce the main features of an experimentally determined regime diagram for primary jet breakup. Here, ODT is used to investigate the interaction of turbulence with an initially planar interface. The notional flat interface is inserted into a periodic box of decaying homogeneous isotropic turbulence, simulated for a variety of turbulent Reynolds and Weber numbers. Unity density and viscosity ratios serve to focus solely on the interaction between fluid inertia and the surface-tension force. Statistical measures of interface surface density and spatial structure along the direction normal to the initial surface are compared to corresponding direct-numerical-simulation (DNS) data. Allowing the origin of the lateral coordinate system to follow the location of the median interface element improves the agreement between ODT and DNS, reflecting the absence of lateral non-vortical displacements in ODT. Beyond the DNS-accessible regime, ODT is shown to obey the predicted parameter dependencies of the Kolmogorov critical scale in both the inertial and dissipative turbulent-cascade sub-ranges. Notably, the probability density function of local fluctuations of the critical scale is found to collapse to a universal curve across both sub-ranges.Comment: 15 pages, 13 figure

    Donor KIR B Genotype Improves Progression-Free Survival of Non-Hodgkin Lymphoma Patients Receiving Unrelated Donor Transplantation

    Donor killer immunoglobulin-like receptor (KIR) genotypes are associated with relapse protection and survival after allotransplantation for acute myelogenous leukemia. We examined the possibility of a similar effect in a cohort of 614 non-Hodgkin lymphoma (NHL) patients receiving unrelated donor (URD) T cell-replete marrow or peripheral blood grafts. Sixty-four percent (n = 396) of donor-recipient pairs were 10/10 allele HLA matched and 26% were 9/10 allele matched. Seventy percent of donors had KIR B/x genotype; the others had KIR A/A genotype. NHL patients receiving 10/10 HLA-matched URD grafts with KIR B/x donors experienced significantly lower relapse at 5 years (26%; 95% confidence interval [CI], 21% to 32% versus 37%; 95% CI, 27% to 46%; P = .05) compared with KIR A/A donors, resulting in improved 5-year progression-free survival (PFS) (35%; 95% CI, 26% to 44% versus 22%; 95% CI, 11% to 35%; P = .007). In multivariate analysis, use of KIR B/x donors was associated with significantly reduced relapse risk (relative risk [RR], .63, P = .02) and improved PFS (RR, .71, P = .008). The relapse protection afforded by KIR B/x donors was not observed in HLA-mismatched transplantations and was not specific to any particular KIR-B gene. Selecting 10/10 HLA-matched and KIR B/x donors should benefit patients with NHL receiving URD allogeneic transplantation

    Primary Postnatal Dorsal Root Ganglion Culture from Conventionally Slaughtered Calves

    Neurological disorders in ruminants have an important impact on veterinary health, but very few host-specific in vitro models have been established to study diseases affecting the nervous system. Here we describe a primary neuronal dorsal root ganglia (DRG) culture derived from calves after being conventionally slaughtered for food consumption. The study focuses on the in vitro characterization of bovine DRG cell populations by immunofluorescence analysis. The effects of various growth factors on neuron viability, neurite outgrowth and arborisation were evaluated by morphological analysis. Bovine DRG neurons are able to survive for more than 4 weeks in culture. GF supplementation is not required for neuronal survival and neurite outgrowth. However, exogenously added growth factors promote neurite outgrowth. DRG cultures from regularly slaughtered calves represent a promising and sustainable host specific model for the investigation of pain and neurological diseases in bovines

    Capecitabine in the treatment of metastatic renal cell carcinoma

    To evaluate the therapeutic effects and systemic toxicities of a capecitabine-based home therapy regimen in patients with metastatic renal cell carcinoma, 30 patients were enrolled in a phase II clinical trial. Treatment consisted of oral capecitabine combined with subcutaneous recombinant human interferon-α 2a, recombinant human interleukin-2 and oral 13-cis-retinoic acid. There were two (7%) complete responses (CRs) and eight (27%) partial remissions (PRs), for an overall objective response rate of 34% (95% CI 17–53%). Except one, all responses are ongoing, with a median duration of 9+ and 8+ months for CRs and PRs, respectively. Additionally, 12 patients (40%) reached stable disease. Eight patients (27%) showed continued disease progression despite treatment. Therapy was well tolerated and was given in the outpatient setting. Capecitabine-related World Health Organization (WHO) grade 2 and 3 toxicities were observed in five and two patients respectively, and were limited to fatigue, nausea/vomiting, diarrhoea, stomatitis, dermatitis and hand-and-foot syndrome. The substitution of capecitabine for 5-FU in the pre-existing biochemotherapy regimen did not result in a reduced therapeutic efficacy and showed significant anti-tumour activity in patients with advanced renal cell carcinoma. © 2000 Cancer Research Campaig

    Intracranial oligodendroglioma with optic nerve infiltration in a Labrador retriever

    A seven-year-old neutered male Labrador retriever was presented with behavioral changes and reduced vision. Neurological examination revealed blindness of the left eye with a positive pupillary light reflex and a decreased mental status. Blood and cerebrospinal fluid analyses showed no abnormalities. MRI revealed a deviant area located in the thalamic, hypothalamic and caudate nuclear regions. Invasion of the left optic nerve was also observed. The dog was euthanized after 38 days of palliative treatment. Necropsy showed a non-encapsulated extruding white lardaceous mass, with a diameter of 2 cm, in the white matter, located in the ventral thalamic region near the chiasma opticum, which extended along the left optic nerve. Histologically, neoplastic cells had a “fried egg” appearance. Immunolabelling for glial fibrillary acidic protein demonstrated the presence of numerous reactive astrocytes. The tumor was diagnosed as a low grade (II) intracranial oligodendroglioma with infiltration of the optic nerve.</jats:p
